Nutritive Value and Utilization of Perennial Grasses Intercropped with Soybean Fodder by Crossbred Heifers in Humid-subtropics of Himachal Pradesh

  • A study was carried out to investigate the nutritive value and utilization of hybrid sorghum and perennial grass species viz. setaria (Setaria anceps) and hybrid napier when intercropped with soybean by growing Jersey crossbred heifers. Fifteen growing crossbred heifers (Jersey${\times}$Red Sindhi) of between 7-10 months age and pre-trial average body weight of 49-50 kg were divided on the basis of weight in to three treatment groups viz. $T_1$-hybrid sorghum+soybean, $T_2$-setaria+soybean and $T_3$-hybrid napier+soybean in a completely randomized block design. Intercropped forages were harvested fresh, chaffed and mixed before they were offered to the heifers. Chemical composition of the herbage, dry matter intake (DMI), body weight gain and nutrient digestibility co-efficients were estimated. The herbage mixtures had crude protein (CP) content in the range of 11.87 to 13.86% and ether extract (EE) contents were 2.91 to 3.11%, respectively. The herbage mixtures were rich in minerals (ash). The gross energy (kcal/g DM) was higher in hybrid napier+soybean, while hybrid sorghum+soybean and setaria+soybean herbage mixtures had lower value for gross energy. The hybrid sorghum+soybean and setaria+soybean herbage mixtures had higher contents of NDF, ADF, cellulose, lignin and silica as compared to that of hybrid napier+soybean herbage mixture. The heifers fed hybrid napier+soybean herbage mixture had significantly (p<0.05) higher $DMI\;g/kg\;W^{0.75}$ ($97.41{\pm}4.34$) as compared to hybrid sorghum+soybean ($88.31{\pm}2.66$) and setaria+soybean ($79.29{\pm}1.06$) herbage mixtures. Nutrients digestibility, DCP percent, DCP intake and nitrogen balance were significantly (p<0.05) higher in the heifers fed on hybrid napier+soybean herbage mixture. There was a significant (p<0.05) difference among different herbage mixtures in TDN. The heifers on setaria+soybean herbage mixture had lower average body weight gain (g/day) than those on hybrid sorghum+soybean and hybrid napier+soybean herbage mixtures. Data obtained in this experiment demonstrated that herbage mixture of hybrid napier+soybean was better than hybrid sorghum+soybean and setaria+soybean herbage mixtures in the nutrition of growing heifers. It had highest nutritive value, better digestibility co-efficients which showed better growth rate and higher feed efficiency. In ranking, hybrid napier+soybean herbage mixture was better followed by hybrid sorghum+soybean and setaria+soybean in nutritive value in the parameters studied. For future wasteland development program in humid-sub tropics of Himachal Pradesh hybrid napier and its intercropping with soybean is recommended for general adoption because of its better adaptability and higher nutritive value.

Influence of Dietary Addition of Dried Wormwood (Artemisia sp.) on the Performance, Carcass Characteristics and Fatty Acid Composition of Muscle Tissues of Hanwoo Heifers

  • An experiment was conducted to examine the performance and carcass characteristics of Hanwoo (Korean native beef cattle) heifers and the fatty acid composition of muscle tissues of the heifers when the animals fed diets containing four levels of dried wormwood (Artemisia sp.). For the experiment the animals were given a basal diet consisting of rice straw and concentrate mixed at 3:7 ratio (on DM basis). The treatments were designed as a completely randomized design with two feeding periods. Heifers were allotted in one of four dietary treatments, which were designed to progressively substitute dried wormwood for 0, 3, 5 and 10% of the rice straw in the basal diet. There was no difference in body weight gain throughout the entire period between the treatment groups. Feed conversion rate was improved (p<0.05) only by the 3% dried wormwood inclusion treatment compared with the basal treatment. Carcass weight, carcass yield and backfat thickness of all treatment groups were not altered by wormwood inclusion. The 5% dried wormwood inclusion significantly increased (p<0.05) the size of loin-eye area over the other treatments. The higher levels (5 and 10%) of dried wormwood inclusion resulted in the higher (p<0.05) water holding capacity (WHC) in loin than the lower levels (0 and 3%) of wormwood inclusion. The redness ($a^*$) and yellowness ($b^*$) values of meat color were significantly lower (p<0.05) in the top round muscle of heifers fed the diet containing 3% dried wormwood. There was a profound effect of the progressively increased intake of dried wormwood led to the linear increase of unsaturated fatty acid content and the linear decrease of saturated fatty acid content in the muscle tissues of Hanwoo heifers. It is concluded that the feeding diets containing dried wormwood substituted for equal weights of rice straw at 5% levels would be anticipated to provide better quality roughage for beef heifer production and economical benefits for beef cattle producers.

