In intestinal helminth infections, Th2 immune respones are generally associated with mucin secretion for worm expulsion from the host intestine. In particular, IL-4 and IL-13 are the important cytokines related with intestinal mucus production via STAT6 signalling in nematode infections. However, this perspective has never been studied in Gymnophalloides seoi infection. The present study aimed to observe the STAT6 signalling and cytokine responses in C57BL/6 mice, a mouse strain resistant to infection with this trematode. The results showed that worm expulsion occurred actively during days 1-2 post-infection (PI), when goblet cells began to proliferate in the small intestine. The STAT6 gene expression in the mouse spleen became remarkable from day 2 PI. Moreover, G. seoi infection induced a significant increase of IL-13 from day 4 PI in the spleen of infected mice. Our results suggested that goblet cell hyperplasia and worm expulsion in G. seoi-infected mice should be induced by STAT6 signalling, in which IL-13 may be involved as a dominant triggering cytokine.
Strongylodiasis is universal in distribution but is most abundant in countries with a tropical climate. Although infestation by Strongyloides stercoralis is usually limited to the intestines, dessemination of this helminth in debilitated host can be lead to death with various clinical disorders, characterized by profound malabsorption, diarrhea, electrolyte imbalance, gram negative or opportunistic fungal sepsis, coma and death. Cell-mediated immunity contributing significantly to the control of helminthic infections, may be suppressed by carcinoma, immunosuppressive chemotherapy and use of corticosteroids. Diagnosis of Strongyloidiasis is achieved by an examination of samples of feces, duodenal aspirates and sputum of patients for Strongyloides stercoralis. Treatment of strongyloidiasis is twofold: correction of the immunosuppressive state by withdrawal of immunosuppressive drug, if possible, and vigorous treatment with thiabendazole. Testing for strongyloidiasis is especially recommanded before treating a patients should be monitored for infection by Strongyloides stercoralis and other opportunistic infection. We are reporting a case patient with Strongyloides stercoralis hyperinfection and pulmonary tuberculosis who had been used corticosteroid for persisting polyarthritis.

A survey was undertaken to evaluate the present status of intestinal infection in army soldiers. Stool specimen of 417 soldiers fixed by SAF solution were collected from a camp located in area of Nakdong river during the period from August to October of 1990. And these stool samples were examined by formalin-ether sedimentation technique once for helminths and protozoan cysts stained with Lugol's iodine solution. The results obtained in this survey were summarized as follows : l) The overall positive rate of intestinal parasite was 18.0%. 2) The egg positive rate of intestinal helminth was 15.1%, : and 11.5% for Clonorchis sinensis. 5.0% for Metagonimus vokogawai, 1.2% for Ascaris lumbricoides 1.7% for Trichiuris trichiura. 0.2% for Taenia sp. 3) The cyst positive rate of intestinal protozoa was 4.1% ; and 1.4% for Entamoeba coli, 1.9% for Giardia lamblia 0.7% for Entamoeba histolytica, 0.5% for Endolimax nana. 4) Most of samples were positive(85.3%) by single species. 10.7% by two species, 2.7% by three species and 1.3% by four species. 5) Infection rate of intestinal parasites among army soldiers decreased distinctly compared with previous data but it is revealed that the infection rate of Clonorchis sinensis among army soldiers in area of Nakdong river is still high in comparison with ever-reported data. 6) SAF fixatives used in this field survey during summertime was useful to conserve protozoan cyst and helminths ova. Also we could examine stool samples directly by formalin-ether sedimentation technique.
Clonorchis sinearis is a liver fluke which is the most prevalent helminth of humans in Korea. The better diagnostic measure of clonorchiasis is required for its nationwide control program. The present study observed antigenic bands of C. sinensis and reacting immunoglobulins in serum of infected residents. Adult C. sinensis were recovered from experimentally infected rabbits and soluble crude extract of the worms was used as the antigen after supplementation of E-64, a cysteine proteinase inhibitor. SDS- PAGE of the crude antigen resolved more than 20 protein bands between 200 and 14 kDa. The sera of infected humans collected at an endemic village showed specific IgG and IgE antibodies but little IgM and IgA antibodies. The protein bands of 94, 80, 72, 68, 52, 47, 43, 37, 34, and 28-25 kDa strongly reacted with serum Ig(GMA) or IgG antibody and 64, 62, 52, 47,44, 34,28, and 26 kDa bands reacted with serum IgE antibody. However, the 94, 80, 72, 68, 64, 62, 52, 47, and 40 kDa bands of C. sinensis antigen were found non specific. The protein bands of 43, 34, and 28-25 kDa of C. sinensis are primary target molecules of further analysis.

Serine proteases form one of the most important families of enzymes and perform significant functions in a broad range of biological processes, such as intra- and extracellular protein metabolism, digestion, blood coagulation, regulation of development, and fertilization. A number of serine proteases have been identified in parasitic helminths that have putative roles in parasite development and nutrition, host tissues and cell invasion, anticoagulation, and immune evasion. In this review, we described the serine proteases that have been identified in parasitic helminths, including nematodes (Trichinella spiralis, T. pseudospiralis, Trichuris muris, Anisakis simplex, Ascaris suum, Onchocerca volvulus, O. lienalis, Brugia malayi, Ancylostoma caninum, and Steinernema carpocapsae), cestodes (Spirometra mansoni, Echinococcus granulosus, and Schistocephalus solidus), and trematodes (Fasciola hepatica, F. gigantica, and Schistosoma mansoni). Moreover, the possible biological functions of these serine proteases in the endogenous biological phenomena of these parasites and in the host-parasite interaction were also discussed.
