This study was performed to examine the hepatoprotective effect of isorhamnetin-3-O-galactoside, a flavonoid glycoside isolated from Artemisia capillaris Thunberg (Compositae), against carbon tetrachloride ($CCl_4$)-induced hepatic injury. Mice were treated intraperitoneally with vehicle or isorhamnetin-3-O-galactoside (50, 100, and 200 mg/kg) 30 min before and 2 h after $CCl_4$ (20 ${\mu}l/kg$) injection. Serum aminotransferase activities and hepatic level of malondialdehyde were significantly higher after $CCl_4$ treatment, and these increases were attenuated by isorhamnetin-3-O-galactoside. $CCl_4$ markedly increased serum tumor necrosis factor-${\alpha}$ level, which was reduced by isorhamnetin-3-O-galactoside. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and heme oxygenase-1 (HO-1) protein and their mRNA expression levels were significantly increased after $CCl_4$ injection. The levels of HO-1 protein and mRNA expression levels were augmented by isorhamnetin-3-O-galactoside, while isorhamnetin-3-O-galactoside attenuated the increases in iNOS and COX-2 protein and mRNA expression levels. $CCl_4$ increased the level of phosphorylated c-Jun N-terminal kinase, extracellular signal-regulated kinase and p38, and isorhamnetin-3-O-galactoside reduced these increases. The nuclear translocation of nuclear factor kappa B (NF-${\kappa}B$), activating protein-1, and nuclear factor erythroid 2-related factor 2 (Nrf2) were significantly increased after $CCl_4$ administration. Isorhamnetin-3-O-galactoside attenuated the increases of NF-${\kappa}B$ and c-Jun nuclear translocation, while it augmented the nuclear level of Nrf2. These results suggest that isorhamnetin-3-O-galactoside ameliorates $CCl_4$-induced hepatic damage by enhancing the anti-oxidative defense system and reducing the inflammatory signaling pathways.
The effect of methanol extract from Agaricus blazei Murill on the hepatotoxicity was investigated $CCl_4$ is one of the oldest and most widely used toxins for the induction of hepatic damages and fibrosis in experimental animals. In this study, male Sprague-Dawley(SD) rats were randomly divided into 6 groups of the control(C), $CCl_4(T),\;CCl_4$ and high fat group(TL) with matching sub-groups of Agaricus blazei Murill extract-fed groups of CA, TA and TLA. Methanol extracts of Agaricus blazei Murill were fed 50 mg/kg B.W daily via drinking water. A 1.2 mL of $CCl_4/kg$ body weight was administered by oral intubation twice a week for total of six times. The levels of total-cholesterol, TG, LDL and LDL-phospholipids were elevated by $CCl_4$ treatment as compared to the control(C). However, Agaricus blazei Murill methanol extract feeding in the group of TA and TLA significantly(p<0.05) decreased TG by 53.1 % and 17.9% compared to the internal control of T and TL, respectively. Triglyceride of TL was increased by 3.33 times(p<0.05) compared to the control(C) with $CCl_4$ and high fat administration from 3.78 mg/g to 12.60 mg/g liver. The extract(CA) also reduced kidney weight compared to the control(C). With the administration of high fat and $CCl_4$(TLA), the extract reduced the organ weight of both liver and kidney and further, significantly reduced TG, total cholesterol and GTP activity. Hepatoprotective effects of Agaricus blazei Murill on GOT, GPT, AP and LDH activities were enhanced by the extract feeding. Electronmicrograph showed that $CCl_4$ deteriorated the structure of cytoplasmic matrix with its uneven distribution. However, the extract reconstituted the damaged cytoplasm and stimulated mitochondriogenesis. The above results suggest that Agaricus blazei Murill may have a possible protective effect against chemically induced liver damage and further help to reduce the symptoms of fatty liver.
