Headspace (HS) and headspace solid-phase microextraction (HS-SPME) were studied for extracting volatile organic compounds (VOCs) from whole blood, with chemical and instrumental variables being optimized for maximum sensitivity: incubation at $60^{\circ}C$, equilibration for 30 min, pH 11, and 2 mL injection volume. Both techniques provided accurate analyses, with detection limits of 0.05-0.1 ng $mL^{-1}$ and 0.05-0.5 ng $mL^{-1}$. HS showed better sensitivity, reproducibility, and analysis times than HS-SPME. Overall levels of chloroform in whole blood were found to be 0.05-5.84 ng $mL^{-1}$; detected levels of benzene were 0.05-2.20 ng $mL^{-1}$.
국내 주요 시판 복분자주 24종을 수거하여 시료의 휘발성 향기성분을 HS-SPME법에 의해 추출한 후, GC-MS를 이용하여 분석 동정하였고 이들 성분의 정량 분석도 실시하였다. 총 43개의 성분이 동정되었고, 화학적 특성에 따라 분류하면 15 esters, 3 ketones, 7 alcohols, 2 aldehydes, 12 terpene류, 4 acids 성분이 동정되었다. ethyl acetate, ethyl octanoate, isoamyl alcohol, phenethyl alcohol이 동정된 성분중 높은 농도를 차지하였고, 그 외 ethyl hexanoate, ethyl decanoate, benzadehyde, dL-limonene, linalool, alpha-terpineol, myrtenol, acetic acid가 주요 성분으로 나타났다. 주성분 분석 결과 향기성분 구성 및 함량에 따라 제조지역별 차이를 나타내지 않았다. 복분자주에 사용되는 복분자 품종이 대개 북미산인 Rubus occidentalis L.으로 동일한 품종이 전국에 재배됨에 따라 향기성분 구성의 차이가 크지 않은 것으로 여겨진다. 또한 복분자주 제조과정의 특성상 발효 후 제성과정을 통해 발효 원주의 함량과 성분에 변화가 생기므로 생산지역보다는 제조공정 및 방법에 더 영향을 받는 것으로 여겨진다. 향후 생산지역별 제조시 사용되는 복분자에 대한 좀 더 자세한 품종 및 재배 특성에 대한 연구와 제조공정에 따른 이화학적 성분 분석이 필요하다. 그러나 향기성분 분포의 경우, 주요 발효산물인 ester, alcohol, acid 성분은 대개 1 사분면에 분포하고, 복분자 과실에서 유래하는 terpene류의 경우 4 사분면에 모두 분포하여 이들 성분간의 차이를 나타내었다. 향후 복분자주 품질관련 주요 지표물질로 활용이 가능하리라 여겨진다.

Array-based gas sensors now offer the potential of a robust analytical approach to odor measurement for medical use. We are developing a fast reliable method for detection of microbial infection by monitoring the headspace from the infected wound. In this paper, we present initial results obtained from wound-state monitoring for burn patients using an electronic nose incorporating an automated solid-phase microextraction (SPME) desorption system to enable the system to be used for clinical validation. SPME preconcentration is used for sampling of the headspace air and the response of the sensor module to variable concentrations of volatiles emitted from SPME fiber is evaluated. Gas chromatography-mass spectrometry studies prove that living bacteria, the typical infectious agents in clinical practice, can be distinguished from each other by means of a limited set of key volatile products. Principal component analysis results give the first indication that infected patients may be distinguished from uninfected patients. Microbial laboratory analysis using clinical samples verifies the performance of the system.
HS-SPME GC-MS에 의한 지표수 중에 37개 휘발성유기물질을 동시에 분석하는 방법을 개발하였다. 이 방법은 물 중 휘발성유기물질을 헤드스페이스 vial에서 $40 ^{\circ}C$에서 30분간 가열 85 ${\mu}m$ carboxen-polydimethylsiloxane에 흡착시키는 방법이다. 4.0 mL의 물 시료를 사용하여 확립한 조건하에서 정량한계는 4.1-96 ng/L의 범위를 나타냈고 이러한 검출한계는 US EPA 또는 독일에서 확립한 준거치의 1/10 이하 값을 만족시켰으며 0.05 ${\mu}g/L$와 0.50 ${\mu}g/L$의 농도에서 정밀도는 15% 이내를 보였다. 이 정량한계는 준거치를 결정하기 위한 조사연구에서 요구하는 최소 감도이다. 확립한 방법을 사용하여 금강물 16개 시료를 분석한 결과 16개 휘발성유기화합물이 검출되었으며 측정값은 US EPA 또는 독일에서 확립한 준거치 이하의 값을 보였다.
