Studies on the Relationship between Number of Repeat and Duration of Estrous Behavior in Hanwoo and Holstein Cattle (한우와 젖소의 발정 행동 반복수와 지속 시간에 관한 연구)

  • The objective of this study was to investigate the relationship between behavioral signs and duration of estrous in Hanwoo and Holstein cattle for the prediction of optical insemination. Fifty two of 58 and 71 of 89 in Hanwoo and Holstein cows or heifers showed estrous exhibition within 72 h after 2 days following $PGF_{2}\alpha$ administration, respectively. The number of mounting and standing heat were 54.2 and 57.6, 59.4 and 53.5, 42.0 and 30.8 and 16.2 and 10.7 times in Hanwoo cows, Hanwoo heifers, Holstein cows and Holstein heifers, respectively. Duration of mounting were no significantly difference for Hanwoo cows ($21.2\pm11.3$), Hanwoo heifers ($19.9\pm11.4$), Holstein cows ($8.7\pm4.4$) and Holstein heifers ($16.9\pm8.0$). Duration of standing heat were significantly (p<0.05) shorter for Holstein cows ($5.4\pm3.4$) than for Hanwoo cows ($17.1\pm9.6$), Hanwoo heifers ($16.5\pm6.3$) and Holstein heifers ($15.0\pm7.2$). Time until mounting after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($56.3\pm11.45$) than for Hanwoo cows ($42.71\pm10.44$), Hanwoo heifers ($36.6\pm8.21$) and Holstein heifers ($40.70\pm6.15$). Time until standing heat after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($61.6\pm8.92$) than for Hanwoo cows ($46.2\pm11.49$), Hanwoo heifers ($42.7\pm6.06$) and Holstein heifers ($44.1\pm6.72$). In the results of this study, duration of standing heat was the shortest in Holstein cows ($5.4\pm3.4$). The estimation of estrous with Holstein cows has more difficulty because to significantly shorter duration of standing heat than for Hanwoo cows, Hanwoo heifers and Holstein heifers. The standing heat can be a good predictor for time of ovulation but it is concluded that mounting behavior could be the best predictor for time of ovulation.

Improvement of conception rate on Hanwoo; The key hormones and novel estrus detector

  • Two field experiments were conducted to improve the conception rate of Hanwoo cow. The first experiment aimed to investigate the physiological condition of Hanwoo cows on estrus, including metabolic profiles and body condition score (BCS). The second experiment investigated the effect of a novel estrus detector on the artificial insemination (AI) conception rate for Hanwoo cows. For the first experiment, 80 Hanwoo cows (2.5 ± 0.10 of parity), approximately one month before estrus, were housed in 16 pens and offered the experimental diets twice daily with free water access. The BCS were recorded, and blood was collected from the jugular veins just before AI. The collected blood was used to measure physiological conditions, such as metabolite and hormone levels. For the second experiment, each cow was equipped with a neck-mounted estrus detector collar, which had a sensor connected through the internet. Approximately one month before estrus, three hundred sixty Hanwoo cows (2.4 ± 0.21 of parity) were assigned into groups with or without W-Tag collar treatments. The animals were managed the same as in the first experiment. The pregnancy rate reached 55% in the first experiment. The concentration of luteinizing hormone (LH) was higher (p < 0.012; 1.56 vs. 1.08 ng/mL) in cows that were not pregnant (NPG) than in cows that were pregnant (PG) after AI. The BCS and other concentrations of metabolites and hormones in the blood were not different in both NPG and PG cows. The ranges of estrogen, LH, and follicle-stimulating hormone for PG cows were 11.9 to 39.0 pg/mL, < 0.25 to 1.98 ng/mL, and < 0.50 to 0.82 ng/mL, respectively. In the second experiment, cows with the estrus detector had lower days open (p < 0.001; 78.1 vs. 84.8 d), insemination frequency (p < 0.001; 1.26 vs. 2.52), and return of estrus (p < 0.001; 70.9 vs. 79.1 d) than those in cows without the estrus detector. In conclusion, the present study indicated that lower LH concentration just before AI potentially increased the pregnancy rate of Hanwoo cows. Furthermore, the application of estrus detectors to Hanwoo cows could improve the conception success rate for AI.