체내에서 영양소의 흡수에 영향을 미치는 에탄올과 섬유소 및 단백질의 급여수준에 따른 이 들 상호작용을 구명하기 위해 에탄올을 투여한 횐쥐에게 단백질 급여 수준을 7%와 20%로 달리 하고 섬유소를 5%와 10% 첨가한 실험식이를 5주간 급여하므로써 이들 영양소가 에탄을 대사효소와 항산화 물질에 미치는 영향을 관찰하였다. Aldehyde dehydrogenase(ADH)와 microsomal ethanol oxidizing system(MEOS) 활성은 에탄을 투여시 대조군에 비해 유의적으로 증가하였으며, 특히 MEOS 활성은 단백질 정상 급여와 정상섬유소 급여시 그 증가가 현저하였다. 에탄을 투여로 감소된 aldehyde de-hydrogenase(AIDH) 활성은 정상섬유소군의 경우 단백질 적정 급여시 유의적인 감소를 나타내었다. Cytoch-rome P-450(P-450) 활성은 에탄을 투여로 증가되었으며 에탄을 투여군에서 단백질 정상급여와 과량의 섬유소 급여로 유의하게 감소되었다. Xanthine oxidase (XO) 활성은 에탄을 투여로 증가하는 경향이었으며, 저 단백군에 비하여 단백질 적정 급여시 유의적인 증가를 나타내었다. 또한 섬유소 급여수준에 따른 XO 활성은 에탄올 투여군 중 단백질 정상군에서만 유의적인 차이가 관찰되었다. 에탄을 투여는 간조직 중의 글루타티온 함량을 유의적으로 감소시켰으며, 단백질 결핍에 따른 영향이 현저하게 나타났다. 간조직 중의 지질과 산화 함량은 단백질 결핍시 에탄올 투여로 증가하였다. 이상의 결과에서 에탄올을 해독키 위하여 간의 ADH와 MEOS 활성이 증가되었으며 이로 인해 생성된 아세트알데히드는 AIDH 보다 XO를 통해 해독된 것으로 나타났다. 또한 에탄올 대사효소 활성에 요구되는 단백질 공급과 10% 즉, 과량의 섬유소 급여 보다 5% 섬유소 급여시 에탄올성 간손상을 경감시킬 수 있을 것으로 사료된다.
본 연구에서는 숙취해소에 좋은 것으로 알려진 식품 소재와 황칠나무 추출물을 복합하여 in vitro 및 in vivo에서 알코올 유도된 간 손상에 대한 보호효과를 검토하였다. HepG2세포에서 300 mM 알코올과 SBJ 혼합물을 처리하였을 때 농도 의존적으로 LDH 수치가 감소하였으나 황칠나무 추출물을 제외한 복합물인 SBJ-복합물에서는 그 효과가 유의적으로 낮아지는 것을 관찰하였다. In vivo에서 SBJ 혼합물의 보호효과를 확인하고자 흰쥐에 알코올과 SBJ 혼합물을 투여한 결과, 알코올 투여 후 1시간까지 급격하게 혈중 알코올 수치가 EtOH군에서 증가하는 것이 관찰되었으며, SBJ 혼합물 투여군은 유의적인 차이로 감소되었음이 관찰되었으며 또한 농도의존적인 경향을 확인하였다. ADH 및 ALDH 활성의 증가는 SBJ 혼합물의 알코올 분해 및 대사산물의 제거 활성에 기여할 것으로 예상된다. 뿐만 아니라, 알코올에 의하여 증가한 LDH의 농도가 대조군과 유사한 수준으로 유지하는 것을 확인하였으며, GST, SOD, GPx 및 reduced glutathione와 같은 항산화 인자 및 효소의 활성은 대조군보다 EtOH군이 유의적으로 감소했으며, SBJ 혼합물에 의해 개선되는 경향을 관찰하였다. 이를 통해 SBJ 혼합물은 ADH, ALDH 활성 증가 및 항산화 방어계를 향상시킴으로써 산화적 스트레스 감소를 통한 간보호 효과가 있는 것으로 생각되며, 이러한 간보호에 미치는 주요한 추출물은 황칠나무 추출물임을 알 수 있었다. 이러한 결과를 토대로 향후 숙취해소 작용을 갖는 신규 식품소재로 활용될 수 있을 것으로 기대된다.
본 연구는 사염화탄소에 의하여 유발되는 지질과산화에 의한 간독성에 대한 흰점박이꽃무지 추출물의 보호효과를 일차배양한 간세포와 간독성 흰쥐에서 GPT, bile acid, bilirubin 등의 활성 지표를 통해 간독성 회복효과를 알아보고자 하였다. 사염화탄소로 유발시킨 일차배양 간세포에 대한 흰점박이꽃무지와 장수하늘소 추출물의 간독성 회복효과는 각각 41.2%와 8.4%로 흰점박이꽃무지 유충 추출물이 간독성 회복 효과가 높게 나타났으며, 용매 추출별로는 흰점박이꽃무지 메탄올 추출물보다는 물 추출물이 농도의존적으로 간독성 회복효과가 있음을 확인하였다. 사염화탄소에 의해 간독성이 유도된 흰쥐에 흰점박이꽃무지 물추출물 분획별로 처리하였을 때, 분자량 낮을수록 효과적이었으며, 분자량이 1,000이하의 분획물에서 32.1%로 간독성 회복 효과가 우수하였다. 또한 흰쥐에 사염화탄소를 복강투여로 간독성을 유발한 후 흰점박이꽃무지 물분획물을 경구투여할 경우에 혈청 GPT, bile acid와 bilirubin의 활성이 정상군보다는 높았지만 사염화탄소군보다 유의적으로 감소하였고, 간섬유화와 관련된 hydroxyproline의 생성 억제에서는 분자량이 1000이하의 분획물에서 사염화탄소군보다 유의성 있게 생성을 억제하여 간섬유화를 예방하는 것으로 확인하였다. 중심정맥 쪽에 아주 경미한 허혈성 변성 이외에는 다른 병변을 찾을 수 없었고 세포내에 organelle이 유지된 편이고 콜라겐 형성이 매우 많이 줄어들어 병리조직학적으로도 간보호 효과가 있었음을 확인하였다. 이에 간독성 지표 활성을 긍정적으로 결과를 확인한 흰점박이꽃무지 추출물의 간질환 치료보조제나 건강기능성식품 소재 개발 가능성을 확인하였다.
