  • Bacillus thuringiensis (Bt) is the most widely used microbial insecticide in the biological control market. Cultivation of the microorganism to high cell densities offers potential for enhancing the rate of formation as well as the concentration of the desired products In the fermentation broths in bioreactor. With this objective, we developed the new bioreactor incorporating ceramic membrane module for the retention of cell mass. Cell yield and spore formation of Bacillus thuringiensis was improved markedly by adopting this new bioreactor based on glucose -limited feeding operation. It was possible to grow the cell and the heat-resistant spore to above $1.2\;{\times}\;10^{10}\;CFU/ml$ density. With glucose-limited operation, we studied the growth behavior of Bacillus thuringiensis during the cell retention culture. Linear growth of Bacillus thuringiensis was observed under glucose-limited culture, which matched well with simple mathematical model of cell retention culture.

The Effect of Sports Drink Containing Schizandra Chinensis on Blood Biochemical Elements, Exercise Performance and HSP70 (오미자를 이용한 스포츠 드링크 섭취가 혈액성분과 운동수행력 및 HSP70에 미치는 영향)

  • OBJECTIVES : Schizandra chinensis is well known for its efficacy at liver function reinforcement, relieving thirst and recovery from fatigue. In this study, we examined the effects of sports drink containing Schizandra chinensis on serum metabolic substrate, electrolyte, stress indicators, related-enzyme and exercise performance, rectal temperature, and heat shock proteinb70 (HSP70). METHODS : Elite long-distance runners (male, 21.3yrs, n=16) were selected and divided into two groups; an experimental group (EXP, n=8) and a control group (CON, n=8). A beverage containing Schizandra chinensis was supplemented 3 times per day to EXP for 4 weeks. Serum biochemical elements (glucose, lactate, total cholesterol, triglyceride, high density lipoprotein cholesterol, glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, creatinine, creatine phosphokinase, lactate dehydrogenase, blood urea nitrogen, Na, K, Cl) were analyzed by auto blood analyzer. Exercise performance was measured by treadmill exercise test, HSP70 was detected by electrophoresis and Western blotting, and rectal temperature was measured by rectal temperature probe. RESULTS : Administration of the beverage increased significantly the rest level of blood Na, Cl and glucose and decreased significantly lactate dehydrogenase, glutamate oxaloacetate transaminase. No difference was found in exercise performance, rectal temperature increment or HSP70 concentration between groups. CONCLUSIONS : Administration of a sports drink containing Schizandra chinensis altered blood glucose, lactate dehydrogenase, glutamate oxaloacetate transaminase, Na and Cl levels.

  • For development of an on-line monitoring unit of fermentation process, an auto dilution unit based on traditional chemical and biological analytical method was developed and the performance was evaluated. The dilution unit was constructed with two syringe pumps and flow direction change valves and fully automated. Total delivery volume of two pumps using distilled water was measured to confirm the operating stability And diluted concentrations of three substrate solutions (glucose, lactic acid, ethanol) were compared with a standard method with a high performance liquid chromatograph (glucose, lactic acid) and gas chromatograph (ethanol). Relative error values of total delivery volume of the pumps were below 3% and standard deviation values were 0.003 (n=5). Relative error values of diluted concentration of the dilution unit measurements were below 2% with 1/10 of dilution ratio and 70, $80{\mu}{\ell}$ of sample volume conditions for glucose and lactic acid, 1/30 of dilution ratio and $70{\mu}{\ell}$ of sample volume conditions for ethanol, respectively. In case of the ethanol, cause of the evaporative characteristics, the relative error values showed over 5% whole experimental conditions.

