Identification of a Novel PGE2 Regulated Gene in SNU1 Gastric Cancer Cell

  • Prostaglandin $E_2$ ($PGE_2$) plays an important role in the regulation of various gastric functions, and the growth-inhibitory activities on tumor cells are studied in vitro and in vivo. Although the mechanisms have attracted many researchers in the past decade, the molecular mechanisms of cell cycle arrest, or induction of apoptosis by $PGE_2$, is unclear. We investigated the effects of $PGE_2$ on the growth of the human gastric carcinoma cell line SNU1 and genes that are regulated by $PGE_2$ and isolated them using differential display RT-PCR (DD RT-PCR). FACS analysis suggested that SNU1 cells were arrested at the G1 phase by $PGE_2$ treatment. This growth inhibitory effect was in a time- and dose-dependent manner. Treatment of SNU1 cells with $10\;{\mu}g/ml$ $PGE_2$, followed by DD RT-PCR analysis, revealed differently expressed bands patterns from the control. Among the differently expressed clones, we found an unidentified cDNA clone (HGP-27) overexpressed in $PGE_2$-treated cells. The full-length cDNA of HGP-27 was isolated using RACE, which consisted of a 30-nt 5'-noncoding region, a 891-nt ORF encoding the 296 amino acid protein, and a 738-nt 3'-noncoding region including a poly(a) signal. This gene was localized on the short arm of chromosome number 11. Using the Motif Finder program, a myb-DNA binding repeat signature was detected on the ORF region. The COOH-terminal half was shown to have similarity with the $NH_3$-terminal domain of thioredoxin (Trx). This relation between HGP-27 and Trx implied a potential role for HGP-27 in modulating the DNA binding function of a transcription factor, myb.

A Study on the Performances of Pullets and Subsequent Egg Production with the Substitution of Ground Sunflower Seeds (해바라기씨앗으로 사육된 병아리의 성장 및 산란계로서의 생산능력에 관한 연구)

  • Ten-week-old layer type pullets were used in this study, housed 10 birds per cage(61$\times$41cm) and were replicated 15 times After raising them on a 19% protein diet up to ten weeks of age they were fed a 12% protein corn-soy control diet or one with half or all of the soybean mea replaced with ground full-fat sunflower seeds. At 19 weeks of age, pulltes were transferred to a layer house and their respective diet was gradually changed to a 13.5% layer diet containing 635% oats. The 19% protein SFS diet in treatment 2 did not show my, adverse effect on body weight or subsequent egg productio. The higher level of SES(38%) appeared to delay sexual maturity somewhat as was indicated by body weight and egg production data. However, the higher replacement of soybean with SFS(38%) in the pullet ration may not be critical for sexual maturity, but beneficial in a full laying cycle. There were no adverse effects on later stages of production.

  • Alterations of cyclin D1, one of the main regulators of the cell cycle, are known to be involved in various cancers. The CCDN1 G870A polymorphism causes production of a truncated variant with a shorter half-life and thus thought to impact the regulatory effect of CCDN1. The aim of the present study was to contribute to existing results to help to determine the prognostic value of this specific gene variant and evaluate the role of CCDN1 G870A polymorphism in brain cancer susceptibility. A Turkish study group including 99 patients with primary brain tumors and 155 healthy controls were examined. Genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism analysis. The CCDN1 genotype frequencies in meningioma, glioma and control cases were not significantly different (p>0.05). No significant association was detected according to clinical parameters or tumor characteristics; however, a higher frequency of AG genotype was recorded within patients with astrocytic or oligoastrocytic tumors. A significant association between AG genotype and gliobilastoma multiforme (GBM) was recorded within the patients with glial tumors (p value=0.048 OR: 1.87 CI% 1.010-3.463). According to tumor characteristics, no statistically significant difference was detected within astrocytic, oligoasltrocytic tumors and oligodentrioglias. However, patients with astrocytic astrocytic or oligoastrocytic tumors showed a higher frequency of AG genotype (50%) when compared to those with oligodendrioglial tumors (27.3%). Our results indicate a possible relation between GBM formation and CCDN1 genotype.

