Iron-fortified recombinant Saccharomyces cerevisiae producing Sus scrofa ferritin heavy-chain recovers iron deficiency in mice

  • In this study, we produced iron-fortified yeast (Saccharomyces cerevisiae) producing Sus scrofa ferritin heavy-chain to provide iron supplementation in anemic piglets. We determined whether iron-ferritin accumulated in recombinant yeasts could improve iron deficiency in mice. C57BL/6 male mice exposed to Fe-deficient diet for 2 weeks were given a single dose of ferrous ammonium sulfate (FAS), ferritin-producing recombinant yeast (APO), or APO reacted with iron ($Fe^{2+}$) (FER). The bioavailability of recombinant yeasts was examined by measuring body weight gain, hemoglobin concentration and hematocrit value 1 week later. In addition, ferritin protein levels were evaluated by western blot analysis and iron stores in tissues were measured by inductively coupled plasma spectrometer. We found that anemic mice treated with FER exhibited increased levels of ferritin heavy-chain in spleen and liver. Consistently, this treatment restored the iron concentration in these tissues. In addition, this treatment significantly increased hemoglobin value and the hematocrit ratio. Furthermore, FER treatment significantly enhanced body weight gain. These results suggest that the iron-fortified recombinant yeast strain is bioavailable.

Effects of Iron Supplementation on Iron Status and Immunity Status of Elite Female Soccer Players (엘리트 여자 축구선수의 철분보충이 체내 철분상태와 면역 및 항산화에 미치는 영향)

  • This study was performed to evaluate the effect of iron supplement for 4 weeks on iron status, immunity, and antioxidant status of national female soccer players (n = 25). This study was performed at summer hard training period right before competition. A single blind design was used to divide the subjects into iron-supplement (IS) or placebo group (P). Iron-supplement group was supplemented with iron (40 mg/d) for 4 weeks. The mean age of the subjects was 23.3 $\pm$ 2.5 years old. Mean height and body weight of the subjects were 164.4 $\pm$ 5.7 em and 57.4 $\pm$ 4.6 kg, respectively. The mean carrier as soccer player was 11.0 $\pm$ 2.6 years and mean training time was 7.0 $\pm$ 1.3 hr/day. The mean hemoglobin, hematocrit, total iron binding capacity and ferritin concentrations before iron supplementation were not different between two groups. After 4 weeks of summer training and iron supplementation, serum ferritin level was significantly increased only in IS group after supplementation. Mean corpuscular volume and total iron binding capacity were significantly decreased in both groups. Meanwhile, hemoglobin and red blood cell count were significantly lowered only in placebo group. The IgM concentration increased significantly in both groups, but IgG concentration had increasing tendency only in IS group (p < 0.064). Therefore, iron supplementation during hard training period may be helpful to improve work capacity of the athletes by improving ferritin status and humoral immune responses.

Repair of Iron Deficiency in Rats by the Intake of Recombinant Yeast Producing Human H-ferritin (훼리틴 생산 재조합 효모의 철분 결핍성 빈혈 개선 효과)

  • This study examined whether or not the iron that is accumulated in the recombinant microbes that produce ferritin is bioavailable to rats with iron deficiency. Rats induced with iron deficiency were treated with iron preparations of $Fe(NH_4)_2(SO_4)_2$, horse spleen ferritin, control yeast, and ferritin-producing recombinant yeast for 14 days. The bioavailability of iron was examined by measuring hemoglobin concentration, hematocrit value, and tissue iron stores. Differences between dietary groups were determined by one-way ANOVA, at the level of significance p<0.05. Based on hemoglobin concentration and hematocrit value, iron in $Fe(NH_4)_2(SO_4)_2$, horse spleen ferritin, and ferritin-producing yeast were bioavailable in rats and cured iron deficiency. The efficacy of ferritin and ferritin-producing yeast was confirmed in establishing tissue iron stores after the induction of iron deficiency. The iron sources of ferritin and the ferritin-producing yeast seemed to be as effective for the recovery from iron deficiency as the iron compounds of ferric citrate and ferrous ammonium sulfate. The results suggest that the iron stored in ferritin of the recombinant yeast is bioavailable, and that the recombinant yeast may contribute widely as a source of iron to resolve the global problem of iron deficiency.

