Studies on the development of the tick (Haemaphysalis longicornis)-vaccine (I) - Immune responses on the crude soluble - (진드기 백신 개발을 위한 기초연구(I) - 수용성 항원에 대한 면역반응에 관하여 -)

  • Haemaphysalis longiscornis is the common cattle tick of great economic importance in Korea. Chemical control using dips or sprays has been the traditional method of attempting to kill these ticks during the infestation period. However, the presence of resistant forms to chemical, the rising costs of acaricides and environmental problems have made it almost impossible to use these chemicals on a regular basis according to the pest problem. For this reason, vaccination against ticks and breeding for host resistance against ticks are being studied. In order to determine the common proteins and antigens according to developmental stages, SDS-PAGE and western blotting were performed. In SDS-PAGE 103.3kD and 98.3kD proteins were observed as common proteins, and these proteins were observed as common antigens in western blotting. Unimmunized rabbits were infestated three times with H longicornis. The weight of the second and the third engorged ticks were 0.153g and 0.104g respectively. This weight is 69% and 47% of the first engorged ticks weight respectively. Immunized rabbits by adult ticks antigen and control were infested with H longicornis. The control taked 3-4 days to fully engorge, but the immunized rabbits taked about 7 days. So adult tick antigen may be effective to render the immunity to host.

  • 진드기의 흡혈(吸血)에 대對하여 경험(經驗)이 없는 가토(家兎)를 실험숙주(實驗宿主)로하여 Haemaphysalis longicornis 진드기를 부착(附着) 흡혈(吸血)시켰을때 형성(形成)된 교상(咬傷)에 대(對)한 조직병리학적(組織病理學的) 변화(變化)를 관찰(觀察)하고, 진드기의 흡혈(吸血)에 경험(經驗)이 있는 감작(感作)된 가토(家兎)에 1개월(個月) 간격(間隔)으로 제2차(第二次) 흡혈(吸血)까지 시도(試圖)하여 가토체내(家兎體內)의 획득면역(獲得免疫)의 형성(形成)과 그에 따른 조직병리학적(組織病理學的) 변화(變化)를 비교관찰(比較觀察)하여 다음과 같은 결론(結論)을 얻었다. 1. H. longicornis 진드기는 구기(口器)를 사용(使用)하여 가토(家兎) 이부위(耳部位)의 피용(皮庸)를 천자(穿刺)하므로서 교상(咬傷)을 일으키며, 흔히 진드기의 구기주변(口器週邊)에는 진드기의 안전(安全)한 부착(附着)을 기도(企圖)하는 편평원추상(扁平圓錐狀)의 시멘트 물질(物質)이 형성(形成)되었다. 2. 진드기 교상(咬傷)에 대(對)한 주요(主要)한 조직병리학적(組織病理學的) 변화(變化)는 국소부위(局所部位)의 출혈성(出血性) 염증성(炎症性) 반응(反應), 광범위(廣範圍)한 부종(浮腫), 백혈구(白血球) 침윤(浸潤), 그리고 세포조직(細胞組織)의 괴사(壞死)로 대징(待徵)지워졌다. 3. 진드기 기생(寄生)의 초기(初期)에 형성(形成)된 병변(病變) 또는 진드기 흡혈(吸血)에 대(對)하여 경험(經驗)이 없는 가토(家兎)의 병변(病變)에서는 호중구(好中球)를 흔히 볼 수 있었으며, 진드기 기생말기(寄生末期) 또는 진드기 흡혈(吸血)에 대(對)하여 경험(經驗)이 있는 감작(感作)된 가토(家兎)의 병변(病變)에서는 호산구(好酸球)가 많이 관찰(觀察)되었다. 4. 진드기의 구기(口器)가 창상내(創傷內)에 잔존(殘存)하는 한, 치유(治癒)은 단시일내(短時日內)에 이루어 지지 않았으며, 증상(症狀)이 진행(進行)됨에 따라 괴사성(壞死性) 염증성(炎症性) 반응(反應)은 더욱 확대(擴大) 되었는데, 이러한 소견(所見)은 감작(感作)되어 있는 숙주(宿主)에서 더욱 확실(確實)히 관찰(觀察)되었다. 5. 최초(最初)(1차(一次)) 흡혈(吸血) 시도후(試圖後) 감작(感作)된 가토(家兎)에 2차적(二次的)으로 부가흡혈(附加吸血)을 시도(試圖)하였을때 볼수 있는 진드기의 흡혈성취도(吸血成就度)의 저하현상(低下現象)은 숙주체내(宿主體內)의 교상병변(咬傷病變)에서 일어나는 조직병리학적(組織病理學的) 소견(所見)과 획득면역(獲得免役)에 깊이 관련(關聯)되어 있는 것으로 사료(思料)된다.