Ovarian Follicular Dynamics, Ovarian Follicular Growth, Oocyte Yield, In vitro Embryo Production and Repeated Oocyte Pick Up in Thai Native Heifers Undergoing Superstimulation

  • The objective of this study was to compare the effectiveness of the protocols for superstimulation of follicular growth in Thai native heifers. Heifers (n = 20) were randomly divided into four groups of five heifers/group. Heifers were given a single dose by i.m. administration of 100 mg Follicle Stimulating Hormone dissolved in polyvinylpyrrolidone (FSHp) at 24 h. Ovum pick up (OPU) occurred at 72 h ($F_{24}O_{72}$ protocol; Group 1) or 96 h ($F_{24}O_{96}$ protocol; Group 2), and at 36 h and OPU at 72 h ($F_{36}O_{72}$ protocol; Group 3) or 96 h ($F_{36}O_{96}$ protocol; Group 4) after follicular ablation. The dynamics of ovarian follicular growth were monitored by twice-daily ultrasonographic examinations. Blood sample collections were performed every 12 h after initiation of treatment for assessment of FSH, E2 and P4 profiles. All heifers were subjected to eight repeated sequential sessions of OPU. The follicular deviation commenced $24{\pm}5.32$ h after follicular ablation in all groups. The circulatory FSH surged quickly from 24 to 36 h (>0.8 ng/ml) after follicular ablation and circulatory estrogen levels steadily increased from 36 h until OPU in all groups. At the end of the OPU sessions, the mean number of aspirated follicles/heifer/session in $F_{36}O_{72}$ protocol (Group 3) and $F_{36}O_{96}$ protocol (Group 4) were higher than in the two other groups (p<0.05). The number of cumulus-oocyte complexes (COCs), cleaved and day 8 blastocysts rates in the $F_{36}O_{72}$ protocol (Group 3) were higher than in the other groups (p<0.05). It can be concluded that a single dose i.m. administration of 100 mg FSHp at 36 h and OPU at 72 h after follicular ablation ($F_{36}O_{72}$ protocol; Group 3) was the most effective protocol for superstimulation of follicular growth for repeated OPU and subsequent in vitro embryo production in Thai native heifers.

Study on Ovum Pick-up(OPU) with Finger-Sensibility using Oocyte Recovery in Holstein Heifers (젖소에서 초음파기기를 이용한 난자 채취에 있어서 손가락 촉지를 이용한 난포란의 채란)

  • This study was conducted to develop an improved method for oocyte pick-up(OPU) with finger-sensibility using ultrasound-guidance from ovarian follicles in Holstein heifers. Oocytes were aspirated from ovarian follicles of clear-outline (>2mm), obscure-outline and invisible($\leq$ 2mm) on ultrasound images with 3 different vacuum pressure(40, 80, 120mmHg). Total number of oocytes recovered/follicles were 309/237(130.4%). 113/80(141.3%) and 107/74(144.6%) with 40, 80 and 120 mmHg of vacuum pressure, respectively. Mean number of oocytes recovered was higher in 2 OPU/week (18.3$\pm$5.3) than 1 OPU/week(14.5$\pm$4.1), but this difference was not statistical1y significant. The recovery rates were not affected by the number of OPU as 135.6%(282 oocytes/208 follicles) in 1~20 OPU, 137.7% (168/122) in 21~40 OPU and 148.4%(92/62) in 41~60 OPU, respectively. The proportions of good oocytes (Grades I) recovered were not significantly different by the number of OPU until 40 OPU(12.4% in 1~20 OPU vs 16.7% in 21~40 OPU). However, a significantly(P<0.05) lower recovery rate resulted from more than 40 OPU compared to less than 40 OPU(7.6%). These results imply that more fertilizable oocytes can be produced from invisible-immature follicles by transvaginal aspiration with finger-sensibility from Holstein heifers.

