Improved Anthocyanin Production in Hairy Root Culture of Daucus carota by Fungal Elicitor (당근의 모상근 배양에서 Fungal Elicitor에 의한 Anthocyanin 생산의 향상)

  • In order to improve anthocyanin production, effects of fungal elicitation in hairy root culture were investigated. fungal elicitor prepared from Fusarium moniliforme was the best in enhancement of anthocyanin production among the eight fungal elicitors tested. The optimum treat-ment time and concentration of treated elicitor for anthocyanin production were 12 hours and 3.28 mg carbohydrates per liter medium. Also, fungal elicitor was treated to hairy root culture in flat-bottomed fluidized-bed bioreactor. The anthocyanin production of elicited culture was enhanced 227% than non-treated.

Anthraquinone Productivity by the Cultures of Adventitious Roots and Hairy Roots from Curled Dock (Rumex crispus) (소리쟁이 (Rumex crispus)의 부정근과 모상근 배양에 의한 Anthraquinone 생산성)

  • In order to survey anthraquinone productivity from in vitro root culture, transformed hairy roots of Rumex crispus were induced from leaf segments by infection with Agrobacterium rhizogenes strain $A_4$ and compared with adventitious roots. The optimum condition of adventitious root formation from leaf segments was 5 $\mu$M NAA added to MS medium. Mannopine was detected in the extract of hairy roots by paper electrophoresis, but not in adventitious roots. Secondary root tips of both adventitious roots and hairy roots elongated without lateral root branching in hormone free MS medium, but primary root tips showed more rapid growth with extensive lateral root branching. MS basal medium was the best for growth of the adventious roots and hairy roots for anthraquinone content. Adventitious root tips and hairy root tips cultured in liquid MS medium supplemented with 0.05 $\mu$M NAA and 0.1 $\mu$M kinetin (contained 5% sucrose) showed the maximal growth and anthraquinone content. Anthraquinone content of hairy roots was increased by the culture periods, but was reduced after 25 days of culture.

  • In this study, the effects of auxin and casein hydrolysate (CH) on the growth of ginseng hairy root was elucidated. Ginseng hairy root was cultured under light and dark conditions in MS solid and liquid medium with various concentrations of auxin and CH for fifty days. After harvesting the cultures, the fresh and dry weight of cultures were examined, respectively. In the MS solid culture, 1 mg/L of IBA was most effective on the growth of ginseng hairy root under the dark condition, whereas IAA and CH did not affect on the growth of ginseng hairy root. In the MS liquid culture, the growth was maintained regularly by the treatments of IAA and NAA. IBA and CH restrained the growth of ginseng hairy root.

  • Ginseng(Panax ginseng C. A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng hairy roots induced by introducing Ri-plasmid of Agrobacterium rhizogenes into genomic DNA of plant cells show vigorous growth, and the hairy roots produce the same or more saponins than natural ginseng roots. Therefore, hairy roots can be used for commercial purposes. The present study was carried out to induce hairy roots with both active growth and high saponin contents. Numerous hairy roots of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes R1000 A4T in dark condition after one month of culture. About 3 hundred lines of hairy roots were selected according as morphological characters on medium with carbenicillin. After pre-selection of fifteen lines of hairy roots with active growth, KGHR-l and KGHR-8 lines were finally selected which had characters of high content of ginsenoside-Rd and ginsenoside-Re, respectively. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark (22 $^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5L Erlenmeyer flasks, lL roller drums, 10L jar-fermenters, and especially in 20L air-lift culture vessels.

  • Induction and culture of hairy roots by Agrobacterium rhizogenes A4 were carried out in Platycodon grandiflorum DC. After 2-4 weeks of inoculation with Agrobacterium rhizogenes hairy roots were formed at root segments in the balloon flower. Optimized growth of hairy roots was obtained in hormone-free MS medium, 6% sucrose and pH 5.8. The pattern of ginsenoside in the transformed roots was not different with that in the ordinary roots.

