Arsenic처리에 따른 흰쥐 혈관의 수축과 heat shock protein 70과의 관계 (Relation between Expression of Heat Shock Protein 70 and Vascular Contractility of Rat Aorta Treated with Arsenic)

  • 외부에서 투여된 열자극, 알콜 및 생리적 염과 같은 환경 스트레스는 체내 각 부위에서 스트레스단백질(열자극단백질, HSP)을 생성하게 된다. 본 연구에서는 비소가 흰쥐 대동맥의 수축에 미치는 영향을 조사하기 위해 스트레스단백질의 발현과 대동맥의 수축력의 변화와 이들과의 관계를 알아보고자 실험을 실시하였다 적출한 혈관은 organ bath에 담가 0, 0.5, 1, 2,및 4 mM As를 처리한 후 1, 3, 및 8시간 뒤에 KCI(55 mM)에 대한 수축반응과 HSP 발현을 각각 생리기록계와 western blotting 을 통해 분석하였다. 비소(4 mM)를 처리한 혈관의 KCI에 대한 수축력은 처리 초기(1, 3시간)에는 효과가 없었고 처리 8시간째 대조군에 비해 39%로 유의적인 증가를 보였다. 또한 비소처리로 혈관의 스트레스단백질 HSP 70의 생성은 비소 처리농도에 따라 증가되었고, 비소 처리 초기에는 변화가 없었으며 비소처리 8시간째 HSP생성이 촉진되었다. HSP는 주로 혈관 평활근세포에서 현저하게 발현되었고 일부 내피세포에서도 발현되었다. 이상의 결과에서 비소 처리에 따른 HSP 70의 발현과 혈관의 수축 증가의 상승되는 현상이 비소처리 8시간째에 유의하게 증가되는 것으로 보아, 비소처리에 의해 혈관의 스트레스단백질 HSP 발현이 증가되고 이로 인해 혈관 수축력을 상승시켜 고혈압 유발과정에 관여하는 것으로 여겨진다.

비소세포 폐암 환자 조직에서 Hsp90α, Hsp90β, GRP94의 발현과 임상병리학적 특성과의 상관관계 분석 (Analysis of the Correlation between Expressions of HSP90α, HSP90β, and GRP94, and the Clinicopathologic Characteristics in Tissues of Non-Small Cell Lung Cancer Patients)

  • 열충격 단백질(heat shock proteins, HSPs)은 다양한 종양에서 과발현 되고, 종양이 형성되는 과정이나 그 예후에 영향을 주며, 분자량에 따라 HSP27, HSP60, HSP90, HSP100 등으로 구분한다. Heat shock protein90은 세포 내 불안정한 단백질을 보호하는 역할을 통해 질병의 유지에 기여하는데, 정상 조직에 비해 종양 세포에서 높은 수준으로 발현된다고 보고되었다. 이에 본 연구에서는 우리나라 사망원인 1위인 폐암 중 비소세포 폐암에서 Heat shock protein90 family 발현과 비소세포 폐암환자의 임상적 특징과의 상관관계를 분석하여 종양의 생물학적 표지자로서의 가능성을 조사하였다. HSP90 family의 발현과 임상병리학적 특성 및 생존율과의 상관관계를 분석한 결과 $HSP90{\alpha}$는 림프혈관강 침윤이 되지 않은 환자에서 높은 발현을 보였고(p=0.014), $HSP90{\beta}$ 은 조직학적 형태에서 편평상피세포 암종에서 높은 발현을 보였으며(p=0.003), GRP94 은 분화도가 낮을수록 높은 발현을 나타내었다(p=0.048). 생존율은 $HSP90{\alpha}$, $HSP90{\beta}$, GRP94 모두 발현 차이에 대한 유의성이 없었다. 본 연구를 통해 miRNA-126, miRNA-155, miRNA-200c의 발현은 비소세포 폐암의 진단을 위한 생물학적 표지자 및 예후 인자로서 사용될 수 있을 것으로 사료된다. 그리고 비소세포 폐암의 치료용으로 HSP90 family가 고려되어야 할 것이며, GRP94가 종양의 예후예측을 위한 중요한 인자라 사료된다.

