Effects of Phenytoin and Diazepam on the Seizure Activity in the Cortical Dysplasia Animal Models

  • Dysplasia-associated seizure disorders are markedly resistant to pharmacological intervention. Relatively little research has been conducted studying the effects of antiepileptic drugs(AEDs)on seizure activity in a rat model of dysplasia. We have used rats exposed to methylazoxymethanol acetate(MAM) in utero, an animal model featuring nodular heterotopia, to investigate the effects of AEDs in the dysplastic brain. Pilocarpine was used to induce acute seizure in MAM-exposed and age-matched vehicle-injected control animals. Field potential recordings were used to monitor amplitude and numbers of population spikes, and paired pulse inhibition in response to stimulation of commissural pathway. Two commonly used AEDs were tested: diazepam 5, 2.5 mg/kg; phenytoin 40, 60 mg/kg. Diazepam(DZP) and phenytoin(PHT) reduced the amplitude of population spike in control and MAM-exposed rats. However, the amplitude of population spike was nearly eliminated in control rats as compared to the MAM-exposed rats. Pharmaco-resistance was tested by measuring seizure latencies in awake rats after pilocarpine administration(320 mg/kg, i.p.) with and without pretreatment with AEDs. Pre-treatment with PHT 60 mg prolonged seizure latency in control rats, but not in MAM-exposed animals. The main findings of this study are that acute seizures initiated in MAM-exposed rats are relatively resistant to standard AEDs assessed in vivo. These data suggest that animal model with cortical dysplasia can be used to screen the effects of potential AEDs.

Effects of Ethosuximide on the Pilocarpine Induced Seizure in Rat Model of Neuronal Migration Disorder

  • Cortical malformation-associated epileptic seizures are resistant to conventional anticonvulsant drugs. Relatively little research has been conducted on the effects of antiepileptic drugs (AEDs) on seizure activity in a rat model of dysplasia. We have used rats exposed to methylazoxymethanol acetate (MAM) in utero, an animal model featuring nodular heterotopia, to investigate the effects of ethosuximide (ETX) in the dysplastic brain. Pilocarpine was used to induce acute seizure in MAM-exposed and age-matched vehicle-injected control animals. Field potential recordings were used to monitor the amplitude and number of population spikes, and paired pulse inhibition in response to stimulation of the commissural pathway. Pharmaco-resistance was tested by measuring seizure latencies after pilocarpine administration (320 mg/kg, Lp.) with and without pre-treatment with ETX. Pre-treatment with 300 mg of ETX significantly prolonged the latency to the status epilepticus (SE) in both control and MAM-treated groups. Pre-treatment with ETX 100mg and ETX 200 mg had little effect in MAMexposed rats. However, ETX 200 mg prolonged the latency to the SE in control groups. Spontaneous field potential and secondary after-discharges were higher for MAM-treated rat in comparison with control rats injects with ETX. The main findings of this study are that acute seizures initiated in MAM-exposed rats are relatively resistant to standard ETX assessed in vivo. These data suggest that ETX do not prolong seizure latencies in MAM-rats exposed to pilocarpine.

  • Site-specific performance is always the real on-site work taking place at the site. Hence, it deals with the reality, in other words, the time of creation and formation. And it creates value and meaning through the interaction and continuous direct communication process between the performers, audience and the local residents. In this performance, the audience's status as the passive observer changes. They become the co-agent who actively lead and complete the performance through their own experience. We have examined the The Working Methods of Site-specific Performance and Aesthetics of Effect through four Korean performances ; Marie, An aesthetic experiment of site as the storyteller; Heterotopia and Urban Movement Research or Play: We Will Move Your Sofa, as performances which have Revealed history, politics, institution engraved in the site ; A Song of Mandala and Miracle, as a ritualistic site-specific performance at the historic site. Some remarks on Site-specific performance ; First, In Site-specific performance, the habitus peculiar to the stage art and the mode of reception are changed. Second, a new mode of theatrical communication requires creator and audience to have a sharp aesthetic sensibility and to change one's perceptual habit. Third, Site-specific performance can act as a demonstration for the viewpoints of political activism through what could be called a dramatic close-up effect. Fourth, Site-specific performance also has the risk of merely becoming an unfocused and scattered performance or degenerating into a pseudo-sightseeing. To avoid this, an in-depth study of the site and its socio-cultural context, and the clear motivation with which one is trying to reveal and tell from the site must be indispensable. As the co-agent, the audience should also be aware and think about what the given performance signifies today.

