This study was conducted to determine changes in th contents and composition of dietary fiber during the growth of soybean sprout. Soybean was soaked in water at $25^{\circ}C$ for 2hrs and cultivated at 2$0^{\circ}C$ for 7 days under dark condition. The soybean sprouts were divided into cotyledon and axis and sampled every 24hrs. The analysis methodlogies used were Van Soest's NDF, AOAC's ADF and lignin and Prosky's IDF, SDF, TDF. The weight of 100 sprouts increased gradually from 20.26g to 90.12g during the growth periods. The weight increased to 344.9% of the original weight. The germination rate was 100% after soaking at $25^{\circ}C$ for 2hrs. Root length increased gradualy from 0.6cm at 1st day to 17.2cm at 7th day. The crude ash and crude fat contents showed no significant change in the cotyledon and axis. The crude protein contents increased in the cotyledon and axis, whereas the total carbohydrate content didn't have general tendency. The insoluble dietary fiber(IDF), soluble dietary fiber(SDF) and total dietary fiber(TDF) contents of cotyledon were no significantly different from 20.01%, 1.45%, 21.46% at 1st day to 22.75%, 2.07%, 24.82% at 7th day on dry basis. In axis those contents increased from 23.19%, 1.97%, 25.16% at 1st day to 32.78%, 3.02%, 35.80% at 7th day, respectively. The neutral detergent fiber(NDF) contents of cotyledon and axis increased from 4.35% to 6.39% and from 6.44% to 26.60% respectively on dry basis. The acid detergent fiber (ADF) contents of cotyledon and axis increased from 2.84% to 4.91% and from 2.5% to 4.7%, but there were no significantly different in the hemicellulose and lignin contents on dry basis. The hemicellulose and lignin contents of axis increased with culture periods from 1.70% to 4.41% and from 0.20% to 2.11%, respectively. The cellulose contents increased from 4.54% to 20.35% on dry basis.

The problems with unsorted municipal waste are always associated with disposal issues as it requires a large area for landfilling or high energy used for incineration. In recent years, an autoclaving technique has been considered a promising approach which could minimize the volume of organic waste from being directly disposed or incinerated. In this work, an attempt was done to study the saccharification potential of organic residues under elevated temperature Thermal treatment involving hot water bath was applied to treat the organic residue ranging from 60℃ to 100℃ for 30 and 60 minutes. The result obtained showed an increasing trend for the concentration of glucose and carbohydrate. However, the result for lignocellulose content which contains various component includes extractive, holocellulose, hemicellulose, cellulose and lignin show variation. Based on the thermal treatment carried out, the result indicated that the trend of glucose and carbohydrate content. The highest percentage of glucose that can be obtained 978.602 ㎍/ml which could be obtained at 90℃ at 60 minutes. The carbohydrate also shows an increasing trend with 0.234 mg/ml as the highest peak achieved at 80℃ for 30 minutes treatment. However, it was found that the lignocellulose content varies with temperature and time. The statistical analysis was carried out using two-ways ANOVA shows an interaction effect between the independent variables (temperature and contact time) and the saccharification effects on the food wastes. The result shows a variation in the significant effect of independent variables on the changes in the composition of food waste.
The production of xylanase and $\beta$-xylosidase was investigated in submerged culture of Aspergillus niger NRC 107. The maximum production occurred when the pH was controlled at 6.0 during the fermentation. Among the various carbon sources investigated, corn-cob xylan (1.5%, w/v) yielded maximal production of the enzymes. The $NaNO_{3}$ was the most favorable nitrogen source for enzyme production and $KH_2P0_4$ concentration at 0.3%(w/v) was found to be optimum. Incorporation of wheat bran to the culture medium improved xylanase production. Addition of L( -) sorbose to the culture medium promoted the secretion of $\beta$-xylosidase. It was possible to increase the production of xylanase (39.43 units/ml) and that of $\beta$-xylosidase (4.2 unitslml) by submerged culturing the A. niger NRC 107 in the modified medium.
