This study aimed to assess the performance of laying hens across twelve crossbreed strains (i.e., CFCK, CFYC, CFYD, CKCF, CKYC, CKYD, YCYD, YCCF, YCCK, YDCF, YDCK, and YDYC) of Korean native chicken (KNC) and compare them with Hy-Line Brown layers. A total of 287 18-week-old laying hens were placed in battery cages by strains (2-5 birds per pen). Results indicated that the YCYD and YDYC strains exhibited numerically heavier body weights than Hy-Line Brown at week 18-64. CKYC and YDYC strains demonstrated more than 94% viability by week 64. The CFYC strain had an age of first egg laying of 127 days, and the YDCF strain reached an age of 50% egg production at 140 days, both earlier than their parent stock. The YDCF strain showed over 70% egg production for up to 60 weeks. Regarding egg quality, the CKCF and YCCF strains had numerically higher egg weights among the KNC groups at week 24-64, with the YDYC strain showing a darker (P<0.05) eggshell color compared to CKCF at week 40. Moreover, KNC crossbreeds showed a higher (P<0.05) egg yolk ratio than Hy-Line Brown. In conclusion, the YDCF and YCCF crossbreeds exhibited the most desirable new synthetic Korean native commercial layer based on egg production and quality parameters. Therefore, these strains could be a viable substitute for Hy-Line Brown layers.
This study was conducted twice to investigate egg purchase behavior and perception on animal welfare of Korean consumers. This study included women, who were the main decision makers and caretakers in the household, and men with one-person household. This survey was conducted with by the Computer Assisted Web Interview and Gang Survey methods. On the key considerations factor, the highest response rate was considered to be 'price', and the response rate of considering 'packing date' increased in the second survey. At a reasonable price based on 10 eggs, the response rate was the highest at 53.8% and 42.9% in both the first and second surveys and the appropriate price averages were 2,482 won and 2,132 won, respectively. The highest rate of purchase of egg consumers from 'Large Mart' followed by 'Medium sized supermarket' and 'Chain supermarket'. As for the awareness about animal welfare, the recognition ratio (73.5%) was higher in the result of the second survey than the first. The cognitive period of animal welfare was 59.0% before the insecticide egg crisis and 41.0% thereafter. Regarding whether or not they have ever seen an animal welfare certification mark and an animal welfare animal farm certification mark, 59.6% of respondents said that they saw it for the first time and 37.6% answered that they knew the animal welfare certification mark. On the animal welfare system, the 'free-range' response rate was the highest at 85.8%. The 'free-range' fit response decreased by 34.2%p, while the 'barn' and 'European type' fit response increased by 13.2%p and 24.1%p, respectively. The number of 'I have never seen' and 'I have ever eaten' responses to the recognition and eating experience of animal welfare certified eggs decreased while the number of those who answered 'Have ever seen' and 'Have eaten' increased. The answer of purchasing animal welfare certified eggs at department stores, organic farming cooperatives, and internet shopping malls was higher than that of buying conventional eggs. Of the total respondents, 92.0% were willing to purchase an animal welfare egg before the price was offered, but after offering the prices of animal welfare eggs, the intention to purchase was 62.7%, which was about 30%p lower than before. The reason for purchasing an animal welfare certified egg was the highest score of 71.0% for 'I think it is likely to be high in food safety', and 38.1% for 'I think the price is high' for lack of intention to purchase. In the sensory evaluation of animal welfare eggs, egg color and skin texture of conventional eggs were significantly higher than those of certified welfare eggs (P<0.05), and boiled eggs showed that egg whites of animal welfare certified eggs were more (P<0.05). As a result, the results of this study will contribute to the activation of the animal welfare certification system for laying hens by providing basic data on consumer awareness to animal welfare certified farmers.
This study was conducted to identify the correlation of bacterial phytase ($Transphos^{(R)}$) to the calcium level in feed. Of all 21-week-old 720 HyLine brown laying hens, 2 birds of similar weight were placed on each individual cage. The experiment was conducted by $3{\times}2{\times}3$ factorial design with including 3 different levels of phytase (0, 300, and 1,000 DPU/kg), 2 different levels of calcium (3.5% and 4.0%), and 3 different levels of no NPP addition 0% (0.095 NPP), 0.5% (0.185% NPP), and 1.0% (0.275% NPP). The feeding trial maintained the ME level of 2,800 kcal/kg and 16% for crude protein. The diet was fed ad libitum and 17 hours of lighting was provided throughout the experimental period. Egg production seemed to increase, in the 300 DPU of bacterial phytase added group and the cracked egg tended to reduce in Transphos added group. The egg productivity between treatment groups did not show significant difference by dietary calcium level, whereas non NPP added group (0.095% NPP) was found to be low compared to NPP added groups (P<0.05). The highest mean egg weight and the highest daily egg mass were detected in 300 DPU phytase added group. Although the mean egg weight was significantly higher in treatment groups fed with 3.5% calcium containing feeds (P<0.05), daily egg mass was no among treatment groups. The mean egg weight and daily egg mass were the lowest in non NPP added group (0.095% NPP) compared to other treatment groups (P<0.05). The feed intake showed similar pattern regardless of the bacterial phytase and calcium levels in the diet. However, the treatment groups fed diets containing NPP level of 0.275% and 0.165% showed significantly higher feed intake than the group fed with 0.095% NPP (P<0.05). Although the feed conversion was not affected by calcium and NPP levels in the diet, the most improved result was obtained from 300 DPU phytase added group (P<0.05). The eggshell breaking strength and thickness increased as dietary calcium level increase the level of calcium increases in diet. The treatment groups fed diet containing 0.275% and 0.165% NPP revealed to show improvement in eggshell breaking strength and yolk color index compared to the NPP non added (0.095% NPP) treatment group. The result of the present study suggests that the appropriate level of microbial phytase is 300 DPU and at this level, tricalciumphosphate supplementation in feed can be reduced to 40% of NRC recommendation. Higher calcium level in feed fail to show synergistic effect by adding microbial phytase.
