The effect of mulberry leaves and yacon tuber extracts (MYE) on antioxidant was tested in this study. The present study investigated the in vivo effects of the anti-oxidative effect of MYE on catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST), glutathione peroxidase (GSH-Px), and thiobarbituric acid reactive substances (TBARS). The seven-day acclimation of the mice was divided into six groups: Normal diet group (NOR), high fat diet group (HFD), high fat diet with 0.5% hydroxycitric acid group diet group for positive group (HHCA), high fat diet with 1% mulberry leaf and 1% yacon diet group (MYE-1), high fat diet with 3% mulberry leaf and 3% yacon group (MYE-3) and high fat diet with 5% mulberry leaf and 5% yacon group (MYE-5). The effect of serum antioxidant in the catalase of MYE-1, MYE-3, and HHCA comparing to HFD by 31.0%, 27.7% and 45.2%, respectively (P<0.05~0.01). The effect on hepatic antioxidant in the catalase of HFD was significantly increased 3.7 (77.3%) times than that of NOR (P<0.01). But, the activities of catalase were decreased significantly in MYE-1, MYE-3, MYE-5 and HHCA by 21.7%, 24.2%, 24.9%, and 28.8% compared to HFD, respectively. GSH-Px was significantly decreased in MYE-1, MYE-3, MYE-5 and HHCA by 15.5%, 37.1%, 23.4%, and 23.7% compared to HFD, respectively (P<0.05). The activities of CAT, SOD, GST, GSH-Px, and TBARS were more significantly decreased in MYE-1 and MYE-3 than those of HFD and HHCA. MYE have shown significant effects on anti-oxidative function against high fat diet.
Baek-kimchi (whitish baechu kimchi) was evaluated for anti-obesity properties and effects on triglyceride (TG) and cholesterol in blood and adipose tissues in rats fed a high fat (20 %) diet, and compared to the similar effects of baechu kimchi. Baek-kimchi does not use red pepper powder but contains higher levels of sliced radish and pear than baechu kimchi. SD rats were raised for four weeks on either a normal diet (ND, based on the AIN-93M diet), high fat diet (HFD, supplemented with 16% lard oil in the ND), or HFD containing 5 % baek-kimchi or 5 % baechu kimchi. Feed consumption was not different among the groups, but weight gains were significantly lower in the groups fed either the normal diet or HFD with baek-kimchi or baechu kimchi diets than the group fed HFD alone. The weights of liver and epididymal and perirenal fat pads in baek-kimchi and baechu kimchi diet groups were lower than those of the HFD groups, but the baek-kimchi diet group had lower epididymal and perirenal fat pad weights than the baechu kimchi diet group (p<0.05). The baechu kimchi dietary group also had significantly lower triglyceride and cholesterol contents in liver and epididymal and perirenal fat, reversing the higher levels seen in HFD. Baek-kimchi and baechu kimchi diets were also effective in lowering serum triglyceride and cholesterol levels (p<0.05). These results suggest that baek-kimchi and baechu kimchi consumption can reverse the effects of HFD on weight gain and blood and tissue lipids, and that baek-kimchi is more effective than baechu kimchi. The greater effect is probably due to the higher content of radish and pear used in baek-kimchi.

Brown alga (Ishige okamurae; IO) dietary supplements have been reported to possess anti-diabetic properties. However, the effects of IO supplements have not been evaluated on glucose metabolism in the pancreas and skeletal muscle. C57BL/6 N male mice (age, 7 weeks) were arranged in five groups: a chow diet with 0.9% saline (NFD/saline group), high-fat diet (HFD) with 0.9% saline (HFD/saline group). high-fat diet with 25 mg/kg IO extract (HFD/25/IOE). high-fat diet with 50 mg/kg IO extract (HFD/50/IOE), and high-fat diet with 75 mg/kg IO extract (HFD/75/IOE). After 4 weeks, the plasma, pancreas, and skeletal muscle samples were collected for biochemical analyses. IOE significantly ameliorated glucose tolerance impairment and fasting and 2 h blood glucose level in HFD mice. IOE also stimulated the protein expressions of the glucose transporters (GLUTs) including GLUT2 and GLUT4 and those of their related transcription factors in the pancreases and skeletal muscles of HFD mice, enhanced glucose metabolism, and regulated blood glucose level. Our results suggest Ishige okamurae extract may reduce blood glucose levels by improving glucose metabolism in the pancreas and skeletal muscle in HFD-induced diabetes.