In this study, dilute acid pretreatment of $Liriodendron$$tulipifera$ was performed for enzymatic hydrolysis. As the pretreatment temperature was increased, enzymatic hydrolysis and enzyme adsorption yield also increased. The highest enzymatic hydrolysis yield was 57% (g/g) and enzyme adsorption was 44% (g/g). Enzymatic hydrolysis yield was determined with weight loss of pretreated biomass by enzyme, and enzyme adsorption was a percentage of enzyme weight attaching on pretreated biomass compared with input enzyme weight. When $L.$$tulipifera$ was pretreated with 1% sulfuric acid at $160^{\circ}C$ for 5 min., hemicellulose was significantly removed in pretreatment, but the lignin contents were constant. Other changes in surface morphology were detected on biomass pretreated at $160^{\circ}C$ by a field emission scanning electron microscope (FESEM). A large number of spherical shapes known as lignin droplets were observed over the entire biomass surface after pretreatment. Hemicellulose removal and morphological changes improved enzyme accessibility to cellulose by increasing cellulose exposure to enzyme. It is thus evidence that enzyme adsorption is a significant factor to understand pretreatment effectiveness.

The objective of this study is to understand how regulatory mechanisms respond to sugar status for more efficient carbon utilization and source-sink regulation in plants. So, we need to identify and characterize many components of sugar-response pathways for a better understanding of sugar responses. For this end, genes responding change of sugar status were screened using Arabidpsis cDNA arrays, and confirmed thirty-six genes to be regulated by sucrose supply in detached leaves by RNA blot analysis. Eleven of them encoding proteins for amino acid metabolism and carbohydrate metabolism were repressed by sugars. The remaining genes induced by sugar supply were for protein synthesis including ribosomal proteins and elongation factors. Among them, I focused on three hydrolase genes encoding putative $\beta$-galactosidase, $\beta$-xylosidase, and $\beta$-glucosidase that were transcriptionally induced in sugar starvation. Homology search indicated that these enzymes were involved in hydrolysis of cell wall polysaccharides. In addition to my results, recent transcriptome analysis suggested multiple genes for cell wall degradation were induced by sugar starvation. Thus, I hypothesized that enzyme for cell wall degradation were synthesized and secreted to hydrolyze cell wall polysaccharides producing carbon source under sugar-starved conditions. In fact, the enzymatic activities of these three enzymes increased in culture medium of Arabidopsis suspension cells under sugar starvation. The $\beta$-galactosidase encoded by At5g56870 was identified as a secretory protein in culture medium of suspension cells by mass spectrometry analysis. This protein was specifically detected under sugar-starved condition with a specific antibody. Induction of these genes was repressed in suspension cells grown with galactose, xylose and glucose as well as with sucrose. In planta, expression of the genes and protein accumulation were detected when photosynthesis was inhibited. Glycosyl hydrolase activity against galactan also increased during sugar starvation. Further, contents of cell wall polysaccharides especially pectin and hemicellulose were markedly decreased associating with sugar starvation in detached leaves. The amount of monosaccharide in pectin and hemicellulose in detached leaves decreased in response to sugar starvation. These results supported my idea that cell wall has one of function to supply carbon source in addition to determination of cell shape and physical support of plant bodies.

Rice straw is an important roughage resource for ruminants in many rice-producing countries. In this study, a rice brittle mutant (BM, mutation in OsCesA4, encoding cellulose synthase) and its wild type (WT) were employed to investigate the effects of a cellulose synthase gene mutation on rice straw morphological fractions, chemical composition, stem histological structure and in situ digestibility. The morphological fractions investigation showed that BM had a higher leaf sheath proportion (43.70% vs 38.21%, p<0.01) and a lower leaf blade proportion (25.21% vs 32.14%, p<0.01) than WT. Chemical composition analysis showed that BM rice straw was significantly (p<0.01) higher in CP (crude protein), hemicellulose and acid insoluble ash (AIA) contents, but lower in dry matter (DM), acid detergent fiber (ADFom) and cellulose contents when compared to WT. No significant difference (p>0.05) was detected in neutral detergent fiber (NDFom) and ADL contents for both strains. Histological structure observation indicated that BM stems had fewer sclerenchyma cells and a thinner sclerenchyma cell wall than WT. The results of in situ digestion showed that BM had higher DM, NDFom, cellulose and hemicellulose disappearance at 24 or 48 h of incubation (p<0.05). The effective digestibility of BM rice straw DM and NDFom was greater than that of WT (31.4% vs 26.7% for DM, 29.1% vs 24.3% for NDFom, p<0.05), but the rate of digestion of the slowly digested fraction of BM rice straw DM and NDF was decreased. These results indicated that the mutation in the cellulose synthase gene could improve the nutritive value of rice straw for ruminants.
