전통 처방의 Protein Tyrosine Phosphatase 1B 저해 활성 검색 및 작감황신부탕(芍甘黃辛附湯) 처방 분석 (Screening of Korean Traditional Prescriptions with Inhibitory Activity against Protein Tyrosine Phosphatase 1B and Analysis of Jakgamhwangsinbu-tang (芍甘黃辛附湯) Prescription)

  • In order to search for protein tyrosine phosphatase 1B (PTP1B) inhibitors as therapy of type 2 diabetes and obesity from Korean traditional prescriptions, we selected 58 traditional prescriptions based on a review of the Korean traditional medicine books. The hot water extracts of Korean traditional prescriptions were screened for the inhibitory activity against PTP1B. Among the tested extracts, water extracts of Jakgamhwangsinbu-tang, Seonbanghwalmyung-eum, and Takreeonjoong-tang showed relatively good inhibitory activity against PTP1B at the concentration of $30{\mu}g/ml$. Additionally, we evaluated PTP1B inhibitory effect for each herbal ingredient and composition in Jakgamhwangsinbu-tang (芍甘黃辛附湯). Of the tested ingredients from this herbal medicine, water extracts of Paeoniae Radix rubra and Rhei Rhizoma, and ethanol extracts of Paeoniae Radix alba, Rhei Rhizoma, Asiasari Radix, and Aconiti Tuber showed good PTP1B inhibitory effect. Herbal compositions composed of these active herbal ingredients exhibited significant activity for PTP1B inhibition over 70% at $7.5{\mu}g/ml$.

광환경 조절이 인삼의 생육과 진세노사이드 함량에 미치는 영향 (Effect of Controlled Light Environment on the Growth and Ginsenoside Content of Panax ginseng C. A. Meyer)

  • Background: The photosynthetic efficiency cool-season, semi-shade ginseng is normal at low morning temperatures, but drops at high afternoon temperatures. Therefore, optimal plant performance would be ensured if it were possible to control daily light transmission rates (LTR). Methods and Results: Plants were grown in a controlled light environment that replicated 11 AM conditions and comparatively analyzed against plant grown under normal conditions. Growth in the controlled light environment resulted in a 2.81 fold increase in photosynthetic efficiency with no change in chlorophyll content, although LTR were high due to low morning temperatures. Increased aerial plant growth was observed in the ginseng plants adapted to the controlled light environment, which in turn influenced root weight. An 81% increase in fresh root weight (33.3 g per plant on average) was observed in 4-year-old ginseng plants grown in controlled light environment compared to the plants grown following conventional practices (18.4 g per plant on average). With regard to the inorganic composition of leaves of 4-year-old ginseng plants grown in controlled light environment, an increased in Fe content was observed, while Mn and Zn content decreased, and total ginsenoside content of roots increased 2.37 fold. Conclusions: Growth of ginseng under a favorable light environment, such as the condition which exist naturally at 11 AM and are suitable for the plant's photosynthetic activity creates the possibility of large scale production, excellent-quality ginseng.

비만에서 장내 미생물 균총의 역할과 발효 한양의 활용 (The Role of Gut Microbiota in Obesity and Utilization of Fermented Herbal Extracts)

  • Complex microbial communities play an important role in the human health and co-evolved with human in the form of symbiosis. Many literatures provide new evidences that the increased prevalence of obesity cannot be attributed solely to changes in the human genome, nutritional habits, or reduction of physical activity in our daily lives. The intestinal flora was recently proposed as an environmental factor responsible for the control of body weight and energy metabolism. A number of studies suggest that the modulation of gut microbiota affects host metabolism and has an impact on energy storage and demonstrated a role for the gut microbiota in weight gain, fat increase, and insulin resistance. Variations in microbiota composition are found in obese humans and mice and the microbiota from an obese mouse confers an obese phenotype when transferred to an axenic mouse. As well, the gut microbial flora plays a role in converting nutrients into calories. Specific strategies for modifying gut microbiota may be a useful means to treat or prevent obesity. Dietary modulations of gut microbiota with a view to increasing bifidobacteria have demonstrated to reduce endotoxemia and improve metabolic diseases such as obesity. The fermentation of medicinal herbs is intended to exert a favorable influence on digestability, bioavailability and pharmacological activity of herbal extract. Therefore we also expect that the fermented herbal extracts may open up a new area to treat obesity through modulating gut microbiota.

