Effects of the Artificial Shrinkage and Assisted Hatching Before Vitrification on the Development of the Vitrified Mouse Expanding Blastocysts (유리화동결 전 인공수축과 보조부화술이 융해 후 생쥐 포배아의 발달에 미치는 영향)

  • Objective: This study was conducted to investigate the effects of the artificial shrinkage and assisted hatching (PZD; patial zona dissetion) before vitrification on the development of vitrified mouse expanding blastocyst. Methods: Mouse 2-cell embryos were collected and cultured in G1.1 and G2.2 to expanding blastocyst. For artificial shrinkage (AS) the micro injection pipette was inserted into blastocoele cavity and blastocoele fluid was aspirated. For assisted hatching (AH) PZD method was used. Control group was -AS/-AH and treatment groups were -AS/+AH, +AS/-AH and +AS/+AH. After AS and AH mouse blastocysts were equillibrated in G10 and G10E20 for 3 mins, respectively, and vitrified in G25E25 after loading on capped pulled-straw. Vitrified mouse blastocysts were thawed and cultured for 24 hrs. The survival and hatching rate was compared among one control and three treatment groups. Results: The survival rates were 99%, 92% in +AS/+AH and +AS/-AH groups and 54%, 58% in -AS/-AH and -AS/+AH group, respectively. The survival rate was significantly higher in +AS group than in -AS group (p<0.01). Hatching rates were 34%, 96% in -AS/-AH and -AS/+AH groups and 41%, 100% in +AS/-AH and +AS/+AH, respectively. The hatching rates was higher in +AH group than in -AH group (p<0.01). After thawing recovery rates were 100%. Loading on capped pulled-straw, that is effective and useful method on vitrification. Conclusion: This study showed that artificial shrinkage of blastocoele cavity and assisted hatching (PZD) significantly improved the development of the vitrified mouse expanding blastocysts.

Effects of Dimethyl-sulfoxide on Sperm Cryopreservation of Grass Carp (Ctenopharyngodon idellus)

  • This study was conducted to investigate the effects of three different concentrations (6%, 8% and 10% final volume) of dimethyl sulfoxide (DMSO) on cryopreserved sperm of grass carp (Ctenopharyngodon idellus). Grass carp sperm was suspended in Kurokura extender #2 and equilibrated at $4^{\circ}C$ for 10 min. French straws (0.25 ml) of sperm were frozen from $4^{\circ}C\;to\;-4^{\circ}C$ at a rate of $4^{\circ}C\;min^{-1}$ and then ken $-4^{\circ}C\;to\;-80^{\circ}C$ at a rate of $11^{\circ}C\;min^{-1}$. The straws were kept at $-80^{\circ}C$ for 10 min and finally stored in liquid nitrogen $(-196^{\circ}C)$. The cryopreserved sperm was thawed in a water bath at $40^{\circ}C$ for 30 sec and fertilization, hatching rate and larval malformation were compared with fresh sperm (control). The fertilization rate of post-thawed sperm was comparable (from 88.21% to 94.30%) to that of fresh sperm. However, hatching rate of all frozen sperm were significantly lower (P<0.05) than that of control. Additionally, the larval abnormality rate of frozen sperm was significantly higher than that of fresh sperm. The results indicate that DMSO could affect the quality of cryopreserved sperm of grass carp, and a freezing program and a proper extender composition should be further studied.

Effects of IGF-I and EGF Supplemented to PZM3 Culture Medium on the Development of Porcine Embryos In vitro

  • This study investigated the effects of IGF-I and EGF on the development of blastocysts or hatched blastocysts during the in vitro culture of embryos from immature porcine oocytes. After the in vitro maturation and fertilization of cumulus-oocyte complexes (COCs) and their culture in vitro in PZM3 medium, we examined the embryo development rate for 168 h. When different concentrations of IGF-I (0, 1, 10, 20 ng/ml) were supplemented to fertilized porcine embryos in vitro, there were no significant differences in cleavage rate, blastocyst development rate or blastocyst hatching rate among the treated groups. On the other hand, when different concentrations of EGF (0, 1, 10, 20 ng/ml) were supplemented to the in vitro culture medium, blastocyst development rate was highest in the group in which EGF was not supplemented and, specifically, it was higher than in the 20 ng/ml treatment group (p<0.05). When 10 ng/ml IGF-I and 1 ng/ml EGF were supplemented separately or simultaneously, there were no significant differences among the treated groups in blastocyst hatching rate and the number of cells in each condition. This study demonstrated that the addition of IGF-I and EGF into PZM3 medium did not enhance development of the blastocyst stage and total cell number in blastocysts.

