면양의 혈절과 혈림프절의 구조 (Structures of hemal nodes and hemolymph nodes in sheep)

  • This study was performed to investigate the gross anatomical features and microscopical structures of the hemal nodes and the hemolymph nodes in the sheep. The hemal nodes and hemolymph nodes were observed mainly in periphery of the thoracic and abdominal aortae of the animals. The size of hemal nodes was generally smaller than that of the hemolymph nodes, and the shape of the both organs was spherical or ovoid. The color of the hemal nodes was red, while that of the hemolymph nodes was grary with red bands. The hemal nodes were surrounded by a relatively thick connective tissue capsule and there were extensive subcapsular sinuses distended by a great number of erythrocytes. Although a few number of lymphatic nodules and small areas of diffuse lymphatic tissues were observed in the parenchyma, no typical cortex and medulla were defined in the hemal node. Small numbers of blood vessels were found at the connective tissue capsule but lymph vessel was not observed microscopically in this organ. The hemolymph nodes were covered by a relatively thick connective tissue capsule and there was a hilus in each node. The parenchyma was divided into cortex and medulla. The cortex was composed of a few numbers of lymphatic nodules and some diffuse lymphatic tissues. The medulla comprised medullary sinuses and cords. Afferent and efferent lymph vessels were observed at the periphery of the capsule adn the hilus, respectively. The subcapsular and medullary sinuses were not extensive but filled with small numbers of erythrocytes. The stroma of hemal node and hemolymph node was composed of reticular cells and fibers, and the capsule and trabecula consisted of collagenous fibers with smooth muscle fibers.

곤충세포 배지 개발을 위한 체액산화지연 돌연변이 누에계통 선발 (Selection of Mutant Silkworm with Oxidation-deficient Haemolymph for Insect Cell Culture)

  • 누에 체액은 공기 중의 산소와 접하게 되면 도파퀴논(dopaquinone)이라는 세포 성장을 저해하는 물질을 생성하게 된다. 체액의 산화를 방지하기 위해서는 일반적으로 열처리에 의해서 효소를 불활성화 시키는 방법이 이용되고 있으나 대량 채혈 시에는 열처리 이전에 이미 산화가 진행되는 문제점이 있다. 이러한 문제점을 해결하기 위하여 누에 중 체액 산화가 더디게 진행되는 누에 계통을 선발하게 되었다. 누에체액을 채취하여 경과시간별로 산화정도(변색)를 육안으로 확인하여 TBO, $Y_4$, 백안$E^b$과잉지 3품종을 선발하고 spectrophotometer를 이용하여 산화반응 속도를 측정하였다. 백옥잠 체액과 각각의 산화지연 누에 체액의 산화속도를 400 nm에서 측정한 결과 백옥잠은 120분인데 비하여 체액산화 지연계통인 $Y_4$는 330분, TBO는 360분 그리고 백안$E^b$과잉지는 450분에 산화가 완료되었다. 산화지연 누에계통의 체액의 곤충세포 증식에 미치는 영향을 조사한 결과 대규모 채혈한 체액산화지연 계통의 체액은 야성형 계통(백옥잠)에서 소규모 채취한 체액에는 미치지 못하였으나 대규모 채취한 체액보다는 우수하였다.

Characterization and Immunological Analysis of Insecticyanin from the Hemolymph of Agrius convolvuli

  • A blue biliprotein, insecticyanin (INS), has been purified from the last instar larval hemolymph of Agrius convolvuli by ultracentrifugation, Sephadex G-100 gel permeation chromatography, and preparative electrophoresis. The molecular mass of INS was estimated to be 26 kDa and the N-terminal sequence of INS revealed high similarity to that of Manduca sexta. Results of Western blotting and autoradiography indicated that INS is synthesized by the epidermis and released into the hemolymph. In contrast to the INS reported in other insects, Agrius convolvuli INS contained a small amount of lipid, predominately consisting of triacylglycerol. Subcellular localization of INS was determined using protein-A gold particles linked to secondary antibodies (anti-rabbit Ig). INS was heavily accumulated in the cytoplasmic inclusion body (CIB). CIBs showed a variety of shapes from rod to globule and generally surrounded the nucleus. They were mostly located near the basement membrane and especially abundant in the intersegmental membrane.

Effect of Systemic Fungicide on Total Hemocyte Count and Hemolymph Biochemical Changes in Silkworm, Bombyx mori L., infected with Beau-veria bassiana

  • Silkworm diseases are major constraint in silk cocoon production. Among silkworm diseases, white muscar-dine is highly contagious and most common in winter and rainy seasons. It is suggested that hemocytes involve in defense against invasion of Beauveria bassi-ana and systemic fungicide/chemicals prevent the proliferation of fungi in the hemolymph or preventing the growth of the fungi in the body cavity through enhancing the hemocyte mediated defense response. In the present study the influence of systemic fungicide on hematological changes in silkworms infected with Beauveria bassiana was reported. It is observed that the total hemocyte counts increased in the hemolymph up to 5 days post inoculation in systemic fungicide treated batches while in the inoculated control the increase was up to 3 days indicating the positive hemocyte mediated response in silkworm treated with systemic fungicide. After 2 days in the inoculated control as the multiplication and growth of mycelia increased, defense capacity of the silkworm was decreased. The biochemical changes were also observed in the hemolymph of silkworm infected with B. bassiana. In silkworm infected with Beauveria bassi-ana, the total protein content increased whereas total carbohydrate and total lipids decreased as the infection progresses. In the case of systemic fungicide treated batches the increase in total protein content was comparatively higher and decrease in total carbohydrate and lipids were comparatively lower than the inoculated control.

