중금속내성균의 중금속 축적에 미치는 외부요인의 영향 (Effect of External Factors on Heavy Metal Accumulation in the Cell of Heavy Metal-Tolerant Microorganisms)

  • 중금속오염폐수처리에 미생물 이용 가능성을 검토하기 위하여 중금속에 강한 내성을 지님과 동시에 균체내 중금속 축적능력이 우수한 중금속 내성균을 분리하여 온도, pH, 및 균체 전배양시간 등과 같은 외부 요인에 따른 균체내 중금속 축적변화를 조사한 결과는 다음과 같다. 중금속화하물 종류에 따른 균체내 중금속 축적량은 $-nitrate(NO_3)$ group을 가진 중금속화합물에서 약간 높았으나 화합물 종류에 따라서 큰 차이는 없었다. 균체 전배양시간에 따른 중금속 축적은 24시간 전배양한 균체에서 높게 나타났으나 전반적으로 전배양시간에 따라서 큰 차이는 없었다. 중금속 내성균의 균체내 중금속 축적은 온도, pH등 외부환경요인에 영향을 받았으며, 중금속 내성균의 중금속 최적축적 온도는 전반적으로 $20{\sim}37^{\circ}C$ 범위였고, 중금속 최적축적 pH는 전반적으로 $6{\sim}8$ 범위였다. 중금속을 처리한 용액중 중금속 내성균의 균체량을 증가시킬수록 균체내 축적된 총 중금속양은 증가되었으나 균체 단위 g당 중금속 축적량은 감소되었으며, 균체량의 증가에 비례해서 중금속 축적량이 증가되지는 않았다.

  • 중금속오염폐수처리에의 미생물 이용 가능성을 검토하기 위하여 중금속에 강한 내성을 지님과 동시에 균체내 중금속 축적능력이 우수한 중금속 내성균을 분리하여 competing ion, 대사저해제 등과 같은 외부 요인에 따른 균체내 중금속 축적변화를 조사한 결과는 다음과 같다. 중금속을 처리한 용액중 양이온인 $Al^{3+}$이온과 음이온인 $CO_3\;^{2-}$$PO_4\;^{2-}$이온이 competing ion으로 존재할 경우 중금속 내성균의 균체내 중금속 축적은 크게 감소되었으나, 그외 다른 양이온들과 음이온들에 대해서는 거의 영향을 받지 않았다. Cd, Zn 및 Cu 내성균주의 Cd, Zn 및 Cu 축적은 대사저해제에 의하여 크게 감소되었으나, Pb 내성균주에 의한 Pb 축적은 거의 영향을 받지 않는 것으로 나타나 Cd, Zn 및 Cu 내성균주의 Cd, Zn 및 Cu 축적은 에너지 의존적인 과정으로서 물질대사와 관련되어 있는 것으로 생각되었으며, Pb 내성균주의 Pb 축적은 에너지 비의존적인 과정으로서 물질대사와 관련이 별로 없는 물리적인 과정에 의하여 축적되는 것으로 생각되었다. 각 중금속 내성균주들은 해당 중금속 축적능력에 비하여 낮았으나 타 중금속들의 축적능력도 있었으며, 여러가지 중금속들을 동일 농도로 복합처리 하였을 경우 해당 중금속들의 축적능력이 타 중금속들에 비하여 매우 높게 나타나 각 중금속 내성균주들의 해당 중금속 축적은 선택적인 과정에 의하여 축적되는 것으로 생각되었다.

광덕산 식물체의 중금속 함량에 관한 연구 (A Study of Heavy Metal Contents in plants from Mt. Kwang-Duk Area)

  • Heavy metal accumulation in living organisms through food-wed can give serious damage on physiological responses for vital activities. The initiation of heavy metal supposed to begin from the bio-accumulation of plants. To establish basic data fur heavy metal contents in plants at the area without artificial contamination, both woody and herb plants at Mt. Kwang-Duk were studied. The content of heavy metals in various organs of plants were analyzed by ICP. The range of heavy metals in plants for Al, As, Cd, Co, Cr, Cu, Fe. Mn, Pb, Se and Zn were 1.019∼257.200ppm, O∼2.929ppm, 0∼0.079ppm, 0∼0.054ppm, 0.023∼3.007ppm, 0∼1.997ppm, 2.031∼148.500ppm, 1.069∼51.320ppm, O∼126.900ppm, 0.708∼4.927ppm and 0.846∼4.949ppm, respectively. The amount of heavy metals in plants are much less than that of soil except some species. In woody plants, it was detected that the metal contents of leaves were higher than that of stems especially in case of Al and Fe with statistical significance. There were significant differences between shoots and roots of herb plants in metal content of Al, Co and Fe. Those metals have more accumulated in roots comparing with shoots. Some species of plants had shown the difference tendency of heavy metal accumulation. Generally, most species had not exceeded over twice of mean value each other, and had various difference according to the kinds of heavy metals.