A role of carboxy-terminal region of Toxoplasma gondii-heat shock protein 70 in enhancement of T. gondii infection in mice

  • We investigated the role of recombinant Toxoplasma gondii heat shock protein (rT.g.HSP) 70-full length, rT.g. HSP70-NH2-terminal region, or rT.g. HSP70-carboxy-terminal region in prophylactic immunity in C57BL/6 mice perorally infected with Fukaya cysts of T. gondii. At 3, 4, 5, and 6 weeks after infection, the number of T gondii in the brain tissue of each mouse was measured by quantitative competitive-polymerase chain reaction (QC-PCR) targeting the surface antigen (SAG) 1 gene. Immunization with rT.g.HSP70-full length or rT.g.HSP70-carboxy-terminal region increased the number of T. gondii in the brain tissue after T. gondii infection, whereas immunization with rT.g.HSP70-NHa-terminal region did not. These results suggest that T.g. HSP70-carboxy-terminal region as well as T.g.HSP70-full length may induce deleterious effects on the protective immunity of mice infected with a cyst-forming T. gondii strain, Fukaya.

  • Bacterial heat shock protein has been one of the components that are responsible to induce autoimmune disease mechanisms in the pathogenesis of atherosclerosis due to high level of homology in sequence with human counterpart. This mechanism may explain how bacterial infectious disease, such as periodontal disease, might contribute to the acceleration of the disease process of atherosclerosis. Porphyromonas gingivalis which is a major periodontal pathogenic bacterial species, has been implicated as one of the pathogenic bacteria playing the role in this context. The present study has been performed to evaluate the anti-atherosclerotic effect of adoptive transfer of Porphyromonas gingivalis heat shock protein epitope-specific T cell lines into severe combined immunodeficiency (SCID) mice. Peptide no. 15 with amino acid sequence VKEVASKTND-specific T cell line was selected for the transfer. When experimental atherosclerosis was induced in SCID mice adoptively transferred either by the T cell lines (experimental group) or by non-specific mouse T cells (control group), there was no significant difference in the severity and extent of the atherosclerosis induced by hypercholesterol diet.

Identification of Genes Encoding Heat Shock Protein 40 Family and the Functional Characterization of Two Hsp40s, MHF16 and MHF21, in Magnaporthe oryzae

  • Magnaporthe oryzae, the causal agent of the rice blast disease, poses a worldwide threat to stable rice production. The large-scale functional characterization of genes controlling the pathogenicity of M. oryzae is currently under way, but little is known about heat shock protein 40 (Hsp40) function in the rice blast fungus or any other filamentous plant pathogen. We identified 25 genes encoding putative Hsp40s in the genome of M. oryzae using a bioinformatic approach, which we designated M. oryzae heat shock protein forty (MHF 1-25). To elucidate the roles of these genes, we characterized the functions of MHF16 and MHF21, which encode type ill and type n Hsp40 proteins, respectively. MHF16 and MHF21 expression was not significantly induced by heat shock, but it was down-regulated by cold shock. Knockout mutants of these genes $({\Delta}$mhf16 and ${\Delta}$mhf21) were viable, but conidiation was severely reduced. Moreover, sectoring was observed in the ${\Delta}mhf16$ mutant when it was grown on oatmeal agar medium. Conidial germination, appressorium formation, and pathogenicity in rice were not significantly affected in the mutants. The defects in conidiation and colony morphology were fully complemented by reintroduction of wild type MHF16 and MHF21 alleles, respectively. These data indicate that MHF16 and MHF21 play important roles in conidiation in the rice blast fungus.

HSP70 and HSC70 gene Expression in Chironomus Tentans (Diptera, Chironomidae) larvae Exposed to Various Environmental Pollutants: Potential Biomarker for Environmental Monitoring

  • In order to identify potential biomarkers of environmental monitoring, we evaluated heat shock genes expressions as effects of various environmental pollutants (nonylphenol, bisphenol-A, 17a­ethynyl estradiol, bis(2-ethylhexyl)phthalate, endosulfan, paraquat dichloride, chloropyriphos, fenitrothion, cadmium chloride, lead nitrate, potassium dichromate, benzo[a]pyrene and carbon tetrachloride) on larvae of aquatic midge Chironomus tentans (Diptera, Chironomidae). Heat shock protein 70 gene expression increased in most of chemicals treated larvae compared to control. The response was rapid and sensitive to low chemical concentrations but not stressor specific. In conjunction with stressor specific biomarkers, heat shock protein 70 gene expression in Chironomus might be developed for assessing exposure to environmental stressors in the fresh water ecosystem. Considering the potential of Chironomus larvae as biomonitoring species, heat shock gene expression has a considerable potential as a sensitive biomarker for environmental monitoring in Chironomus.

