Detection of Porcine Stress Syndrome from Genomic DNA of Hair Follicle by PCR-RFLP in Breeding Pig (종돈의 모근 Genomic DNA를 이용한 스트레스 증후군 검색)

  • This study was carried out to investigate PSS (Porcine Stress Syndrome) with the PSE (Pale, Soft, Exudative) in 319 different pigs(Yorkshire 150; Landrace 89 and Duroc 80). The PCR-RFLP method was adapted to detect the ryanodine receptor (RYR 1) gene mutation and to estimate the genotype frequency of the RYR1 gene in breeding pig population. The DNA samples were collected from hair follicles of pigs of Yorkshire, Landrace and Duroc. After DNA amplification by PCR, the PCR products were digested by restriction enzyme, Cfo I. Primary PCR products of ryanodine receptor gene were length of 659 bp in hair follicle and their second PCR products were length of 522 bp in hair follicle. The exon region (522 bp) including point mutation ($C \arrow T; Arg \arrow Cys$) in the porcine ryanodine receptor gene, which is a causal mutation for PSS, was digested with Cfo I restriction enzyme. The RYR1 gene was classifed into three genotypes by agarose gel electrophoresis. The normal homozygous (NN) individuals showed two DNA fragments consisted of 439 and 83 bp. The mutant homozygous (nn) individuals showed only one DNA fragment 522 bp. In addition, all three fragments (522, 439 and 83 bp) were showed in heterozygous (Nn) carrier animals. The normal homozygous (NN), heterozygous (Nn) and mutant homozygous (nn) were 98.00, 2.00 and 0.00% in Yorkshire pigs, 87.64, 11.24 and 1.12% in Landrace, 100.00, 0.00 and 0.00% in Duroc, respectively. The gene frequencies of N and n were 0.990 and 0.010 in Yorkshire pigs, 0.933 and 0.067 in Landrace, 1.000 and 0.000 in Duroc, respectively.

  • The origin of natural planting, which is getting the spotlight in modern time facing natural and environmental problems, can be found from wild gardens. They were started by William Robinson and concretely embodied by Gertrude Jekyll. It is worth shedding new light on wild gardens, as they served as a pathbreaker for ecological design and an important foundation for the specialization of naturalism, which are part of the most important topics in modern gardens. This study aimed to infer the conceptual model of wild gardens and identify their historic significance by comparatively analyzing Robinson's Gravetye Manor and Jekyll's Munstead Wood. The results are: Firstly, they inherited inspirations for spatial organization from basic cottage gardens and introduced informal forms. Secondly, in terms of the use of materials, they had observed various climates in their journeys so that they could use both native and naturalized plants based on their understanding of the plants' hardiness and exotic species. They also displayed interests in plants in the woodlands and forests. Thirdly, in terms of design techniques, they investigated the colors and textures of individual plants and their relationships to produce a variety of views that resembled nature in microcosm. Fourthly, in terms of maintenance, their basic orientation was the minimum maintenance to allow plants to live according to their nature.

Genetic Variation of Pinus densiflora Populations in South Korea Based on ESTP Markers (ESTP 표지를 이용한 국내 소나무 집단의 유전변이)

  • Genetic diversity and genetic differentiation of thirteen Pinus densiflora populations in South Korea were estimated using nine ESTP (Expressed Sequence Tag Polymorphism) markers. The numbers of allele and the effective allele were 2.2 and 1.8, respectively. The percentage of polymorphic loci (P) was 98.8%. The observed and the expected heterozygosity were 0.391 and 0.402, respectively, and the eleven populations except for Ahngang and Gangneung population were under Hardy-Weinberg equilibrium state. The level of genetic differentiation (Wright’s FST = 0.057) was higher than those of isozyme or nSSR markers. We could not find out any relationship between the genetic distance and geographic distribution among populations from cluster analysis. Also, the genetic differentiation between populations was not correlated with the geographic distance (r = 0.017 and P = 0.344 from Mantel test). From the result of FST-outlier analysis to identify a locus under selection, six loci were detected at confidence interval of 99% by the frequentist’s method. However, only three loci (sams2+AluⅠ, sams2+RsaⅠ, PtNCS_p14A9+HaeⅢ) were presumed as outliers by Bayesian method. The sams2+AluⅠ and sams2+RsaⅠlocus were originated from the sams2 gene and seemed to be the loci under balancing selection.