This study evaluated the effect of Allium hookeri roots on lipid metabolism of the serum, liver, and adipose tissues induced by a high-fat diet in male Sprague-Dawley rats. The rats were divided into four groups with 8 rats per group for 4 weeks: the normal-diet group (N), the high-fat diet (HFD) group, the HFD containing 3% Allium hookeri (HFD-A3) group, and the HFD containing 5% Allium hookeri (HFD-A5) group. The results showed that the body weight gain and food intake of rats in the HFC-A3 and HFC-A5 groups were significantly decreased compared with those in the HFD group. The epididymal adipose tissue weight in the HFD-A5 group was significantly decreased compared with those in the HFD group, and adipose tissue weights of liver and mesenteric adipose tissues in the HFD-A3 and HFD-A5 groups were significantly decreased compared with those in the HFD group. Serum triglyceride, total cholesterol and LDL cholesterol concentrations, atherogenic index, and cardiac risk factor were significantly lower in the HFD-A3 and HFD-A5 groups than in the HFD group. Serum lipid profiles, as well as ALT and AST activities did not show any difference in all groups. Serum ALP and LDH activities were suppressed in the HFD-A5 group compared with those in the HFD group. The hepatic triglyceride and total cholesterol levels of rats in the HFD-A5 group was significantly lower than those in the HFD group. Moreover, triglyceride and total cholesterol in the epididymal and mesenteric adipose tissues were significantly lower in the HFD-A5 group than in the HFD group. These results demonstrated that the intake of Allium hookeri showed a hypolipidemic effect, changing the lipid metabolsim of a high-fat diet induced rats.
Obesity is the most common nutritional disorder in the developed world and has become a global epidemic in recent years. In this study, Zanthoxylum planispinum extracts (ZPE) were evaluated on the effect on inhibition of pancreatic lipase and lipid metabolism by oral treatment for 2 months in high-fed diet obesity-induced Balb/c mice. The ZPE showed pancreatic lipase inhibitory activity with $IC_{50}$ of $0.3{\mu}g/ml$. No significant difference in feed intake was observed among the groups. The high-fat diet-treated Z. planispinum extracts groups (HFD+ZPE, 100mg/kg) significantly decreased body weight compared to the high-fat diet vehicle groups (HFD, p<0.05). The high-fat diet-treated Xenical$^{(R)}$ groups (HFD+Xenical, n=10, 30mg/kg) also showed a significant reduction of body weight compared to HFD (p<0.05). Biochemical parameters (triglyceride, total cholesterol, and high-density lipoprotein cholesterol) in HFD plus ZPE diet groups were significantly lower than those of the HFD groups (p<0.05). These results indicated that ZPE more effectively suppressed the effects of HFD on body fat gain with the inhibitory effect on pancreatic lipase.
Purpose: Obesity is associated with alterations in vitamin D metabolism and elevation of parathyroid hormone (PTH). Increased PTH level in obesity is likely one of the factors contributing to the dysregulation of vitamin D metabolism. We investigated the effects of lowering the PTH level in high-fat diet-induced obese mice on vitamin D metabolism. Methods: Five-week-old male C57BL/6N mice were fed either with control (10% energy as fat) or high-fat (60% energy as fat) diets ad libitum for 12 weeks, and vehicle or cinacalcet HCl (30 ㎍/g body weight) was gavaged daily during the final week of the experiment. The following groups were studied: CON (control diet + vehicle), HFD (high-fat diet + vehicle), and HFD-CIN (high-fat diet + cinacalcet HCl). PTH, 1,25-dihydroxyvitamin D (1,25[OH]2D), 25-hydroxyvitamin D (25[OH]D), calcium, and phosphate levels in circulation, and the expression of genes related to vitamin D metabolism in the liver and kidneys were determined. Results: Renal 1α-hydroxylase expression in the HFD group was higher than that in the CON group despite the lack of a difference in the PTH levels between the 2 groups. The plasma PTH level in the HFD-CIN group was 60% lower than that in the HFD group (p < 0.05). In parallel, the HFD-CIN group had lower adipose tissue amount (9% lower), renal 1α-hydroxylase expression (48% lower), and plasma 1,25(OH)2D concentration (38% lower) than the HFD group. Conclusion: Lowering the PTH levels in high-fat diet-induced obese mice recovered the expression of renal 1α-hydroxylase and might be associated with lower amounts of white adipose tissue.
Objectives This study aimed to investigate the effect of Bangkibokryeong-tang (BBT, Fangjifuling-tang) on blood glucose and body fat in high-fat diet-induced obese mice. Methods The experimental animals were divided into five groups- normal diet-fed control (ND), high-fat diet-fed control (HFD), HFD+BBT 75, HFD+BBT 150, and HFD+olistat as a positive drug control group. Markers of obesity, such as body weight, organ weight, diet efficiency, and serum levels of total cholesterol, triglycerides, lipid content, leptin, adiponectin, glutamic oxaloacetic transaminase (GOT)/glutamic pyruvic transferase (GPT)/lactate dehydrogenase (LDH), blood glucose, and insulin, were measured. Furthermore, results of the oral glucose tolerance test and ${\alpha}-glucosidase$ inhibition activity were examined in obese mice. Results Mice treated with BBT demonstrate lower body and organ weight, and reduced weight gain and food efficiency than that in the HFD-only control group. In addition, BBT decreased lipid accumulation in the liver and the levels of enzymes such as GOT, GPT, and LDH in the serum. Furthermore, the levels of triglycerides, total cholesterol, low density lipoprotein (LDL), and leptin were decreased in the serum but the levels of high density lipoprotein (HDL) and adiponectin were increased in the BBT-treated group compared with the control group. The BBT-treated group also demonstrated decreased blood glucose and insulin concentrations induced by feeding on a high-fat diet and improved glucose tolerance. Conclusions Based on the results above, BBT may reduce body fat and hyperglycemia in HFD-induced obesity. This suggests that BBT may be clinically useful in the treatment of obesity.