  • Filtering is necessary to reduce statistical noise and to increase image quality in SPECT images. Noises controled by low-pass filter designed to suppress high spatial frequency in SPECT image. Most SPECT filter function control the degree of high frequency supression by chosing a cut of frequency. The location of cut off frequency determines the affect image noise and spatial resolution. If select the low cut off frequency, its provide good noise suppression but insufficient image quantity and high cut off frequencies increase the image resolution but insufficient noise suppression. The purpose of this study was to determines the optimam cut off level with comparison of FWHM according to cut off level in each kiters-Band-limited, Sheep-logan, Sheep-logan Hanning, Generalized Hamming, Low pass cosine, Parazen and Butterworth filter in SPECT camera. We recorded image along the X, Y, Z-axis with $^{99m}TcO_4$ point source and measured FWHM by use profile curve. We find averaged length is $9.16\;mm{\sim}18.14\;mm$ of FWHM in X, Y, and Z-axis, and Band-limited and Generalized Hamming filters measures 9.16 mm at 0.7 cycle/pixel cut off frequency.

SOME PHYSIOLOGICAL STUDIES ON THE UTILIZATION OF ORGANIC SUBSTRATES BY EUGLENA GRACILIS VAR. BACILLA 10616 IN LIGHT AND IN DARKNESS ("유-그레나"의 명암배양에 따르는 유기질의 이용과 호흡 및 생장에 대하여)

  • 1) The comparative studies of the quantitative measurement of growth characteristics and utilization of substrates by Euglena gracilis var. bacilla 10616 in the light and in darkness have been carried out. Eodogenous respiration, effect of respiratory inhibitors and responses to the added substrates for the exogenous respiration are also investigated. 2) All cultures are grown in the open air under the continuous illumination of fluorescent light of 3500 lux at room termperature, the growth rate of the culture in the basal medium added 0.5% lactate is found to be the highest. The growth rate decreases successively for the cultures of 0.5% sucinate, 0.5% Na-acetate, 0.5% malate, and control. There is no growth in the basal meidum added 0.5% butyrate and 0.5% hydroquinone. The similar results are obtained for the mentioned cultures in the darkness. However, the growth rate in basal medium added 0.5% glucose and 0.5% sucrose does seem to increase in the darkness unlike the illumination. 3) The endogenous rate of respiration for the organism cultured photosynthetically is about 12.94ul 02/mg/hr, in basal medium and the respiratory quotient is about 0.84. The rate is decreased by starvations to 6.5ul 02/mg/hr, about to a half, but the respiratory quotient does net change. 4) The oxygen consomption during initial 2 hours in suspending solution ranging from pH 4.5 to pH 9.3 is highest at pH 4.5 in which the algae had grown, at pH 5.5 and at pH 6.9. 5) Endogenous respiration of the cells is strongly inhibited by 0.1M of potassium cyanide, malomic acid, sodium fluoride and iodo-acetic acid. It is also strongly inhibited by 0.01M of potassium cyanide. 6) The respiratory response to added substrates for the exogenous respiration in the organism is coincided with the rate in the basal medium added the substrate in light and in darkness, whether the cells are fed or starved. 7) According to the results of this study, there seems to be the flexibility of the interconversion between photosynthesis and chemosynthesis, heterotropic mode of metabolism, in Euglena gracilis var. bacillaris, and that this organism utilizes the lactate most. It also may be suggested that the enayme systems linked in the each steps of Embden-Myerhof-Parnas path way and TCA cycle seem to exist in this organism.

  • Recently, Both rapid economic growth and high-quality native finishing materials demand in buildings such as local infrastructure facilities and cultural facilities have increased along with local quarries. So, increasing local quarries and environmental pollution occurred in quarries get the eyes to damaged area of the surroundings. As an example, carcinogen such as solid formed to fixing asbestos and dust have damaged to local resident. Especially, Radon gas released from asbestos can exist everywhere on earth, released soil and rock as radioactive substances, can be caused lung cancer followed by a smoking. When pollution source to indoor air quality that lacking ventilation rate of the residential building moved in a cycle, human responses such as headache, dizziness, etc. get appear, so on it threatened resident's physical condition. Thus, we need to urgent attention to reduction harmful substance. In the case of radon gas of the pollution source to indoor air quality in housing, it has characteristic that keep on going through half-life released from source, we need to control radon gas source than source removal. We set on vermiculite addition ratio to 10% which has harmful substances adsorption performance, proceed experiment to basalt waste rock addition ratio 50, 60, 70, 80(%). The result of an experiment, based on 'KS F 4035, precast terrazzo', we can be obtainable in the best terrazzo at basalt waste rock addition ratio 70%.