Analysis of factors on the asymbiotic germination of white lady's slipper orchid(Cypripedium macranthos Sw. albiflorum) (멸종위기 흰복주머니란 종자발아에 미치는 요인 분석)

  • Cypripediums, popularly called lady's slippers or moccasin flowers, are the showiest and most sought after hardy terrestrial orchids, collected and grown by orchid and alpine plant enthusiasts alike. In Korea, 4 species of cypripedium are reported as Cypripedium japonicum, C. macranthos, C. guttatum, and C. calceolus. We had already reported the feasibilities of C. macranthos and C. guttatum with in vitro germination methods from immature seeds. The seeds of white lady's slipper orchid (Cypripedium macranthos Sw. alba) were collected 65 days after pollination in 2018. The green pods were sterilized with flame and sowed immediately on the POM(Phytomax orchid maintenance media(R), Sigma) supplemented with BAP 0, 0.5, 1.0 mg/L and NAA 0, 1, 2mg/L. The germination of seed was observed 90 days after sowing, and the plantlets were subcultured to the same media according to the size of the protocorm with 1~2, 2~3, 3~4, 5~6, 7~8mm. The time of the subculture to the new media seems to be critical factors of forming rhizoids which is the hairy root of the cypripediums. As a results, the protorms of the white lady's slipper orchid was successfully germinated in the POM media supplemented BAP 0.5 and NAA 1.0 mg/L. The roots and rhizoids were formed in 5~6mm protocorms subculture over 95% survival ratio. We also tried to subculture to liquid medium without activated charcoal, however the browning or malformation of the roots was observed in the root. The formation of shoots from the protocorm was effectively enhanced in the POM media with non-additives of plant growth regulators. These results indicate the possibility of high and stable production and practical industrialization of endangered white lady's slipper orchids.

Agrometeorological Analysis on the Freeze Damage Occurrence of Yuzu Trees in Goheung, Jeonnam Province in 2018 (2018년 전라남도 고흥 유자나무 동해 발생에 대한 기상학적 구명)

  • In 2018, severe diebacks have occurred on yuzu trees cultivated in Goheung, Jeonnam Province. On-farm surveys at 18 randomly selected orchards revealed the dieback incidence ranged from 7.5% to 100% with an average of 43.6%, and 56.6% of the affected yuzu trees were eventually killed. In order to find the reason for this sudden epidemic, we investigated the weather conditions that are exclusively distinct from previous years, hypothesizing that certain weather extremes might have caused the dieback epidemic on yuzu trees. Since different temperatures can cause freeze damage to plants depending on their dormancy stages, we investigated both periods when yuzu becomes hardy under deep dormancy (January-February) and when yuzu loses its cold hardiness (March-April). First, we found that daily minimum air temperatures below $-10^{\circ}C$ were recorded for 7 days in Goheung for January and February in 2018, while no occasions in 2017. In particular, there were two extreme temperature drops ($-12.6^{\circ}C$ and $-11.5^{\circ}C$) beyond the yuzu cold hardiness limit in 2018. In addition, another occasion of two sudden temperature drops to nearly $0^{\circ}C$ were occurred right after abnormally-warm-temperature-rises to $13^{\circ}C$ of daily minimum air temperatures in mid-March and early April. In conclusion, we estimated that the possible damages by several extreme freeze events during the winter of 2018 could be a major cause of severe diebacks and subsequently killed the severely affected yuzu trees.