Enzyme-Free Glucose Sensing with Polyaniline-Decorated Flexible CNT Fiber Electrode (Polyaniline을 이용한 CNT fiber 유연 전극 기반의 비효소적 글루코스 검출)

  • As the demand for wearable devices increases, many studies have been studied on the development of flexible electrode materials recently. In particular, the development of high-performance flexible electrode materials is very important for wearable sensors for healthcare because it is necessary to continuously monitor and accurately detect body information such as body temperature, heart rate, blood glucose, and oxygen concentration in real time. In this study, we fabricated the nonenzymatic glucose sensor based on polyaniline/carbon nanotube fiber (PANI/CNT fiber) electrode. PANI layer was synthesized on the flexible CNT fiber electrode through electrochemical polymerization process in order to improve the performance of a flexible CNT fiber based electrode material. Surface morphology of the PANI/CNT fiber electrode was observed by scanning electron microscopy. And its electrochemical characteristics were investigated by chronoamperometry, cyclic voltammetry, electrochemical impedance spectroscopy. Compared to bare CNT fiber electrode, this PANI/CNT fiber electrode exhibited small electron transfer resistance, low peak separation potential and large surface area, resulting in enhanced sensing properties for glucose such as wide linear range (0.024~0.39 and 1.56~50 mM), high sensitivity (52.91 and 2.24 ㎂/mM·cm2), low detection limit (2 μM) and good selectivity. Therefore, it is expected that it will be possible to develop high performance CNT fiber based flexible electrode materials using various nanomaterials.

  • Characteristics of ethanol fermentation were investigated during the stationary culture of a killer yeast, Saccharomyces cerevisiae B15-1. Specific ethanol production rate reached the maximum level, 1.203 g-EtOH/g-cell-hr, at 150 g/l of the initial glucose concentration. No big differences were obtained in ethanol fermentability based on the initial sugar concentration below 150 g/l. When 200 g/l of sugar was used, fermentability dropped significantly. Although the final cell mass and the amount of ethanol produced were increased, their increase rates were declined according to the increase of initial sugar concentration. It was found that most of the sugar used below 150 g/l of concentration could be changed to ethanol. However, when 200 g/l of sugar was used, some of them remained in the media even after increase of cell mass and fermentation stopped. The ethanol yield was decreased when initial sugar concentration was high, and were increased when the amount of ethanol produced was increased and finally reached the plateau over 60 g/l of ethanol concentration.

Poly(3-hydroxybutyrate) Extrusion by Cells of Recombinant Escherichia coli

  • Poly(3-hydroxybutyrate) (PHB) was synthesized and accumulated intracellularly to a high concentration (7 g/l) by cultivating recombinant Escherichia coli XL1-Blue (pSYLl05) in a complex medium containing 20 g/l glucose. The morphology of PHB granules was examined by transmission electron microscopy. The PHB granules synthesized in recombinant E. coli were much larger than reported values for wild type microorganisms, and were often irregularly shaped. Some cells were apparently extruding PHB into the medium, which suggests that PHB granules maintain some fluidity and cells become fragile due to PHB accumulation.

  • Streptozotocin (STZ) is a naturally occurring nitrosoamide used extensively to produce diabetes in experimental animals. Since streptozotocin reportedly decrease deformability of red blood cells (RBC), we sought to investigate its potential toxicity at RBC. In addition to elevation blood glucose, 100mg/kg iv streptozotocin caused significant RBC hemolysis in female Sprague-Dawley at 48 hrs post treatment. Streptozotocin induced hemolysis was found to be dose and time-dependent` complete hemolysis required a relatively high streptozotocin concentration (500 mM, 4 hrs incubation), which is much more than in vivo dose.

