수컷 흰쥐 생식기관에서의 세로토닌 수용체 아형 유전자 발현 (Expression of Serotonin(5-HT) Receptor Isotypes in Reproductive Organs of Male Rat)

  • 세로토닌(serotonin, 5-HT)은 아민류 가운데 카테콜아민과 더불어 중요한 체네 생리 및 행동 조절을 담당하는 신경 전달물질이다. 특히 'Prozac'으로 잘 알려진 항우울제는 5-HT의 세포내 재흡수를 선택적으로 억제하는 약물(selective serotonin reuptake inhibitor, SSRI)로써 광범위하게 사용중인 약물인데, 알려진 부작용으로 사정 능력의 변화가 관찰된다. 이는 5-HT에 의한 성 행동 또는 성 기능 조절기작이 있음을 의미한다. 본 연구에서는 수컷 흰쥐의 성 행동 조절에 관여함이 잘 알려진 5-HT의 중추 신경계 수준에서의 작용 외에 말초 조직 수준에서 직접 작용이 가능한가의 여부를 검증하기 위해서 정소, 정관, 저정낭 등 수컷 흰쥐의 생식 기관들을 대상으로 5-HT 수용체 아형들의 유전자 발현 여부를 조사하였다. 적출된 뇌, 정소, 정관, 저정낭, 음경해면체 등의 장기에서 추출한 total RNA로 5-HT 수용체 각 아형들에 대한 RT-PCR을 시행한 결과, 대조군인 뇌는 물론 모든 생식 기관에서 다양한 5-HT수용체 아형들의 mRNA발현을 확인할 수 있었다. 특히 사정 반응을 담당하는 최종 장기인 저정낭과 정관에서, 지금까지 여러 연구 결과에서 사정 현상에 관여할 것으로 추정되어 온 5-H $T_{1A}$, 5-H $T_{1B}$, 5-H $T_{2C}$ 수용체 아형들이 실제로 발현됨을 관찰하였다. 이 결과는 5-HT에 의한 성 행동 조절이 중추신경계 수준에서는 물론 말초 조직 수준에서도 직접 가능함을 시사하는 것이다. 본 연구의 시도와 같이 5-HT의 다양한 작용기전에 대한 조사는 포유동물 성 행동에 대한 이해 증진과 더불어 성기능 이상의 부작용이 적은 새로운 항우울제의 개발로 연결될 수 있다고 판단된다.다.다.다.ized freezing방법간에 큰 차이가 없음을 볼 수 있었다.유의 경우 $World-Labs^(R)$ 처리구가 가장 높았다.상으로 면역활성 실험을 실시한 결과, 장관면역 활성은 젖산균의 세포질 획분에서 양성 대조군으로 사용한 LPS와 같은 수준의 면역활성을 나타내었고, 특히 Lb. acidophilus속 균주들의 세포벽 획분의 장관 면역활성이 다른 균주보다 높게 나타났다. 한편 젖산균주의 세포질과 세포벽 성분에 대한 비장 림프구의 증식능은 대조군과 비슷한 수준으로 낮게 관찰되었으나, 이들의 대식 세포증식능은 세포벽 및 세포질 모두의 획분에서 대조군보다 높았으며 특히 세포벽 획분의 경우에는 양성대조군인 LPS 보다 높거나 유사한 정도의 높은 활성을 확인할 수 있었다. 이상의 결과로부터 시험 균주 중에서 Lb. acidophilus 균주인 DDS-1과 B-3208이 프로바이오틱스로서 요구되는 조건을 충족시킨다는 것을 알 수 있었으며, 이들 균주의 상업적 이용 가능성을 재차 확인할 수 있었다./TEX>, 함량은 가공된 냉동감자보다 높은 것으로 나타났다. 상온 냉장저장 감자의 vitamin $B_1$, $B_2$ 함량은 각각 6, 4주차 이후부터 급격한 감소를 보였으나, 냉동저장 감자의 경우 48주(12개월)저장 중 함량변화가 없었다. 물성적 품질변화 평가에서 상온 냉장저장 감자의 무게와 부피는 24주간 다소 감소하는 경향을 보였으나 통계적인 유의차는 없었다. 냉동저장 감자의 무게와 부피 변화는 48주간 전혀 없었다. 상온 냉장저장 감자의 조직의 강도(hardness)는 24주간 다소 감소하는 경향을 보였으나 냉동저장 감자는 48주간 hardness의 변화가 전혀 없었다. P3HT:PCBM 활성층을 갖는 유기 박막태양전지의 후속 열처리 효과 (The Post Annealing Effect of Organic Thin Film Solar Cells with P3HT:PCBM Active Layer)