물리화학적 전처리에 의한 중금속 내성세균의 균체내 중금속 축적 변화 (Heavy Metal Accumulation in Cell of Heavy Metal-Tolerant Bacteria by Some Physical and Chemical Treatments)

  • 광산폐수, 산업폐수등으로 부터 Cd, Pb, Zn 및 Cu등의 중금속에 강한 내성을 지니고 있을 뿐만 아니라 균체내 중금속 축적능력이 우수한 중금속 내성 미생물 균주 Pseudomonas putida(Cd), Pseudomonas aeruginosa(Pb), Pseudomonas chlororaphis(Zn) 및 Pseudomonas stutzeri(Cu)를 각각 분리하여, 여러가지 물리화학적인 방법으로 세포를 전처리하여 세포구성성분을 인위적으로 조절한 후 세포내 중금속이온의 흡수 거동 및 조단백질 함량과 중금속 축적관계를 조사한 결과는 다음과 같다. 세포를 알카리로 전처리하였을 경우 세포내 중금속 축적은 매우 감소되었으며, 메탄올과 클로포름으로 전처리하였을 경우에는 중금속 축적에 큰 영향을 미치지 않았으나, 메탄올과 클로로포름으로 전처리한 후 다시 알카리로 재차 처리하였을 경우에는 중금속 축적이 매우 감소되었다. 전처리된 세포내 중금속 축적은 용출되지 않고 남아있는 조단백질 함량이 감소됨에 따라 더 얼마나 크게 감소되었으므로 세포 구성물질중 단백질이 중금속 축적에 중요한 역할을 하는 물질인 것으로 판단되었다.

공장지역 도로변 야생식물들의 중금속 축적 (Heavy Metal Accumulation in Wild Plants on the Roadside of Industrial Areas)

  • This study was conducted to investigate the heavy metal accumulation in wild plants. The samples from 5 plant species on 14 sites were taken on the roadside where urban and industrial regions in northern Gyeonggi do. Artemisia princeps var. orientalis, Chenopodium album L. var. centrobrum, Erigeron canadensis L., Rumex crispus L. and Taraxacum platycapum H. Dahlst were taken from different parts of the plants gathered in surveyed sites for analysing. This study classified which species were suitable to accumulators, excluders, or indicators which were provided as Phytoremediation for heavy metals(Cd, Cr, Ni, Pb and Zn) and considered the applicability of that work. 1. The mean accumulation of 6 heavy metals in plants was as follows; Zn> Cu> Cr> Pb> Ni> Cd in rows except for Ni and Pb in Erigeron. 2. The highest heavy metal accumulators were Taraxacum and Artemisia. 3. The comparison of heavy metal accumulations between the tops and roots in the plants was as follows; Zn> Cd> Pb> Ni> Cu> Cr. 4. Comparison of heavy metal accumulations between tops and roots, root parts were higher than the top parts in most of the plants. Especially, in Taraxacum, a ratio was over 1.0 in Cr, Ni, and Zn. Therefore, Taraxacum was an accumulator for 3 heavy metals. And Erigeron, Chenopodium, Rumex, Artemisia were good for excluders which had lower than 1.0.

Level of Heavy Metals in the Onsan Bay in Korea and Involvement of Metal Binding Protein in the Accumulation of Cadmium in Littorina brevicula

  • The heavy metal concentrations in seawater and winkles (Littorina brevicula) collected from the Onsan bay area in southeast of Korea were analyzed. The heavy metal concentrations in the seawater obtained from the most polluted site showed approximately 189. 205. and 110 fold higher cadmium, copper. and zinc concentrations, respectively. than the uncontaminated control site. The contamination levels of these metals in winkles were 11.08 - 2.35, 334.5 - 212.5, and 426.0 - 499.2$\mu\textrm{g}$ per gram dry body weight. respectively. The concentrations of all three metals in both the seawater and winkles decreased gradually with increasing distance from Daejeong stream, suggesting the stream being the major source of heavy metal input into the bay. Among the four body parts of digestive gland and gonad. gill. kidney, and remaining tissue in contaminated winkles, kidney showed the highest accumulation level of cadmium: copper and zinc, however. were more or less distributed among the four body parts. Upon gel filtration chromatography of the cytosol from the kidney of cadmium induced winkles, one cadmium peak corresponded to the elution peak of horse kidney metallothionein.