Effects of Repeated High Ambient Temperature on Performance in Broilers Heat-Conditioned at an Early Age (반복적인 고온환경이 사전고온 적응한 육계의 생산성에 미치는 영향)

  • Heat conditioning at an early age has been known to help chickens cope with heat stress later in life. The present study was conducted to determine the effects of heat conditioning at 5 days of age in broilers repeatedly exposed to high ambient temperature later in life. A total of 256 day-old Arbor Acre boiler chicks were housed in two identical rooms with a 23-h light/1-h dark cycle and provided with feed and water ad libitum. At 5 days of age, the birds in one room were exposed to $37^{\circ}C$ for 24 hours, while those in the other room served as controls. On day 21, half of the birds in each room were moved into the other room so that each room contained both control and heat-conditioned birds. After a 7-day adaptation period, the birds in one room were exposed to high ambient temperature ($21^{\circ}C{\rightarrow}31^{\circ}C$) for 3 days, whereas those in the other room were kept at normal temperature. The same 3-day exposure to high ambient temperature was repeated two weeks later. Hence, there were four treatment groups (CON+CON: control+control; CON+HS: control+high ambient temperature; HC+CON: heat conditioning+control; and HC+HS: heat conditioning+high ambient temperature). Repeated heat stress resulted in decreased feed intake, water intake, body weight gain, and spleen weight (p<0.05) and increased rectal temperature (p<0.05), mortality, and plasma corticosterone concentrations. The relative weight of the spleen was increased in the heat-conditioned group (p<0.05). Plasma biochemicals were also influenced by high temperature. Thus, no beneficial effects of heat conditioning at an early age were detected in broilers repeatedly exposed to high ambient temperature later in life.

COATED PARTICLE FUEL FOR HIGH TEMPERATURE GAS COOLED REACTORS

  • Roy Huddle, having invented the coated particle in Harwell 1957, stated in the early 1970s that we know now everything about particles and coatings and should be going over to deal with other problems. This was on the occasion of the Dragon fuel performance information meeting London 1973: How wrong a genius be! It took until 1978 that really good particles were made in Germany, then during the Japanese HTTR production in the 1990s and finally the Chinese 2000-2001 campaign for HTR-10. Here, we present a review of history and present status. Today, good fuel is measured by different standards from the seventies: where $9*10^{-4}$ initial free heavy metal fraction was typical for early AVR carbide fuel and $3*10^{-4}$ initial free heavy metal fraction was acceptable for oxide fuel in THTR, we insist on values more than an order of magnitude below this value today. Half a percent of particle failure at the end-of-irradiation, another ancient standard, is not even acceptable today, even for the most severe accidents. While legislation and licensing has not changed, one of the reasons we insist on these improvements is the preference for passive systems rather than active controls of earlier times. After renewed HTGR interest, we are reporting about the start of new or reactivated coated particle work in several parts of the world, considering the aspects of designs/ traditional and new materials, manufacturing technologies/ quality control quality assurance, irradiation and accident performance, modeling and performance predictions, and fuel cycle aspects and spent fuel treatment. In very general terms, the coated particle should be strong, reliable, retentive, and affordable. These properties have to be quantified and will be eventually optimized for a specific application system. Results obtained so far indicate that the same particle can be used for steam cycle applications with $700-750^{\circ}C$ helium coolant gas exit, for gas turbine applications at $850-900^{\circ}C$ and for process heat/hydrogen generation applications with $950^{\circ}C$ outlet temperatures. There is a clear set of standards for modem high quality fuel in terms of low levels of heavy metal contamination, manufacture-induced particle defects during fuel body and fuel element making, irradiation/accident induced particle failures and limits on fission product release from intact particles. While gas-cooled reactor design is still open-ended with blocks for the prismatic and spherical fuel elements for the pebble-bed design, there is near worldwide agreement on high quality fuel: a $500{\mu}m$ diameter $UO_2$ kernel of 10% enrichment is surrounded by a $100{\mu}m$ thick sacrificial buffer layer to be followed by a dense inner pyrocarbon layer, a high quality silicon carbide layer of $35{\mu}m$ thickness and theoretical density and another outer pyrocarbon layer. Good performance has been demonstrated both under operational and under accident conditions, i.e. to 10% FIMA and maximum $1600^{\circ}C$ afterwards. And it is the wide-ranging demonstration experience that makes this particle superior. Recommendations are made for further work: 1. Generation of data for presently manufactured materials, e.g. SiC strength and strength distribution, PyC creep and shrinkage and many more material data sets. 2. Renewed start of irradiation and accident testing of modem coated particle fuel. 3. Analysis of existing and newly created data with a view to demonstrate satisfactory performance at burnups beyond 10% FIMA and complete fission product retention even in accidents that go beyond $1600^{\circ}C$ for a short period of time. This work should proceed at both national and international level.