Ticks Collected from Wild and Domestic Animals and Natural Habitats in the Republic of Korea

  • Ticks were collected from 35 animals from 5 provinces and 3 metropolitan cities during 2012. Ticks also were collected by tick drag from 4 sites in Gyeonggi-do (2) and Jeollabuk-do (2) Provinces. A total of 612 ticks belonging to 6 species and 3 genera were collected from mammals and a bird (n=573) and by tick drag (n=39). Haemaphyalis longicornis (n=434) was the most commonly collected tick, followed by H. flava (158), Ixodes nipponensis (11), Amblyomma testudinarium (7), H. japonica (1), and H. formosensis (1). H. longicornis and H. flava were collected from all animal hosts examined. For animal hosts (n>1), the highest Tick Index (TI) was observed for domestic dogs (29.6), followed by Siberian roe deer (17.4), water deer (14.4), and raccoon dogs (1.3). A total of 402 H. longicornis (adults 86, 21.4%; nymphs 160, 39.8%; larvae 156, 38.9%) were collected from wild and domestic animals. A total of 158 H. flava (n=158) were collected from wild and domestic animals and 1 ring-necked pheasant, with a higher proportion of adults (103, 65.2%), while nymphs and larvae only accounted for 12.7% (20) and 22.2% (35), respectively. Only 7 A. testudinarium were collected from the wild boar (6 adults) and Eurasian badger (1 nymph), while only 5 I. nipponensis were collected from the water deer (4 adults) and a raccoon dog (1 adult). One adult female H. formosensis was first collected from vegetation by tick drag from Mara Island, Seogwipo-si, Jeju-do Province.

  • Severe fever with thrombocytopenia syndrome virus (SFTSV) is a zoonotic, tick-borne RNA virus of the genus Bandavirus (Family Phenuiviridae), mainly reported in China, Japan, and the Republic of Korea (Korea). For the purpose of this study, a total of 3,898 adult and nymphal ticks of species Haemaphysalis longicornis (94.2%), Haemaphysalis flava (5.0%), Ixodes nipponensis (0.8%), and 1 specimen of Ixodes ovatus, were collected from the Deogyusan National Park, Korea, between April 2016 and June 2018. A single-step reverse transcriptase-nested PCR was performed, targeting the S segment of the SFTSV RNA. Total infection rate (IR) of SFTSV in individual ticks was found to be 6.0%. Based on developmental stages, IR was 5.3% in adults and 6.0% in nymphs. The S segment sequences obtained from PCR were divided into 17 haplotypes. All haplotypes were phylogenetically clustered into clades B-2 and B-3, with 92.7% sequences in B-2 and 7.3% in B-3. These observations indicate that the Korean SFTSV strains were closer to the Japanese than the Chinese strains. Further epidemiological studies are necessary to better understand the characteristics of the Korean SFTSV and its transmission cycle in the ecosystem.