Effects of Silkworm Hemolymph and Cartilage-specific Extracellular Matrices on Chondrocytes and Periosteum-derived Progenitor Cells

  • In order to enhance the repair of defects in articular cartilage via cell therapy with autologous chondrocytes, as well as with periosteum-derived progenitor cells (PDPCs), silkworm hemolymph (SH) and a variety of cartilage-specific extracellular matrices (ECMs) including type II collagen, proline, chondroitin 4-sulfate, and chondroitin 6-sulfate were assessed with regard to their efficacy as media supplements. SH, a known anti-apoptotic agent, was found to enhance cell growth, as was shown by the results of a 3-(4,5-dimethylthiazol-2yl)-2,5-diphenyltetrazolium bromide (MTT) assay. According to the results of reverse transcriptase polymerase chain reaction (RT-PCR) analyses, the cartilage-specific ECMs were found to stimulate the expression of hyaline cartilage-specific genes, most notably type II collagen and Sox9, in monolayer cultures of PDPCs.

Anti-Apoptosis Engineering Using a Gene of Bombyx mori

  • We have previously shown that the addition of silkworm hemolymph to a culture medium increases the longevity of insect and mammalian cells by inhibiting apoptosis. This indicates that the component which inhibits apoptosis is contained in the silkworm hemolymph, The apoptosis-inhibiting component was isolated from silkwonn hemolymph and characterized in our previous study. A database search using the N-terminal amino acid sequence of this component as a template resulted in a 95% homology with a low molecular weight lipoprotein, the so called ’30K protein' of unknown function. In this study, the 30K protein gene was expressed in mammalian and insect cells to confirm the apoptosis-inhibiting effect. The overexpression of 30K protein in mammalian cell inhibited the staurosporin-induced apoptosis by the prevention of the activation of caspase 3. Using an Autographa californicanuclear polyhedrosis virus (AcNPV) system, the 30K protein was overexpressed also in insect cells. The expression of the 30K protein increased the longevity of baculovirus-infected insect cells by inhibiting apoptosis. These results suggest that the 30K protein is a novel anti-apoptotic protein.

매미나방(Lymantria dispar L.)의 Lipophorin의 정제 및 특성 (Isolation and Characterization of Lipophorin from Lymantria dispar L.)

  • Lipophorin of Lymuntrn'adispor L. has been purified by KBr density gradient ultracentrifugation. The properties and synthetic site of lipophorin and quantitative change of lipophorin during development have been determined using electrophoresis and immunological analysis. Lipophorin is composed of ho subunits. apo-Lpl (230.000), ago-Lpll (49,000). ann contains carbohydrates and lipids. Anti-lipophorin showed positive reactions with fat body extract and ovary extract but not with gut extract. The concentration of lipophorin in hemolymph showed gradual decrease during larval and pupal stages. Also. fat body released lipophorin into medium. Immunological test showed some partial identity between lipophorin of Lymantpia dispar and hemolymph proteins (probably lipophorin) of Hyphontria cuneo and Galleria metlonefla.

고지방식이 비만 유도 mouse에서 누에체액의 항비만 및 LDL-cholesterol 저하효과 (Anti-obesity and LDL-cholesterol lowering effects of silkworm hemolymph in C57BL/6N mice fed high fat diet)

  • 본 연구에서는 고지방식이로 유도한 비만 C57BL/6N 마우스 모델에서 누에체액 투여가 마우스의 체중 감소 및 혈중 지질 개선 효과를 보이는지 평가하였다. 실험동물은 정상군 (N), 고지방대조군 (HFC), 누에체액투여군 (HFS-1, HFS-5, HFS-10)으로 분리하여 12주 동안 열량비 10% 지방식이나 45% 지방식을 섭취시키고 고지방식이 급여군들의 일부에 누에체액을 체중 대비 1%, 5%, 10% 경구투여 하였다. 그 결과 체중과 부고환지방 및 총지방의 무게가 유의하게 감소하였으며 혈장 LDL-cholesterol 농도가 고지방대조군 (HFC)군에 비하여 HFS-1, HFS-5, HFS-10군에서 유의하게 감소하였다. 혈중 ALT와 AST의 활성은 고지방대조군에 비하여 HFS-5, HFS-10 군에서 유의하게 저하되었고 누에체액을 투여한 모든 군에서 HFC군과 비교하여 혈장 leptin 수준이 유의하게 감소하였다. 비록 누에체액의 유효성분을 동정하고 그 특성을 규명하기 위해서는 더 많은 연구를 필요로 하지만 본 연구 결과는 누에체액이 비만을 예방할 수 있는 잠재적인 천연소재로서 가능성을 보여준다.

