Aging of hair follicle stem cells and their niches

  • Hair follicles in the skin undergo cyclic rounds of regeneration, degeneration, and rest throughout life. Stem cells residing in hair follicles play a pivotal role in maintaining tissue homeostasis and hair growth cycles. Research on hair follicle aging and age-related hair loss has demonstrated that a decline in hair follicle stem cell (HFSC) activity with aging can decrease the regeneration capacity of hair follicles. This review summarizes our understanding of how age-associated HFSC intrinsic and extrinsic mechanisms can induce HFSC aging and hair loss. In addition, we discuss approaches developed to attenuate ageassociated changes in HFSCs and their niches, thereby promoting hair regrowth.

The Efficacy of Shampoo Containing Ginseng Radix on Preventing Hair Loss and Promotion Hair Growth (홍삼사포닌 Rg3가 함유된 샴푸의 탈모방지와 양모개선 효과)

  • Recently, hair growth effect of red ginseng was proved by many in vitro experiments, but there are not enough clinical studies. In the present study, we evaluated the efficacy of Rg3 hair shampoo (Somang Co., Korea) containing Rg3 0.003% extracted from ginseng radix on preventing hair loss and promoting hair growth by clinical study. 42 volunteers were enrolled present study and devided into two groups; one group used the Rg3 hair shampoo and another group used shampoo not containg Rg3. Before and after 8 and 16 weeks use of Rg3 hair shampoo, we measured the density, thickness and growth rate of hair. At the same time, an assessment of improvement of hair loss and subjective satisfaction of patients were conducted by clinicians and patients. Results obtained from the group used Rg3 hair shampoo showed statistically significant improvement in the density, thickness and growth rate of hair. Results from the clinicians, patients and subjective satisfaction of patients also showed better responses for the group used Rg3 hair shampoo. In conclusion, Rg3 hair shampoo is an effective product on preventing hair loss and promoting hair growth.

Hair-Growth Effect and Single Dose Oral Toxicity Test of Illite Powder (Illite 분제 원액의 육모 활성 시험 및 단회 투여 경구 독성시험)

  • The hair-growth effect of Illite was suggested by some people who were using Illite as a beautifying material. We investigated the hair-growth effect of Illite powder. The hair-growth effects were investigated by two methods; the activity of hair-growth after shaving the hairs on the black mouse (C57BL/6) and the recovery activity of hair-growth after hair-loss induced by cyclophosphamide treatment. Suspension of Illite powder was applied to the back of the black mouse by method of skin paste. Illite promoted significantly the hair growth of mouse in both conditions of shaving and hair-loss. And then we investigated the toxicity which may be induced by Illite when it was administrated orally as a single dose. We could not fond out any significant toxicity induced by single dose oral administration of Illite.

  • Objectives : With increasing interests in hair cosmetic to keep their scalp and hair healthy we must have much more information about the oriental medical cosmetic goods. Methods : Shampoos which contain herbal ingredients are commonly used to stopping their hair loss, alopecia. On the basis of oriental pharmacology, we analyzed several shampoos which have herbal ingredients and thought about the oriental cosmetic methods for a hair loss. Results : We found that many herb based shampoos are developed on the basis of the principle of oriental medicine. In addition, the interest in oriental medical cosmetic methods that used the principles of the oriental medical science has been increased and cosmetic goods based on the oriental medicine have a positive market. Conclusions : With many possibilities for the oriental medical cosmetic methods, clinical records to prove scientific facts are urgently needed. In addition, the oriental medical cosmetic methods based on oriental medicine to examine scalp and hair should be offered.

ER71/ETV2 Promotes Hair Regeneration from Chemotherapeutic Drug-Induced Hair Loss by Enhancing Angiogenesis

  • Chemotherapy-induced alopecia and hair loss can be stressful in patients with cancer. The hair grows back, but sometimes the hair tends to stay thin. Therefore, understanding mechanisms regulating hair regeneration may improve the management of chemotherapy-induced alopecia. Previous studies have revealed that chemotherapeutic agents induce a hair follicle vascular injury. As hair growth is associated with micro-vessel regeneration, we postulated that the stimulation of angiogenesis might enhance hair regeneration. In particular, mice treated with 5-fluorouracil (5-FU) showed delayed anagen initiation and reduced capillary density when compared with untreated controls, suggesting that the retardation of anagen initiation by 5-FU treatment may be attributed to the loss of perifollicular micro-vessels. We investigated whether the ETS transcription factor ETV2 (aka ER71), critical for vascular development and regeneration, can promote angiogenesis and hair regrowth in a 5-FU-induced alopecia mouse model. Tie2-Cre; Etv2 conditional knockout (CKO) mice, which lack Etv2 in endothelial cells, presented similar hair regrowth rates as the control mice after depilation. Following 5-FU treatment, Tie2-Cre; Etv2 CKO mice revealed a significant reduction in capillary density, anagen induction, and hair restoration when compared with controls. Mice receiving lentiviral Etv2 injection after 5-FU treatment showed significantly improved anagen induction and hair regrowth. Two-photon laser scanning microscopy revealed that enforced Etv2 expression restored normal vessel morphology after 5-FU mediated vessel injury. Our data suggest that vessel regeneration strategies may improve hair regrowth after chemotherapeutic treatment.