동의보감(東醫寶鑑) 월국환가미방(越鞠丸加味方)의 방제학적(方劑學的) 분석(分析) (Analysis of Herbal formulation on Wolgug-hwan Series)

  • Objectives : This study was performed to analysis the composition of herbal formulations about a series of Wolgug-Hwan(WGH, formed by Gukchur-won and Sagung-san) in Dongeuibogam written by Heo Jun. Methods : The series of WGH in Dongeuibogam were selected by 'Prescription Lineage Graph' (http://164.125.206.43/PrescriptionLineageGraph.aspx) established by prof. Lee at Pusan National University. In the analysis of a series of WGH, we used some formulation books including classics. Results : Gaeuldogi-tang(GDT) is formed by WGH, Yeonggangchulgam-tang(YGT), Yugil-san (YIS) and Gungchul-san. Cheongul-san(CUS) is formed by WGH, Ijin-tang(IJT), YGT and Haegeum -hwan(HGH). Sosikcheongul-san(SCS) is formed by CUS and IJT. Jeungmiijin-tang(JIT) is formed by WGH, IJT, Sambaek-tang and Gyullyeonjichul-hwan. Sihoeokgan-tang(SET) is formed by WGH, Sihosogan-san(SSS) and Jigolpi-eum. Wolgukbohwa-hwan(WBH) is formed by WGH, Bohwa-hwan, Mokyangjichul-hwan and Hwangnyeon-tang(HNT). Chimsa-hwan(CSH) is formed by WGH, Injinho-tang(IHT), Yungmisamneung-hwan, Jeungmisamul-tang and Sinseongugi-tang. Conclusions : SCS, CUS, or JIT has been used frequently in case of phlegm-fire. These are formulated by the combination of WGH and IJT with Samseon-san or HGH or Jichul-hwan according to the special symptoms, respectively. SET which is formulated by WGH and SSS has been used in case of stagnation of liver-qi. GDT, WBH or CSH has been used in damp-heat. These are formulated by the combination of WGH with YIS, HNT, or IHT, respectively.

버섯 병재배 배지재료의 이화학성과 활용 (Physico-chemical Characteristics and Utilization of Raw Materials for Mushroom Substrates)

  • 본 시험은 버섯 병재배 농가에서 사용하고 있는 배지재료와 혼합배지의 수분함량, pH, 전탄소(T-C), 전질소(T-N), C/N율, 인산, 13종의 양이온 함량 등 이화학적 특성을 파악하여 기초자료로 활용하고자 수행하였다. 전국의 병재배 농가에서 임의로 선정한 35농가 중 팽나무버섯 13개소, 큰느타리버섯 14개소, 느타리버섯 8개소에서 배지재료 143점과 혼합배지 35점의 시료가 수집되었다. 배지재료의 성분함량은 동일한 명칭의 재료라도 2~5배로 차이가 많은 경우도 있었다. 그리고 농가에서 수집한 버섯재배용 혼합배지의 C/N율은 팽나무버섯과 큰 느타리버섯 배지가 $28{\pm}4$, 느타리버섯 병재배용 배지는 $21{\pm}2$의 범위이었으며 중금속 등 특정성분에 특이사항은 없었다. 그리고 배지재료별 성분함량표를 이용하여 버섯 재배용 혼합배지의 성분조성을 산출하기 위한 계산식을 제시하였다.