Enhanced Hatching Rate of Bovine IVM/IVF/IVC Blastocyst by Laser Drilling (Laser Drilling 처리를 받은 체외생산된 소 배반포기배의 부화율 제고)

  • 본 실험은 체외 생산된 소 배반포기배에 대한 laser drilling 처리가 배의 부화율에 미치는 영향을 조사하고자 실시하였다. 그 결과는 다음과 같다. 소 수정란의 체외 발달율을 조사하였던 바, 82.3%의 난할율( 2-세포기)과 체외수정 후 배양 7일째에 32.6%의 배반포 발달율을 나타내었다. 이렇게 생산된 배반포기배에 laser drilling 효과를 조사하였던 바, 처리 후 24시간째의 부화진행율(90.0%)은 대조군(44.4%)보다 유의하게 높게 나타났다(p<0.0001). 또한, 처리 후 48시간째의 부화율(68.0%)도 대조군(33.3%)보다 유의하게 높게 나타났다. 이러한 결과는 laser drilling이 체외 생산된 소 배반포기배의 부화진행율과 부화율을 유의하게 증가시킬 수 있다는 것을 알 수 있었다(p<0.001).

A Study on Survival Ability of Refrozen-Thawed Bovine IVF Embryos (한우 체외수정란의 재동결에 관한 연구)

  • This study was conducted to investigate the survival and hatching rates after refrozen-thawed bovine IVF blastocysts. The survival rates after refrozen-thawed bovine IVF blastocysts produced on day 7, day 8 and day 9, were 66.6%(16/24), 62.5%(15/24) and 65.3%(17/26), respectively. The survival and hatching rates after the first frozen-thawed bovine JVF blastocysts were 90.0%(27 /30) and 70.0%(21 /30), but in refrozen-thawed bovine IVF blastocysts were 66.2%(49 /74) and 45.9%(34 /74), respectively. The results of this study were suggest that refrozen-thawed bovine IVF embryos had survival ability.

Exposure to Triclosan Induces Mortality through Oxidative Stress and DNA Damage in the Java Medaka Oryzias javanicus

  • To understand the detrimental effects of triclosan on Java medaka (Oryzias javanicus) embryos, fertilized embryos were exposed to different concentrations (1, 10, 50, 100, 200, 400, 600, 800, and 1,000 ㎍ l-1) of triclosan until hatching. Then, we examined the survival rate and developmental parameters as well as alterations in antioxidant constituents and DNA damage markers. The results showed dose-dependent mortality, hatching delays, and developmental abnormalities in the embryos. Additionally, there were significant increases in oxidative stress parameters and antioxidant responses, along with elevated DNA damage. These findings suggest that sublethal concentrations of triclosan induce toxic effects through oxidative stress on Java medaka embryos, as evidenced by changes in in vivo parameters and biochemical constituents.

Mode of Transmission of a Newly Discovered Microsporidian and Its Effect on Fecundity and Hatching in Silkworm, Bombyx mori L.