Effect of Parity of Hanwoo Cow on Physico-Chemical, Sensory and Nutritional Characteristics of Sullungtang (한우 암소의 산차가 설렁탕의 이화학적, 관능적 및 영양적 특성에 미치는 영향)

  • This study was performed to provide scientific information for consumers and to increase the quality and consumption of Sullungtang prepared with bone from Hanwoo. The physico-chemical, sensory, nutritional properties of Sullungtang by different parity were investigated. The extracting extents of blood were significantly higher for heifer among Hanwoo cows with different parity(none, the second or the fourth) and for back bones among different bone portions (round, hind shank, arm, fore shank or back bone) (p〈0.05). For Sullungtang extracted from bones of heifer was higher in collagen contents than those extracted from cows with the second or the fourth parity. Sullungtang extracted from bones of heifer and cows with the second parity had significantly higher contents of condroitin and viscosity than those of cows with the fourth parity, however there was no significant difference between heifer and cows with the second parity. In color, L values were significantly high, and a and b values were significantly low for Sullungtang extracted from bones of heifer when compared to those extracted from the other cows with the second parity. The contents of total nitrogen, sodium, or calcium were higher for Sullungtang extracted from heifer than those from the cows with the second or the fourth parity. In amino acid compositions, the percentages of glycine were highest and proline and glutamic acid were followed fro all Sullungtang samples from Hanwoo. The results showed that the overall quality of Sullungtang significantly decreased as the parity increased for Hanwoo cows. The Sullungtang extracted from bones of heifer had the best sensory scores as well as nutritional quality when compared to those extracted from the cows with parity. Therefore, the labeling and price for cow bones should be differently evaluated by their parity and age in marketing system.

Genetic Parameter Estimates for Ultrasonic Meat Qualities in Hanwoo Cows

  • Real time ultrasound data was generated on 10,596 live Hanwoo cows to study genetic variation on ultrasonic beef quality traits and to assess the best model to estimate genetic parameters on these traits. Pedigree stacking and data validation was done using the SAS statistical software and the genetic parameter estimates were obtained by EM-REML algorithm. Out of the five different multi-trait mixed animal models constructed, the optimal model included fixed effects of herd, year-season-appraisal, body condition score, linear and quadratic covariates for chest girth, the linear covariate effect of age and the random animal and residual effect of the five models studied. The heritability of longissimus muscle area (LMA), $12^{th}$ rib measurement of back fat thickness (BF) and marbling score (MS) was 0.11, 0.17 and 0.15, respectively. Genetic correlation of LMA vs. BF, LMA vs. MS and BF vs. MS was -0.15, 0.06 and 0.61, respectively. The results showed presence of genetic variation in these ultrasonic beef quality traits in Hanwoo cows and suggest that the selection of Hanwoo cows may be possible by performing ultrasonic scans on live animals, which will ultimately be helpful in reducing the generation interval and the cost of selection procedure.

Study on Growth Curves of Longissimus dorsi Muscle Area, Backfat Thickness and Body Conformation for Hanwoo (Korean Native) Cows

  • The objective of this study was to estimate the parameters of Gompertz growth curves with the measurements of body conformation, real-time ultrasound longissimus dorsi muscle area (LMA) and backfat thickness (BFT) in Hanwoo cows. The Hanwoo cows (n = 3,373) were born in 97 Hanwoo commercial farms in the 17 cities or counties of Gyeongbuk province, Korea, between 2000 and 2007. A total of 5,504 ultrasound measurements were collected for the cows at the age of 13 to 165 months in 2007 and 2008. Wither height (HW), rump height (HR), the horizontal distance between the top of the hips (WH), and girth of chest (GC) were also measured. Analysis of variance was conducted to investigate variables affecting LMA and BFT. The effect of farm nested in location was included in the statistical model, as well as the effects of HW, HR, WH, and GC as covariates. All of the effects were significant in the analysis of variance for LMA and BFT (p<0.01), except for the HR effect for LMA. The two ultrasound measures and the four body conformation traits were fitted to a Gompertz growth curve function to estimate parameters. Upper asymptotic weights were estimated as $54.0cm^2$, 7.67 mm, 125.6 cm, 126.4 cm, 29.3 cm, and 184.1 cm, for LMA, BFT, HW, HR, WH, and GC, respectively. Results of ultrasound measurements showed that Hanwoo cows had smaller LMA and greater BFT than other western cattle breeds, suggesting that care must be taken to select for thick BFT rather than an increase of only beef yield. More ultrasound records per cow are needed to get accurate estimates of growth curve, which, thus, helps producers select animals with high accuracy.