Effect of BIS depletion on HSF1-dependent transcriptional activation in A549 non-small cell lung cancer cells

  • The expression of BCL-2 interacting cell death suppressor (BIS), an anti-stress or anti-apoptotic protein, has been shown to be regulated at the transcriptional level by heat shock factor 1 (HSF1) upon various stresses. Recently, HSF1 was also shown to bind to BIS, but the significance of these protein-protein interactions on HSF1 activity has not been fully defined. In the present study, we observed that complete depletion of BIS using a CRISPR/Cas9 system in A549 non-small cell lung cancer did not affect the induction of heat shock protein (HSP) 70 and HSP27 mRNAs under various stress conditions such as heat shock, proteotoxic stress, and oxidative stress. The lack of a functional association of BIS with HSF1 activity was also demonstrated by transient downregulation of BIS by siRNA in A549 and U87 glioblastoma cells. Endogenous BIS mRNA levels were significantly suppressed in BIS knockout (KO) A549 cells compared to BIS wild type (WT) A549 cells at the constitutive and inducible levels. The promoter activities of BIS and HSP70 as well as the degradation rate of BIS mRNA were not influenced by depletion of BIS. In addition, the expression levels of the mutant BIS construct, in which 14 bp were deleted as in BIS-KO A549 cells, were not different from those of the WT BIS construct, indicating that mRNA stability was not the mechanism for autoregulation of BIS. Our results suggested that BIS was not required for HSF1 activity, but was required for its own expression, which involved an HSF1-independent pathway.

  • Mitochondrial heat shock protein 75 (mtHSP75) is a member of the HSP90 family and plays essential roles in refolding proteins of the mitochondrial matrix. Mitochondria provide energy in the form of ATP and generate reactive oxygen species (ROS). Heat shock proteins (HSPs) are activated in response to stress, and protect cells. In this study, we characterized the mtHSP75 of the big-belly seahorse Hippocampus abdominalis. The protein (BsmtHSP75) is encoded by an open reading frame (ORF) of 2,157 nucleotides, has 719 amino acids (aa), and is of molecular mass 82 kDa. BsmtHSP75 has two functional domains, a histidine kinase-like ATPase (HATPase_c) domain (123-276 aa) and an HSP90 family domain (302-718 aa). BsmtHSP75 was expressed in all tested tissues of healthy seahorses. The ovary contained the highest transcription level, followed (in order) by the blood, brain, and muscle. Pouch tissue showed the lowest expression level. The expression of BsmtHSP75 was significantly (P<0.05) up-regulated on viral or bacterial challenge, suggesting that BsmtHSP75 plays a role in the immune defense against bacterial and viral pathogens.

Molecular Chaperonic Function of C-Reactive Protein Induced by Heating in HT-29 Human Colon Carcinoma Cells

  • The effects of heat shock, or all-trans retinoic acid, on the expression of the C-reactive protein mRNA in the HT-29 human colon carcinoma cells, as well as the functional role of the C-reactive protein as a molecular chaperone, were studied. The expression level of the C-reactive protein mRNA in the HT-29 cells was increased time-dependently when exposed to heat-shock, and dose-dependently when treated with all-trans retinoic acid. The activities of transglutaminase C and K in the HT-29 cells were significantly increased when treated with all-trans retinoic acid. The C-reactive protein prevented thermal aggregation of the citrate synthase and stabilized the target enzyme, citrate synthase. The C-reactive protein promoted functional refolding of the urea-denatured citrate synthase up to 40-70%. These results suggest that the C-reactive protein, which is induced in human colon carcinoma cells, when heated or treated with all-trans retinoic acid has in a part functional activity of the molecular chaperone.

The Heat Shock Protein 27 (Hsp27) Operates Predominantly by Blocking the Mitochondrial-Independent/Extrinsic Pathway of Cellular Apoptosis

  • Heat shock protein 27 (Hsp27) is a molecular chaperone protein which regulates cell apoptosis by interacting directly with the caspase activation components in the apoptotic pathways. With the assistance of the Tat protein transduction domain we directly delivered the Hsp27 into the myocardial cell line, H9c2 and demonstrate that this protein can reverse hypoxia-induced apoptosis of cells. In order to characterize the contribution of Hsp27 in blocking the two major apoptotic pathways operational within cells, we exposed H9c2 cells to staurosporine and cobalt chloride, agents that induce mitochondria-dependent (intrinsic) and -independent (extrinsic) pathways of apoptosis in cells respectively. The Tat-Hsp27 fusion protein showed a greater propensity to inhibit the effect induced by the cobalt chloride treatment. These data suggest that the Hsp27 predominantly exerts its protective effect by interfering with the components of the extrinsic pathway of apoptosis.