울산 달천광산 인근의 식물체 중금속 축적에 관한 연구

  • This study was investigated the contents of heavy metals in plants at Dalcheun mine area, in order to offer basic data for phytoremedation on soil contamination by heavy metals. As the results of this study, the contents of Ni, As, Cd in plants were the highest at the west area of Dalcheun mine area. But the content of Zn was similar in all of Dalcheun mine area. For the herbaceous plants, the contents of heavy metals were higher in plants which were included in Gramineae and Compositae than any other families. And then the contents of Ni, Zn, Cd in root were higher $1.4{\sim}1.8$ times than stem, especially the content of As was higher 4 times. For the trees, the contents of heavy metals were usually high in Zanthoxylum piperitum, Pine tree, Chestnut tree, Quercus acutissima and Lindera obtusiloba. The contents of heavy metals with parts of tree were almost same range. All experimental result, we paid attention on two conclusions. First, the contents of heavy metals in herbs was higher than in trees. And second, if there were some spices that the content of heavy metal in leave were higher than in root, they could accumulate that kind of heavy metal. Therefore, It is above 2 times which the content of heavy metal in the leave is higher than in the root about herbs, that kind of plants was regarded as a accumulation species. According as the heavy metals, accumulation species were selected. We think that the accumulation species are able to absorb the heavy metals from the soil, they will make use of phytoremedation on contaminated soil by heavy metals.

Foliar Transfer of Dust and Heavy Metals on Roadside Plants in a Subtropical Environment

  • In this study, the contents of dust and associated heavy metals on roadside plants were investigated to assess their foliar transfer. The study was conducted at six different locations (four roadside and two industrial) near an industrial area in Bilaspur (Chhattisgarh), India. Six metals (Fe, Mn, Pb, Cu, Cr, and Cd) were examined in this study. The concentrations of heavy metals in foliar dust were found to be in the order of Fe>Mn>Pb>Cu>Cr>Cd. However, this relative order changed in the case of leaf concentrations to Fe>Mn>Cd>Cu>Pb>Cr. The metal concentrations in the dust and leaves can be attributed mainly to industrial and vehicular emissions. In contrast to other metals, Cd showed significant accumulation in the leaves compared to the respective dust samples. This study showed different patterns in the distributions of heavy metals between the dust deposited on the leaves and the metal accumulated in the leaves. These results suggest that the dust retention and heavy metal accumulation in native plant species should be explored in an attempt to manage these hazardous metallic elements.

  • This study was performed to investigate the effects of peking-duck extracts added with medicinal herbs(DJ) on the intoxication of heavy metals (Hg, Pb, Cd, As) in rats. Sprague-Dawley rats weighing 150$\pm$15g, were randomly assigned to groups: basal diet only in normal control group(NCG): basal diet and heavy metals without DJ injection in heavy metal control group(HMC); basal diet, heavy metals and DJ(3mg/ml) injection in heavy metal low duck-juice group(HMLD), basal diet, heavy metals and DJ(30mg/ml) injection in heavy metal middle duck-juice group(HMMD): basal diet, heavy metal and DJ(300mg/ml) injection in heavy metal high duck-juice group(HMHD). Hg and As was injected by 50ppm and Cd and Pb by 25ppm for 17days. Also DJ oral feeding was conducted for 28days. The result of this study were as follows: Food intake and body weight gain in heavy metal administered groups were lower than those of NCG. Liver, kidney and testis weights were not significantly different among 5 groups. GOT, GPT and BUN activities were significantly reduced in DJ treated groups as compared to HMC. DJ showed the suppressing effect on the accumulation of Hg, Pb and Cd in serum, liver and kidney. Fecal Hg and Cd excretions increased with DJ feeding. The results suggested that DJ may have some protective effects on Hg, Cd and Pb intoxication by reducing the accumulation in tissues and increasing excretion. This study also showed the effective way of using duck-extract and its application to the oriental medicine.