The prevalence of liver and intestinal fluke infections was determined by surveying inhabitants of Hengxuan, Fusui, and Shanglin villages which were known to be endemic for liver flukes in Guangxi, China in May 2010. A total of 718 people were examined for helminth eggs by the Kato-Katz thick smear technique, ultrasonography, immunoaffinity chromatography, and DNA sequencing. The overall egg positive rate was found to be 59.6% (28.0-70.6%) that included mixed infections with liver and intestinal flukes. Cases showing higher than 20,000 eggs per gram of feces (EPG) were detected between 1.3% and 16.2%. Ultrasonographic findings exhibited overall 28.2% (72 of 255 cases) dilatation rate of the intrahepatic bile duct. Clonorchis sinensis infection was detected serologically in 88.3% (38 of 43 cases) among C. sinensis egg positive subjects by the immunoaffinity chromatography using a specific antigen for C. sinensis. For differential diagnosis of the liver and intestinal flukes, more precise PCR and nucleotide sequencing for copro-DNA were performed for 46 egg positive cases. Mixed infections with C. sinensis and Metagonimus yokogawai were detected in 8 of 46 egg positive cases, whereas 29 specimens were positive for Haplorchis taichui. Ultrasonographic findings and immunoaffinity chromatography results showed usefulness, even in a limited way, in figuring out of the liver fluke endemicity.
Distribution of a monogenean helminth Microcotyle sebastis on the gills of cultured Korean rockfish (Sebastes schlegeli) was investigated with regard to gill arches, sides of gill hemibranches (anterior or posterior), and their sections (dorsal, medial and ventral). M. sebastis has a significant preference for the second and third pair of gills, and shows marked affinity for anterior hemibranches of each gill branch, and medial sections of each gill hemibranch. The results suggest that the larger volume of water flows and surface area of the second and third pair of gills might affect the distribution of M. sebastis, and the concentrated distribution of M. sebastis on the anterior medial section of gills would be related with the increasing chances of mating by niche restriction.
Fecal samples of cats purchased in Seoul were examined for helminth ova or protozoan oocysts from December 1987 to March 1988. Out of the 41 samples,31 (75.6%) were positive and 60 (146.3%) were cumulative positive for parasites. The followings were identified In the samples: Eggs of Toxocarn cacti. Clonorchis sinensis, Metoeonimn sp., Phnrvngostomum cordntum, Spirometra erinocei, Tcenia toenicejormis and oocysts of Isosporn sp. From nine autopsied cats, larvae of Anisakis simplex, adults of C. sinensis, M. yokogawai. P cordatum, S. erinacei and T. tqeniaejormis were identified. This is the first report on the detection of Anisakis larvae from cats In Korea. The possible role of cats as a source of human infection with each parasite was discussed. A small-scale epidemiological survey was undertaken on the residents along the Mekong River near Pakse, Laos, to know the status of helminthic infections. A total of 137 fecal samples were collected from the staffs of the provincial government, their family, and primary schoolchildren in Pakse City, Champassak Province, and examined by Kato- katz smear technique. The overall helminth positive rate was 75.9%, and the helminths detected were Opisthorchis viuerrini (43.8%), Asccris lumbricoines (26.3%), fichuris trichiurn (19.0%), hookworms (19.0%), Strongyle컸os stercoraLis (2.2%), Taenio sp. (0.7%), and Schistosoma mekongi (1.5%). To obtain the adult worm of the liver fluke, three infected persons were treated with praziquantel and purged with magnesium sulfate. Five, 10, and 395 adult flukes, respectively, were collected from their diarrheic stools, all of which were morphologically identified as 0. viueni,ni. The results represent that the liver fluke and soil-transmitted helminths are highly prevalent, and the life cycle of 5. mekowi is likely to be maintained in this area.
Many immune down-regulatory molecules have been isolated from parasites, including cystatin (cystain protease inhibitor). In a previous study, we isolated and characterized Type I cystatin (CsStefin-1) of the liver fluke, Clonorchis sinensis. To investigate whether the CsStefin-1 might be a new host immune modulator, we induced intestinal inflammation in mice by dextran sodium sulfate (DSS) and treated them with recombinant CsStefin-1 (rCsStefin-1). The disease activity index (DAI) increased in DSS only-treated mice. In contrast, the DAI value was significantly reduced in rCsStefin-1-treated mice than DSS only-treated mice. In addition, the colon length of DSS only-treated mice was shorter than that of rCsStefin-1 treated mice. The secretion levels of IFN-${\gamma}$ and TNF-${\alpha}$ in the spleen and mesenteric lymph nodes (MLNs) were significantly increased by DSS treatment, but the level of TNF-${\alpha}$ in MLNs was significantly decreased by rCsStefin-1 treatment. IL-10 production in both spleen and MLNs was significantly increased, and IL-$10^+F4/80^+$ macrophage cells were significantly increased in the spleen and MLNs of rCsStefin-1 treated mice after DSS treatment. In conclusion, rCsStefin-1 could reduce the intestinal inflammation occurring after DSS treatment, these effects might be related with recruitment of IL-10 secreting macrophages.