Systems for Production of Calves from Hanwoo IVM/IVF/IVC Blastocyst. IV. Direct Transfer of Vitrified and One-Step Diluted Hanwoo Blastocysts

  • This study was to examine whether the vitrified, one-step diluted and direct transferred Hanwoo IVM/IVF/IVC blastocysts can be successfully survived in vivo and they were succeeded into the live birth. For vitrification, blastocysts were serially exposed in glycerol (G) or/and ethylene glycol (EG) mixtures [10% (v/v) G for 5 min, 10% G plus 20% EG (v/v) for 5 min, and 25% G plus 25% EG (v/v) for 30 sect] which is diluted in 10% FBS added D-PBS. Thawing of straw was carried out in air for 10 sec and then in water bath of $25^{\circ}C$ for 20 sec. One-step dilution within the straw was done in water bath of $25^{\circ}C$ for 1 min. Vitrified and one-step diluted embryos were directly transferred into 36 (natural or hormone induced synchronized) recipient cows in 6 areas of Kyungsang Buk-Do. Pregnancies were confirmed at first when recipient cows did not return to the subsequent estrus cycle, and later by manual palpation per rectum on day 45, 90 and then living calves were derived into parturition. Overall pregnancy was 33.3%(12/36), However, higher pregnancy was obtained when the recipients exhibited estrus one day earlier than the age of transferred embryos (53.3 vs 25.0-27.3%), irrespective of synchronization methods. Also, parous recipients became pregnant higher than nulliparous heifers, And, there were not different in pregnancy rates by the aspect of corpus luteum (CL) quality of recipients (good, 29.4; fair, 37.5; poor, 33.3%). One hundred eight of frozen-thawed Hanwoo blastocysts were directly transferred into 36 recipient cows. In 12 of pregnant cows, 3 cows were aborted and 9 cows were calved [single, 66.7% (6/9): twin, 33.3% (3/9)]. Total embryo implantation rate was 11.1% (12/108). However, 9 Hanwoo calves were lived. Therefore, these results demonstrate that direct transfer technique of vitrified and one-step diluted bovine blastocysts can be applied easily and effectively with the higher pregnancy rate on field trial without the equipment and embryological skills.

Effects of feeding sodium metabisulfite-treated fruit and vegetable discards to Hanwoo heifers and cows

  • Objective: Two series of experiments were conducted to determine how the incremental levels of sodium metabisulfite (SMB)-treated fruit and vegetable discards (FVD) in diet of Hanwoo heifers and cows affect their performance and health. Methods: In Exp. 1, 36 Hanwoo heifers were stratified by age (13.3±0.83 mo) and initial body weight (305±19.7 kg), and divided randomly to one of three diets containing 0%, 10%, or 20% SMB-treated FVD (as-fed basis). The experiment lasted 110 d, including 20 d of adaptation. In Exp. 2, 24 multiparous Hanwoo cows were divided into three groups based on age (48.2±2.81 mo) and initial body condition score (2.64±0.33). Cows in each block were assigned randomly to one of three diets containing 0%, 11%, or 22% SMB-treated FVD (as-fed basis). The experiment lasted 80 d, including a 20-d adaptation period. In both experiments, SMB-treated FVD was used as a replacement for wet brewers grain in total mixed ration (TMR). Results: Growing heifers exhibited no differences in their daily feed intake (6.58±0.61 kg/d dry matter [DM]), average daily gain (0.60±0.07 kg/d), and body condition score when they consumed the incremental levels of SMB-treated FVD. Although most blood metabolites were unaffected by treatments, blood urea-N and β-hydroxybutyrate levels decreased linearly as the SMB-treated FVD level increased in TMR. Similar to Exp. 1, minor differences were found in daily feed intake (8.27±0.72 kg DM/d) and body condition score of Hanwoo cows. Most blood metabolites remained unaffected by treatments, but blood urea-N decreased as the SMB-treated FVD level in TMR increased. Conclusion: Our findings suggest that SMB-treated FVD could be safely incorporated into the diet of Hanwoo heifers and cows, potentially improving N-use efficiency in the body while not impairing performance or health.