Chemotherapy-induced side effects affect the quality of life and efficacy of treatment of cancer patients. Current approaches for treating the side effects of chemotherapy are poorly effective and may cause numerous harmful side effects. Therefore, developing new and effective drugs derived from natural nontoxic compounds for the treatment of chemotherapy-induced side effects is necessary. Experiments in vivo and in vitro indicate that Panax ginseng (PG) and its ginsenosides are undoubtedly non-toxic and effective options for the treatment of chemotherapy-induced side effects, such as nephrotoxicity, hepatotoxicity, cardiotoxicity, immunotoxicity, and hematopoietic inhibition. The mechanism focus on anti-oxidation, anti-inflammation, and anti-apoptosis, as well as the modulation of signaling pathways, such as nuclear factor erythroid-2 related factor 2 (Nrf2)/heme oxygenase-1 (HO-1), P62/keap1/Nrf2, c-jun Nterminal kinase (JNK)/P53/caspase 3, mitogen-activated protein kinase (MEK)/extracellular signal-regulated kinases (ERK), AMP-activated protein kinase (AMPK)/mammalian target of rapamycin (mTOR), mitogen-activated protein kinase kinase 4 (MKK4)/JNK, and phosphatidylinositol 3-kinase (PI3K)/AKT. Since a systemic review of the effect and mechanism of PG and its ginsenosides on chemotherapy-induced side effects has not yet been published, we provide a comprehensive summarization with this aim and shed light on the future research of PG.
Purpose: Acetaminophen (APAP) is a widely available drug responsible for a large part of drug-induced hepatotoxicity in developed countries. Although acetaminophen overdose cases in Korea are being continuously reported, there are no reports related to the level of this drug in the patient's blood or of laboratory analysis at emergency departments (ED). This study sought to analyze the acetaminophen overdose cases at a toxicological laboratory and to survey APAP analysis services offered at select EDs. Methods: We analyzed the demographic and analytic data at a toxicological laboratory run by the National Emergency Medical Center (NMC) in 2019-2020. We surveyed the APAP laboratory service in the 38 regional emergency medical centers (EMCs) and 68 local EMCs near the toxicological laboratory. Results: We studied 175 acute poisoning cases (112 women) with positive blood APAP results (mean age 47.0±24.1 years). Suicide attempts comprised 40.0% of the cases and 30.3% APAP overdose events. In the univariate analysis, we observed that patients were significantly younger, with fewer underlying medical diseases. There were a higher number of APAP overdose events, more favorable initial mental status, more toxic quantity intake in the above treatment line group (p<0.05), In multivariate analysis, the toxic amount intake was significantly more frequent in the above treatment line group (p<0.01). Hospital APAP analysis services were available in six EMCs (3/38 regional and 3/68 local). The hospital blood APAP level reporting intervals were shorter than outside-hospital laboratory services (p<0.01, regional 7.0±3.0 vs. 40.6±27.5, local 5.3±3.1 vs. 57.9±45.1 hours). The NMC toxicological laboratory reporting interval was shorter than the other outside-hospital laboratories (p<0.01, regional 5.7±0.6 vs. 50.2±22.7 local 7.5±3.0 vs. 70.5±41.5 hours). Conclusion: Over the treatment line group, toxic amount intake was significantly more frequent. Only six of 106 EMCs have their own APAP analysis service in their hospitals.