The Heterotopiatic Placeness of North Korea and a Priming Effect: The Case of The Korean-American (북한의 헤테로토피아적 장소성과 점화 효과: 재미교포를 대상으로)

  • The purpose of this study is to prove double faced heterotopiatic placeness of North Korea which can be highlighted to one aspect through priming test. Korean-Americans are found to have a perception of North Korea as a closed society where political leader cult is practiced, Pyongyang as a display city. They have Christian nostalgia toward it as a place where Christianity was first introduced to. North Korea's heterotopiatic placeness is sharing the 5,000 years of history of the Korea peninsula on the one hand and being a closed dictatorship place on the other. North Korea is kept isolated and closed but has had a liminal space through the intentional open system like special economic zone. Pyongyang is the city for specific class where it shows the heterotopiatic character. Priming is found effective in Yeongbyun, a place of extreme mixtures placeness as being the hometown of the beloved Korean poet Kim So-Wol and the site of nuclear weapon experiment but Korean-Americans have not found any priming effect regarding the Geumgang mountain tour. As to the Arirang performance, a man-made landscape expressing North Korea's sense of value and ideology, priming resulted in preference. This study raises the needs for understanding North Korea as a multifaced placeness and it can purposely be emphasized and changed to contribute the two Korea's unification.

Der anti-theatralische Topos im modernen Theater -Die Ästhetik der Abwesenheit und des Webens- (현대 연극의 반-연극적 지형 -부재와 직조의 미학-)

  • Die heutigen Experimente, die seit fast 20 Jahren in den Theaterbereiche $durchgef{\ddot{u}}hrt$ werden, sind so verschieden, $da{\ss}$ wir gar nicht voraussehen $k{\ddot{o}}nnen$, wieweit sie ihre Grenze mehr erweitern werden. Sie brechen alle vertrauten theatralischen Konventionen, ob diese Konventionen mit Text, Schauspielkunst oder Raum usw. zu tun haben. Und sie fragen sich selbst, was Theater in der heutigen simulierten Gesellschaft bedeuten soll. Die vorliegende Arbeit untersucht anhand einigen $Auff{\ddot{u}}hrungen$, die von solchen Selbstbefragen ausgehen, wieweit sie sich von den tradtionellen Theaterabeiten entfernen und welche neue $Theater{\ddot{a}}sthetik$ sie herzustellen versuchen, um der $gegenw{\ddot{a}}rtigen$ Gesellschaft, besonders nach 1990 Jahre, zu entsprechen. Nach dem Zusammenbruch des Ost-Blocks hat sich die Welt radikaler denn je $ver{\ddot{a}}ndert$. Durch die $revolution{\ddot{a}}ren$ Technologien wie Mobilfunk und Internet verwischen sich die Grenzen zwischen Wirklichkeit und Fiktion, zwischen Original und Nicht-original mehr und mehr. Die letztliche globale Finanz- und Wirtschaftskriese hat die ganze Welt in Schockzustand versetzt, aber man scheint bei den verschiedenen Schreckenzeneraien und den negativen Meldungen daraus noch keine "Lehre" gezieht zu haben. Die Experimente, die in dem Bereich der zeitigen Theaterarbeiten gewagt werden, sind die Versuche, die Theater selbst macht, um eine den radikalen gesellschaflichen $Ver{\ddot{a}}nderungen$ entsprechende neue $k{\ddot{u}}nstliche$ Aufgabe zu suchen. In dieser Arbeit $m{\ddot{o}}chte$ ich sie, $ant{\acute{i}}-theatralische$ Strategien nennen, weil sie jenseits der konventionellen theatralischen Arbeitsweise liegen. Diese $R{\ddot{a}}ume$, wo solche $Auff{\ddot{u}}hrungen$ gespielt werden und $nat{\ddot{u}}rlich$ auch den Zuschauerraum $einschlie{\ss}en$, sind, die $R{\ddot{a}}ume$ des Abwesenheit und des Abweichens,,, weil sie dem $gew{\ddot{o}}hnlichen$ Wunsch des Publikums, alles zu sehen, entgegenkommen. Sie fordern dem Zuschauer, der auch zu ihren wichtigen theatralische Element $geh{\ddot{o}}rt$, $da{\ss}$ er an ihren $Auff{\ddot{u}}hrungen$ mit den ganz anderen Sichtweise als den ${\ddot{u}}berlieferten$ aktiv, teilnehmen, $mu{\ss}$. Sie wollen ihre $Auff{\ddot{u}}hrungen$ zu den $Erfahrungsr{\ddot{a}}umen$ machen, wo der Zuschauer ${\ddot{u}}ber$ sich und sein eigenes Leben selbst reflektieren kann, indem er die wahrgenommenen Ereignisse zu seinen eigenen Erfahrungen $zusammenf{\ddot{u}}gt$. Dadurch wird die Dichtonomie zwischen den Sichtbaren und den Nicht-Sichtbaren $entsch{\ddot{a}}rft$.