수온 증가에 따른 말전복, Haliotis gigantea과 둥근전복, Haliotis discus discus (Reeve, 1846) hemolymph의 생리학적 변화 (Influence of elevated temperatures on the physiological response of hemolymph from two species of abalone, Haliotis gigantea and Haliotis discus discus (Reeve, 1846))

  • 본 연구에서는 수온 증가에 따른 말전복 (Haliotis gigantea) 과 둥근전복 (H. discus discus) 헤모림프의 생리 및 면역학적 변화를 관찰하기 위하여 위 두 전복을 20, 22, 24, 26 및 $28^{\circ}C$ 수온에 각각 4 일간 노출시켰다. 노출 결과, 헤모림프의 totalo-protein (TP), glucose, calcium (Ca) 은 둥근전복 (H. discus discus)이 말전복 (H. gigantea) 보다 높은 값을 보였으나, magnesium (Mg), alkaline phosphatase (ALP) 두 전복에서 유사한 값을 보였다. 그리고 면역인자인 PO에서는 둥근전복 (H. discus discus)이 더 높게 나타났다. 수온 증가에 따른 헤모림프의 TP, glucose, Mg, AST, ALT 및 ALP는 유의적인 변화가 나타나지 않았다. 하지만 헤모림프의 Ca 농도는 둥근전복 (H. discus discus)에서 $24^{\circ}C$ 이상, 말전복 (H. gigantea)에서는 $26^{\circ}C$ 이상의 수온에서 유의적 증가를 보였다. 그리고 면역인자인 phenoloxidase (PO) 는 둥근전복 (H. discus discus)은 $22^{\circ}C$ 이상 말전복 (H. gigantea) 에서는 $26^{\circ}C$ 이상의 수온에서 유의적 감소를 나타내었다. 결론적으로 말전복 (H. gigantea)은 $24^{\circ}C$ 이상에서 혈액학적 변화를 보였고, $26^{\circ}C$ 이상에서 면역학적 변화를 보였다. 둥근전복 (H. discus discus)은 $26^{\circ}C$ 이상에서 혈액학적 변화를 보였고, $22^{\circ}C$ 이상에서 면역학적 변화를 나타내었다.

${\cdot}$단명 누에 품종의 체액단백질의 전기영동상 (Electrophoretic Patterns of Hemolymph Proteins of Varieties with Long and Short Life Span in the Silkworm Bombyx mori L.)

  • 장명품종(SA10, NTZN, 잠109, J037 및 설악)과 단명품종[한강, 괴축, PR, 천문 및 Daizo(sdi)]의 幼蟲, 용, 成蟲의 발육단계별 및 암수별 체액단백질의 변화과정을 전기영동상의 패턴에 의해 비교 분석하였다. Native 전기영동상 분석 결과로 볼 때, 용 1일째의 體液 蛋白質은 MHP-a, b, c 패턴에 따라 長命特異群(MHP-Ⅰgroup), 短命特異群(MHP-Ⅱgroup) 및 張${\cdot}$短命混合群(MHP-Ⅲgroup)등 3개의 단백질 유형으로 분류할 수 있었다. MHP-Ⅰgroup에는 SA10, 잠109, J037이, MHP-Ⅱgroup에는 한강, 천문, Daizo(sdi)가, MHP-Ⅲgroup에는 NTZN, 설악, 괴축 및 PR의 품종들이 각각 분포하고 있었으며, 이들 group내 소속 품종의 MHP패턴은 거의 유사하였다. 그리고 이들 MHP-a, b, c는 용의 발육에 따라 영동상의 변화를 보였다. 최단명품종인 Daizo(sdi)의 경우, 성충 1일 암컷과 수컷 모두 MHP-c가 뚜렷하게 존재하는 반면 최장명품종인 J037에서는 이 단백질이 관찰되지 않는 등 성충 체액 단백질 패턴에 있어서 J037과 Daizo(sdi)의 품종간에 뚜렷하게 차이가 인정되었다. 그러나 이들 단백질 밴드상의 수명의 장${\cdot}$단과 직접적인 관계가 있는 것인지 또는 수명과 관련된 특정 단백질 밴드가 존재하는지 등에 대해서는 앞으로 추구할 연구과제로 남는다. 한편, 최장명 J037품종과 최단명 Daizo(sdi) 품종의 체액단백질에 대한 SDS-PAGE상의 영동패턴을 비교 분석 결과, 幼蟲期, 용期 및 成蟲期에 있어서 장${\cdot}$단명 품종간에 단백질 패턴의 차이보다는 양적인 변화가 뚜렷하였다.