Objectives : This study was undertaken to verify the effects of Massa Medicata Fermentata (MMF) on nonalcoholic fatty liver disease (NAFLD) using high fat diet-fed male mice. Methods : Fifty four male C57BL/6N mice (age matched) were used for all experiments. Nine standard chow diet-fed mice were used as normal group and forty five high fat diet-fed obese mice were randomly divided into 5 groups: control, atorvastatin-10mg/kg, MMF(1)-62.5mg/kg, MMF(2)-125mg/kg and MMF(3)-250mg/kg. After all groups were treated with several kinds of diets for 8 weeks, we measured body weight gain, adipose tissue weights, plasma lipid and glucose metabolism, visceral organ weights, histological analysis for liver on the mice. Results : MMF-treated mice had lower body weight gain compared with controls. Among MMF-treated mice, the effect was magnified in MMF(2). MMF(3)-treated mice had lower blood plasma total cholesterol (TC) and glucose level compared with controls. MMF decreased hepatic lipid accumulation, liver fibrosis and liver inflammation of mice compared with controls. The effects was maximized in MMF(2) and atorvastatin. Blood plasma aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyltransferase (${\gamma}$-GT) concentrations tends to be decreased by MMF compared with controls. Blood plasma AST, ALT, ${\gamma}$-GT concentrations and organ weights were not changed by MMF, indicating that all three kinds of MMF do not show any hepatotoxicity. Conclusions : These results suggest that MMF improves NAFLD by reducing body weight gain, hepatic lipid accumulation, liver fibrosis, liver inflammation.
Objectives : Non-alcoholic fatty liver disease (NAFLD) is characterized by the accumulation of hepatic triglycerides (TG) that leads to inflammation and fibrosis. Crataegi Fructus ethanol extract (CE) is a korean traditional herb that used for digestive diseases. It has been investigated that CE has the effect that prevent hepatotoxicity caused by CCl4 or GaIN and regulate the inflammatory in several organs. However, a hypolipidemic effect of CF has not been reported. Methods : The purpose of this study is that examine the lipid accumulation inhibitory effect of CE on NAFLD. We checked the body and liver weight change of MCD-diet induced mice with/without administration of CE. The blood lipid levels of C57BL/6J mice were checked by biochemistry. Also we observed the liver histology of MCD-diet induced mice and investigate the molecular mechanisms in MCD-diet-induced NAFLD in C57BL/6J mice. Results : CE improved MCD-diet-induced lipid accumulation and TG and TC levels. Also, CE decreased hepatic lipogenesis such as SREBP-1, $C/EBP{\alpha}$, $PPAR{\gamma}$, ACC and FAS. Besides, we also found out that CE increased AMPK phosphorylation. These results indicated that CE has the same ability to activate AMPK and then reduce SREBP-1, and FAS expression, finally leading to inhibit hepatic lipogenesis and hepatic antioxidative ability. Conclusions : In this report, we found CE exerted a regulatory effect on lipid accumulation by decreasing lipogenesis in MCD-diet induced NAFLD model. Therefore, CE extract may be active in the prevention of fatty liver.
본 연구는 투어마린 소재와 전기적인 방전을 통해 물의 성질을 변화시킨 투어마린이온활성수를 이용하여 간보호 효과, 비만 예방 효과, 숙취제거 효과를 확인하기 위해 시행되었다. 고지방식이로 비만을 유도한 ICR 마우스에서 투어마린이온활성수를 급이한 실험군은 정수물을 급이한 대조군과 비교하여 사료섭취량이 비슷하고 음수량이 더 적었지만 체중증가는 대조군에 비하여 억제되어 13주에는 유의한 차이를 보여줌으로써 비만 예방 효과를 나타내었다(P<0.05). 사염화탄소로 간독성을 유발시킨 생쥐를 이용하여 간보호 효과를 관찰한 실험에서, 실험군의 aspartate aminotransferase (AST)와 alanine aminotransferase (ALT)는 대조군과 비교하여 평균적인 차이를 나타내었으며, 간의 조직학적 관찰에서 괴사를 비롯한 병리학적 병변이 감소한 것으로 나타났다. S-D rats을 이용한 숙취 실험에서는 실험군이 대조군에 비해 혈중알콜농도가 유의하게 낮았으며(P<0.01), 알콜 대사와 관련된 항산화 효소인 glutathione-S transferase (GST)는 활성이 증가한 것으로 관찰되었고(P<0.05), 간기능 지표인 AST와 ALT는 상대적인 감소현상을 보였다. 이와 같은 결과는 투어마린이온활성수의 음용이 동물의 항산화 체계에 영향을 미침으로써 간조직을 보호하고 알콜 분해를 향상시킨 것으로 판단되며, 그 결과 숙취해소에 어느 정도 효과가 있을 것으로 판단된다. 투어마린이온활성수의 효능과 관련된 기전은 아직 과학적 검증에 어려움이 있으나, 물에 전달된 투어마린의 원적외선에너지나 전기적 영향으로 인하여 물분자의 결합력과 분자 구조 또는 클러스터의 크기에 변화를 일으켰기 때문으로 판단된다. 사용된 투어마린이온활성수는 음용안전성이 확보된 물로서 효과적인 기능수로 개발될 가능성을 가지며, 유효성과 관련된 기전은 더 연구될 필요성이 있다.

서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