Association analysis of polymorphisms of G protein-coupled receptor 54 gene exons with reproductive traits in Jiaxing Black sows

  • Objective: The aim of this study was to detect single nucleotide polymorphisms (SNP) of G protein-coupled receptor 54 (GPR54) gene and explore association of this candidate gene with reproductive traits in Jiaxing Black sows. Methods: Six pairs of primers of the gene were designed to amplify all exons thus sequences of which were detected by means of direct sequencing and then SNP loci were scanned. The effects of SNPs on total number of piglets born (TNB), number of piglets born alive (NBA), number of still born piglets (NSB), and litter weight at birth (LWB) of Jiaxing Black sows were analyzed. Results: Three SNP loci, including T3739C, C3878T and T6789C, were identified via comparison of sequencing and two genotypes (AB, BB) at each SNP site were observed. T3739C resulted in the change of amino acid ($Leu{\rightarrow}Pro$) in corresponding protein, and C3878T resulted in synonymous mutation ($Ile{\rightarrow}Ile$). Statistical results demonstrated that allele B was the preponderant allele at the three SNP loci and Genotype BB was the preponderant genotype. Meanwhile, Chi-Square test of these three SNPs indicated that all mutation sites fitted in Hardy-Weinberg equilibrium (p>0.05). For GPR54-T3739C locus, Jiaxing Black sows with genotype BB had 1.23 TNB and 1.28 NBA (p<0.01) that were more than those with genotype AB, respectively. Jiaxing Black sows that had the first two parities with genotype BB had additional 2.23 TNB, 2.27 NBA (p<0.01), and 1.94 LWB (p<0.05) compared to those with genotype AB, respectively. However, for other two loci, no significant difference was found between TNB, NBA, NSB, and LWB, and different genotypes of Jiaxing Black sows. Conclusion: In conclusion, the polymorphisms of GPR54-T3739C locus were significantly associated to TNB, NBA, and LWB and could be used as a potential genetic marker to improve reproductive function of Jiaxing black sows.

Association of polymorphisms in bone morphogenetic protein receptor-1B gene exon-9 with litter size in Dorset, Mongolian, and Small Tail Han ewes

  • Objective: The present study was to investigate the association of polymorphisms in exon-9 of the bone morphogenetic protein receptor-1B (BMPR-1B) gene (C864T) with litter size in 240 Dorset, 232 Mongolian, and 124 Small Tail Han ewes. Methods: Blood samples were collected from 596 ewes and genomic DNA was extracted using the phenol: chloroform extraction method. The 304-bp amplified polymerase chain reaction product was analyzed for polymorphism by single-strand conformation polymorphism method. The genotypic frequency and allele frequency of BMPR-1B gene exon-9 were computed after sequence alignment. The ${\chi}^2$ independence test was used to analyze the association of genotypic frequency and litter size traits with in each ewe breed, where the phenotype was directly treated as category. Results: The results indicated two different banding patterns AA and AB for this fragment, with the most frequent genotype and allele of AA and A. Calculated Chi-square test for BMPR-1B gene exon-9 was found to be more than that of p value at the 5% level of significance, indicating that the population under study was in Hardy-Weinberg equilibrium for all ewes. The ${\chi}^2$ independence test analyses indicated litter size differences between genotypes was not the same for each breed. The 304-bp nucleotide sequence was subjected to BLAST analysis, and the C864T mutation significantly affected litter size in singletons, twins and multiples. The heterozygosity in exon-9 of BMPR-1B gene could increase litter size for all the studied ewes. Conclusion: Consequently, it appears that the polymorphism BMPR-1B gene exon-9 detected in this study may have potential use in marker assisted selection for litter size in Dorset, Mongolian, and Small Tail Han ewes.

Investigation of mulberry farm's soil properties and mulberry leaf nutritive components in local areas of Korea