D-glucose-glycine 계를 사용하여 Maillard 반응에 의하여 생성되는 저분자 휘발성성분을 비롯하여 amide 화합물의 생성 및 그 기구를 검토하였다. 그 결과, 동정된 저분자 휘발성성분 중에서, headspace gas 중의 휘발성성분은 furan, acetone, 2-methylfuran, 2,5-dimethylfuran 2-butanone 2,3-pentanedione, diacetyl 등이었다, 이 중에서 diacetyl의 생성량이 가장 많아, 전 peak 면적의 약 $70\%$를 차지하였다. 또한, 에테르 추출물중의 주요반응생성물은 초산, furfuryl alcohol 2,5-dimethylpyrrole 2-acetylpyrrole 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one 등이었고, ethyl acetate 로 추출한 산성획분중에는 N-acetyl-glycine와 N-methylacetamide 등 2종류의 amide 화합물의 생성이 밝혀졌다. 이들 amide 화합물적 생성기구를 밝히기 위하여, Mailiard 반응초기생성물인 diacetyl 및 glyoxal을 각각 butylamine과 반응시킨 결과, Schiff 염기의 산화적 분해로 N-butylacetamide 및 N-butylformamide의 생성이 인정되었다. 따라서 N-acetylglycine 및 N-methylacetamide는 glucosylamine의 2,3-enol 화 및 ${\beta}-elimination$에 의한 탈수의 진행으로 생성된 dicarbonyl 화합물이 glycine과 반응하여 Schiff 염기를 형성하고, 이 Schiff 염기가 산화적분해를 받아서 N-acetylglycine이 생성되고, N-methylacetamide는 N-acetylglycine의 탈탄산에 의해서 생성된다고 생각한다.

Production of good Koji primarily depends upon the selection of substrate materials and fermentative microflora, which together influence the characteristic flavor and aroma. Herein, we performed comparative metabolomic analyses of volatile organic compounds (VOCs) and primary metabolites for Koji samples fermented individually with Bacillus amyloliquefaciens and Aspergillus oryzae. The VOCs and primary metabolites were analyzed using headspace solid phase microextraction (HS-SPME) followed by gas chromatography time-of-flight mass spectrometry (GC-TOF-MS). In particular, alcohols, ketones, and furans were mainly detected in Bacillus-fermented Koji (Bacillus Koji, BK), potentially due to the increased levels of lipid oxidation. A cheesy and rancid flavor was characteristic of Bacillus Koji, which is attributable to high content of typical 'off-flavor' compounds. Furthermore, the umami taste engendered by 2-methoxyphenol, (E,E)-2,4-decadienal, and glutamic acid was primarily detected in Bacillus Koji. Alternatively, malty flavor compounds (2-methylpropanal, 2-methylbutanal, 3-methylbutanal) and sweet flavor compounds (monosaccharides and maltol) were relatively abundant in Aspergillus-fermented Koji (Aspergillus Koji, AK). Hence, we argue that the VOC profile of Koji is largely determined by the rational choice of inocula, which modifies the primary metabolomes in Koji substrates, potentially shaping its volatolome as well as the aroma characteristics.
Two different extraction procedures, SDE and SPME, were employed to determine a comprehensive and efficient fragrance profile of Korean Rosa hybrida. Both extraction methods could compensate for each other, covering compounds with diverse boiling point, polarities, and chemical properties. A total of 46 compounds were identified in Mi-hyang. The identified compounds were composed of 17 alcohols, 14 carbonyls, 7 aliphatic hydrocarbons, 2 terpene hydrocarbons, 4 benzenes, 1 ester, and 1 miscellaneous compound. Quantitatively, carbonyls($12.96{\sim}21.79%$ in essential oils of SDE and $2.89{\sim}8.44%$ in SPME headspace) and alcohols($7.98{\sim}11.73%$ in essential oils of SDE and $3.39{\sim}17.35%$ in SPME headspace) were dominant in Mi-hyang's volatiles.
Headspace solid phase trapping solvent extraction (HS-SPTE) and GC-MS was applied for the characterization of volatile flavors from fennel, anise seed, star-anise, dill seed, fennel bean, and Ricard aperitif liquor. Tenax was used for HS-SPTE adsorption material. Recoveries, precision, linear dynamic ranges, and the limit of detection in the analytical method were validated. There were some similarities and distinct differences between fennel-like samples. The Korean and the Chinese fennels contained trans-anethole, (+)-limonene, anisealdehyde, methyl chavicol as major components. The volatile aroma components from star anise were characterised by rich trans-anethole, (+)-limonene, methyl chavicol, and anisaldehyde. Additionally, principal component analysis (PCA) has been used for characterizing or classifying eight different fennel-like samples according to origin or other features. A quite different pattern of dill seed was found due to the presence of apiol (dill).
수용액 시료 중 휘발성 지방산을 HS (headspace)-SPME (Solid phase microextraction)를 이용하여 간편하고 빠르게 분석하는 방법에 대하여 연구하였다. 1-Pyrenyldiazomethane (PDAM)을 이용한 화이버 내 유도체화를 통하여 휘발성 지방산의 검출감도를 향상시킬 수 있었으며, SPME 추출조건으로 pH, 염 효과 및 초음파 추출에 대하여 조사하였다. 본 연구에서 개발된 방법을 기반으로 실제 폐수 중 휘발성 지방산을 추출하여 기체크로마토그래피/질량분석기-선택이온검색법으로 정량 분석하였다.
Volatile flavor compounds from milk were analyzed and identified by using the analysis methods of headspace solid phase microextraction gas chromatography/mass spectrometry (HSPME-GC/MS) and electronic nose (E-Nose) system. About 30 volatile compounds were identified by HSPME-GC/MS for the fresh and off-flavor milk samples. Also, the correlation between rancidity and ageing days of milk was obtained by the aid of principal component analysis algorithm. It shows that the E-Nose system can identify the various types of milk flavor. These results imply that the analysis method based on the E-nose system can apply to the quality control of milk flavor and the rancidity.