  • The mode of transmission, effect on fecundity, hatching and tissues specificity of a microsporidian $(Lb_{ms})$ recovered from Lamerin breed of the silkworm Bombyx mori L. was studied and compared with standard strain Nosema bombycis. Peroral inoculation of $Lb_{ms}$ or N. bombycis to zeroday of $4^{th}$ instar larvae of silkworm was the most suitable method for producing information on development of stage specific mortality, pupation and obtaining infected adults for transovarial transmission studies. It was observed that pupal mortality, the percentage of moths emerged and the percentage of moths infected were significantly high in N. bombycis infected batches as compared $(Lb_{ms})$ in all the three tested breeds of the silkworm. However no significant difference was observed in larval mortality. The fecundity and hatchability was not affected significantly in $(Lb_{ms})$ infected adults, however significant reduction in egg production, fecundity, hatchability and increased egg retention was observed in mother moths infected with N. bombycis. The $(Lb_{ms})$ is transmitted both horizontally and vertically at lower rate due to its low rate of proliferation. The trans ovarial transmission of $(Lb_{ms})$ to the $F_1$ progeny generation through eggs averaged only $61.33\pm5.10\%$ whereas N. bombycis was transmitted at $100\%$. The $(Lb_{ms})$ had low oral infectivity and low transovarial transmission in silkworm B. mori.

Studies on the teratogenicity of folpet in the developing chick embryo (계태아를 이용한 농약(folpet)의 기형독성 연구)

  • A teratogenicity test of 'folpet' was carried out in the developing chick embryos to investigate and validate the safety of rural environmental hazardous materials. Folpet was administered to chick embryos' yolk sac at a rate of 0.1mg and 0.01mg per SPF eggs at 96 hours of incubation. The morphological changes were examined. Fertility ratio of SPF eggs used was 94.9%. Hatching rate of untreated control group was 74.4% and the group dosed with 100ul of corn oil into the yolk sac was 70.0%. The $LD_{50}$ of folpet was 0.663mg/100ul/egg. After hatching, mean body weight, body length, claw length and beak length of high and low dose administered groups were not significantly different from untreated and vehicle control group. There was no abnormal appearence in all the groups. Therefore it seems that, within the doses applied, folpet dose not induce potential teratogenicity in the developing chick embryos.

Cyclopamine, an Antagonist of Hedgehog (Hh) Signaling Pathway, Reduces the Hatching Rate of Parthenogenetic Murine Embryos

  • Hedgehog (Hh) pathway plays a key role in development from invertebrate to vertebrate. It is known to be involved in cell differentiation, polarity, proliferation, including the development of vertebrate limb and the establishment of flies' body plan. To investigate how the regulation of Hh pathway affects the development of parthenogenetic murine embryos, the parthenogenetically activated murine embryos were treated with either cyclopamine (Cyc), an antagonist of Hh pathway, or purmorphamine, an agonist of Hh pathway. While Cyc did not affect the blastocyst formation and its total cell number, the chemical reduced the hatching rate of embryos and the expression levels of Fn1 mRNA. The results of the present study show the possibility that Cyc may affect the development of embryos at blastocyst stage by blocking Hh pathway and this may cause detrimental effect to the embryos at peri-, and post-implantation stages.

Effect of Cow Manure and Saw Dust as the Additive Materials for Efficient Vermistabilization

  • The influence of additive materials including saw dust and cow manure in the ripening of leather sludge for the efficient vermisabilization was studied. The ripening characteristics of the leather sludge, the growth of earthworms in the growth bed and the by-products like cast were observed according to the mixing ratio of additive materials. When the mixed leather sludge of which the additive materials contents were over 20% were ripened for 50 days, the values of ORP and alkalinity were changed to the favorable ranges for earthworms, the positive (+) and below 1,000 mg/l as $CaCO_3$, respectively. The velocity of ripening improving the characteristics of the leather sludge was faster in the sludge mixed with cow manure than saw dust. This results could also be verified from the monitoring of the states of earthworm growth using the survival rate, the increase rate of lifeweight and the hatching rate of earthworm cocoon in the their growth bed packed with the ripened sludge. The values of CEC in cast, the vermi-stabilized sludge, were increased with the mixing ratio of additive materials, but the content of heavy metals was drcreased by the dilution effect and accumulation in the earthworm body. Above results show that the leather sludge might be efficiently stabilized by earthworms through the ripening for 50 days using additive materials. Cow manure can be used as a good additive materials for leather sludge as much as saw dust, and the optimum content of additive materials in the well-ripened leather sludge was about 30%.
