Tensile Strength of Post-Installed High-Shear Ring Anchors (HRA) After Shear Loading (전단 하중을 경험한 후설치 고전단 링앵커의 인장 강도)

  • Tensile load tests were conducted on High-Shear Ring Anchors (HRAs) after shear load had been applied to the HRAs, which had been developed to reduce the number of the anchors. Test variables include the embedment length of the rod and the width of the specimens and a total of 12 specimens were tested. Test results show that the HRAs pulled out due to bond failure or steel failure occurred in case that the HRAs were installed to the members with 300mm or greater width and the embedment length of 160mm (the actual embedment of rod is 140mm) or deeper. Except 4 HRAs showing steel failure of rod, the minimum and average of test-to-prediction by ACI 318-14 ratios are 1.18 and 1.79, respectively. The tensile strength of HRAs, after shear load was applied to the HRAs, can be safely evaluated by the minimum among the concrete breakout strength and bond strength with the actual embedment length of the rod.

Identification of a Potential Anticancer Target of Danshensu by Inverse Docking

  • Objective: To study potential targets of Danshensu via dual inverse docking. Method: PharmMapper and idTarget servers were used as tools, and the results were checked with the molecular docking program autodock vina in PyRx 0.8. Result: The disease-related target HRas was rated top, with a pharmacophore model matching well the molecular features of Danshensu. In addition, docking results indicated that the complex was also matched in terms of structure, H-bonds, and hydrophobicity. Conclusion: Dual inverse docking indicates that HRas may be a potential anticancer target of Danshensu. This approach can provide useful information for studying pharmacological effects of agents of interest.

Compound HRAS/PIK3CA Mutations in Chinese Patients with Alveolar Rhabdomyosarcomas

  • The rhabdomyosarcoma (RMS) is the most common type of soft tissue tumor in children and adolescents; yet only a few screens for oncogenic mutations have been conducted for RMS. To identify novel mutations and potential therapeutic targets, we conducted a high-throughput Sequenom mass spectrometry-based analysis of 238 known mutations in 19 oncogenes in 17 primary formalin-fixed paraffin-embedded RMS tissue samples and two RMS cell lines. Mutations were detected in 31.6% (6 of 19) of the RMS specimens. Specifically, mutations in the NRAS gene were found in 27.3% (3 of 11) of embryonal RMS cases, while mutations in NRAS, HRAS, and PIK3CA genes were identified in 37.5% (3 of 8) of alveolar RMS (ARMS) cases; moreover, PIK3CA mutations were found in 25% (2 of 8) of ARMS specimens. The results demonstrate that tumor profiling in archival tissue samples is a useful tool for identifying diagnostic markers and potential therapeutic targets and suggests that these HRAS/ PIK3CA mutations play a critical role in the genesis of RMS.

Changes in the Expression of Ras-family Genes in Rats Exposed to Formaldehyde by Inhalation

  • Exposure to formaldehyde(FA) is closely associated with adverse health effects such as irritation, inflammation, and squamous cell carcinomas of the nasal cavities. Owing to its rapid metabolism and elimination, exposure to FA does not always result in an increased concentration in blood or urine of animals and humans. Therefore, the development of biomarkers for FA exposure is necessary for risk assessment. In the present study, the effects of FA were investigated on the expression of genes involved in the MAPK pathway in vitro and results confirmed in rats exposed to FA by inhalation. Treatment of Hs 680.Tr human tracheal epithelial cells with FA induced gene expression for PDGFA, TNFSF11, SHC1, and HRAS. HRAS expression was also increased in tracheas of rats exposed to FA. In addition, FA exposure induced the expression of RASSF4, a member of the Rasassociation domain family of Ras effectors, in rat tracheas. In conclusion, data showed FA-inducible expression of genes involved in the MAPK pathway occurred and increased expression of HRAS and RASSF4 was noted in rat tracheas subchronically exposed to FA by inhalation. These genes may serve as molecular targets of FA toxicity facilitating the understanding of the toxic mechanism.

New Gene Profiling in Determination of Breast Cancer Recurrence and Prognosis in Iranian Women

  • Breast cancer (BC) is the second most common cancer in the world and by far the most frequent cancer among women, with an estimated 1.67 million new cancer cases diagnosed in 2012 (25% of all cancers). Polygene expression analysis is used to predict the prognosis and determine the most appropriate treatment regimen. The objective of this study was to examine the gene expression profiles of SIRT3, HRAS, LSP1, SCUBE2 and AP2A2 in Iranian women with BC.A total of 136 patients including healthy controls were categorized into three groups based on the relapse of the disease. Expression of desired genes in formalin-fixed, paraffin embedded tissues collected from all groups of participants was analyzed via the RT PCR method. RNA extraction and cDNA synthesis were performed then real-time quantitative PCR was carried out. Gene expression analysis revealed that the expression of SIRT3 was equal among patient and control groups. LSP1 was down regulated in all patient groups relative to controls but reduced expression in the metastatic group relative to the non-metastatic one was not significant. HRAS was significantly overexpressed in total and metastatic tumor samples versus normal but not in non-metastatic cases. SCUBE2 expression showed significant over-expression in both overall tumor samples and the non-metastatic group as compared to normal tissues. Gene expression level of AP2A2 in all groups was not detectable. Our data are compatible with a tumor suppressor role of LSP1 related to potential prognostic factor for tumor recurrence and outcome. This study for the first time assayed the prognostic value and changes in the expression of SIRT3, LSP1, HRAS, SCUBE2 and AP2A2 genes in women with breast cancer in the Iranian population and findings confirmed potential biomarker and prognostic capability of these genes. Such expression profiling data can critically improve prognosis and treatment decisions in cancer patients.