전탕 방법 및 전탕 시간에 따른 십전대보탕 전탕액 비교 연구 (The Compositional Differences of Sipjeondaebo-tang(Siquandabu-tang) Decoctions Extracted by Different Extraction Method and Extraction Time)

  • Objectives: This study was performed to compare the differences between decoctions extracted by different extraction method and extraction time. Methods: Decoctions were prepared with pressurized or non-pressurized extraction for 60, 120 and 180min. The yield of extract, total soluble solid content, hydrogen ion concentration(pH) and the content of reference compound in Sipjeondaebo-tang (Siquandabu-tang) were investigated. Results: While yields and the total soluble solid of decoction were higher in pressurized method proportional to extraction time, pH values were lower in pressurized method and showed decreasing values with increasing extraction time. Albilflorin, ferulic acid, nodakenin, coumarin, cinnamaldehyde and glycyrrhizin were contained in decoctions by pressurized extraction method more than non-pressurized method whereas the content of coumarin was higher in decoctions by non-pressurized extraction method. In addition, coumarin was extracted increasingly with extraction time in decoctions by pressurized method, however, nine compounds except cinnamaldehyde showed the tendency of increasing proportional to extraction time. Conclusions: The results show that extraction methods including pressurized or non-pressurized extraction, and extraction time could affect the physicochemical characteristic or composition of Sipjeondaebo-tang(Siquandabu-tang) decoction.

사군자탕(四君子湯)에서 군약(君藥)의 변화에 따른 네트워크 약리학적 분석 결과 비교 (Comparison of network pharmacology based analysis results according to changes in principal herb in Sagunja-tang)

  • Objectives : The purpose of this study was to confirm whether Codonopsis Radix(CR) could be used in the same way for expected indications or diseases of adaptation instead of Ginseng Radix(GR), which acts as a principal herb in Sagunja-tang. Methods : The Traditional Chinese Medicine Systems pharmacology(TCMSP), a database for the study of systems biology related to Chinese medicine, screened potential active compounds in each quartet. By searching for all the proteins that each compound provides, the target of Sagunja-tang with GR(GRST) and the target of Sagunja-tang with CR(CRST) were compared using the network analysis method, and the top ranked target of each serving was selected. Results : Through TCMSP, a Chinese medicine database, the potential effective ingredients of GRST or CRST screened, and the target proteins related to these substances were found to be the most affected by Glycyrrhizae Radix et Rhizome, an herbal medicine mixed in Sagunja-tang, and the target diseases were the same. And the same were found for the target protein, gene and target diseases of GRST and CRST. Conclusions : The prescription with similar composition is likely to have similar network pharmacology analysis results, and the analysis result may be controlled by the herbal medicines which are assumed to be the main function. Therefore, rich and reproducible basic studies is more important because network pharmacological studies can be dominated by data that has been done a lot of previous studies.

DNA 염기서열과 미각패턴 분석을 이용한 사상자와 벌사상자의 감별 (Comparison between Torilis japonica and Cnidium monnieri Using DNA Sequencing and Taste Pattern Analysis)

  • Objectives : Cnidii Fructus is prescribed as the fruit of Cnidium monnieri (L.) Cusson or Torilis japonica (Houtt.) DC. in Korea pharmacopoeia. Although there are differences in the composition of useful components, two species have been used without distinction. In order to discriminate them, DNA sequencing and taste pattern analysis were used in this study. Methods : Primers ITS 1 and ITS 4 were used to amplify the intergenic transcribed spacer(ITS) region of nuclear ribosomal DNA from seven T. japonica and six C. monnieri samples. Taste pattern of samples were measured by using taste-sensing system SA402B equipped with five foodstuff sensors(CT0, C00, AAE, CA0, and AE1). The five initial taste(sourness, bitterness, astringency, umami, and saltiness) and three aftertaste(aftertaste of bitterness, astringency, and umami) of two species were compared. Results : According to the results of ITS region sequence analysis, two species showed 94 base pairs differences. The similarity of two sequences was 85%. From the taste pattern analysis, sourness, bitterness, aftertaste of bitterness(aftertaste-B), and umami showed a different pattern. Especially, bitterness and aftertaste-B of C. monnieri were significantly higher than T. japonica. In addition, two species were shown to have two markedly different clustering by these two flavors. Conclusion : T. japonica and C. monnieri were effectively discriminated using DNA sequencing and taste pattern analysis. These methods can be used to identify the origin of traditional medicine in order to maintain therapeutic efficacy.

