Laminariae Thallus has been known to be effective for the treatment of obesity. Te show the effectiveness of Laminariae Thallus in a more scientific way, Laminariae Thallus extract was prepared and evaluated in high fat diet rats by measuring the changes of body weight and lipid metabolism as described briefly below. 200 g of crushed Laminariae Thallus was extracted with methyl alcohol. The extract was evaporated under reduced pressure to give 14.4g. For 10 weeks, control group rat were ingested high fat diet, while the test group rat were fed with a fat diet pius Laminariae Thallus extract. Normal group was fed with normal diet. 150 mg of Laminariae Thallus extract per 1 kg of body weight was added to the diet in a test group rats. Control group rats on a high fat diet gained weight significantly, whereas the test group rats on a high fat diet plus Laminariae Thallus extract gained weight less. Significant increase of liver weight caused in a high fat diet was also inhibited by the Laminariae Thallus extract treatment. Total lipid, triglyceride, phospholipid and total cholesterol levels of serum in the high fat diet rats were remarkably increased, whereas their levels on a high fat diet plus Laminariae Thallus extract were less increased. While serum HDL-cholesterol levels was remarkably decreased in a high fat diet, its level was less decreased in a high fat diet plus Laminariae Thallus extract. Furthermore, we observed that the activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase increased in a high fat diet, while their activities in a high fat diet plus Laminariae Thallus extract were getting back nearly to the normal levels of normal diet rats. These result showed that the obesity caused by a high fat diet was effectively inhibited by a Laminariae Thallus extract. Our results also showed that the abnormal lipid metabolism caused by a high fat diet was effectively cured by adding Laminariae Thallus extract.
Objectives : This experimental study was designed to investigate the effect of SBY-III extract on the weight, cell size of epididymal fat-pad, fat accumulation area in liver, serum lipid level and UCP1 mRNA in brown adipose tissue of high fat diet-fed obese rats continued by high fat diet and regulated by normal Diet. Methods : The body weight gain, weight of the internal organs(epididymis, liver, brown adipose tissue), insulin, triglyceride, total cholesterol, total lopod, free fatty acid, expression of UCP1 mRNA were measured in high fat diet-fed obese rats continued by high fat diet and regulated by normal diet. The experimental study are divided into exp-I and exp-II. Each study was administered normal diet, high fat diet and SBY-III according to each situation. Normal group is normal diet for 8 weeks. Exp-I are divided into control group(high fat diet for 8 weeks) and sample group(high fat diet for 8 weeks and SBY-III for last 2 weeks). Exp-II are divided into control group(high fat diet for 6 weeks and normal diet for 2 weeks) and sample group(high fat diet for 6 weeks and normal diet with SBY-III for 2 weeks). These were then compared mutually. Results : 1. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease of weight gain than control group and diet control-fed sample group with SBY-III showed the more effective decrease of weight loss including weight gain than control group. 2. Irrespective of diet control, sample group taken SBY-III showed the more effective decrease cell size of epididymal fat-pad, fat accumulation area in liver than control group. 3. Non diet control-fed sample group taken SBY-III showed the more effective decrease of serum triglyceride, total lipid, free fatty acid than control group and diet control-fed sample group taken SBY-III showed the decrease of serum triglyceride, free fatty acid than control group. 4. Only diet control-fed sample group taken SBY-III showed the decrease of UCP1 volume. Conclusions : These results shows that SBY-III has effects on anti-obesity, especially keeping pace with diet control.
This study was performed to observe the effects of dietary fat levels and sources on lipids contents and cellularities of liver, brain, and adipose tissue of early weaned rats. Male Sprague-Dawley rats were prematurely weaned from postnatal 17th day with the experimental diets differ in fat levels : low(5%), medium(10%), high(20%) and fat sources : butter, soybean oil, butter+ soybean oil. On the postnatal 29th day, contents of total lipid, triglyceride, cholesterol and phospholipid of serum, liver, brain and adipose tissue were determined, and DNA was determined to assess the cell growth. Rats early weaned fed high fat diet showed lower total lipid and triglyceride levels in serum and liver than those fed medium or low fat diet Rats early weaned fed high fat diet had adipocytes of fewer number, but larger size than those of rats fed low or medium fat diets. Rats early weaned fed soybean oil diet had more adipocytes thu those fed butter diet. Rats normally weaned to commercial chow diet showed lower total lipid, triglyceride, and cholesterol levels in serum and liver, had fewer adipocytes than all early weaned rats except for rats fed high fat-butter diet. These results suggest that high fat-butter diet is ideal weaning diet at early weaning.
