Effects of Acute Oral Administration of Bisphenol A on the Immune Function in Mice (Bisphenol A의 급성노출이 마우스의 면역기능에 미치는 영향)

  • In order to investigate the effects of bisphenol A (BPA) on immune system in mice we examined the various immunological parameters. After single oral administration of BPA to female ICR mice, the weights of bodies and lymphoid organs, splenic cellularity and hematological parameters were examined on day 2 and 7. Among them WBC and splenic cellularity were slightly decreased on day 2. To assess the effects of BPA on humoral immune responses, splenic IgM plaque forming cell (PFC) and serum IgM were assayed. When BPA was administered after immunization with SRBC, but not before immunization, IgM PFC against SRBC was significantly lowered in a dose dependent manner. Serum IgM level was also decreased on day 4 when high dose (2000 mg/kg) of BPA was administrated after injection of OVA-antigen. The indexes of splenocyte proliferation (SP) to concanavalin A (Con A) and bacterial lipopolysaccharide (LPS) were measured in vitro by MTT assay. At low concentration BPA slightly increased splenocyte proliferation but at higher concentration it showed significant inhibitory effects on cell proliferation. Mitogen-stimulated SP was also determined with spleen cells from BPA treated mice. Con A-induced SP was slightly decreased and LPS-induced SP was especially inhibited at 1000 mg/kg and 2000 mg/kg of BPA. These results indicate that BPA is able to acutly evoke humoral and cell mediated immune suppression in mice.

Immunosuppressive Effect of the Intraperitonially Injected Pine Needle Distillate in Mice

  • This study examined the effect of pine needle distillate (Pinus densiflora Sieb. et Zucc) on the immune system and hematological parameters. C57BL/6 male mice weighing 20 ~21 g were divided into 3 groups and intraperitonially injected with either 200 $\mu$L of saline (control), 50% diluted (P50) or 100% pine needle distillate (P100) once a day for 24 days. At the end of the experiment, the mice were anesthetized by ether and peripheral blood was collected from the femoral artery and the spleen was excised. Spleen weight decreased significantly (p<0.001) in the pine needle groups compared to the control group. The blood was used for a complete blood count and flow cytometrical analysis after immunofluorescence staining. The pine needle distillate dose-dependently decreased the CD4$^{+}$/CD8 sup +/ ratio (p <0.05), and showed a tendency to increase the mean FSC (forward scatter) values of the CD8$^{+}$T cells, while decrease the values of the CD4$^{+}$T cells. There were no significant differences in WBC, RBC and platelet counts among the three groups, but hemoglobin and hemoglobin-related parameters and platelet volume increased and red blood cell volumes decreased with the administration of the pine needle distillate. These results suggest that the pine needle distillate may have immunosuppressive effects.

Erythrocyte Sedimentation Rate: Its Determinants and Relationship with Risk Factors Involved in Ischemic Stroke

  • Erythrocyte sedimentation rate (ESR) evaluation is a useful tool for monitoring disease activity in various inflammatory and non-inflammatory conditions. ESR is known to be influenced by a multitude of confounding factors. The present study aimed to assess the possible determinants of the ESR and its relationship with various risk factors involved in ischemic stroke. ESR and other hematological and biochemical parameters were investigated in 163 ischemic stroke patients (107 males and 56 females) selected based on imaging techniques including magnetic resonance imaging (MRI) or computed tomography (CT) scans. Statistical analysis was performed using the SPSS 16.0 software. Linear regression analysis showed a significant inverse relationship of hemoglobin (Hb) and hematocrit or packed cell volume (PCV) (P<0.001 for females; P<0.01 for males) with the ESR. It was observed that the red blood cell (RBC) count was not strongly correlated with the ESR (P<0.05 for both males and females). It was also observed that sex significantly affected the variables determining the ESR levels, whereas age had no effect. Gender differences were also observed with respect to Hb, RBC, PCV, mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), and ESR. The possible determinants of higher ESR levels in ischemic stroke may be sex, Hb, hematocrit, and RBC count, but the role of other clinical and laboratory parameters cannot be underestimated.

Changes in water quality and hematological parameters according to the stocking density of olive flounder, Paralichthys olivaceus raised in bio-floc environment (바이오플락 양식기술 적용 넙치, Paralichthys olivaceus 수용밀도에 따른 수질변화 및 혈액학적 성상 변화)

  • The Paralichthys olivaceus (mean weight 34.3±3.5 g) was used in the conduction of density experiment in this study to establish the optimal density determination for a period of 13 weeks. The density consisted of four groups. These were group 1 (500 fish, 40 fish m-2), group 2 (750 fish, 60 fish m-2), group 3 (1,000 fish, 80 fish m-2) and group 4 (1,250 fish, 100 fish m-2), each in 12.56 ㎡ circular water tanks. In the high density groups, such as groups 1 and 2, the nitrite was increased to over 21 mg L-1 (group 3) and 25 mg L-1 (group 4). The experiment of the groups was terminated by skin ulcers and mortality after 49 days for group 3 and 43 days for group 4. The water quality environment, such as the water temperature, dissolved oxygen, salinity and pH, remained constant. The ammonia and nitrite in groups 1 and 2 remained stable after ammonia and nitrite peak. However, the experiments in groups 3 and 4 were terminated due to the mortality induced by high nitrite. Hematological parameters, such as hemoglobin and hematocrit, were significantly decreased in the high density groups. The plasma components were significantly changed in the high density groups, such as groups 3 and 4. The mean weight of groups 1 and 2 after 13 weeks were 91.3 g and 83.7 g, respectively. The survival rates were 99.4% and 98.2%, respectively. The final FCR was 0.6 in both groups. The results of this study show that the density of 80 fish m-2 or more induce mortality due to high nitrite with hematological changes. Additionally, they indicate that the 60 fish m-2 indicate proper density in bio-floc environment in olive flounder weighing less than 100 g.