BACKGROUND/OBJECTIVES: Changes in nutritional status during gestation and lactation have detrimental effects on offspring metabolism. Several animal studies have shown that maternal high-fat diet (HFD) can predispose the offspring to development of obesity and metabolic diseases, however the mechanisms underlying these transgenerational effects are poorly understood. Therefore, we examined the effect of maternal HFD consumption on metabolic phenotype and hepatic expression of involved genes in dams to determine whether any of these parameters were associated with the metabolic outcomes in the offspring. MATERIALS/METHODS: Female C57BL/6 mice were fed a low-fat diet (LFD: 10% calories from fat) or a high-fat diet (HFD: 45% calories from fat) for three weeks before mating, and during pregnancy and lactation. Dams and their male offspring were studied at weaning. RESULTS: Dams fed an HFD had significantly higher body and adipose tissue weights and higher serum triglyceride and cholesterol levels than dams fed an LFD. Hepatic lipid levels and mRNA levels of genes involved in lipid metabolism, including $LXR{\alpha}$, SREBP-2, FXR, LDLR, and ABCG8 were significantly changed by maternal HFD intake. Significantly lower total liver DNA and protein contents were observed in dams fed an HFD, implicating the disturbed liver adaptation in the pregnancy-related metabolic demand. HFD feeding also induced significant oxidative stress in serum and liver of dams. Offspring of dams fed an HFD had significantly higher serum cholesterol levels, which were negatively correlated with liver weights of dams and positively correlated with hepatic lipid peroxide levels in dams. CONCLUSIONS: Maternal HFD consumption induced metabolic dysfunction, including altered liver growth and oxidative stress in dams, which may contribute to the disturbed cholesterol homeostasis in the early life of male mice offspring.
Objectives : Samhwangsasim-tang (SST), Hwangryeonhaedok-tang (HHT), Ukgan-san (UGS), Onjunghwadam-hwan (OHH) and Samul-tang (SMT) have been used for the treatment of various diseases. This study was performed to compare the anti-obesity effects of 5 herbal formulas in high fat diet-(HFD) induced obese mice. Methods : The mice were randomly divided into seven groups that were fed a normal diet (ND), a HFD, a HFD plus SST (HFD + SST), a HFD plus HHT (HFD + HHT), a HFD plus UGS (HFD + UGS), a HFD plus OHH (HFD + OHH), or HFD plus SMT (HFD + SMT) at 300 mg/kg/day for 7 weeks. All groups were assayed for body weights, food efficiency ratio (FER), final liver and fat weight and blood biochemical parameters. Results : The increased body weights, food efficiency ratio (FER), and serum total triglyceride were decreased in HFD + OHH group relative to the same measurements in HFD group. Furthermore, the HFD + SST group significantly reduced FER, liver and abdominal subcutaneous fat weight gains, and serum total triglyceride, whereas HDL-cholesterol level was increased compared to HFD group. Conclusions : These results suggested that HFD + OHH and HFD + SST exert anti-obesity effects in HFD-induced obese mice.
This study investigated the effects of onion juice on the serum lipid components and blood pressure in obese rats fed high fat diets. Thirty-nine of Sprague-Dawley rats were divided into six groups, and were treated for 8 weeks: (1) normal diet (ND); (2) high fat diet (HFD); (3) HFD for first 4 weeks and high fat diet with 40% onion juice for the last 4weeks (H-H+O); (4) HFD with 40% onion juice for 8 weeks (H+O); (5) HFD for first 4 weeks and ND for the last 4 weeks (H-N); (6) HFD for first 4 weeks and ND with 40% onion juice for the last 4 weeks (H-N+O). The rates of increasing body weight were reduced in H+O and H-H+O groups compared with HFD group. The levels of triglyceride, low density lipoprotein-cholesterol and total cholesterol in blood serum were significantly decreased in the H+O and H-H+O groups compared with the HFD group. Administration of onion reduced the size of adipocyte, steatosis, and serum hyperlipidemia in obese rats fed HFD. Moreover, the antihypertensive effects of onion were observed in obesity rat fed HFD. Overall results suggest that onion reduces the serum lipid components and improves hypertension in obese rat fed HFD.
