A Cytoplasmic Polyhedrosis Virus Isolated from the Oriental Tobacco Budworm, Heliothis assulta Guenee (Lepidoptera: Noctuidae) (담배나방 세포질다각체병 바이러스의 동정 및 병원성에 관한 연구)

  • A cytoplasmic polyhedrosis virus isolated from the oriental tobacco budworm, Heliothis assulta (HaCPV), was studied on morphology of the polyhedron and virus particles, analysis of viral protein and nucleic acid, and bioassay of the HaCPV to determine the feasibility of application as a microbial control agent. The shape of polyhedron was hexagonal ranging 0.5-3.7 ${\mu}m$ and the virus particles were icosahedral outline measured 55 nm in diameter. Polyhedral protein was composed of a major polypetide of 24.3 Kd and 5 minor components and virus particle had seven polypeptides ranging in 28.0 Kd-133. 6 Kd by the SDS-P AGE. The genome of virus was segmented with 10 double stranded RNA in the total mol. wt. of 18.08 Md ranging in 0.65 Md -2.79 Md. The $LC_{50}$ values of the HaCPV to the 3rd instar of H. assulta larvae were calculated to $2.895{\times}10^5PIBs/ml$. The $LT_{50}$ values in the concentration of $5.0{\times}10^{6}PIBs/ml$ was 16.4 days.

  • The behavior and circadian rhythm of emergence, copulation and oviposition in the Oriental tobacco budworm (Heliothis assulta Guenee) were studied to obtain the following results. More than three quarters of adults emerged within 3 hours after the light-off. It took about 33 min for an adult to expand and tan its wings after emergence. Adult male showed a sequential pattern of mating behavior, such as antennal movement, wing elevation and vibration, exten¬sion of hairpencils, and tapping of female ovipositor leading to a copulation. However, adult female revealed a rather simple behavior, such as protraction and retraction of her terminal abdomen and vibration of wings. All of the mating took place during the early part of the scotophase, with the peak from 1 hr to 2 hrs after the light-off. But the mating peak in virgin females tended to advance with age. Adult females showed the highest rate of mating among 24 hr-olds and the rate declined with age. The rate of first matings in males gave a very similar pattern to that of females. The duration of copulation was $77\pm$14min. Oviposition occurred throughout the entire scotophase, with the peak during the first 2 hrs. The most nu¬mber of eggs deposited daily was seen on the 3rd day after the mating. Mating rate, number of eggs laid and longevity we~e compared under different sex ratio. Unmated adults lived longer than mated adults.

Structure of sensilla on the Antenna and Mouthparts of the Oriental Tobacco Budworm(Heliothis assulta Guenee) Larvae (담배나방의 유충의 촉각 및 구기 감각기의 구조)

  • Studies were carried out to describe the structure and distribution of various sensilla on the antenna and mouthparts of the Oriental tobacco bud worm (H. assulta) larvae by light and scanning electron microscopy. The larval antenna has 3 segments. The second segment has 2 hairs and 3 pegs, while the third segment shows 3 regular pegs and a segmented peg (styloconicum type). Each mandible bears 2 aporous hairs on the lateral surface. The labrum bears 12 aporous hairs on its external surface. and 6 aporous hairs, 4 coeloconica-like sensilla and 2 campaniformia-like sensilla on its epipharynx. The stipes has 3 hairs, and the galea has 3 pegs. 2 papillae and 2 domes. The maxillary palpus has only a digitiform sensillum on its 1st segment but 11 sensilla of 4 different types on its 2nd segment. Maxillary sensilla represent almost 70% of total number of larval sensilla on the mouthparts. Labial pal pus has a single segment bearing a large segmented uniporous peg and a small peg. Finally their possible fun¬ctions were suggested.

