Change of Fibrinolytic and Antioxidative Activities of Codonopsis lanceolata According to Various Storage Conditions, and Heat or Salt Treatments

  • We investigated the changes of fibrinolytic and antioxidative activities of wild or cultivated Codonopsis lanceolata, which were packed in woven polypropylene (WP) film or low density polyethylene (LDPE) film and stored for 0, 15, and 30 days at refrigerated $(2\~4^{\circ}C)$ or room $(l8\~20^{\circ}C)$ temperature (So we have 16 samples, and wrote them as the abbreviated words such as W-WP-RE-15, W-WP-RO-15, W-LDPE-RE-15, W-LDPE-RO-15, W-WP-RE-30, W-WP-RO-30, W-LDPE-RE-30, W-LDPE-RO-30, C-WP-RE-15, C-WP-RO-15, C-LDPE-RE-15, C-LDPE-RO-15, C-WP-RE-30, C-WP-RO-30, C-LDPE-RE-30, C-LDPE-RO-30). Fibrinolytic activity of fresh Codonopsis lanceolata cultivated in the mountain or field were 0.8 unit (plasmin unit/ml) or 7.3 units, respectively. In descending order, the activities of wild Codonopsis lanceolata stored for 15 days were as followed; LDPE-RE (0.70 unit), WP-RO (0.52 unit), WP-RE (0.45 unit), and LDPE-RO (0.30 unit). After 30 days, fibrinolytic activities of them decreased to 0.47 unit (LDPE-RE), 0.28 unit (WP-RO), 0.21 unit (WP-RE), and 0.30 unit (LDPE-RO). Considering from the point of fibrinolytic activity, the optimal storage condition of wild Codonopsis lanceolata was packing with LDPE film and storing at $4^{\circ}C$. The change of fibrinolytic activities of Codonopsis lanceolata cultivated in the field revealed the similar trend as wild samples, but the fibrinolytic activities of 30 days-stored samples were maintained better than the wild Codonopsis lanceolata. Fibinolytic activity of wild Codonopsis lancealata was increased by heating for 5 min at $100^{\circ}C$ and decreased by addition of NaCl. Antioxidative activities of Codonopsis lanceolata were also compared from the electron donating activity. Fresh Codonopsis lanceolata had about $70\%$ of electron donating activity. Independent of cultivation area, electron donating activity dropped to $19\~74\%$ (wild Codonopsis lanceolata) and $27\~59\%$ (cultivated Codonopsis lanceolata) during 15 days storage. But after 30 days storage, we obtained the unexpected results, which meant that the activities were higher activities than 15 days-stored samples or even though the fresh samples. In general, Codonopsis lanceolata could maintain antioxidative activities most strongly with LDPE film and chilled condition.

Characteristics of Al-Pillared Clay Synthesized from Bentonite and the Adsorption Properties for Phosphate Ion (벤토나이트로부터 합성한 Al-층간가교점토의 특성과 인산이온의 흡착성)

  • Al-pillared clay was synthesized by constructing pillars of aluminum oxides at the interlayer of montmorillonite in bentonite. XRD, DTA and chemical analyses of Al-pillared clay were performed to examine mineralogical properties. Batch adsorption experiments were also conducted to determine the adsorption properties of this synthesized clay for phosphate ions. XRD analyses showed that the interlayer space of Al-pillared clay expanded to 18.03 $\AA$ at room temperature and shifted to $17 \AA$ after heating to $550^{\circ}C$. A small change in interlayer space after heating indicates high thermal stability. The interlayer expansion by glycerol was also very small. From DTA analyses, pillared clay showed the characteristic endothermic peaks at 270 and $420^{\circ}C$ , which might be caused by dehydration in framework of pillars between interlayers. Adsorption experiment revealed that Al-pillared clay had an excellent adsorption capacity to the phosphate ions, whereas montmorillonite had very low adsorption capacity to phosphate ions. In phosphate solution concentration up to 300 mg/L, 2 g of pillared clay could uptake almost 100% of phosphate ions from 20 mL of solution. After heat treatment of the phosphate adsorbed pillared clay at 50$0^{\circ}C$ to remove phosphate, the calcined pillared clay could adsorb phosphate ions with a little decreased adsorption efficiency. This fact indicates that Al-pillared clay can be recycled for the adsorption of phosphate ions.