A Study on Iron Nutritional Status of Girls at Puberty in Kangnung Area (강릉지역 일부 사춘기 소녀의 철분 영양상태에 관한 연구)

  • This study was designed to assess the iron nutritional status of girls at puberty in Kangnung area. The subjects consisted of 161 adolescents in sixth-grade in primary school and first-grade in middle school girls. Anthropometric measurements were taken for body weight, height, percentage of body fat, and circumferences of waist and hip. Nutrient intakes were assessed by modified 24-hour recall method. Food models and other measuring tools were also used. Fasting blood samples were obtained and analyzed for hemoglobin(Hb) concentration, hematocrit(Hct), serum iron(FE) and total iron binding capacity(TIBC). Mean values for Hb, Hct, Fe, TIBC, TS and serum ferritin were $13.6{\pm}0.9g/dl$, $39.6{\pm}3.9%, 91.3{\pm}36.3{\mu}g/dl$, $327.9{\pm}45.2{\mu}g/dl$, $28.3{\pm}11.8%$ and $37.4{\pm}24.2ng/ml$, respectively. Prevalence of iron deficiency greatly varied by indices from 4.8% when judged by Hb to 18.4% by serum Fe concentration. The Hb concentration was positively correlated with Hct(r=0.641), serum iron(r=0.266) and transferrin saturation(r=0.237)(p<0.05). On the other hand, serum ferritin concentration showed significantly negative correlation with TIBC(r=-0.572). Mean daily intake of iron was 14.94mg and heme iron intake was 1.13mg and which was 7.6% of total iron intake. Total absorbable iron calculated by the method of Mosen was 1.38mg and bioavailability of dietary iron was 9.3%. These results suggest that the prevalence of iron deficiency of pubertal girls is very high, therefore the guidelines for diet and social supports, such as, school food service system should be provided to improve their iron status in middle school students.

Effects of Aspirin on Nitrite Toxicity in Cultured Eel (Anguilla japonica) (양식 뱀장어 (Anguilla japonica)의 아질산 독성에 미치는 aspirin의 투여 효과)

  • The effect of aspirin on the recovery of nitrite-induced methemoglobinemia in cultured eels (Anguilla japonica) was studied. Methemoglobinemia was induced by exposing eels to nitrite (120 ㎎ NO2-N/ℓ) for 24 hr. The nitrite exposed eels were bathed in 20 ppm aspirin solution (Aspirin), 0.8 % NaCl solution (NaCl), 20 ppm aspirin plus 0.8% NaCl solution (NaCl + Aspirin) and 50% nitrite free water(control) for 24 hr to recover from nitrite toxicity. Peripheral blood was taken from the arterious bulb from all groups to analyse hematocrit value, hemoglobin concentration, and nitrite concentration of the blood. Histopathological features of gill were also observed. Aspirin and control groups were more effective than NaCl and NaCl + Aspirin groups in recovery of hematocrit value and hemoglobin concentration, methemoglobin rate and nitrite concentration. The histopathological features on the gill of aspirin group were similar to those of normal eels, but other groups showed focal hyperemia in the lamellar carpillaries, epithelial hyperplasia. These results suggested that aspirin was very effective to recover from methemoglobinemia in nitrite-induced cultured eels.

Improvement of Lipid Metabolism and Antihyperglycemic by Lentinus edodes in High Fat-fed and Streptozotocin-treated Rats (고지방과 streptozotocin으로 유도한 제 2형 당뇨에서 표고버섯이 지질대사와 항당뇨 효능에 미치는 영향)

  • Abnormal regulation of glucose and impaired lipid metabolism that result from a defective or deficient insulin are the key etiological factor in type 2 diabetes mellitus (T2DM). The our study evaluated the beneficial effect of diet supplementation with Lentinus edodes on hyperglycemia and lipid metabolism in normal and type 2 diabetic rats. The animals were divided into 4 groups: group I(control) rats were fed standard diet (12% of calories as fat); group II (T2DM) rats were fed HFD (40% of calories as fat) for 2 weeks and then injected with STZ (50 mg/kg); group III and group IV rats were continually fed a diet containing 1% and 10% Lentinus edodes for 4 weeks after T2DM induction, respectively. After 4 weeks we determined biochemical parameters such as glucose, insulin concentration, serum total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL), and glycosylated hemoglobin (HbA1c) concentration were also measured. There was a significant reduction in serum TC and TG in the Lentinus edodes supplement groups. The Lentinus edodes diet supplementation were found to have a potent lipid metabolism improvement as well as LDL concentration decreased and HDL concentration was increased. Concentrations of blood glucose and HbA1c in the experimental groups II were significantly decreased after 4 weeks compared with the control group. The Lentinus edodes diet supplementation is useful in regulating the glucose level, improves the insulin, HbA1c, serum lipid metabolism in experimental diabetic rats. We suggest that Lentinus edodes supplementation may have the control effects of diabetes mellitus by improving blood glucose control and lipid metabolism.