  • Embryonic developmental rates of the Oriental tobacco budworm, Heliothis assulta Guenee, were compared at various constant temperatures and 16 hours of light, and detailed embryogenesis was also studied at $25^{\circ}C$. The egg was nearly globular in form and had an average equatorial diameter of 0.53mm. A single micropyle was in the center of the circular area at the anterior pole of the egg. Durations of embryogenesis at 20, 25, 30, and $35^{\circ}C$ were 147, 81, 61, and 67hrs, respectively. Embryonic death was especially higher at $35^{\circ}C$ than any other temperatures investigated. Embryogenesis progressed with changes in color and pattern, which are quite characteristic at each developmental stage of the embryo. At $25^{\circ}C$, organogenesis began in 14hrs after oviposition, formation of gut completed in 44hrs and eclosion occurred in 80hrs. The embryo formed along the long axis at early developmental stage, moved towards the equatorial plane in 24hrs, and made a half-turn on the plane in 36hrs. In 40hrs, head was oriented to the anterior pole of the egg until eclosion.

  • Field experiments on red-pepper were conducted in Suweon area during 1972-1974 to determine the seasonal fluctuation of Heliothis assulta H. using black light traps and direct counts. Adult moths emerged in late May, and showed three peaks of fluctuation from late June to middle July, from middle August to late August, and in mid-September. Eggs were first discovered in the field in early July, with peak incidence Iron late August to early September. Larval peaks occurred in late July to early August, in late August and mid-September to mid-October Initial hatching in the field occurred in early July. The numbers of the first larval generation were the highest. The parasitization ratio of Trichogramma spp. on eggs averaged 51 percent during mid-July to mid-September. The numbers of Trichogramma spp. emerging from one tobacco bud-worm egg ranged from one to four, but in most cases one or two egg parasitizing wasps emerged.

  • Expression of insecticidal protein by fusion product of truncated HD-1[CryIA(a)] N-terminal and HD-73[CryIA(c)] C-Terminal fragment of Bacillus thruingiensis subsp. kurstaki was investigate. Immunological analysis of transformants by using polyclonal antisera raised against the whole-crystal protein of HD-1 revealed that SK4 and SK5 were observed cross-reaction with polypeptides of 77-kDa and 105-kDa, respectively. Bioassay of the transformant pSK5 to Plutella maculipennis and Heliothis assulta were 96% and 97%, respectively.

  • The oriental tobacco budmoth, Heliothis assulta Guenee were reared under various temperatures; $20^{\circ}C,\;25^{\circ}C,\;30^{\circ}C$ and the control effects of Thuricide $HP^{(R)}$ were examined. The results obtained were as fellows: 1. The adult longevity of oriental tobacco budmoth was 11.35 days, and 3.00 days for preovipositional period, 4.75 days for ovipositional Period, and 3.50 days for postovipositional period. 2. The total number of eggs laid by a female were 307 at $20^{\circ}C$, 413 at $25^{\circ}C$ and 189 at $30^{\circ}C$. The number of eggs per female per day were 64.05 in average. 3. The average egg Periods were 7.71 days at $20^{\circ}C$, 4.12 days at $25^{\circ}C$ and 3.58 days at $30^{\circ}C$ and the hatchiabilities were $71.25\%,\;78.49\%\;and\;81.05\%$ at the respective incubation temperatures. 4. The larval developmental periods were 43.51 days at $20^{\circ}C$, 21.79 days at $25^{\circ}C$ and 18.05 days at $25^{\circ}C$ and the mortalities were $80.70\%,\;95.93\%$ and $87.01\%$ at the respective temperatures. 5. The pupal developmental periods were 24.22 days at $20^{\circ}C$, 12.36 days at $25^{\circ}C$ and 11.50 days at $30^{\circ}C$ and the mortalities at the respective temperatures were $18.18\%,\;42.11\%\;and\;40.00\%$. 6. The calculated threshold temperatures for the development were $11.61^{\circ}C$ for the eggs, $11.96^{\circ}C$ for the larvae, and $10.06^{\circ}C$ for the pupae. The estimated total effective temperatures were 60.41 day degrees for e eggs, 319.35 day degrees for the larvae, 222.66 day degrees for the pupae, and overall total effective temperatures, however, would be ranged 640-660 day degrees if the reproductive period of the adult was considered. 7. The relationship between the overall developmental periods and the rearing temperature could be Y=-4.272X+155.39 (r=0.9105), where Y; number of days required to complete the life cycle, X; treated temperatures. 8. The control effects of Thuricide $HP^{(R)}$ were $73.43\%$ for spray and $58.22\%$ for bait applications.