Properties and Activities of Nireogenase System of Azospirillum amazonensa Kp1 (Azospirillum amazonense Kp1의 질소고정효소계의 활성 및 특성)

  • The maximum nitrogen fixation activity of the associative, microaerobic and acid tolerant bacteria, Azospirillum amazonense Kp1 was obtained with 0.2Kpa of $O_{2}$ and showed a reversible inhibition by the higher concentrations. Ammonium treatment caused a gradual inhibition of the activity up to 350mM. The nitrogenase systems were purified by gradient chromatography on DEAE-52 cellulose, heat treatment and preparative PAGE. The MoFe protein showed molecular weight of 210,000 including two nonidentical subunits with apparent molecular weights of 55,000 and 50,000 and an isoelectricpoint of 5.2 and contained 2, 24 and 28 atoms of Mo, Fe and acid labile S per molecule. The Fe protein revealed molecular weight of 66,000 including two types of subunits with molecular weights of 35,000 and 31,000 and an isoelectric point of 4.6, and contained 4 atoms of Fe and 6 atoms of S per molecule. The maximum specific nitrogenase activity attained 2,200 and 1,700nM $C_2H_4mg^{-1} min^{-1}$, respectively for MoFe and Fe proteins at pH7 and $35^{\circ}C$. The activity was lost after 10 and 30 days under the cold room ($4^{\circ}C$) condition for Fe and MoFe proteins, respectively.

Evaluation for Mechanical Properties of High Strength Concrete at High Temperature by Stressed Test and Unstressed Test (설계하중 사전재하 및 비재하방식에 의한 고강도콘크리트의 고온특성 평가)

  • Recently, the effects of high temperature on compressive strength, elastic modulus and strain at peak stress of high strength concrete were experimentally investigated. The present study is aimed to study the effect of elevated temperatures ranging from 20 to 700 on the material mechanical properties of high strength concrete of 40, 60, 80 MPa grade. In this study, the types of test were the stressed test and stressed residual test that the specimens are subjected to a 25% of ultimate compressive strength at room temperature and sustained during heating and when target temperature is reached, the specimens are loaded to failure. And another specimens are loaded to failure after 24 hour cooling time. Tests were conducted at various temperatures ($20{\sim}700^{\circ}C$) for concretes made with W/B ratios 46%, 32% and 25%. Test results showed that the relative values of compressive strength and elastic modulus decreased with increasing compressive strength grade of specimen and the axial strain at peak stress were influenced by the load before heating. Thermal strain of concrete at high temperature was affected by the preload level as well as the compressive strength. Finally, model equation for compressive strength and elastic modulus of heated high strength concrete proposed by result of this study.

Establishment of location-base service(LBS) disaster risk prediction system in deteriorated areas (위치기반(LBS) 쇠퇴지역 재난재해 위험성 예측 시스템 구축)

  • This study uses beacons and smartphone Global Positioning System (GPS) receivers to establish a location-based disaster/hazard prediction system. Beacons are usually installed indoors to locate users using triangulation in the room, but this study is differentiated from previous studies because the system is used outdoors to collect information on registration location and temperature and humidity in hazardous areas. In addition, since it is installed outdoors, waterproof, dehumidifying, and dustproof functions in the beacons themselves are required, and in case of heat and humidity, the sensor must be exposed to the outside, so the waterproof function is supplemented with a separate container. Based on these functions, information on declining and vulnerable areas is identified in real time, and temperature/humidity information is collected. We also propose a system that provides weather and fine-dust information for the area concerned. User location data are acquired through beacons and smartphone GPS receivers, and when users transmit from declining or vulnerable areas, they can establish the data to identify dangerous areas. In addition, temperature/humidity data in a microspace can be collected and utilized to build data to cope with climate change. Data can be used to identify specific areas of decline in a microspace, and various analyses can be made through the accumulated data.

Characteristics of ${\beta}-galactosidase$ activity in Lactobacillus plantarum from kimchi (김치에서 분리한 유산균 Lactobacillus plantarum의 ${\beta}-galactosidase$ 특성에 관한 연구)

  • To investigate a possible application of three strains of lactic acid bacteria(strain No. 49. No. 61. No. 75) from kimchi in milk fermentation industry, the optimal condition for production of intracellular ${\beta}-galactosidase$ from Lactobacillus(L.) plantarum and its enzymatic properties were examined. The preferable carbon source of the medium for strain No. 49 in production of ${\beta}-galactosidase$ was MRS broth with 1.0% lactose instead of dextrose of pH 65. for strain No. 75 with 1.0% galactose and for strain No. 61 with 3.0% lactose at pH 7.5, respectively. The maximum enzyme production from strain No. 49, No. 75 was observed after 48 hours culture at $30^{\circ}C$ in a medium containing the appropriate carbon source, from strain No. 61 after 48 hours culture at room temperature. The optimum temperature for ${\beta}-galactosidase$ activity from L. plantarum was $60^{\circ}C$ for strain No. 49, $37^{\circ}C$ for strain No. 61 and $50^{\circ}C$ for strain No. 75, respectively. The heat stability of enzyme activities for all three strains remained 90% at $45^{\circ}C$. The optimal pH was pH 6.5 and enzyme activities were most stable at pH for all three bacteria.

