Antioxidative and Anti-aging Effects of Sancho (Zanthoxylum schinifolium) Extract in Rats Fed High Fat Diet

  • This study was performed to investigate antioxidative and anti-aging action of extracts from Sancho (Zanthoxylum schinifolium) leaves. Two extracts were obtained by 80% methanol extraction followed by subsequent fractionation with methylene chloride (MC) and n-butanol (B) and fed at one or three levels to rats on normal level (5%) of fat (control) and high fat(20%) in diets. Male Sprague-Dawley rats weighing about 100 g were divided into ten groups such as control diet group(C), control diet+0.50%B group (CB), control diet+0.50%MC group (CMC), high-fat diet group (HF), high-fat diet+0.25%B group (HBL), high-fat diet+0.50%B group (HBM), high-fat diet+0.75%B group (HBH), high-fat diet+0.25%MC group (HMCL), high-fat diet+0.50%MC group (HMCM) and high-fat diet+ 0.75%MC group (HMCH) and fed each diet for four weeks. The effects of the extracts on antioxidant enzyme activities and indices of lipid peroxidation and aging were seen only in high fat diet groups. Hepatic superoxide dismutase and aryleaterase activities were not changed by Sancho extracts. But glutathione peroxidase, catalase and paraxonase activities were significantly restored by both MC and B at the level of 0.75% lipid peroxide which was increased by high fat diet was significantly reduced by B and MC at the level of 0.25% and over. Lipofuscin fluorescence and cabonyl value were increased by high fat diet were reduced by B and MC at the level of 0.5% and over. It is concluded that the Sancho extracts can be utilized as functional ingredients of health foods for reducing oxidative stress.

Effect of Silybin on Body Weight and Glucose Tolerance in High-fat-diet Induced Obese Mice (고지방 식이로 유도된 비만 쥐에서 실리빈 (Silybin)이 체중 및 내당 능에 미치는 영향)

  • In this study, we investigated the effects of silybin on body weight and glucose tolerance in mice fed high fat diet mice. We found that body weight, plasma TG contents, fat size, glycerol, 3-hydroxybutyrate and total cholesterol were significantly decreased in silybin (500 mg/kg) supplemented groups compared to high fat diet group. Whereas, total food intake was not changed between high fat diet group and high fat diet plus silybin group. Futhermore, supplement of high fat elevated the glucose intolerance and was improved in silybin supplement group. Finally, we examined the effect of silybin on circulating adipocytokine level to explore the possible mechanism by which silybin improves high fat diet-induced obesity and diabetes. The silybin supplement significantly reduced the level of adipocytokine, such as leptin, resistin, IL-6, and MCP-1 induced by high fat diet. These results suggest that silybin can be used to improve obesity and diabetes.

Effects of Indigestible Dextrin on Lipid Metabolism in Rats Fed Normal or High Fat Diet (식이지방 수준이 다른 식이를 섭취한 흰쥐의 체내 지질대사에 대한 난소화성 덱스트린의 섭취 효과)

  • The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. These effects are mediated by the increase of facal cholesterol and bile acid excretion. (Korean J Nutrition31(6) : 981-990, 1998)

The Effects of Ginsenoside Re on High-Fat Diet induced Insulin Resistance in Muscle (Ginsenoside Re가 골격근의 고지방식 유도 인슐린 저항성에 미치는 영향)