Objective: The objective of this study was to determine the digestible energy (DE) and metabolizable energy (ME) of yellow dent corn sourced from different meteorological origins fed to growing pigs and develop equations to predict the DE and ME of yellow dent corn from southwestern China. Methods: Sixty crossbred barrows were allotted to 20 treatments in a triplicate 20×2 incomplete Latin square design with 3 replicated pigs per dietary treatment during 2 consecutive periods. Each period lasted for 12 days, and total feces and urine during the last 5 days of each period were collected to calculate the energy contents. Results: On dry matter (DM) basis, the DE and ME in 20 corn grain samples ranged from 15.38 to 16.78 MJ/kg and from 14.93 to 16.16 MJ/kg, respectively. Selected best-fit prediction equations for DE and ME (MJ/kg DM basis) for yellow dent corn (n = 16) sourced from southwestern China were as follows: DE = 28.58-(0.12×% hemicellulose)+(0.35×% ether extract)-(0.83×MJ/kg gross energy)+(0.20×% crude protein)+(0.49×% ash); ME = 30.42-(0.11×% hemicellulose)+(0.31×% ether extract)-(0.81×MJ/kg gross energy). Conclusion: Our results indicated that the chemical compositions, but not the meteorological conditions or physical characteristics could explain the variation of energy contents in yellow dent corn sourced from southwestern China fed to growing pigs.
This study is for the effective pretreatment and saccharification of lignocellulosic biomass for a transport fuel receiving attention. The waste water during the pretreatment of biomass is major factor for determining the price of biofuel. Therefore, we conducted high concentration of organosolv pretreatment for decline waste water and reusing the solvent. We confirmed effect of organosolv pretreatment by components analysis and enzymatic hydrolysis of pretreated biomass. The corn stover was used for and 99.5 wt% of ethanol as a organosolv pretreatment. The pretreatment condition was varied 130 to $190^{\circ}C$ during the designated reaction times and the effect of pretreatment was investigated by enzymatic hydrolysis. The highest glucose conversion was more than 68% the pretreatment condition of $190^{\circ}C$ for 70 min or more. The solid remaining was more than 70% and almost of cellulose and hemicellulose were survived.
A method for acid-hydrolysis of agricultural wastes and its utilization was investigated. In order to obtain fermentable sugar solution from cellulosic wastes such as cereal straws and hulls, in particular, of rice, barley and wheat, the chemical compositions were analyzed and optimum conditions of hydrolysis determined. The cereal straws contain 42 to 55 % of crude cellulose including hemicellulose. On the hydrolysis with 1% of sulfuric acid at 40 psig, 35.6% of the reducing sugar based on the weight of dry matter was formed from rice straw, (variety Chinheung) in 30 min. More powerful condition of hydrolysis would appear to decompose the sugar formed into other compounds, for instance, furfural. Under atmospheric pressure with 5% of the acid, rice straw was hydrolyzed to 35% of reducing sugar content in 3 hours. Candida utilis could assimilate the sugars in the hydrolyzate up to more than 97%, and a yield of the yeast cells reached 55% to the utilized sugars.
Pretreatment of cellulosic biomass is necessary before enzymatic saccharification and fermentation. Extrusion is a well established process in food industries and it can be used as a physicochemical treatment method for cellulosic biomass. Aqueous ammonia soaking treatment at mild temperatures ranging from 60 to $80^{\circ}C$ for longer reaction times has been used to preserve most of the cellulose and hemicellulose in the biomass. The objective of this study was to evaluate the effect of extrusion treatment on aqueous ammonia soaking method. Extrusion was performed with miscanthus sample conditioned to 2mm of particle size and 20% of moisture content at $200^{\circ}C$ of barrel temperature and 175rpm of screw speed. And then aqueous ammonia soaking was performed with 15%(w/w) ammonia solution at $60^{\circ}C$ for 1, 2, 4, 8, 12 hours on the extruded and raw miscanthus samples respectively. In the combined extrusion-soaking treatment, most compositions removal occurred within 1~2 hours and on a basis of 1 hour soaking treatment values, cellulose was recovered about 85% and other compositions, including hemicellulose, are removed about 50% from extruded miscanthus sample. The combined extrusion-soaking treated and soaking only treated samples were subjected to enzymatic hydrolysis using cellulase and ${\beta}$-glucosidase. The enzymatic digestibility value of combined extrusion-2 hours soaking treated sample was comparable to 12 hours soaking only treated sample. It means that extrusion treatment can shorten the conventional long reaction time of aqueous ammonia soaking. The findings suggest that the combination of extrusion and soaking is a promising pretreatment method to solve both problems for no lignin removal of extrusion and long reaction time of aqueous ammonia soaking.