  • This study includes the taxonomy and occurrence of ticks and mites from Jinju, Kyogsangnamdo province, Korea. Materials examined were attraccted to carrions of chicken and cattle flesh from September 2002 to December 2003. As a result, the following seven species were identified. Suborder Ixodida Family Ixodidae 1. Haemaphysalis longicorttis Neumann, 1901 Suborder Mesostigmata Family Machrochelidae 2. Macrocheles muscaedomesticae (Scopoli, 1772) 3. Holostaspella ornata (Berlese, 1904) Family Ascidae 4. Protogamasellus micus (Athias-Henriot, 1961) 5. Lasioseius sugawarai Ehara, 1964 6. Proctolaelaps sp. Suboder Prostigamta Family Erythraeidae 7. Abrolophus sp. Of these, M. muscaedomesticae and Protogamasellus micus are dominant species. Holostaspella ornate(Berlese, 1904) and Protogamasellus mica(Athias-Henriot, 1961) are new to Korean fauna.

Distribution of ticks carrying Severe Fever with Thrombocytopenia Syndrome Virus (SFTSV) around Jiri walking trails of Jeollanam-do, Korea (지리산둘레길에서 채집한 참진드기의 분포와 참진드기에서의 중증열성혈소판감소증후군바이러스 검출)

  • Severe fever with thrombocytopenia syndrome (SFTS) is an emerging disease characterized by fever and thrombocytopenia. Haemaphysalis longicornis ticks comprise the major population of ticks in the environment and have been considered as the main vector for SFTS virus (SFTSV). Here we investigated the distribution of ticks carrying SFTSV collected from the environment using the dragging or sweeping methods during April~October 2015 in Jeollanam-do, Korea. Sampling was taken from Songjeong, Omi, Bangkwang, Sandong areas in Jiri walking trails. Among the total 3,869 ticks collected, 3,823 ticks (98.8%) were H. longicornis, 41 (1.1%) were Amblyomma testudinarium, and 5 (0.1%) were Ixodes nipponensis. Classification results by regional groups of H. longicornis indicated that 1,613 ticks were collected in Sandong, 1,190 ticks in Omi, 603 ticks in Bangkwang, and 417 ticks in Songjeong. In monthly distributional studies of H. longicornis based on the developmental stages, nymph (325 ticks) was collected from May to October, 94% of larvae from April to June, and 94% of adult from June to August. These results showed the different dominant stage of ticks according to seasons. However, no SFTSV-specific gene was detected in 3,823 ticks of H. longicornis, 41 of A. testudinarium and 5 of I. nipponensis.

Rhipicephalus sanguineus (Latreille 1806); A new Record of Male Tick Identified with Scanning Electron Microscopy in Korea (Rhipicephalus sanguineus에 대한 최초(最初)의 국내보고(國內報告)와 수컷에 대한 주사전자현미경적관찰(走査電子顯微鏡的觀察))

  • Rhipicephilus sanguineus (Latreille 1806) the brown dog tick, is one of the most widely distributed ixodid ticks in the world, however, there was no report for the discovery of the species in Korea. Specimens unfed or partly fed were collected from the dogs rearing in the livestock farms and the pastures near the farms located in Kyunggi-Do, Chungchong-Namdo, Cholla-Pukdo and Cheju-Do, in summer seasons during 1981 to 1984. The specimens were examined at the Institute of Veterinary Research in Anyang and were identified as R. sanguineus. The morphological characteristics observed by means of the scanning electron microscope were presented with some SEM microphotographs. As. R. sanguineus is firstly discovered to occur in Korea, a total of 20 species of ticks belonging to E: genera, such as, Argas, Amblyomma, Boophilus, Dermacentor, Haemaphysalis, Hyalomma, Ixodes and Rhipicephilus, has now been recorded.

Changes in Protein Phosphorylation during Salivary Gland Degeneration in Haemaphysalis longicornis

  • The ticks feed large amount of blood from their hosts and transmit pathogens to the victims. The salivary gland plays an important role in the blood feeding. When the female ticks are near engorgement, the salivary gland gradually loses its functions and begins to rapidly degenerate. In this study, data-independent acquisition quantitative proteomics was used to study changes in the phosphorylation modification of proteins during salivary gland degeneration in Haemaphysalis longicornis. In this quantitative study, 400 phosphorylated proteins and 850 phosphorylation modification sites were identified. Trough RNA interference experiments, we found that among the proteins with changes in phosphorylation, apoptosis-promoting Hippo protein played a role in salivary gland degeneration.