  • 본 연구에서는 P3HT와 PCBM 물질을 전자도너와 억셉터 광활성층 물질로 사용하여 벌크이종접합 구조를 갖는 Glass/ITO/PEDOT:PSS/P3HT-PCBM/Al 구조의 유기박막태양전지를 제작하였다. P3HT와 PCBM은 각각 0.5 wt%의 농도로 톨루엔 용액에 용해하였다. 광활성층 농도를 최적화하기 위하여 P3HT:PCBM= 3:4, 4:4, 4:3 wt%의 농도비로 소자를 제작하고, 농도비에 따른 전기적 특성을 조사하였다. 또한 활성층의 후속열처리 온도가 소자의 전기적 특성에 미치는 영향을 조사하였다. P3HT와 PCBM의 농도비가 4:4 wt%의 비율에서 가장 우수한 전기적 특성을 나타내었으며, 이때 단락전류밀도 ($J_{SC}$), 개방전압 ($V_{OC}$), 및 충실인자 (FF)는 4.7 $mA/cm^2$, 0.48 V 및 43.1%를 각각 나타내었다. 또한 전력변환효율(PCE)은 0.97%의 값을 얻었다. 최적화된 농도비를 갖는 태양전지 소자에 대해 $150^{\circ}C$에서 5분, 10분, 15분, 20분간 후속 열처리를 실시한 결과 P3HT 전자도너의 흡광계수가 증가하는 경향을 보였다. 후속 열처리 조건이 $150^{\circ}C$에서 15분인 경우 전기적 특성이 열처리 하지 않은 소자에 비해 특성이 개선되었다. 즉, 이때의 전기적 특성은 $J_{SC},\;V_{OC}$, FF, PCE의 값이 각각 7.8 $mA/cm^2$, 0.55 V, 47%, 2.0%를 나타내었다.

  • Objectives : This research was performed to investigate the effect of acupuncture at the $KI_2\;KI_{10}\;HT_8\;HT_3$, on Aquaporin-2(AQP2) expression related with the renal functions in rats. Methods : Acupuncture was performed during 100-seconds, 6-times, at 150-seconds intervals under anesthesia in rats. We observed rats' mean arterial pressure(MAP), heart rate(HR), renal sympathetic nerve activity(RSNA) during acupuncture and AQP2 expression by western blot method and atrial natriuretic peptide(ANP), renin, norepinephrine of plasma after decapitation. Results : The AQP2 expression was significantly increased in $HT_8$ group, but decreased in $KI_{10}$ group. Average MAP during 6-times acupuncture was significantly increased in $HT_8$ group. Average HR was significantly increased in $HT_8$ group, Average RSNA was increased in $KI_{10}$ group, but that was marginally increased in $KI_{10}$ group. Plasma renin concentration was increased in $KI_2,\;HT_3$, groups. Plasma ANP show a tendency to decrease in $KI_10\;HT_3$ groups, increased in $KI_2,\;HT_3$ but not significant. Plasma norepinephrine concentration was significantly decreased in $KI_{10},\;HT_3$ groups. Conclusions : These results suggested that acupuncture at $HT_8$ activate renal function to reuptake, but $KI_{10}$ show a decline on effect of AQP2 expression, blood pressure, nerve activity and renin.