Comminuted sausages with reduced fat (25-0%) and increased moisture (19-44%) combinations were manufactured, and their chemical composition, and physico-chemical, textural and sensory properties were determined for the selection of the most palatable sausage treatment. The moisture and fat contents of the comminuted sausages varied from 55-79% and 26.4-2.14%, respectively, whereas, the protein content was relatively constant at 13.5-14.5%. Expressible moisture (EM, %) decreased with increased fat addition, and the high-fat control sausage (~25% fat) had lower EM than those with lower than 15% fat addition. Increased fat level also increased Hunter L values (lightness) of sausage samples taken from the core, and differences in lightness were observed between 15 and 25% fat level at the initial mixture. Sausages without fat addition had different textural characteristics from high-fat control sausages in most texture profile analysis (TPA) values. However, no differences in TPA values were observed among treatments with added fat (<25%) in the formulation. Results of the sensory evaluation showed that the most appropriate fat content of comminuted sausages to have better sensory properties ranged from 15-20% of added fat at the initial mixture. These results also indicated that decreased fat and increased moisture contents produce sausages with higher EM and lower lightness. Comminuted sausages without fat addition had different textural characteristics from the high-fat control.
Anti-obesity effect of biocellulose and diet formulation containing it was evaluated using obese rats. Thirty male Sprague-Dawlely (SD) rats were randomly assigned to high-fat diet group (CON), high-fat diet group containing 5% biocellulose (BIO), and high-fat diet group containing 5% dietary formulation (DF). After 10 weeks, body weight gains of BIO and DF groups were reduced by 15.1 and 6%, respectively, as compared with CON group. Epididymus, parirenal, and visceral fat pads of BIO and DF groups were lower than those of CON group. Weight of interscapular brown adipose tissues increased slightly in BIO group and significantly in DF group. Size of adipocyte in rats decreased in BIO and DF groups. Results indicate biocellulose-containing diet has anti-obesity effect by reducing body weight gain and white adipocytes depots in rats fed high-fat diet.
The purpose of this research was to find effects of high fat diet and endurance exercise on mitochondria biosynthesis and insulin resistance in male Wister rat. Th rats were randomly separated into 4 groups: A, Isocaloric high fat diet group (50% of calories from fat), B. Chow group, C. Isocaloric high fat diet with endurance exercise (treadmill running, slop 8%, 23 m/min, 120 min/day, 5 days per week), D. Chow group with endurance exercise. Both Isocaloric high fat diet group and chow group were given an equal caloric composition with 3.2kcal/g. equal amount of food were checked every day and given to both groups. 4 weeks of high fat diet did not show any change in body weight and amount of body fat. Further the level of glucose and insulin in blood, and insulin-stimuilated glucose transport rate in epitrochlearis muscle was not affected by Isocaloric high fat diet. However, the endurance exercise showed statistically significant change in the level of insulin in blood. Although either Isocaloric high fat diet or endurance exercise alone did not change on mitochondria biogenesis marker, Isocaloric high fat diet with endurance exercise could induce the increased level of marker (p<0.05). Also, plasma free fatty acids were increased in this group. From this investigation, Isocaloric high fat diet with moderate-intensity endurance exercise is effective way to induce mitochondrial biogenesis.
This study was carried out to investigate the effect of body weight reduction of patty containing glucomannan in high fat diet-induced obese rats. Male Spraque-Dawley rats were randomly assigned to control and high fat diets groups for 4 weeks. After 4 weeks, the control and high fat diet groups were rearranged into 5 groups by different levels of patty containing glucomannan (60%). After 5 weeks, serum and whole blood levels were analyzed. Obesity index was significantly lower in the group fed patty containing glucomannan than that of high fat diet groups. The status of white blood cells in hematological examination was significantly higher in rats fed high fat diet and not significantly different from those fed patty. Serum albumin levels were not significantly different, while serum glucose levels were significantly different among groups. Serum Triglyceride and total cholesterol levels were the highest in rats fed high fat diets, while the lowest in rats fed patty containing glucomannan.