This study was conducted to investigate the effects of housing systems on the productivity and physiological response as stress indicators in White Leghorn chickens. The chickens subjected to the conventional cages had a significantly lower viability, hen-housed egg production, egg weight and body weight compared with those to the floor pens. However, the hens housed in the conventional cages had a shorter day of the first egg and a greater egg quality compared with those housed in the floor pens. In addition, this study was also investigated to identify biological markers for assessing the physiological response of chickens under stress conditions. As biological markers, the amount of telomeric DNA was analyzed by quantitative fluorescent in situ hybridization on the nuclei of cells. The DNA damage rate of lymphocytes was also quantified by the comet assay. The amount of telomeric DNA of the lymphocytes, kidney and spleen was significantly higher in the chickens under floor pens than those under conventional cages. The DNA damage also increased in chickens raised under conventional cages, as compared to the chickens under floor pens. As results, we conclude that the chickens housed in conventional cages have a greater stressful status than those housed in floor pens.
Follicle-stimulating hormone (FSH) is a pituitary glycoprotein hormone that is encoded by separate alpha- and betasubunit genes. It plays a key role in stimulating and regulating ovarian follicular development and egg production in chicken. FSH signal transduction is mediated by the FSH receptor (FSHR) that exclusively interacts with the beta-subunit of FSH, but characterization of prokaryotic expression of the FSHb gene and its effect on the expression of the FSHR gene in local chickens have received very little attention. In the current study, the cDNA fragment of the FSHb gene from Dagu chicken was amplified using reverse transcription polymerase chain reaction (RT-PCR), and inserted into the pET-28a (+) vector to construct the pET-28a-FSHb plasmid. After expression of the plasmid in E. coli BL21 (DE3) under inducing conditions, the recombination protein, $FSH{\beta}$ subunit, was purified and injected into the experimental hens and the effect on the mRNA expression levels of the FSHR gene was investigated. Sequence comparison showed that the coding region of the FSHb gene in the local chicken shared 99%-100% homology to published nucleotides in chickens; only one synonymous nucleotide substitution was detected in the region. The encoded amino acids were completely identical with the reported sequence, which confirmed that the sequences of the chicken FSHb gene and the peptides of the $FSH{\beta}$ subunit are highly conserved. This may be due to the critical role of the normal function of the FSHb gene in hormonal specificity and regulation of reproduction. The results of gene expression revealed that a recombinant protein with a molecular weight of about 19 kDa was efficiently expressed and it was identified by Western blotting analysis. After administration of the purified $FSH{\beta}$ protein, significantly higher expression levels were demonstrated in uterus, ovary and oviduct samples (p<0.05). These observations suggested that the expressed $FSH{\beta}$ protein possesses biological activity, and has a potential role in regulation of reproductive physiology in chickens.
An experiment was conducted to elucidate the effect of graded levels of vitamin $D_3$ in White Leghorn (WL) layer breeders on egg production, shell quality, hatchability of eggs and juvenile performance of offspring during their late laying period (72-88 wk). White Leghorn breeder females were randomly divided into 5 groups of 50 each and were housed in individual California cages in an open-side housing system. Considering birds in five cages as a replicate, 10 such replicates were randomly allotted to each treatment. A basal diet was formulated containing all the nutrients as recommended for WL layers except vitamin $D_3$, which served as control. Another, four diets were formulated by supplementing graded levels of feed grade crystalline cholecalciferol to the basal diet that contained 300, 600, 1,200 and 2,400 ICU of vitamin $D_3$ per kg. Each diet was offered ad libitum to one of the above five treatment groups. The egg production, egg weight, daily feed consumption and the feed intake per dozen eggs or kg egg mass of the birds fed diet without any supplemental vitamin $D_3$ was comparable with those of supplemental groups. Similarly, the level of vitamin in the diet did not have any effect on any of the above parameters. However, the specific gravity of eggs laid by the birds fed the diet without supplemental vitamin $D_3$ was comparable with either 600 or 2,400 ICU supplemental groups but significantly higher when compared to the 300 and 1,200 ICU groups. The egg -shell breaking strength was significantly lowered in the 600 ICU supplemental groups as compared to the strength of other dietary groups. The Haugh unit, egg shell weight, shell thickness, tibia breaking strength, bone ash and calcium content were not influenced by vitamin $D_3$ concentration in the diet. Serum Ca concentration was influenced by vitamin $D_3$ level in the diet. The serum Ca concentration of birds fed either control or the vitamin supplemented diet up to 1200 ICU/kg diet was comparable. However, increasing the concentration of vitamin $D_3$ to 2,400 ICU/kg diet significantly enhanced the concentration of Ca in the serum, which was significantly higher compared to other dietary groups. The serum concentration of P and protein, however, was not influenced by level of vitamin $D_3$ in the diet. Neither fertility nor hatchability was influenced by vitamin $D_3$ concentration in the diet. Feeding a vitamin $D_3$ deficient diet or supplementation of vitamin to hens did not have any influence on their progeny chicks. It can be concluded that dietary supplementation of vitamin $D_3$ may not be essential for optimum production, shell quality, hatchability, and juvenile performance of WL breeders during 72 to 88 weeks of age.