Anti-tumor Activity of Acanthospermum hispidum DC on Dalton Ascites Lymphoma in Mice

  • The present study investigated the anti-tumor activity of ethyl acetate extract of Acanthospermum hispidum DC against daltons ascites lymphoma in mice. The extract was prepared by cold maceration with ethyl acetate for 3 - 7 days and evaporated in vacuum to dry. (Yield : 14.2 g, 1.42% w/w). The extract was fractionated by column chromatography by using gradient elution technique and the diterpenes fraction isolated (0.649 g). Both extract and the fraction were administered as oral suspension with tween 20 in water to tumor bearing mice (DAL) and changes in dead cell count, histopathology of tumor cells, hematological parameters and median survival time (MST) were examined and compared with that of tumor control or 5-Fluorouracil (5-FU). The results indicate that both ethyl acetate extract and fraction possess anti-tumor activity. The study suggests that Acanthospermum hispidum DC seems promising as a source of diterpenes for potential anti-tumor activity.

Toxic Effect of Combination of Buprofezin and Carbaryl in Rats (Buprofezin과 Carbaryl의 복합독성에 관한 연구)

  • In this study, it was examined the toxic effects of combination of buprofezin and carbaryl on hematological, biological and enzymetic parameters in rats. The administration of buprofezin or carbaryl both induced the tissue content of cytochrome P-450 and furthermore, the combination of the both increased significantly the liver content of cytochrome P-450 in rat. But cytochrome P-450 and NADPH -cytochrome c reductase activities in kidney were slightly increased. Administration of carbaryl and combination of the both also significantly increased hepatic aniline hydroxylase activity. In addition, in the combination group, glucose-6-phosphatase and lipid peroxidase activities were changed in the rat liver. Furthermore, cholinesterase was inhibited in rats treated with carbaryl or the combination of buprofezin and carbaryl. The above results suggested that the combined administration of buprofezin and carbaryl can induce more toxic effects than the single administration of buprofezin or carbaryl.

Four-Week Oral Toxicity Study of Gamma Irradiated Chikens in Mice (감마선 조사계육의 아급성 독성평가)

  • In order to evaluate their possible subacute toxicity, groups of 40 male and female ICR mice were given to the diet with chickens irradiated up to 30 kGy for four weeks. During the experimental periods, appearance, behavior, mortality, food and water consumption of mice fed irradiated chickens were not affected compared to the control. In urine analysis, hematological as well as in serum biochemical findings, no significant differences were found between the control (non-irradiated) and the irradiated chickens. Although minor changes in biochemical parameters were observed, they were in the normal range and were not dose dependent. Spotty necrosis was found in the male liver administered with 30 kGy irradiated chicken. However, it seems not related with gamma irradiation, because it lacks either the dose dependency and the secondary changes accompanied. Our results indicate that the adverse effect of gamma irradiated chicken can be observed at more than 30 kGy level.

Biological Contents of Velvet Antler and Femoral Venous Blood in Farmed Elk(Cervus canadensis) (사육 엘크의 녹용혈과 체녹혈의 혈액성분)

  • This study was performed to investigate the blood contents of velvet antler and femoral venous blood in farmed elk (Cervus canadensis). Blood samples were collected from 12 young adult elk (3.5-4.5 years old) with a mixture of fentanyl-azaperone-xylazine (Fentazine$^{(R)}$). Values for hematological and clinicochemical components of the clinical healthy elk were determined. The results were grouped for comparison according to velvet antler and femoral venous blood. Lower mean platelet counts and fibrinogen values were found in the velvet antler than femoral venous blood (p<0.01). White blood cell count of velvet antler was significantly higher than that of femoral venous blood 0<0.05). The clinicochemical parameters (total protein, albumin, globulin, alanine aminotransferase, aspartate aminotransferasei blood urea nitrogens calciums magnesium and phosphonls) obtained in this study far farmed elk are consistent with the values between velvet antler and femoral venous blood.

Subacute Oral Toxicity of DWP-311 in Sprague-Dawley Rats (DWP-311의 랫드에 대한 아급성경구독성시험)

  • The subacute oral toxicity study of DWP-311 was carried out in Sprague-Dawley rats of both sexes. We daily examined clinical signs, body weights, hematological and biochemical parameters, and histopathological examinations for 30 days after administration of DWP-311 with different dose levels (0, 0.04, 0.2, and 1.0 g/kg). There were no clinical signs and pathological changes compared with control group except slight decreases in spontaneous motor activities and locomotions at high dose group of DWP-311. Body weights were not significantly changed in animals treated with DWP-311, In histopathological examinations, there were 2 cases of pneumonia in control group for one male and one female, but it was not directly related to DWP-311. These results indicate that subacute oral toxicities of DWP-311 were low and the no-observed a dverse effect level (NOAEL) was considered to be 1.0 g/kg in rats.

Subacute Oral Toxicity of KDRD-010 in Rats (랫드에 대한 KDRD-010의 아급성경구독성시험)

  • The subacute toxicity was investigated in Sprague-Dawley rats orally treated with KDRD-010 at the doses of 0.056, 0.28, and 1.4 g/kg for one month. There were no clinical signs and pathological changes compared with control group. Body weights were not significantly changed between control and treatment groups. In hematological and biochemical serum parameters, all mean values appear to be within the normal range. In pathological examinations, hemorrhages of lung was observed in one male rat at low dose group and one female rat at high dose group of KDRD-010, but it was not considered to be caused by KDRD-010. These results suggest that KDRD-010 dose not induce any significant subacute oral toxicities in Sprague-Dawley rats.