  • Mulberry is a hardy, perennial, deep-rooted plant capable of thriving under diverse agroclimatic conditions. The selection of suitable land and appropriate variety can help the sustainable mulberry field. However, no conclusive and comprehensive investigation has been conducted on the mulberry soil properties and nutritional composition of mulberry cultivars from Korea local areas in previous studies. In our study, soil properties and mulberry leaf components of Korea local mulberry farms were briefly investigated. In result, the soil organic matter (OM) content was significantly high in Buan (6.81%) and Jangseong (6.14%). In contrast, available phosphate (P2O5) was different in each local area. To investigate relationship between soil property and nutritive component of mulberry leaf, Cheongil leaf samples from 8 local areas were analyzed. Among the macrominerals (K+, Ca2+, Na+, and Mg2+), the concentration of K varied from (1884±9.36) mg/100 g to (2685±11.5) mg/100 g. The potassium (K+) of Cheongil leaf in Sangju was the highest at 2685 mg/100 g. Besides macrominerals, flavonoids, total dietary fiber contents and moisture of Cheongil leaf samples were studied in the 8 local areas. In terms of these contents, the variation was largely depending on the local areas. This study provides a possible industrial use of mulberry, and holds promise to enhance the overall profitability of sericulture.

Association of CAPN10 gene (rs3842570) polymorphism with the type 2 diabetes mellitus among the population of Noakhali region in Bangladesh: a case-control study

  • Type 2 diabetes mellitus (T2DM) is a multifactorial, polygenic, and metabolically complicated disease. A large number of genes are responsible for the biogenesis of T2DM and calpain10 (CAPN10) is one of them. The association of numerous CAPN10 genetic polymorphisms in the development of T2DM has been widely studied in different populations and noticed inconclusive results. The present study is an attempt to evaluate the plausible association of CAPN10 polymorphism SNP-19 (rs3842570) with T2DM and T2DM-related anthropometric and metabolic traits in the Noakhali region of Bangladesh. This case-control study included 202 T2DM patients and 75 healthy individuals from different places in Noakhali. A significant association (p < 0.05) of SNP-19 with T2DM in co-dominant 2R/3R vs. 3R/3R (odds ratio [OR], 2.7; p=0.0014) and dominant (2R/3R) + (2R/2R) vs. 3R/3R (OR, 2.47; p=0.0011) genetic models was observed. High-risk allele 2R also showed a significant association with T2DM in the allelic model (OR, 1.67; p=0.0109). The genotypic frequency of SNP-19 variants showed consistency with Hardy-Weinberg equilibrium (p > 0.05). Additionally, SNP-19 genetic variants showed potential associations with the anthropometric and metabolic traits of T2DM patients in terms of body mass index, systolic blood pressure, diastolic blood pressure, total cholesterol, and triglycerides. Our approach identifies the 2R/3R genotype of SNP-19 as a significant risk factor for biogenesis of T2DM in the Noakhali population. Furthermore, a large-scale study could be instrumental to correlate this finding in overall Bangladeshi population.

Fruit Characteristics of New Cultivar 'Autumn sense' of Hardy Kiwi (Actinidia arguta) by Stem Pruning (전정에 의한 신품종 다래 '오텀센스'의 과실 특성)

  • In this study, we examined the changes of fruit number and fruit weight according to the length of bearing branch of Actinidia arguta and identified the correlation the length of bearing branch and fruit characteristics. The fruit weight and the fruit number of A. arguta bearing branch which length are below 15 cm and over 30 cm were 11.7 g, 4.3 and 12.3 g, 13.8, respectively. From the results, the bearing brach which length was below 15 cm must be removed in winter season pruning. The pruning experiment was conducted to examine the effects on fruit quality and yield of A. arguta. Total fruit yield of heavy-pruning was $14.3{\pm}1.5kg/tree$. The production of fruits over 15 g wight was $8.2{\pm}0.9kg/tree$, that of fruits between 10 g to 15 g was $4.0{\pm}0.7kg/tree$, and that of fruit below 10 g was $2.1{\pm}0.3kg/tree$, respectively. Average fruit yield of nonpruning was $26.7{\pm}2.1kg/tree$, fruit yield over 15 g, between 10 g and 15 g, and below 10 g were $2.5{\pm}0.5kg/tree$, $19.2{\pm}1.4kg/tree$, and $5.0{\pm}0.6kg/tree$, respectively. Distribution of high quality fruit (over 15 g) showed that non-pruning was almost 15~16 g but pruning was evenly distributed between 15 g and 20 g. According to the survey, The high quality fruit (over 15 g) would not be harvested if the winter pruning is not applied in the A. arguta cultivation.