  • We evaluated the effect of the ginsenoside Re on insulin resistance of glucose transport in muscles of rats made insulin resistant with a high fat diet. After a week of adaptation period to the laboratory environment, 40 male wistar rats were randomly assigned into 2 groups (Chow diet group; CD, n = 20, High fat diet group; HFD, n = 20). After 5-week of high fat diet, Food was removed after 6:00 PM the day before the experiment. The following morning, rats were anesthetized by an intraperitoneal injection of pentobarbital sodium (50 mg/kg body wt), and the soleus muscles were removed. Before incubation, the soleus muscle was split longitudinally into strips with an average weight of 15~20 mg. After the muscle dissection was completed, the abdominal cavity was opened, and the epididymal, mesenteric, and retroperitoneal fat pads were removed and weighed. Treatment of muscles with ginsenoside Re alone had no effect on glucose transport. The high fat diet resulted in ~50% decreases glucose transport rate in soleus muscles. Treatment of muscles with ginsenoside Re in vitro for 90 min completely reversed the high fat diet-induced insulin resistance of glucose transport in soleus muscles. This effect of ginsenoside Re is specific for insulin stimulated glucose transport, as Re treatment did not reverse the high fat diet-induced resistance of skeletal muscle glucose transport to stimulation by contraction. Our results show that the ginsenoside Re induces a remarkably rapid reversal of high fat diet-induced insulin resistance of muscle glucose transport.

Effect of a five-week high-fat diet on serum-lipid composition and LPL mRNA expression in rat skeletal muscles

  • Our purpose was to investigate the effect of a 5-week high-fat diet on the body weight, blood components [triglycerides (TGs), free fatty acids (FFAs), and glucose], and lipoprotein lipase (LPL) mRNA expression in the skeletal muscles of rats. Body weight increased overtime in experimental and control groups without significant differences. In terms of the blood components, the density of TG was significantly lower in the high-fat diet group compared to the control, whereas FFA and glucose levels were similar in the two groups. Although the levels of glyceraldehyde 3-phosphate dehydrogenase(GAPDH) mRNA expression were similar between the groups, LPL mRNA expression was significantly higher in the high-fat diet group than that in the control group. The results of this study suggest that a high-fat diet enhances LPL mRNA expression, as well as possibly increases fat metabolism. For a better understanding of the relationship between diet patterns and fat metabolism, further analysis of genes related to the fat metabolism is warranted.

Effects of taeeumjoweetang Herbal-Acupuncture on plasma and liver lipid composition and antioxidative capacity in rat fed high fat diet (태음조위탕(太陰調胃湯) 약침(藥鍼)이 고지방 급여 흰쥐의 혈장 및 간장의 지질구성과 항산화능에 미치는 영향)

  • Objectives : In order to measure the Effects of taeeumjoweetang Herbal- Acupuncture at joksamri(ST36) and kwanweun(CV4) on liver and plasma lipid composition and antioxidative capacity in rat fed high fat diet. Results : Concentrations of plasma triglyceride, total cholesterol and LDL-cholesterol showed a tendency to increase in the high fat diet group. However Herbal-Acupuncture groups showed a lower values than control groups. HDL-cholesterol showed a tendensy to decrease in high 131 fat diet groups and in high fat diet groups, these values showed no significantly different. Liver total cholesterol values showed no significantly different in all treatment groups. Triglyceride concentration showed a high value in control group and other treatment groups showed no significantly different. plasma GOT and GPT values showed a tendency to increase in high fat diet group. however these values decreased in Herbal-Acupuncture group. The concentration of TBARS in liver and plasma showed a high values in high fat diet group, however these values showed a tendency to decrease in aqua- acupuncture group. Glutathione peroxidase, superoxide dismutase and catalase activity values showed a low values in high fat diet group, however these values showed a tendency to increase in Herbal-Acupuncture group.

Effect of Ginseng on Fat Accumulation in the Obese Rats Induced by High Fat Diet (인삼이 고지방식이에 의한 비만유도 Rat에서 지방축적에 미치는 영향)

  • Obesity is common disease resultly accumulated excess fat. In the model for obesity induced by high fat diet contains 30% fat, administration of ginseng extract inhibited increment of body weight, epididymal fat pads and enlargement of fat cell size. This was as the result of inhibition of lipogenesis in the liver and fat accumulation in the adipose tissues.

