Objective : Recently, hair has become one of the important aspects of beauty. So, there are many studies about prevention and treatment of alopecia. Finasteride and minoxidil have been known to drug for alopecia treatment. However, these agents have side effects such as irritation, pruritus, and erythema when they were used for a long time. So, we assessed hair growth effect on herbal mixture (RAA) include in Rehmanniae Radix Preparata, Achyranthis Radix, and Acanthopanacis Cortex. Methods : 6 weeks aged C57BL/6 mice were divided three treatment groups. : CON group (vehicle solution), MNXD group (positive control, 3% minoxidil), RAA group (15% RAA solution). And we applied 200 ㎕ of three groups to shaved dorsal skin every day. Hair growth effects of treatment were determined through phototrichogram by folliscope and hair follicle morphometry by H&E staining. And we assessed hair growth-related gene (VEGF, IGF-1, TGF-β1) expressions by western blot and 5-α reductase II analysis of dermal (skin) or internal organ (prostate gland). Results : Hair density and hair follicle size in MNXD or RAA group was enhanced compared to those of CON group, respectively. Also, the protein expression levels in dermal of VEGF, IGF-1 increased but TGF-β1 decreased in RAA and MNXD group, compared to CON group, respectively. 5-α reductase II levels of tissues in MNXD or RAA group significantly decreased compared to those of CON group, respectively. Conclusion : These results suggest that RAA has the promoting effect on hair growth.

Gray hair is a representative sign of aging. Intrinsic aging, stress, and the external environment cause hair graying. Stress is known to be a major factor in the early onset of hair graying. We previously found that Pueraia lobata root extract (PLRE) can prevent hair graying by promoting melanin formation. However, it remains unknown whether PLRE can prevent hair graying induced by conditions of stress. In this study, we confirmed the effect of PLRE on stress-induced hair graying. A reporter cell line was newly constructed to confirm the expression of microphthalamia-associated transcription factor (MITF), the main transcription factor for melanin production. MITF expression and melanin pigmentation were reduced in human hair follicle tissue treated with the stress hormone cortisol or H2O2 to induce oxidative stress. PLRE treatment restored MITF expression and increased the amount of melanin pigment in the hair follicle. The expression of Tyrosinase related proteins-2 (TRP-2), a melanin synthesis enzyme in the hair follicle, also increased. In conclusion, PLRE can effectively prevent the inhibition of melanin synthesis by stress hormones and oxidative stress.

The effect of a chronic programme of either low- or moderate-to-high-intensity treadmill running on the activation of the Extracellular-signal regulated protein kinase (ERK1/2), Phosphorylated ERK 1/2(pERK1/2) and the Phosphorylated c-Jun N-terminal kinase(pJNK) pathways was determined in rat Back skin Hair follicle. Sprague-Dawley rats were assigned to one of three groups: (i) sedentary group(NE; n=10); (ii) low-intensity exercise group (Bm/min; LIE; n=10); and (iii) moderate-high-intensity exercise group(28m1min; HIE; n=10). The training regimens were planned so that animals covered the same distance and had similar utilization for both LIE and HIE exercise sessions. The report runs as follows; A single bout of LIE or HIE following 4 weeks of exercise led to a twofold increase in the phosphorylation of ERK2, pERK2 and a threefold increase in pJNKl, pERKl. ERKI phosphorylation in LIE Back skin sampled and pJNK2 in HIE Back skin sampled 48h after the last exercise bout was similar to sedentary values, while pJNK2 phosphorylation in LIE Back skin sampled was 70-80% lower than sedentary. 48h after the last exercise bout of LIE or HIE increased ERK2, pERKl and pJNKl expression, with the magnitude of this increase being independent of prior exercise intensity or duration. PERK1/2, pJNKl expression was increased Three- to fourfold in Back skin Hair follicle sampled 48h after the last exercise bout irrespective of the prior exercise programme, but ERKI expression in HIE Back skin sampled was approximately 90% lower than sedentary values. In conclusion, exercise-training of different jntensities/durations results in selective postexercise activation of intracellular signal pathways, which may be one mechanism regulating specific adaptations induced by diverse training programmes.

The effects of four extracts of medicinal herbs, Rubus coreanum, Acorus calamus, Morus alba and Rehmannia glutinosa on hair growth activity and acnes control were investigated. In the course of screening natural extracts for hair growth, we found that the extract of dried root of Rubus coreanum has the hair growth promoting effect. After topical application of these extracts to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by treatment of these extracts. Furthermore these extracts do not possesspotent inhibitory effect on $5{\alpha}-reductase$ I and II activity and anti-bacterial effect on Escherichia coli , Propionibacterium acnes, Pityrosporum ovale, Staphylococcus aureus, Staphylococcus epidemidis, and Candida albicans. RT-PCR analysis showed that these extracts did notinduce mRNA levels of growth factors such as insulin-like growth factor-I, keratinocyte growth factor, hepatocyte growth factor and vascular endothelial growth factor in dermal papilla cells. These results suggest that Rubus coreanum has hair growth promoting effect. However, the effects of these materials on the hair growth promotion are not mediated through inhibition of $5{\alpha}-reductase$ I and II activity, stimulation of hair follicle cells and expression of growth factors in the dermal papilla cells.