Apparent and standardized ileal nutrient digestibility of broiler diets containing varying levels of raw full-fat soybean and microbial protease

  • Background: Although soybean meal (SBM) is excellent source of protein in diets for poultry, it is sometimes inaccessible, costly and fluctuates in supply. The SBM can partially be replaced by full-fat SBM, but the meals prepared from raw full-fat soybean contain antinutritional factors. To avoid the risk of antinutritional factors, heat treatment is always advisable, but either excessive or under heating the soybean could negatively affect the quality. However, the potential for further improvement of SBM by supplementing with microbial enzymes has been suggested by many researchers. The objective of this study was to evaluate the performance and ileal nutrient digestibility of birds fed on diets containing raw soybeans and supplemented with microbial protease. Methods: A $3{\times}2$ factorial, involving 3 levels of raw full-fat soybean (RFFS; 0, 45 or 75 g/kg of diet) and 2 levels of protease (0 or 15,000 PROT/kg) was used. The birds were raised in a climate-controlled room. A nitrogen-free diet was also offered to a reference group from day 19 to 24 to determine protein and amino acid flow at the terminal ileum and calculate the standardized ileal digestibility of nutrients. On days 10, 24 and 35, body weight and feed leftover were recorded to calculate the body weight gain (BWG), feed intake (FI) and feed conversion ratio (FCR). On day 24, samples of ileal digesta were collected at least from two birds per replicate. Results: When RFFS was increased from 0 to 75 g/kg of diet, the content of trypsin inhibitors was increased from 1747 to 10,193 trypsin inhibitors unit (TIU)/g of diets, and feed consumption of birds was also reduced (P < 0.05). Increasing RFFS level reduced the BWG from hatch 0 to 10 d (P < 0.01) and hatch to 24 d (P < 0.05). The BWG of birds from hatch to 35 was not significantly (P = 0.07) affected. Feed intake was also reduced (P < 0.05) during 0 to 35 d. However, protease supplementation improved (P < 0.05) the BWG and FCR during 0 to 24 d. Rising levels of RFFS increased the weight of pancreas (P < 0.001) and small intestine (P < 0.001) at day 24. Except for methionine, apparent and the corresponding standardized ileal digestibility of CP and AA were reduced (P < 0.01) by increasing levels of RFFS in diets. Conclusion: This study showed that some commercial SBM could be replaced by RFFS in broiler diets, without markedly compromising productivity. The AID and SID of CP and lysine were slightly improved by dietary supplementation of microbial protease.

Electrical properties of a resistive SFCL with shunt resistor (분로저항을 가진 저항형 초전도 한류기의 전기적 특성)

  • We fabricated a resistive SFCL having a shunt resistor parallel to it in order to bifurcate the transient current at faults. The SFCL consists of a YBCO film coated with an Au layer (10 ${\omega}$ at room temperature), which is to disperse the heat generated at hot spots in the YBCO film, and the 5 ${\omega}$ shunt resistor. The minimum quench current of the SFCL was found to be 12.2 A$_{peak}$. This SFCL successfully controlled the fault current below 23 A$_{peak}$ which is otherwise to increase up to 113 A$_{peak}$. Bifurcation of the current resulted in the temperature rise of the YBCO/Au film 3 times slower than without the shunt, protecting the SFCL at high currents.

  • As a fundamental study to control Alopeuclus aequalis Sobol. var. amurensis(Kom.) Ohwi., on dominant weed in barley and dry seeded rice cultivation on paddy fields, several factors which would have closer relations to its germination and emergence, were examined. The results are as follows. The dormancy of seeds were broken at dry-heat treatment of $30^{\circ}C$. The germination rate of the seeds was high in order of 15>10>20> $5^{\circ}C$ and was very low at more than $25^{\circ}C$. The emergence of A. aequalis was influenced little for the light, but had a tendency to be good on the dark condition. The seeds dried on room temperature germinated few but them treated on $30^{\circ}C$ for 24 hours were germinated over 80%. The A. aequalis occured few in the saline soil of salt concentration of more than 0.25% and the germination rate of seeds was very low on the over 0.5% solution of NaCl. On the solution of pH 6.0~12.0, the germination was not effected for pH but was very few on pH 2.0. At the paddy-lowland which have cultivated the soybean for three years A. aequalis emerged a few.

  • As a carbon-free, green growth alternative, internal and external interest in hydrogen energy and technology is growing. Hydrogen was added to co-axial methane, methane-propane, and methane-propane-ethane diffusion flames, which are the main ingredients of LNG, to evaluate its effect on flame formation and combustion products. The variation in combustion products produced by adding hydrogen gradually to diffusion pyrolysis at room temperature and normal pressure conditions was observed experimentally by using a gas analyzer, and the shape of diffusion pyrolysis was observed step by step using a digital camera. The experimental results showed that the production volume of nitrogen oxides tended to increase and became close to linear as hydrogen was added to the diffusion pyrotechnic. This is because the relatively high temperature of heat insulation and fast combustion speed of hydrogen facilitated the production of thermal NOx. On the other hand, CO2 production tended to decrease as hydrogen was added to reduce the overall carbon ratio contained in the mixed diffusion flame of methane, methane-propane, and methane-ethane-propane. This means that the mixed fuel use of LNG-hydrogen in ships may potentially reduce emissions of CO2, a greenhouse gas.