Iron Nutritional Status of Female Students in Kangnung National University (강릉대 일부 여대생의 철분영양상태에 관한 연구)

  • To evaluate iron nutritional status of female college students, fasting blood samples were taken from 76 female students of Kangnung National University. Hemoglobin(Hb), hematocrit(Hct), serum iron(Fe), total iron binding capacity(TIBC) and serum ferritin concentrations were measured and transferrin saturation was calculated. Mean values for Hb, Hct, Fe, TIBC, TS and serum freeitin were 13.64$\pm$1.42g/dl, 40.99$\pm$4.31%, 103.0$\pm$33.3$\mu\textrm{g}$/이, 395.3$\pm$9.07$\mu\textrm{g}$/dl, 26.58$\pm$9.07$\%$and 26.76$\pm$17.5ng/ml, respectively. Prevalence of iron deficiency greatly varied by indices from 6.8% when judged by Hct to 26.0$\%$ by serum ferritin concentration. The Hb concentration was positively correlated with hematocrit (r=0.5402), serum iron(r=0.2819) and transferrin saturation(r=0.2777)(p<0.05). on the other hand, serum ferritin concentration showed significantly negative correlation with TIBC(r=-0.3196). Two-day dietary intake records were collected from subjects to estimate mean daily iron intake and bioavailability of dietary iron. Mean daily intake of iron was 13.15mg and heme iron intake was 0.83mg which was 6.4% of total iron intake. Total absorbable iron calculated by the method of Monsen was 1.27mg and bioavailability of dietary iron was 9.6%. In the light of high prevalence of iron deficiency based of serum ferritin concentration and low bioavailability of iron in the diet, guidelines about diet should be made to increase the content and bioavailability of iron in the diet if female college students.

  • Failure Renal is the function of the kidneys to remove waste products and keep them on the periphery. and minerals for the body. Chronic renal failure is a syndrome characterized by a slow, irreversible deterioration of renal function due to the slow destruction of renal parenchyma. Calcium is one of the important minerals that the body contains in the blood and important tissues, and it has an important role in vital processes such as muscle contraction, nerve impulse conduction, the efficiency of heart muscle work, and blood clotting processes. The aim of the study is to study and compare calcium levels in men and women. It includes studying abnormal levels of calcium that cause many diseases, including chronic renal failure, and studying changes associated with renal failure. The method of this study was conducted on patients with chronic renal failure at Murjan Teaching Hospital in Babylon city during the period. The study included a sample of 70 patients (40 males, 30 females) with chronic renal failure, their ages ranged from 30-65, and 60 (30 males, 30 females) healthy without the disease of the same age. The result was a significant decrease in the number of red and white blood cells, hemoglobin concentration, hematocrit and platelets in patients with chronic renal failure, The result has been showed significant level in enzymes activity for transfer of amine group (alanine amino transferase, aspartate amino transferas) and phosphatase alkaline and also concentration of total bilirubin in patient with compare with healthy, Significantly increases, were found in the concentration of urea, uric acid and creatinine, as well as the concentration of calcium and phosphorous ions in the blood serum of patients compared to healthy controls.

The Oxidative Stress in Cigarette Smokers and Antioxidant Vitamins

  • The purpose of this study was to find the extent of lipid peroxidation of erythrocytes in cigarette smokers, and to determine the relative effectiveness of $\beta$-carotene, canthaxanthin , and $\alpha$-tocopherol as antioxidants. Thirty smokers and 30 nonsmokers participated in this study . No significant differences according to age, sex, and height were shown. Cigarette smokers in this study had higher hemoglobin concentrations and more oxidation of hemoglobin than non-smokers. In addition, the erythrocytes of cigarette smokers had significantly higher MDA concentrations than crythrocytes of nonsmokers, which suggests that smokers may have tocopherol were studied in vitro by measuring the concentration of malondialdehyde(MDA) and precent hemolysis of erythrocytes. The addition of any antioxidant to erythrocytes significantly decreased MDA concentrations(p<0.05) while antioxidants showed nonsignificant inhibition of hemolysis. Among the antioxidant used in this study, canthaxanthin showed the greatest inhibition of both lipid peroxidationand hemolysis. Meanwhile, $\alpha$-tocopherol showed potent inhibition of lipid peroxidation, but not of hemolysis.

Nutritional Status in Healthy Elderly Koreans from Urban Households (도시 지역에 거주하는 노인의 영양상태에 관한 연구)

  • Nutritional Status of 74 elderly Koreans(35 men and 39 women) from middle-income households in Incheon were evaluated in terms of dietary nutrient intakes, anthropometric measurements and biochemical blood analysis. The mean consumptions of all nutrients were lower than R.D.A except iron, thiamine and niacin. However, 25.7% of men and 33.3% of women consumed less amount of iron than 75% of R.D.A. Average body mass index of women was sightly but significantly higher than that of men (P<0.05). Obesity rate and triceps skinfold thickness of women were much higher than those of men (23.3% vs 5.4% and 14.9mm vs 9.6mm, respectively). By judging from amounts of hemoglobin, hematocrit, serum iron, total iron binding capacity, mean corpuscular hemoglobin concentration and transferrin saturation of the elderly, 5.6-33.3% of men and 5.0-33.3% of women seem to be possibly anemic.