Bioceramic Effects to Enhance Secondary Metabolites Production in Tissue Culture of Some Medicinal Plants

  • We have investigated that a couple of soft ferrite ceramic powders having a spinal structure have shown the effect on growth and secondary metabolites production of some medicinal plants cultured in vitro. The addition of the ceramic powders as bare state to culture medium has stimulated the growth of Achyranthes japonica callus and plantlet, adventitious root of Hyoscyamus niger and Platycodon grandiflorum hairy root about 65, 75, 150 and 50%, respectively. Whereas Centella asiatica callus and plantlet, Scopolia parviflora hairy root, and Hyoscyamus albus adventitious root were not affected markedly. Moreover, the ceramic powder has enhanced the growth of H. niger adventitious roots even under conditions of irradiating alone without any direct contact between ceramic powder and media. Based on growth stimulation effect, the ceramic powders have enhanced the gross production of tropane alkaloid in H. niger adventitious root, and polyacetylene in P. grandiflorum hairy root about 35 and 30%, respectively.

Isoflavones and biotransformed dihydrodaidzein in hairy roots of Korean wild arrowroot

  • Pueraria lobata is a perennial legume plant that produces a variety of isoflavones, such as puerarin, daidzin and daidzein. These are metabolized to equol via dihydrodaidzein and tetrahydrodaidzein by the bacterial fermentation of natural isoflavone sources in the human intestines. In this study, we described the growth and accumulation of isoflavone in the hairy root of the Korean wild arrowroot according to the culture period, as well as dihydrodaidzein biosynthesis in hairy root extracts fermented with Pediococcus pentosaceus. Daily proliferation was best in DY1 cultured for 1 week. DY1 showed significant differences in daily production of puerarin and daidzin+daidzein, as compared to DJ7; furthermore, both were best in DY1 cultured for 1 week. The hairy root extract was fermented successfully with P. pentosaceus with confirmed production of dihydrodaidzein, an equol precursor formed by biotransformation. The results indicated that the growth of hairy roots and isoflavone accumulation in the hairy roots is best 1 week after culture. These results are expected to contribute to the mass production of hairy root and isoflavones as equol precursors from the Korean wild arrowroot and provide a basis for equol production by biotransformation in vitro.

Hairy Root Culture of Daucus carota for Anthocyanin Production in a Fluidized-bed Bioreactor (유동층 생물반응기에서 anthocyanin 생산을 위한 당근의 모상근 배양)

  • Hairy root culture of Daucus carota was investigated for anthocyanin production in a fluidized-bed bioreactor. The growth of hairy roots in this bioreactor increased 2.5 fold while anthocyanin production was lower. However, the anthocyanin production of hairy roots in a fluidized-bed bioreactor was enhanced 2.3 fold in response to the treatment of the fungal elicitor.

  • Plant-derived natural products have been and will continue to be valuable sources. Elicitors have been employed to modify cell metabolism in order to enhance the productivity of useful metabolites in plant cell/tissue cultures. In this study, several elicitors were used to improve the productivity of useful metabolites and to reduce culture time for archiving high concentration in P. ginseng hairy root cultures. The addition of chitosan, chitosan oligosaccharide and alginate oligosaccharide to the culture of P. ginseng hairy roots caused growth to be inhibited with the increase in elicitor concentration. The usage of the chitosan elicitor and D-glucosamine caused a slight decrease in hairy root growth, whereas total ginseng saponin accumulated slightly with the increase in elicitor concentration. When gel beads were added to the culture medium at the initial period, hairy root growth was enhanced. The maximum growth was 1.35 times higher than that of the control at $1\%$ (w/v). Total ginseng saponin content decreased due to the addition of alginate beads. This would result in consistent diffusion of lower levels of calcium ions during the culture period that promotes biomass growth.

The Production of Ginsenosides from Ginseng Hairy Root by Treatment of the Chitin and Chitosan (Chitin과 Chitosan 처리에 의한 인삼모상근으로부터 Ginsenosides 생산)

  • To elucidate the effect of chitin and chitosan on the production of ginsenosides and growth, ginseng hairy root was cultured on the 1/2 MS medium supplemented with chitin and chitosan of various concentrations and culture period. The highest growth was obtained with 1 mg/L of chitin. However, the growth was inhibited by 20 mg/L or above. The contents and productivity of ginsenosides were the highest when ginseng hairy roots were cultured on 40 mg/L chitin and applied of the third-weeks of culture period. Ginseng hairy root culture with 1 mg/L of chitosan resulted in the best growth, but the highest ginsenosides level was appeared in 30 mg/L chitosan. Ginsenosides content was increased when it was treated at the forth-week after culture as 30 mg/L of chitosan.