H. pylori is known to be a key pathogen of chronic gastric and duodenal ulcers. Bacterial adhesion to hosts is an essential step for bacterial infection and the inhibition of this adhesion provides a possible method for the treatment of the infection. The inhibitory effect of antibody lgY, produced from immunized hens with H. pylori antigen, was studied in vitro. The inhibition of H. pylori adhesion to AGS was as high as 90% using 0.5mg/ml of lgY, and almost 80% of the detachmentwas also achieved. The inhibitory effect of adhesion-inhibition candidates was investigated. Additives in combination with lgY increased the adhesion-inhibiting effect by about 30-50%. However, the adhesion molecules of H. pylori were varied and complex, therefore the further studies are necessary to develop an adhesion inhibitor and effective enough to be employed for the treatment of H.pylori, in vivo.
The records on 787 white Leghorn hens from two different strains, A and K were wed in this study to find out the pattern of increase in egg weight with age and the optimum age of early selection for maximizing the genetic gain. The pattern of increase in egg weight with age in two lines which was expressed on graph showed that the amount of increase was larger in the early part of the laying year and it decreased gradually. Regression equations suggested that egg weight increased linearly with age at the rate of 0.46 gram per week for line A and 0.83 gram for line K. And in the regression analyses of each period mean egg weight on annual mean egg weight, the coefficients of EW 30 that is mean egg weight from 29 weeks of age to 32 weeks of age were 0.86 for line A, 0.88 for line K, and those were the highest figures among the all regression coefficients of each mean egg weight on annual mean egg weight. In estimating the relative selection efficiency of each period mean egg weight on annual mean egg weight in EW 46, selection on EW 34 for line A and on EW 30 for line K was the most efficient for improving egg weight. These results suggested that one periods from 29 weeks of age to 36 weeks of age could be used for the best predictable time for improving annual mean egg weight.
This experiment was conducted to study the optimum energy feeding regimens fur broiler breeders peaked in winter season with 400 caged hens of Ross strain. Four energy supply regimens which were different in daily energy allotment during laying period were employed for 40 weeks from 24 to 64 weeks of age. All experimental diets were formulated to contain 2,750 kcal ME/kg with adjustments made in total feed allotment to provide the desired energy levels. Total consumption of the feed would provide 20 g of protein, 4 g of calcium and 0.35 g of available phosphorus. There were no difference in hen-day egg production and average egg weight among the regimens of energy supply. Feed, ME and feed cost required per egg or per kg egg were significantly increased as the level of energy allotment increased(p<0.05). It was concluded that the energy supply regimen, which supplied 280 kcal ME per day at the age of 24 weeks and then increased the energy supply up to 400 kcal ME per day at the peak period of 30∼34 weeks of age, was superior in feed, ME and feed cost required per egg or per kg egg without any adverse effect on eg production and egg weight.
This study was carried out to investigate the effect of feeding fermented earthworm cast additives (FEC) on egg production, egg yolk fatty acids, blood lipid, cecal microorganism and fecal odor. The completely randomized experiment was applied, and the treatments were designed as 2 feeding regimens (control vs. FEC feeding), and FEC contained 3.5% top dressing to common diet. Egg production and egg weight of FEC group were significantly higher than that of control (p<0.05). Ratios of n-6/n-3 fatty acids in the egg yolk of FEC group were significantly higher than those in control group (p<0.05). Concentrations of HDL-C in FEC group was significantly higher than control group (p<0.05), but not triglyceride and total cholesterol. The caecal Lactobacillus of FEC group was significantly higher in FEC group than control group (p<0.05). $NH_3$ concentrations from poultry house were significantly lower in FEC group than control (p<0.05). In conclusion, these study results showed that the addition of FEC at 3.5% to the diet has reduced odor of poultry house and improved the n-6/n-3 fatty acid of egg yolk and egg production in laying hens.