5-Hydroxytryptamine의 장억제작용(腸抑制作用) (The Inhibitory Effects of 5-Hydroxytryptamine on the Intestine)

  • The inhibitory effect of 5-hydroxytryptamine (5-HT) on the isolated intestinal strips of the tortoise (Amyda japonica), rat, rabbit and guinea pig was investigated. 1) The strips from the middle or lower part of the tortoise intestine responded with relaxation to 5-HT $(10^{-9}{\sim}10^{-5}g/ml)$, and the magnitude of the relaxation was proportional to the dose of 5-HT. The rectal part of the tortoise intestine, in contrast, showed contraction, the magnitude of which also was proportional to the dose of 5-HT. 2) Various blocking agents such as methysergide, morphine, tetracaine, nethalide, bretylium, hexamethonium, mecamylamine and chlorisondamine, showed no selective blocking activity on the relaxant effect of 5-HT on the tortoise intestine. The inhibitory effect of isoproterenol on the tortoise intestine, however, was selectively blocked by nethalide, and the stimulatory effect of 5-HT on the rectal part of the tortoise was blocked by methysergide. 3) In the presence of 5-HT, the stimulatory effect of DMPP on the tortoise intestine was remarkably attenuated, whereas that of acetylcholine and $BaCl_2$ was little affected. In the presence of isoproterenol, the stimulatory effect of acetylcholine and $BaCl_2$ were affected, but that of DMPP was little affected. 4) Large dose of 5-HT($10^{-4}$g/ml) produced inhibitory effect on the strips from the distal part of the isolated colon of the rat, rabbit and guinea pig, when the strips had been exposed to 5-HT($10^{-4}$g/ml), methysergide or phen`oxybenzamine. 5) Bretylium, as well as nethalide, abolished or remarkably reduced, in a few cases of the experiments, the inhibitory effect of the large dose of 5-HT on the distal part of the colon, whereas morphine did not affect it. 6) The ileal strips of the guinea pig also showed relaxation, as in the colonic strips, having been exposed to the large dose of 5-HT or phenoxybenzamine. This effect, however, was not obsered in the case of the rabbit ileum. 7) The property of the action-site of 5-HT in the tortoise intestine seemd to be different from the 5-HT receptors which have been revealed by several investigators. 8) Adrenergic component seemed to be participated in the inhibitory effect of 5-HT on the colon of the rat and rabbit.

남자 정신분열병 환자에서 성기능장애에 대한 검토 (Review of Sexual Dysfunction in Male Schizophrenics)

  • 1) 성기능장애 빈도는 정신분열병 환자군이 80%로 대조군 42%에 비해 유의하게 높았다(p<0.001). 성반응 주기에 따른 장애유형은 성적욕망장애 76%, 발기장애 75%, 발기유지장애 75%, 성적극치감의 질적감소 61%, 지루 50%, 사정시 정액량의 감소 44% 순서로 많았다. 성적극치감 횟수의 감소는 32%로 다른 장애유형에 비해 적었으며, 조루를 호소하는 경우도 15%나 되었다. 2) PRL 및 5-HT 측정치는 환자군에서 $28.5{\pm}20.6ng/ml$, $298.5{\pm}89.1ng/ml$으로서 대조군 $10{\pm}5.6ng/ml$, $169.2{\pm}37.8ng/ml$보다 유의하게 높았다(p<0.001). TST 측정치는 환자군이 $4.3{\pm}1.5ng/ml$, 대조군이 $4.5{\pm}1.2ng/ml$로 양군 모두 유의한 차이가 없었다. 3) 성기능장애를 증가시키는 변인은 결혼(미혼), 초발연령, 이병기간, 입원기간, 총약물투여기간 그리고 5-HT 측정치이었다. 특히 5-HT 비정상치군이 정상치군보다 성기능장애가 유의하게 높았다. 그러나 연령, 교육연한, 종교, 경제상태, PRL 및 TST 측정치, 항정신병약물용량, 약물역가, benztropine, ACSE, CGI, BPRS, PANSS, PANSS-CF, MMSE-K 점수와 성기능장애와는 무관하였다. 성기능장애는 정신분열병군에서 높았고, 성기능장애 유형별로는 성적욕망장애와 발기장애가 많았다. 미혼이거나, 초발연령이 높을수록, 질병에 이환된 기간이 길수록, 5-HT 농도가 높을수록 성기능장애가 증가하였다. 비록 혈중 5-HT 농도가 뇌 5-HT 활성을 어느정도 반영하는지는 알 수 없으나, 정신분열병 환자의 일차 병인인 과도한 5-HT 활성으로 인한 직접적인 성기능 억제효과 때문에 장애가 초래한다고 생각된다.