Effect of Increasing Dietary Fat on Plasma Lipoprotein in Young Korean Women (지방 섭취 증가가 일부 젊은 여성의 혈장 지단백 조성에 미치는 영향)

  • This study was conducted to investigate the effect of increasing dietary fat on plasma lipoprotein in young Korean women. During the three weeks of experimental periods, 8 healthy college women consumed a regular diet, a medium fat diet and a high fat diet for one week, consecutively. The percentages of fat from energy intake in the subjects on the regular, medium, and high fat diets were 20, 29 and 38%, respectively. Our data showed that the dietary intakes of fat and cholesterol had an influence on the plasma lipoprotein. The medium fat diet enhanced the blood glucose concentration and the plasma HDL, and the high fat diet resulted in increases in the blood glucose concentration and the plasma LDL as well as HDL. Also the type of diets led to changes in the composition of lipoprotein. When on the medium fat diet, HDL and LDL had higher phospholipid content. On the high fat diet, LDL had higher phospholipid, cholesterol and triglyceride contents, and HDL included higher cholesterol content.

Effects of Porphyran and Insoluble Dietary Fiber Isolated from Laver, Porphyra yezoensis, on Lipid Metabolism in Rats Fed High Fat Diet (김에서 분리한 포피란과 불용성 식이섬유가 고지방식이 섭취 흰쥐의 지질대사에 미치는 영향)

  • This study was conducted to investigate the hypocholesterolemic effects of porphyran and insoluble dietary fiber isolated from laver in rats fed high fat diet containing 1% cholesterol, 0.25% sodium cholate and 12% lard. Rats were fed, ad libitum, diets containing 5% diet fiber as cellulose(normal control or high fat control), porphyran or insoluble dietary fiber for 4 weeks. Among the groups fed high fat diet, liver weight was significantly lower in high fat porphyran group than high fat control. Plasma GOT, GPT, total cholesterol, cholesteryl ester, LDL-cholesterol and liver total cholesterol concentration were significantly lower in high fat porphyran group than high fat control. The feeding of porphyran significantly increased fecal cholesterol and bile acid excretion. The feeding of insoluble dietary fiber had no significant effect on either plasma or liver cholesterol levels, although fecal cholesterol level in the insoluble dietary fiber group was significantly higher than that in the high fat control. The results indicate that porphyran isolated from the laver may exert their hypocholesterolemic effect by increasing excretion of fecal bile acid and cholesterol.

Effect of n-3 Polyunsaturated Fatty Acids on Glucose Uptake of Soleus Muscle in NIDDM Diabetic Rats (NIDDM 당뇨병 흰쥐에서 n-3 다가불포화지방산이 가자미근의 Glucose Uptake에 미치는 영향)

  • The purpose of this study was to investigate the effects of n-3 polyunsaturated fatty acids(PUFA) on glucose and lipids metabolism in high-fat diet rate. Rats were randomly assigned to normal, high-fat with n-3 PUFA and high-fat dietary groups. Experiments were carried out after 5 weeks feeding with prescriptive diets following 7 hrs fasting. Body weight gains tended to be higher in high-fat fed rats than normal. Blood glucose was increased (p<0.05) by high-fat diet compared with normal diet, and decreaseed (p<0.05) to normal level by n-3 PUFA. Plasma insulin level was significcantly higher (p<0.01) in high-fat diet rats than that of normal-diet rats, and also decreased (p<0.01) by n-3 PUFA. Glucose up take of soleus muscle in vitro was decreased markedly in high-fat fed rats than normal diet rats at 0, 1, 10, and 100nM insulin concentration. Therefore insulin sensitivity and responsiveness were decreased by high-fat diet. Omega-3 PUFA made a recover(p<0.01) insulin sensitivity to almost normal level, and improved (p<0.05) insulin responsiveness in some extent. In conclusion, the results suggest that metabolic disorder of glucose and insulin resistance of skeletal muscle are caused by high-fat diet and n-3 PUFA can ameliorate metabolic disorder and insulin resistance.