Cortex Mori radicis has been used as a components of antidiabetics, antiasthma and diuresis in Oriental Medicine. This experiment examined the effect of an extracts, obtained from the acetone(CM-A) and aqueous(CM-W) extracts of Cortex Mori radicis, on hair growing activity of the C57BL/6N mice after topical application to skin. We investigated the number of hair follicle and mast cells, and changes of subpopulation of splenocytes and thymocytes in skin for 16 day. The results were as follows : The Hair growing effect in experimental groups was more increased in 85%(CM-A) and 90%(CM-W) than control group(10%) in hair depilated area. The number of hair follicle in experimental groups(CM-A and CM-W) was more increased than control group. Splenic B/T lymphocytes of CM-A group were decreased compare to control group. CD4/CD8 positive TH cells in splenic T lymphocytes of CM-W group were increased compare to control group. These results suggest that CM-A and CM-W may be used in treatment of alopecia areata.

Background: The human hair follicle undergoes cyclic phases-anagen, catagen, and telogen-throughout its lifetime. This cyclic transition has been studied as a target for treating hair loss. Recently, correlation between the inhibition of autophagy and acceleration of the catagen phase in human hair follicles was investigated. However, the role of autophagy in human dermal papilla cells (hDPCs), which is involved in the development and growth of hair follicles, is not known. We hypothesized that acceleration of hair catagen phase upon inhibition of autophagy is due to the downregulation of Wnt/β-catenin signaling in hDPCs, and that components of Panax ginseng extract can increase the autophagic flux in hDPCs. Methods: We generated an autophagy-inhibited condition using 3-methyladenine (3-MA), a specific autophagy inhibitor, and investigated the regulation of Wnt/β-catenin signaling using the luciferase reporter assay, qRT-PCR, and western blot analysis. In addition, cells were cotreated with ginsenoside Re and 3-MA and their roles in inhibiting autophagosome formation were investigated. Results: We found that the unstimulated anagen phase dermal papilla region expressed the autophagy marker, LC3. Transcription of Wnt-related genes and nuclear translocation of β-catenin were reduced after treatment of hDPCs with 3-MA. In addition, treatment with the combination of ginsenoside Re and 3-MA changed the Wnt activity and hair cycle by restoring autophagy. Conclusions: Our results suggest that autophagy inhibition in hDPCs accelerates the catagen phase by downregulating Wnt/β-catenin signaling. Furthermore, ginsenoside Re, which increased autophagy in hDPCs, could be useful for reducing hair loss caused by abnormal inhibition of autophagy.

Cytokeratin 19 (CK19) expressed in epidermis of skin, bulge region of hair follicle, outermost layer of outer root sheath and proximal and distal to bulge. Vimentin is a fibrous protein that localized in cytoplasm of fibroblast and forms cytoskeleton to maintain shape of cell and nucleus. In this study, CK19 and vimentin in skin were confirmed with light, fluorescence and transmission electron microscope. As a result, CK19 was localized epidermis, hair follicles, outer root sheath and nucleus of Merkel's cell. However, vimentin was localized some epidermis, dermis, hypodermis and nucleus of Merkel's cell. The role of CK19 is self-renewal and homeostasis in skin. Also, hair follicle regeneration and hair growth is known to be related. It is supposed that required of structural proteins that make up cytoskeleton is increased. Thereby, expression of CK19 is increased. It is considered that vimentin localized in order to stabilize structure of cell and cytoskeleton of fibroblasts. Also, CK19 and vimentin present in nuclei of Merkel's cell, and to act as a fibrous protein that make up end of a nerve fiber present in Merkel's cell and paracrine function of Merkel's cell.
The recent development of methods for culturing hair follicles in vitro has proved an important tool to investigate many aspects of drug screening. Hair follicles develop as a result of epithelial-mesenchymal interactions between epidermal keratinocytes and dermal cells. We isolated some follicle cells using explantation and enzymatic digestion method from human scalp hair follicles. So we could culture some follicular cells, such as outer root sheath (ORS) cells, dermal papilla (DP) cells, dermal sheath (DS) cells, matrix cells and melanocytes. To induce hair morphogenesis in vitro the cells were 3-D cultured as skin structures. Moreover, to develop hair follicel organ culture model, we applied dermal equivalent (DE) to culturing hair follicles to expand hair growth period.

Many investigators have tried to develop models to prolong of the period of hair elongation in vitro. However these are limited in submerged culture, which don't work due to the lack of cell-cell interactions which are abundant in vivo environment. So we applied dermal equivalent (DE) to culturing flair follicles to prolong hair growth period.