The effect of exercise and/or high fat diet on carnitine status and carnitine palmitoyltransferase I(CPT I) level were investigated in Weanling Sprague Dawley rats. The rats were fed an AIN 76 diet or a modified high fat AIN diet, supplemented with 35% corn oil, for 31 days. During the 31 day period half of the animals in each dietary group were exercised on a treadmill for 90 minutes per day. Carnitine concentrations were determined in plasma and liver and CPT I mRNA levels were measured by Northern blot analysis with CPT I cDNA probe in livers of rats. Exercise rats gained less weight than non exercised rats during the study for high fat diet group. Exercise rats had a higher plasma acid soluble acylcarnitine and acid insoluble acylcarnitine concnetrations than non exercised rats for normal diet group. Exercise or high fat diet increased liver carnitine concentration, but a mixed effect was not shown. In exercised rats, CPT I mRNA levels increased significantly relative to those of nonexercised rats. CPT I mRNA levels also increased when compared high fat fed rats with those of normal diet fed rats. These data suggest that there is a correlation between carnitine concen trations and CPT I mRNA levels and that CPT I can be regulated at the transcriptional level by exercise and/or high fat diet.

Objective: The objective of the current study was to find out the independent and interactive effects of prilled fat supplementation with protein on the production performance of early lactating Nili Ravi buffaloes. Methods: Sixteen early lactating buffaloes (36.75±5.79 d in milk; mean±standard error) received 4 treatments in 4×4 Latin-square design according to 2×2 factorial arrangements. The dietary treatments were: i) low protein low fat, ii) low protein high fat, iii) high protein low fat, and iv) high protein high fat. The dietary treatments contained 2 protein (8.7% and 11.7% crude protein) and fat levels (2.6% and 4.6% ether extract) on a dry matter basis. Results: The yields of milk and fat increased with increasing protein and fat independently (p≤0.05). Energy-, protein-, and fat-corrected milk yields also increased with increasing protein and fat independently (p≤0.05). Increasing dietary protein increased the protein yield by 3.75% and lactose yield by 3.15% and increasing dietary fat supplies increased the fat contents by 3.93% (p≤0.05). Milk yield and fat-corrected milk to dry matter intake ratios were increased at high protein and high fat levels (p≤0.05). Milk nitrogen efficiency was unaffected by dietary fat (p>0.10), whereas it decreased with increasing protein supplies (p≤0.05). Plasma urea nitrogen and cholesterol were increased by increasing protein and fat levels, respectively (p≤0.05). The values of predicted methane production reduced with increasing dietary protein and fat. Conclusion: It is concluded that prilled fat and protein supplies increased milk and fat yield along with increased ratios of milk yield and fat-corrected milk yields to dry matter intake. However, no interaction was observed between prilled fat and protein supplementation for production parameters, body weight, body condition score and blood metabolites. Predicted methane production decreased with increasing protein and fat levels.
Objectives: Samjunghwan has been known to be effective for the treatment of obesity. To show the effectiveness of Samjunghwan in a more scientific way, Samjunghwan extract was prepared and evaluated in high-fat diet rats by measuring the changes of body weight and lipid metabolism as described below. Methods: 245 g of crushed Samjunghwan was extracted with methyl alcohol. The extract was evaporated under reduced pressure to give 21.8 g. For 30 days, the control group rats were given a high fat diet, while the test group rats were given a high fat diet plus Samjunghwan extract. The normal group rats were given a normal diet. 50 mg of Samjunghwan extract per 1 kg of body weight was added to the diet for the test group rats. Results: The control group rats on a high fat diet gained weight by about 27-28% as compared to the normal group, whereas the test group rats on a high fat diet plus Samjunghwan extract lost weight about 6-8% as compared to the control group. A significant increase of liver weight caused by a high fat diet was also inhibited by the same Samjunghwan extract administration. Similar inhibitory effects on the food intake and on the epididymal adipose tissue weight were observed in the high fat diet rats by the administration of Samjunghwan extract. Serum and liver total lipid levels in the control group on a high fat diet increased significantly as compared to the normal group, whereas their serum and liver levels increased less on a high fat diet plus Samjunghwan extract administered test group than the control group. Impressively, serum leptin levels in the test group decreased almost to the level of the normal group, which was well in accordance with the decreased fat contents in the test group rats. Furthermore, the activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase were increased in the control group, while their activities in the test group on a high fat diet plus Samjunghwan extract decreased nearly to the levels of normal group rats on a normal diet. Conclusions: These results showed that the obesity caused by a high fat diet was effectively inhibited the administration of Samjunghwan extract. Our results also showed that the abnormal lipid metabolism caused by a high fat diet was effectively cured by the administration of Samjunghwan extract.