Pituitary and Gonadal Response to GnRH in Prepubertal Buffaloes (Bubalus bubalis)

  • The objective of this study was to investigate the responsiveness of hypophysis and gonads to synthetic GnRH among prepubertal buffalo heifers at 12 months of age. Peripheral plasma FSH, LH, estradiol and progesterone level were measured in blood samples collected at 1 hr before and up to 18 days subsequent to the administration of $200{\mu}g$ GnRH (n=6) or saline (n=6) in Murrah buffalo heifers. The pretreatment peripheral plasma FSH, LH, estradiol and progesterone among GnRH treated heifers were $7.35{\pm}0.45ng/ml$, $1.08{\pm}0.3ng/ml$, $22.93{\pm}1.06pg/ml$ and $0.27{\pm}0.04ng/ml$ respectively. A quick elevation (p < 0.01) of FSH and LH within five min of GnRH administration was observed in all geifers. Although the peak FSH $(89.57{\pm}23.43ng/ml)$ and LH $(7.52{\pm}3.08ng/ml)$ reached by 10 min of GnRH administration, yet the animals differed both in terms of their amplitude response of FSH and LH release as well as in terms of time which animals took to exhiit maximum response to GnRH administration. The GnRH administration did not cause alteration in plasma estradiol and progesterone level. The present study suggests that the pituitary of 12 month buffalo heifers has capacity to synthesize and store of gonadotropin and have developed receptors for GnRH for a spike of gonadotropin release.

Conception Rate of Pabna Cows and Heifers Bred Under Artificial Insemination and Natural Service

  • A study on the conception rate of 885 cows and heifers bred under artificial insemination (AI) and natural service was carried out at AI sub-centers. A high conception rate was observed at first service in all breeding methods. It was revealed that the conception rates in AI with liquid semen (LS) were found to be 25.08, 18.58, 12.69, and 2.48% after 1, 2, 3 and 4 services, respectively. Inseminations of cattle with frozen semen (FS) were found to achieve 42.15, 14.46, 5.85, and 2.46% conception rates after 1, 2, 3, and 4 services, respectively. In natural service, conception rates were found to be 62.45, 6.75, 5.91, and 4.64%, respectively, after 1, 2, 3, and 4 services. Insemination of cows and heifers at mid-cycle had significantly (p < 0.05) higher conception rate as compared to early and late cycles. The existing AI machineries and breeding management need to be improved to achieve higher conception rate of dairy cattle under AI.

EFFECT OF SUPPLEMENTATION WITH PROTEIN MEAL ON THE GROWTH OF CATTLE GIVEN A BASAL DIET OF UNTREATED OR AMMONIATED RICE STRAW

  • A 98 d feeding trial carried out to study liveweight change and rumen metabolites in heifers weighing initially 275 kg and given either untreated or ammoniated rice straw supplemented with 0, 0.4, 0.8 or 1.2 kg protein meal consisting of cottonseed meal (60). All 32 animals received 0.6 kg rice polishings/hd/d and had continuous access to molasses/urea block-licks containing 15% urea. The effects on growth rates of treatment of the straw with ammonia and of supplementation with bypass protein were additive. The heifers fed ammoniated straw grew 267 g/hd/d (p<0.001) faster and consumed 11% (p<0.05) more straw than the heifers on untreated straw. The mean growth response to bypass protein was 0.37 kg gain/kg protein meal supplied. Supplementation with protein meal tended (p=0.06) to depress intake of straw, but straw intakes of the unsupplemented groups were high. Small changes in the composition of the block-licks that were fed throughout the feeding trial led to changes in block intake and in intake of untreated straw. Increasing quantities of protein meal fed were associated with linear increase in concentrations of ammonia (p<0.05) and in molar percentages of iso-butyrate (p<0.01), iso-valerate (p<0.01) and valerate (p<0.01) in the rumen fluid of the heifers on a basal diet of untreated straw. However, in the rumen fluid of the heifers given ammoniated straw, the levels of these metabolities were not affected by the quantity of protein meal given.