Inhibition of Proinflammatory Cytokine-induced Invasiveness of HT-29 Cells by Chitosan Oligosaccharide

  • The effect of chitosan oligosaccharide (COS, 1 kDa${\gamma}$, 10 ng/ml IL-$1{\alpha}$, and 25 ng/ml TNF-${\alpha}$) in HT-29 cells. Inducible nitric oxide synthase (iNOS) expression induced by these cytokines was inhibited by COS. COS pretreatment inhibited the invasiveness of cytokines-treated HT-29 cells through Matrigel-coated membrane in a dose-dependent manner. COS also inhibited cytokines-induced matrix metalloproteinase (MMP)-2 activity. This study shows that proinflammatory cytokines induce NO production, iNOS expression, and invasiveness of human colorectal adenocarcinoma HT-29 cells. COS pretreatment inhibited cytokines-mediated NO production, iNOS expression, and invasiveness of HT-29 cells. These results provide sufficient information for the further development of COS as an antitumor metastatic agent for the treatment of colon cancer.

수종생약의 향 세로토닌작용 (Antagonistic Activities of Several Medicinal Plants on Serotonin-Mediated Actions)

  • Serotonin (5-hydroxytryptamine, 5-HT) has been known to have peripheral actions as well as central actions. Recently physiological roles of 5-HT as a central neurotransmitter have been studied in detail and this is a subject of the present study. Plausible central actions mediated by 5-HT receptor seem to be a control of appetite, bring about psychiatric disorders such as anxiety or dementia, and migraine headache. In this study, thirty-five medicinal plants were tested for their antagonistic activities to 5-HT mediated actions. Through the preliminary screening, methanol extracts of three medicinal plants (Saussureae Radix, Sanguisorbae Radix and Xanthii Fructus) were shown to possess relatively specific antagonistic activities to 5-HT mediated ileal contraction over acetylcholine. To luther investigate the central antagonistic activities of the selected plant in vivo, m-chlorophenylpiperazine (mCPP) which is known to be a 5-H $T_{1c}$ receptor agonist was injected to mouse to induce an anxious and/or hypolocomotion states, and also social interaction test, which was based on the method described by File (S.E. File, 1980), was performed to see whether ethylacetate fraction of Sanguisorbae Radix methanol extract possessed a specific anxiolytic activity.y.

  • High temperature proton exchange membrane fuel cell (HT-PEMFC) has been regarded as a promising clean energy sources. In this study, a quasi-three-dimensional dynamic model of HT-PEMFC has been developed and the local dynamic characteristics are investigated. The model has the geometrical simplification of 2+1D reduction (quasi-3D). The one-dimensional model consists of nine control volumes in cross-sectional direction to solve the energy conservation and the species conservation equations. Then, the one-dimensional model is discretized into 25 local sections along the gas flow direction to account for gas and thermal transport in channels. With this discretization, the local characteristics of HT-PEMFC such as species conservation, temperature, and current density can be captured. In order to study the basic characteristics of HT-PEMFC, it is important to investigate the local dynamic characteristics. Thus, the model is simulated at various operating conditions and the local dynamic characteristics related to them are observed. The model is useful to investigate the distribution of HT-PEMFC characteristics and the physical phenomena in HT-PEMFC.