Single Hair Transplantation for Cicatrical Eyebrow Alopecia (흉터성 눈썹소실환자에 실시한 단일모 이식술)

  • Purpose: Single hair transplantation for eyebrow loss has been considered as a good method of achieving natural appearance of eyebrow. To date, however, no study has reported the survival rate following single hair transplantation for cicatricial eyebrow alopecia. Methods: We performed single hair transplantation using Choi hair transplanter for 49 cases of scarring eyebrow loss from March 2003 to March 2006. Of these cases, 33 could be under a follow-up postoperatively for more than seven months. These 33 cases comprised 20 men and 13 women. Mean follow-up period was 12 months (7-36 months). In 17 patients(51.5%), 100 to 200 hairs were unilaterally transplanted. Results: Following initial hair transplantation, the surgical outcome was excellent in 11 cases(33.3%), good in 12 cases(36.4%), fair or poor in the remaining ten cases(30.3%). All procedures were done under intravenous sedation and local or regional block anesthesia. It took about one and a half hours for us to transplant 200 hairs. The shape of eyebrow was aesthetically acceptable with a good direction and an ideal inclination of hairs. No patients developed notable complications, while 30 percent had a low survival rate. Conclusion: Single hair transplantation using Choi hair transplanter is a safe, effective surgical method for scarring eyebrow loss.

The Preclinical and Clinical Effects of Herbal Product Containing Rosa mutiflora Roots Extracts as a Main Component on the Hair Growth Promotion (찔레뿌리 추출물을 주성분으로 하는 천연물 조성물의 발모촉진 효과에 관한 전임상 및 임상시험 연구)

  • In this study, to develop hair growth agents using natural products which has excellent ability to promote hair growth effect and fewer side effect, animal experiment and clinical trials was performed to evaluate hair growth promotion effect of herbal product containing $Rosa$ $mutiflora$ roots extracts as a main component (RMHP). 4-week-old male C57BL/6 mice were removed the dorsal hair using thioglycolate, and applied 15% EtOH solution as a negative control, 5% minoxidil as a positive control and RMHP to dorsal skin. In the results of macroscopy and photo-interpretation, RMHP group recorded 100% (+++++) of hair growth was proved to significantly stimulate hair growth against 80% (++++) negative control group. 37 patients were treated with RMHP and evaluated the therapeutic effect at 16 weeks. Hair density was significantly increased at 16 weeks after applying RMHP ($125.0{\pm}4.9\;FU/cm^2$) compared to before treatment ($104.3{\pm}4.7\;FU/cm^2$, p < 0.05), and hair thickness were also significantly increased ($0.066{\pm}0.003$ mm) than before treatment ($0.055{\pm}0.002$ mm, p < 0.05). The result of clinical photo-interpretation using 7-point rating scale assessment, after 16 weeks clinical symptoms were evaluated to significantly improve with $1.23{\pm}0.05$ (p < 0.05). Therefore, the results of this study were observed that RMHP have hair loss prevention effect and hair growth promotion effect to hair loss patients.

Wnt/β-catenin signaling activator restores hair regeneration suppressed by diabetes mellitus

  • Diabetes mellitus is one of the most prevalent diseases in modern society. Many complicationssuch as hepatic cirrhosis, neuropathy, cardiac infarction, and so on are associated with diabetes. Although a relationship between diabetes and hair loss has been recently reported, the treatment of diabetic hair loss by Wnt/β-catenin activators has not been achieved yet. In this study, we found that the depilation-induced anagen phase was delayed in both db/db mice and high-fat diet (HFD) and streptozotocin (STZ)-induced diabetic mice. In diabetic mice, both hair regrowth and wound-induced hair follicle neogenesis (WIHN) were reduced because of suppression of Wnt/β-catenin signaling and decreased proliferation of hair follicle cells. We identified that KY19382, a small molecule that activates Wnt/β-catenin signaling, restored the capabilities of regrowth and WIHN in diabetic mice. The Wnt/β-catenin signaling activator also increased the length of the human hair follicle which was decreased under high glucose culture conditions. Overall, the diabetic condition reduced both hair regrowth and regeneration with suppression of the Wnt/β-catenin signaling pathway. Consequently, the usage of Wnt/β-catenin signaling activators could be a potential strategy to treat diabetes-induced alopecia patients.

Promoting Effect of a Mixture of 8 Herbal Extracts (SPELA 707) on Hair Growth

  • In Korean folk medicine, several herbs, Glycyrrhizae Radix, Persicae Semen, Salviae Radix, Angelicae Gigantis Radix, Zanthoxyli Fructus, Ginseng Radix Alba, Cnidii Rhizoma, and Carthami Flos, are known to enhance blood circulation and have wound healing or anti-inflammatory effects. These pharmacological actions prompted us to investigate whether these herbs might stimulate hair growth. Thus, using a mixture of their extracts called SPELA 707, we investigated their effects and found that SPELA 707 possessed significant hair cycle converting activity from the telogen phase to the anagen phase in C3H mice. Furthermore, we found that SPELA 707 enhanced the hair density in subjects with hair loss and also promoted the conversion of hair into the anagen phase in subjects with androgenetic alopecia. In addition, hair growth promotion effect of SPELA 707 occurred through inhibition of steroid $5{\alpha}$-reductase activity, which is known to block hair growth. Taken together, these results suggest that SPELA 707 has a potential to be used for the treatment of hair loss.

The Hairless Gene: A Putative Navigator of Hair Follicle Development

  • The Hairless (HR ) gene regulates the expression of several target genes as a transcriptional corepressor of nuclear receptors. The hair follicle (HF), a small independent organ of the skin, resides in the epidermis and undergoes regenerative cycling for normal hair formation. HF development requires many genes and signaling pathways to function properly in time and space, one of them being the HR gene. Various mutations of the HR gene have been reported to cause the hair loss pheno-type in rodents and humans. In recent studies, it has been suggested that the HR gene is a critical player in the regulation of the hair cycle and, thus, HF development. Furthermore, the HR gene is associated with the Wnt signaling pathway, which regulates proliferation and differentiation of cells and plays an essential role in hair and skin development. In this review, we summarize the mutations responsible for human hair disorders and discuss the roles of the HR gene in HF development.