A Study on the Principle of Making-Music of the Chaegut ("Stroke Music") in Farmers' Band Music (풍물굿 채굿 가락의 형성원리에 관한 연구)

  • Farmers' band music is a religious ritual in Korea. It is a solemn ritual to exhibit the auspicious holiness of the people to the God. Most of the ritual is accomplished by music. The music of the farmers 'band, in this sense, has its meaning as the expression of holiness by sound. In this paper, I will explore the principle to make various rhythms of farmers' band music, especially focusing on the chaegut (lit. "stroke music"). It is very symbolic because the name of the music shows the strokes of the gong, called jing, in a rhythmic phrase. In this sense, the chaegut is man-made music based on the specific principles in accordance with the strokes of the gong. Among many chaegut rhythms, samchae (lit. "three strokes") to chilchae (lit. "seven strokes") are the main rhythms. They were made after the principle of 'forward with drums, backward with gongs' in Five way procession. It the basic principle of military procession from the early Joseon Dynasty. The procession follows with the sound of the drums and gongs. There was always a principle of "five strokes" to control the procession. The "five strokes" became the basis of the making of chaegut rhythms. The rhythms of the samchae to chilchae are based on the rhythm of samchae. The samchae has three gong strokes in a cycle, which exhibits the chaotic moment with the rhythmic noise. The name of the rhythm exhibits the correspondence of the signifiant, that is the name "three strokes" and the signifié, that is the real three strokes of gongs in music. Other four rhythms has made up from the samchae with half cycles are added in accordance with the strokes of the gongs. In this way, the chaegut shows the principle of "five strokes" in the military procession. The rhythm of ochae jilgut is a mixture of ochae (lit. "five strokes") and jilgut (lit. "road music") which is usually performed on the road to a mountain shrine. The musical structure of ochae jilgut corresponds to the colotomic structure of Southeast Asian music and the 15th-century music of old musical scores. The rhythm of gilgunak chilchae is a mixture of gilgunak (lit. "road military music") and chilchae (lit. "seven strokes"). The rhythmic structure is similar to other regional music, sijo ("short song") of the literati music and norae garak (lit. "some melody") of the shaman music. In sum, the chaegut is very artistic music made from the military procession of the Joseon Dynasty. The name of the rhythm corresponds with the strokes of the gong in a cycle. In this way, the chaegut shows the principle of music-making to exhibit the ritual characteristics of the Korean people.

Evaluation of $^{14}C$ Behavior Characteristic in Reactor Coolant from Korean PWR NPP's (국내 경수로형 원자로 냉각재 중의 $^{14}C$ 거동 특성 평가)

  • This study has been focused on determining the chemical composition of $^{14}C$ - in terms of both organic and inorganic $^{14}C$ contents - in reactor coolant from 3 different PWR's reactor type. The purpose was to evaluate the characteristic of $^{14}C$ that can serve as a basis for reliable estimation of the environmental release at domestic PWR sites. $^{14}C$ is the most important nuclide in the inventory, since it contributes one of the main dose contributors in future release scenarios. The reason for this is its high mobility in the environment, biological availability and long half-life(5730yr). More recent studies - where a more detailed investigation of organic $^{14}C$ species believed to be formed in the coolant under reducing conditions have been made - show that the organic compounds not only are limited to hydrocarbons and CO. Possible organic compounds formed including formaldehyde, formic acid and acetic acid, etc. Under oxidizing conditions shows the oxidized carbon forms, possibly mainly carbon dioxide and bicarbonate forms. Measurements of organic and inorganic $^{14}C$ in various water systems were also performed. The $^{14}C$ inventory in the reactor water was found to be 3.1 GBq/kg in PWR of which less than 10% was in inorganic form. Generally, the $^{14}C$ activity in the water was divided equally between the gas- and water- phase. Even though organic $^{14}C$ compound shows that dominant species during the reactor operation, But during the releasing of $^{14}C$ from the plant stack, chemical forms of $^{14}C$ shows the different composition due to the operation conditions such as temperature, pH, volume control tank venting and shut down chemistry.