흰쥐 해마절편에서 포도당/산소 고갈에 의한 5-hydroxytryptamine 유리변동에 미치는 Adenosine의 영향 (Effect of Adenosine on the Release of $[^3H]-5-hydroxytryptamine$ during Glucose/Oxygen Deprivation from Rat Hippocampal Slices)

  • The effects of adenosine, adenosine A1 receptor antagonist (DPCPX), or NMDA receptor antagonist (APV) on the spontaneous release of $[^3H]-5-hydroxytryptamine$ ($[^3H]-5-HT$) during normoxic/normoglycemic or hypoxic/hypoglycemic period were studied in the rat hippocampal slices. The hippocampus was obtained from the rat brain and sliced $400\;{\mu}m$ thickness with the tissue slicer. After 30 min's preincubation in the normal buffer, the slices were incubated for 30 min in a buffer containing $[^3H]-5-HT$ ($0.1\;{\mu}M,\;74{\mu}Ci/8\;ml$) for uptake, and washed. To measure the release of $[^3H]-5-HT$ into the buffer, the incubation medium was drained off and refilled every ten minutes through sequence of 14 tubes. Induction of glucose/oxygen deprivation (GOD; medium depleting glucose and gassed with 95% $N_2/5%\;CO_2$) was done in 6th and 7th tube. The radioactivities in each buffer and the tissue were counted using liquid scintillation counter and the results were expressed as a percentage of the total radioactivities. When slices were exposed to GOD for 20 mins, the spontaneous release of $[^3H]-5-HT$ was markedly increased and this increase of $[^3H]-5-HT$ release was blocked by adenosine ($10\;{\mu}M$) or DL-2-amino-5-phosphonovaleric acid (APV; $30\;{\mu}M$). Adenosine $A_1$ receptor specific antagonist, 8-cyclopentyl-1,3-dipropylxanthine (DPCPX) exacerbate GOD-induced increase of spontaneous release of $[^3H]-5-HT$. These results suggest that Adenosine may play a role in the GOD-induced spontaneous release of $[^3H]-5-HT$ through adenosine $A_1$ receptor activity.

Resveratrol Affects Protein Kinase C Activity and Promotes Apoptosis in Human Colon Carcinoma Cells

  • Background: Resveratrol has been reported to have potential chemopreventive and apoptosis-inducing properties in a variety of tumor cell lines. Objective: In this study, to investigate the effects of resveratrol on protein kinase C (PKC) activity and apoptosis in human colon carcinoma cells, we used HT-29 cells and examined the $PKC{\alpha}$ and ERK1/2 signaling pathways. Methods: To test the effects of resveratrol on the growth of HT-29 cells, the cells were exposed to varying concentrations and assessed with the the MTT cell-viability assay. Fluorescence-activated cell sorter (FACS) analysis was applieded to determine the effects of resveratrol on cell apoptosis. Western blotting was performed to determine the protein levels of $PKC{\alpha}$ and ERK1/2. In inhibition experiments, HT-29 cells were treated with G$\ddot{o}$6976 or PD98059 for 30 min, followed by exposure to $200{\mu}M$ resveratrol for 72 h. Results: Resveratrol had a significant inhibitory effect on HT-29 cell growth. FACS revealed that resveratrol induced apoptosis. Western blotting showed that e phosphorylation of $PKC{\alpha}$ and ERK1/2 was significantly increased in response to resveratrol treatment. Pre-treatment with $PKC{\alpha}$ and ERK1/2 inhibitors (G$\ddot{o}$6976 and PD98059) promoted apoptosis. Conclusion: Resveratrol has significant anti-proliferative effects on the colon cancer cell line HT-29. The PKC-ERK1/2 signaling pathway can partially mediate resveratrol-induced apoptosis of HT-29 cells.