Effect of Cassia tora Ethanol Extract on the Lipid Levels of Serum and Liver in Rats Fed High Cholesterol Diet (결명자 에탄올 추출물이 고콜레스테롤 식이를 섭취한 흰쥐의 체내 지질함량에 미치는 영향)

  • This study was undertaken to investigate the effect of Cassia tora ethanol extract on the lipid levels in serum and liver of rats fed high cholesterol diet. Experimental rats were divided into the following 4 groups: normal diet group, high cholesterol diet group, high choleslerol-0.25% C. tora ethanol extract group and high cholesterol-0.5% C. tora ethanol extract group. After 4 weeks, rats were sacrificed and analyzed the serum lipid profiles, activities of serum alanine aminotransferase (AST), aspartate aminotransferase (ALT), hepatic glucose-6-phosphate dehydrogenase (G6PDH) and malic enzyme (ME). It was also determined the contents of total cholesterol, triglyceride and thiobarbituric acid reactive substances (TBARS) in liver. There was no difference in weight gains between experimental groups. The concentrations of serum total cholesterol, free cholesterol, triglyceride and free fatty acid were tended to be decreased in C. tora groups compared with control group. HDL-cholesterol concentration was significantly decreased in high cholesterol diet group and slightly increased by C. tora ethanol extract feeding. The contents of liver cholesterol and triglyceride were higher in high cholesterol diet group than normal group, but significantly decreased by feeding of C. tora ethanol extract. Supplementation of 0.5% C. tora extract decreased significantly the activities of hepatic G6PDH and ME. Activities of serum AST, ALT and contents of liver TBARS were tended to be increased with high cholesterol diet and reduced by C. tora ethanol extract supplementation but had not significance. These results suggest that C. tora ethanol extract may exert a lipid lowering effect in serum and liver of rats.

Identification and Cultivation of Pseudomonas fluorescens Antagonistic to Pseudomonas tolaasii (Pseudomonas tolaasii 길항세균인 Pseudomanas fluorescens의 분리 및 배양)

  • Pseudomanas fluorescens was selected from mushroom and studied in both batch and continuous culture in order to find out optimum conditions for cultivation. P. fluorescens is an aerobic bacterium and antagonistic to Pseudomonas tolaasii which causes blotch disease on the mushroom cap. Cells of P. fluorescens were grown well on medium containing 30g/L of glucose, whereas the growth was inhibited with the glucose concentration at higher than 30g/L. The highest value of specific growth rate and productivity were obtained when using 10g/L of yeast extract. Optimum concentrations of $NH_4Cl$ and $(NH_4)_2SO_4$ for culture were found to be 1.0g/L and 0.1g/L respectively. Optimum concentration of $MgSO_4{\cdot}7H_2O$ used as a sulfur source was 1.0g/L. It was also found that the cell concentrations were at the maximum level when grown on the medium containing 1.0g/L of $KH_2PO_4$ and 0.1g/L of $CaCl_2$. Also, the optimum culture conditions were $30^{\circ}C$ and pH 6.0. Cultivation of P.fluorescens at high initial dissolved oxygen (D.O) value led to a decrease of bacterial productivity in batch culture. Maximum productivity was achieved at 68 for the initial D.O value.

  • Carob waste is a useful raw material for the second-generation ethanol because 50% of its dry weight is sucrose, glucose, and fructose. To optimize the process, we have studied the influence of the initial concentration of sugars on the fermentation performance of Saccharomyces cerevisiae. With initial sugar concentrations (S0 ) of 20 g/l, the yeasts were derepressed and the ethanol produced during the exponential phase was consumed in a diauxic phase. The rate of ethanol consumption decreased with increasing S0 and disappeared at 250 g/l when the Crabtree effect was complete and almost all the sugar consumed was transformed into ethanol with a yield factor of 0.42 g/g. Sucrose hydrolysis was delayed at high S0 because of glucose repression of invertase synthesis, which was triggered at concentrations above 40 g/l. At S0 higher than 250 g/l, even when glucose had been exhausted, sucrose was hydrolyzed very slowly, probably due to an inhibition at this low water activity. Although with lower metabolic rates and longer times of fermentation, 250 g/l is considered the optimal initial concentration because it avoids the diauxic consumption of ethanol and maintains enough invertase activity to consume all the sucrose, and also avoids the inhibitions due to lower water activities at higher S0 .