The Studies on Embryo Production from Hanwoo Cows with High Offspring Meat Quality Grade and Embryo Transfer (후대의 육질등급이 우수한 한우 암소의 수정란 생산 및 이식에 관한 연구)

  • These studies were conducted to evaluate developmental competence of follicular oocyte collected from the ovaries of Hanwoo cows with the high offspring meat quality (1++ and 1+ grade). Cumulus oocyte complexes from individual cows were matured, fertilized and cultured using protocols of in-vitro maturation (IVM), invitro fertilization (IVF) and in-vitro culture (IVC). The rates of blastocyst development from Hanwoo cows with the offspring meat quality grades of 1++ and 1+ were 18.6 and 21.2%, respectively. The rates of blastocyst development were 26.3, 20.7, 20.7, 17.2 and 31.2% from Hanwoo cows with the meat quality grades of 1++, 1+, 1, 2 and 3, respectively. Fiftyseven transferable embryos were recovered from 11 Hanwoo donor cows (5.2/head) with the high offspring meat quality grades of 1++ and 1+ in vivo, and the pregnancy rate after embryo transfer was 61.1%. In conclusion, these results suggest that in vitro embryo production from the ovaries of cows with the high meat quality grades using individual culture system can be used an efficient method for livestock improvement. In addition, for the successful industrialization of embryo transfer, conception rate should be improved.

A study on the correlation between vaginal conductivity and estrus in Hanwoo (Bos taurus coreanae) cows

  • Background: Estrus in cows can be detected through vaginal electrical resistance or conductivity. However, there are no studies measuring vaginal electrical resistance in Hanwoo cows. This study aims to measure the vaginal electrical resistance value in Hanwoo cows and compare it with estrus and ovulation. Methods: Vaginal electrical resistance values of 73 Hanwoo cows were measured before and after estrus at the Gyeongsangbuk-do Livestock Research Institute. Measurements were taken on days -6, -3, -2, -1, 0, 1, 2, 3, and 6 of artificial insemination. Large follicles and ovulation were confirmed using transvaginal ultrasonography. Results: The vaginal electrical resistance averaged 225.6 ± 6.3 Ω days before the artificial insemination date, decreasing until the day of artificial insemination. The average vaginal electrical resistance was 163.7 ± 4.6 Ω on the date of artificial insemination, and 188.8 ± 4.3 Ω one day after artificial insemination, when large follicles were observed. In addition, on the 6th day after artificial insemination, the vaginal electrical resistance averaged 231.4 ± 5.5, which was similar to the 6th day before artificial insemination (222.5 ± 6.3). Transvaginal ultrasonography showed that most of the cows ovulated one day after artificial insemination. Conclusions: The accuracy of estrus is high if the vaginal electrical resistance is measured for cows with confirmed estrus, making is a potentially useful for determining the timing of artificial insemination.

Improvement of pregnancy rate after deep uterine artificial insemination with frozen-thawed cauda epididymal spermatozoa in Hanwoo cattle

  • In the present study, we examined if deep uterine artificial insemination (DUAI) can improve the pregnancy rate of artificial insemination (AI) using epididymal spermatozoa (ES) in Hanwoo cattle. The estrus cycles of 88 Hanwoo cows were synchronized, and 17 cows were artificially inseminated using the DUAI method with ES, 20 cows were artificially inseminated via the uterine body (BUAI) method with ES, and as a control, 51 cows were inseminated by using the BUAI method with ejaculated spermatozoa from 1 proven bull after frozen thawing. The pregnancy rate of the DUAI method (58.8%) was higher than that of the BUAI method (25.0%, p = 0.0498). The motility of ES was examined immediately after thawing and after 3 and 6 h of incubation. The rapid progressive sperm motility of the control group was significantly higher than that of the ES group immediately after thawing and after 3 and 6 h of incubation (p < 0.05). The straight line velocity and average path velocity of the ES group after 6 h of incubation were significantly lower than those in the control group (p < 0.05). The linearity and amplitude of lateral head of ES were lower than those at 6 h (p < 0.05). The flagellar beat cross frequency and hyperactivation of ES were lower than the control spermatozoa immediately after thawing and at 3 h (p < 0.05). These motility parameters suggested that ES had a low motility and fertilization ability compared to the control spermatozoa. After frozen-thawing and 3 h of incubation, the percentage of live spermatozoa with intact acrosomes in the ES was significantly lower than that in ejaculated spermatozoa (p < 0.05). Our findings suggested that the DUAI method can overcome the low pregnancy rate of ES, despite the low motility, viability, and fertilization ability of ES.