Derivation of Necessary Items for Implementation of Gardens in Urban Agricultural Parks

  • Background and objective: This study was conducted to obtain empirical data for deriving necessary items for the creation and management of gardens in urban agricultural parks while maintaining the publicness of the place by examining the difference in perception among park visitors about the gardens in the public parks. Methods: A survey was conducted on users of urban agricultural parks in 6 locations and 113 copies of the questionnaire were collected. After understanding the demographic characteristics and the current use of the garden, we identified the importance of the necessary items for the public gardens. Results: 108 subjects(95.6%) responded that gardens are needed in urban parks, for psychological and emotional health (26.2%) and for interaction and friendship with family and neighbors(23.2%). For use of garden crops, most were private sales(96 subjects, 64.4%), and both sales preferred to partially donate their crops. Most used communal gardens operated by public institutions(30.1%). It was found that 96.4% of the respondents were satisfied with gardening activities, and 107(94.7%) of them showed their intention to participate in the gardening in the future. The Kaiser Meyer Olkin value was .848 and the significance level was .001, proving the validity of factor analysis. The factors were named composition elements(Factor 1), management items(Factor 2), convenience elements(Factor 3), and operational facilities(Factor 4). In the survey on the creation and management of gardens in urban agricultural parks, there were no statistically significant differences, but all items had correlations. Conclusion: The results have reflected the needs of actual users in establishing the plans to operate urban gardens, thereby having great utility value as the basic data for continuous garden management. Further research can be conducted to derive detailed elements that can guarantee sustainability of urban gardens and suggest high-quality data for management of gardens in urban agricultural parks.

마황성분 체중감량 한약제제가 심혈관계에 미치는 영향: 3차원 맥영상 검사 연구 (Hemodynamic Effects of Herbal Prescription Containing Ephedra on Weight Loss: A 3-Dimensional Radial Pulse Tonometry Device Study)

  • Objectives: This study was aimed to evaluate the hemodynamic feasibility using pulse parameters as a way to establish safe dose guidelines for herbal prescription containing Ephedra herb (Ephedra intermedia Schrenk & C.A.Mey) on weight loss, and to provide a foundation for developing evidence-based guidelines for clinical use. Methods: Forty-two volunteers were recruited to participate in a study examining the changes in pulse wave characteristics following the ingestion of Gambi-hwan, a herbal prescription containing ephedra, over a period of 4 weeks, and pulse wave measurements were taken before and after the administration. Pulse wave parameters were measured in this study using a 3-dimensional radial pulse tonometry device (DMP-Lifeplus). In addition, questionnaire, blood pressure, temperature, and body composition were also measured as secondary measures. Results: Fifteen minutes after administration of Gambi-hwan, the non-adverse event group (non-AE) exhibited a statistically significant increase in several power and pressure-related parameters, including h1, h5, systolic area, pulse area, and pulse width, while the AE group showed a trend of decreasing stroke volume and increasing Radial Augmentation Index (RAI), w, and w/t. After 4 weeks of administration, both groups exhibited significant changes in pulse rate, w/t, RAI, t3/t, stroke volume (SV), and stroke volume Index (SVI). Notably, there are significant differences between AE group and non-AE group in w/t, SV, and SVI. Conclusions: These findings suggest that pulse parameters may be a useful way to establish safe dosing guidelines for weight loss herbal prescription containing ephedra. Further research is needed to confirm these results and to develop evidence-based guidelines for clinical use.