Effects of by-product feed-based silage on feeding, rumination, and excretion in growing Hanwoo heifers

  • This study investigated the effects of feeding by-product feed (BF)-based silage on the behavior of growing Hanwoo heifers. Twelve Hanwoo heifers (13.2 months-old, 315 kg body weight; four heifers per pen) were assigned to three diets: a rice straw (RS) diet (concentrate mix and free access to RS), a RS and BF-based silage (RSBFS) diet (concentrate mix and free access to RS and BF-based silage), and a BF-based silage (BFS) diet (concentrate mix and free access to BF-based silage). Behavior was recorded for 5 days using camcorders. Compared to the RS group, the BFS group showed 21.7% higher dry matter intake, shorter feeding, rumination, and chewing times, as well as longer resting time (p < 0.05). Although all groups exhibited similar drinking, urination, and defecation frequencies, the BFS group exhibited higher feeding rates, rumination efficiency, and chewing efficiency than the RS group (p < 0.05). Compared to the BFS group, the RSBFS group showed higher $peNDF_{8.0}$ intake (15.2% vs. 25.0% dry matter intake), longer feeding and sitting times, lower defecation frequency (p < 0.05), and similar rumination efficiency. In conclusion, complete replacement of conventional RS with BF-based silage reduced rumination and chewing activity in growing Hanwoo heifers, and BF-based silage feeding with large-particle straw is an effective approach in improving heifer behavior.

Investigation on Ultrasonographic Observation of Recipient Ovaries, Serum Progesterone Level Measurement and Their Relationship with Pregnancy Rate (수란우의 초음파진단에 의한 난소 상태 및 혈청 Progesterone 농도가 수정란이식 후 수태율에 미치는 영향)

  • The ovaries of 178 Holstein heifers or cows (heifer; 41, 1 parity; 72, 2$\leq$ parity; 65) on Day 6 or 7 (Day 0=day of estrus) were examined by transrectal ultrasonography. Diameter of corpus luteum (CL) and large follicle ( $\geq$ 10 mm), and luteal tissue area were determined by ultrasound system with a 5 MHB rectal probe. Blood samples were taken to progesterone analysis. After selection of recipients, frozen Holstein embryos were thawed and directly transferred to recipients non-surgically. The diameter of CL and luteal tissue area was greater (P<0.01) on Day 7 than on Day 6 in heifers, 1 parity or 2 $\leq$ parity cows, respectively, although progesterone concentrations were not different. The presence of fluid-filled luteal cavitied or multiple CL (2 or more) did not affect serum progesterone concentration. A large follicles were observed in 67.4% of heifers or cows and the average diameter was 14.1 mm. Greater luteal tissue area attributed higher pregnancy in heifers, but not in cows, although there were no difference on pregnancy rate according to progesterone concentration in heifers or cows. The pregnancy rate of recipients contained a large follicle at embryo transfer was lower than that of recipients not contained. These results show ultrasonic assessment of ovaries in Holstein recipients is a reliable tool to determine the follicle and CL for recipient selection.

Effects of Growth Hormone Gene Polymorphism on Lipogenic Gene Expression Levels in Diaphragm Tissues of Japanese Black Heifers

  • Two SNPs, i.e. L127V and T172M, of bovine growth hormone (GH) causing the presence of GH gene haplotypes A, B, and C was previously shown to alter intramuscular fatty acid (FA) composition in Japanese Black (JB) heifers. To determine the SNP effect on somatotropic hormone concentration and lipogenesis, we measured plasma GH, insulin, and insulin-like growth factor-1 (IGF-1) concentrations. We also measured mRNA levels of fatty acid synthase (FASN), stearoyl-coA desaturase (SCD), and sterol regulatory element binding proteins-1 (SREBP-1) and FA composition in diaphragm tissues. Heifers with genotype CC had the lowest plasma insulin concentration and FASN and SCD mRNA levels among genotypes. FASN mRNA levels in haplotype A tended to positively correlate with saturated FA (SFA) content and negatively correlated with C18:2 and unsaturated FA (USFA) contents. SCD mRNA levels in haplotype A positively correlated with monounsaturated FA (MUFA) contents and negatively correlated with C18:0 content. They also tended to positively correlate with C16:1, C18:1, and USFA contents and USFA/SFA ratio and negatively correlate with SFA content. Taken together, GH gene polymorphism affects the lipogenic genes expression levels and their relationships with fatty acid compositions in diaphragm tissues of JB heifers at 31 months of age.
