한국산 진드기의 분류학적 연구(III) (Haemaphysalis 속) (A Taxonomical Study on Ticks in Korea(III) The Genus Haemaphysalis)

  • 한국산 Haemaphysalis 속 진드기는 Ogura and Takada (1927), Akasawa (1938), Jishida(1936), Itagaki Noda, Yamaguchi (1944) 등에 의하여 Haemaphysalis bispinosa, H. campamulata, H. flava의 3 종이 보고되었다. 저자가 1963년 6월 이후 채집한 진드기류를 정리한 결과 Haemaphysalis cornigera , H. Kutchensis 가 한국미기록종임이 밝혀졌으므로 이에 발표하는 바이다.

Status of Haemaphysalis tick infestation in domestic ruminants in Iran

  • The geographical distribution and ecological preferences of Haemaphysalis in domestic animals in Iran were studied 4 times a year from April 2003 to March 2005. A total of 1,622 ixodid tick specimens were collected from 3 different zones. Among them, 108 (6.7%) Haemaphysalis ticks, consisting of 6 species, were identified; H. punctata (3.4%), H. parva (0.5%), H. sulcata (0.6%), H. choldokovskyi (1.7%), H. concinna (0.06%) and Haemaphysalis sp. (0.6%). H. punctata was the most abundant species, whereas H. concinna was the rarest species collected in humid and sub-humid zones on cattle, sheep and goats. H. choldokovskyi was principally collected from sheep and goats grazed in cold mountainous areas. The infested areas consisted of Caspian Sea (Guilan, Mazandaran, Golestan, and central provinces), mountainous (Azarbaiejan, Ardebil, Kohgilouyeh, and Kordestan) and semi-dessert (Khorasan, Semnan, Herman, Sistan, and Baluchestan) zones. The Caspian Sea zone (23.6%) was the most highly infested region. The results show that various species of Haemaphysalis ticks infest domestic ruminants in Iran and each tick species show characteristic geographical distributions.

북한산 둘레길에서 참진드기의 계절적 발생과 중증열성혈소판감소증후군 바이러스 검출 (Seasonal Prevalence of Ticks at Bukhansan Dullegil and Detection of Severe Fever with Thrombocytopenia Syndrome Virus)

  • 2019년 4월부터 10월까지 북한산 둘레길 12구간(지축동), 14구간(울대리), 18구간(호원동)에서 참진드기를 채집해서 계절적 분포 조사와 매개체의 SFTS 바이러스 감염 여부를 확인하였다. 천젓기와 천끌기 방법으로 개피참진드기, 일본참진드기와 중증열성혈소판감소증후군의 주된 매개체로 알려진 작은소피참진드기 3종이 채집되었다. 작은소피참진드기는 채집비율이 지축동 91%, 울대리 94%, 호원동 98%를 차지하여 우점종이었다. 작은소피참진드기의 성숙단계에 따른 계절별 채집비율을 보면 성충의 경우 세 지역 모두 9월에 최고치를 보였고 약충의 경우에는 4월에서 6월 사이에 주로 채집되었다. 유충의 경우에는 9월과 10월에 최고치를 보였다. 채집된 작은소피참진드기 1,158개체를 성숙단계별로 2× OneStep RT-PCR과 nested PCR로 SFTS 바이러스 검사를 한 결과 양성은 나타나지 않았다. 2010년 개장 이후 이용객이 늘어나는 북한산 둘레길에서 기후 온난화로 증가가 예상되는 참진드기 매개 질환에 대한 위험분석과 효율적인 예방에 본 조사 결과가 활용될 수 있다.

Molecular Detection of Toxoplasma Gondii in Haemaphysalis Ticks in Korea

  • Toxoplasma gondii are intracellular protozoa that can cause neurological disease or death in fetuses and even in immunocompromised human adults. Ticks are recognized as vectors of many microorganisms including viruses, bacteria, and protozoa. Recent studies detected T. gondii in various tick species in many countries. In this study, we performed PCR detection of the T. gondii B1 gene from Haemaphysalis ticks collected from vegetation in 4 localities, Wonju, Gunsan, Miryang, and Yangsan, in Korea. We analyzed DNA from 314 ticks (268 Haemaphysalis longicornis and 46 Haemaphysalis flava) and the B1 gene of T. gondii was detected in 13 of these. The detection of T. gondii in ticks differed significantly by region (P=0.021). T. gondii was detected in the following percentages of collected ticks: 3.7% (7 of 189) in Gunsan, 10% (5 of 50) in Wonju, 16.7% (1 of 6) in Yangsan, and 0% (0 of 69) in Miryang. The detection of T. gondii in ticks was not associated with tick species or development stage. This is the first report of T. gondii detection in ticks in Korea. Our results provide important information necessary to understand toxoplasmosis transmission.

Immunization effect of recombinant P27/30 protein expressed in Escherichia coli against the hard tick Haemaphysalis longicornis (Acari: Ixodidae) in rabbits

  • We investigated the induction of resistance to Haemaphysalis longicornis infestation in rabbits that had been immunized with recombinant H. longicornis P27/30 protein. The success of immunological control methods is dependent upon the use of potential key antigens as tick vaccine candidates. Previously, we cloned a gene encoding 27 kDa and 30 kDa proteins (P27/30) of H. longicornis, and identified P27/30 as a troponin I-like protein. In this study, rabbits that were immunized with recombinant P27/30 expressed in Escherichia coli showed the statistically significant longer feeding duration for larval and adult ticks (P<0.05), low engorgement rates in larval ticks (64.4%), and an apparent reduction in egg weights, which suggest that H. longicornis P27/30 protein is a potential candidate antigen for a tick vaccine. These results demonstrated that the recombinant P27/30 protein might be a useful vaccine candidate antigen for biological control of H. longicornis.

No Detection of Severe Fever with Thrombocytopenia Syndrome Virus from Ixodid Ticks Collected in Seoul

  • Larvae, nymphs, and adult stages of 3 species of ixodid ticks were collected by tick drag methods in Seoul during June-October 2013, and their infection status with severe fever with thrombocytopenia syndrome (SFTS) virus was examined using RT-PCR. During the period, 732 Haemaphysalis longicornis, 62 Haemaphysalis flava, and 2 Ixodes nipponensis specimens were collected. Among the specimens of H. longicornis, the number of female adults, male adults, nymphs, and larvae were 53, 11, 240, and 446, respectively. Ticks were grouped into 63 pools according to the collection site, species, and developmental stage, and assayed for SFTS virus. None of the pools of ticks were found to be positive for SFTS virus gene.

Resistance and control of cypermethrin and chlorpyrifos as acaricide for control of hard tick Haemaphysalis longicornis (acari: ixodidae)

  • Chemotherapeutic treatment is still the foundation of tick control programs. This study investigated the acaricidal efficacy of cypermethrin alone and in combination with chlorpyrifos against Haemaphysalis (H.) longicornis. Unfed larval ticks were exposed to 0.1, 1.0, and 10 mg/mL cypermethrin for 60 min, after which the acaricidal efficacy was examined based on tick mortality. All compounds showed similar suppression curves, with the best control being achieved by cypermethrin and chlorpyrifos (1 : 1 ratio) at 10 mg/mL. Effective cypermethrin concentrations for tick control were two to seven times higher than the recommended doses, indicating resistance by H. longicornis.

  • A chitinase cDNA named CHT1 was cloned from the hard tick, Haemaphysalis longicornis, and the enzymatic properties of its recombinant proteins were characterized. The CHT1 cDNA encodes 930 amino-acid (aa) residues including a 22 aa putative signal peptide, with the calculated molecular mass of the putative mature protein 104 kDa. The E coli-expressed rCHT1 exhibited weak chitinolytic activity against $4MU-(GlcNAc)_3$. The rCHT1 protein with higher activity was obtained using recombinant Autographa californica multiple nuclear polyhedrosis virus (AcMNPV), which expresses rCHT1 under polyhedrin promoter. These findings suggest that the rCHT1 expressed in baculovirus-mediated Sf 9 cells has a high activity than E coli-expressed rCHT1.

너구리에 기생한 개피참진드기(Hoemaphysalis flava)에 대한 재기술 및 너구리참진드기(Irones tcnuki)의 국내 보고 (Redescription of Haemaphysalis flava and Ixodes tanuki collected from a raccoon dog in Korea)

  • 1995년 3월 전북 완주군 소재 모악산에서 포획된 한 마리의 너구리(Nyctereutes procyonoides koreensis)로부터 총 135개체의 참진드기를 채집하여 2속 2종을 동정하였는데, 개피참진드기(Haemaphysalis flava) 117개체와 너구리참진드기(Ixodes tanuki) 18개체를 확인하였다. 그 중 너구리참진드기(신청)는 국내에 서식하는 것으로 처음확인되었다.

관악산에서 참진드기 조사 및 중증열성혈소판감소증후군 바이러스 검출 (Prevalence of severe fever with thrombocytopenia syndrome virus among ticks surveyed at Mt. Gwanak, Korea)

  • This study was performed to investigate the distribution of ticks and the rate of infection with severe fever with thrombocytopenia syndrome (SFTS) virus in ticks collected at Mt. Gwanak and the Seoul National University campus, Korea. Ticks (n = 273) were collected from May to October and included 76 Haemaphysalis longicornis (4 adult females, 72 nymphs), 49 Haemaphysalis flava (9 adult females, 3 adult males, 37 nymphs), and 148 Haemaphysalis spp. larvae. SFTS virus detection was performed by using one-step RT PCR and nested PCR. The SFTS virus was detected in 7 samples (1 Haemaphysalis longicornis nymph, 3 Haemaphysalis flava nymphs, and 3 Haemaphysalis spp. larva). The overall minimum field infection rate was 2.6%, whereas the minimum field infection rates of adult, nymphal, and larval ticks were 0%, 3.2%, and 2.0%, respectively. For a more accurate indication of the prevalence of SFTS virus in Korea, further in-depth investigations of tick species and SFTS virus occurrence over a larger area and longer period are needed.