Mutational Analysis of Key EGFR Pathway Genes in Chinese Breast Cancer Patients

  • Background: The epidermal growth factor receptor (EGFR) is a potential therapeutic target for breast cancer treatment; however, its use does not lead to a marked clinical response. Studies of non-small cell lung cancer and colorectal cancer showed that mutations of genes in the PIK3CA/AKT and RAS/RAF/MEK pathways, two major signalling cascades downstream of EGFR, might predict resistance to EGFR-targeted agents. Therefore, we examined the frequencies of mutations in these key EGFR pathway genes in Chinese breast cancer patients. Methods: We used a high-throughput mass-spectrometric based cancer gene mutation profiling platform to detect 22 mutations of the PIK3CA, AKT1, BRAF, EGFR, HRAS, and KRAS genes in 120 Chinese women with breast cancer. Results: Thirteen mutations were detected in 12 (10%) of the samples, all of which were invasive ductal carcinomas (two stage I, six stage II, three stage III, and one stage IV). These included one mutation (0.83%) in the EGFR gene (rs121913445-rs121913432), three (2.50%) in the KRAS gene (rs121913530, rs112445441), and nine (7.50%) in the PIK3CA gene (rs121913273, rs104886003, and rs121913279). No mutations were found in the AKT1, BRAF, and HRAS genes. Six (27.27%) of the 22 genotyping assays called mutations in at least one sample and three (50%) of the six assays queried were found to be mutated more than once. Conclusions: Mutations in the EGFR pathway occurred in a small fraction of Chinese breast cancers. However, therapeutics targeting these potential predictive markers should be investigated in depth, especially in Oriental populations.

Recovering the Energy Potential of Sewage as Approach to Energy Self-Sufficient Sewage Treatment (하수처리장 에너지 자립화를 위한 하수 에너지 잠재력 회수 기술)

  • Domestic sewage treatment plants (STPs) consume about 0.5 % of total electric energy produced annually, which is equivalent to 207.7 billion Korean won per year. To minimize the energy consumption and as a way of mitigating the depletion of energy sources, the sewage treatment strategy should be improved to the level of "energy positive". The core processes for the energy positive sewage treatment include A-stage for energy recovery and B-stage for energy-efficient nitrogen removal. The integrated process is known as the A/B-process. In A-stage, chemically enhanced primary treatment (CEPT) or high rate activated sludge (HRAS) processes can be utilized by modifying the primary settling in the first stage of sewage treatment. CEPT utilizes chemical coagulation and flocculation, while HRAS applies returned activated sludge for the efficient recovery of organic contents. The two processes showed organic recovery efficiencies ranging from 60 to 70 %. At a given recovery efficiency of 80 %, 17.3 % of energy potential ($1,398kJ/m^3$) is recovered through the anaerobic digestion and combustion of methane. Besides, anaerobic membrane bioreactor (AnMBR) can recover 85% of organic contents and generate $1,580kJ/m^3$ from the sewage. The recovered energy is equal to the amount of energy consumption by sewage treatment equipped with anaerobic ammonium oxidation (ANAMMOX)-based B-stage, $810{\sim}1,620kJ/m^3$. Therefore, it is possible to upgrade STPs as efficient as energy neutral. However, additional novel technologies, such as, fuel cell and co-digestion, should be applied to achieve "energy positive" sewage treatment.

  • In this paper, a probability-based reliability analysis was proposed based on a finite element method-based random vibration analysis and serviceability limit state of structures. The limit state model defined for the study is a serviceability limit state in terms of the more realistic crack failure that might cause the emission of radioactive materials. The SAP V-2 is used for a three-dimensional finite element analysis of concrete containment structure, and the reliability analysis is carried out by modifying HRAS reliability analysis program for this study. In this study, the load factors for the design of reinforced concrete cointainment structures in Korea are proposed by considering appropriate load combination criteria for design, and the results are compared with the present ASME code. The proposed load factors were proved to be in accordance with a set of code performance objective and showed consistency in the limit state probability.

Costello syndrome: three sporadic cases (Costello 증후군 3례)

  • Costello syndrome (CS) is a rare multiple congenital abnormality syndrome characterized by a typical coarse face, developmental delay, psychomotor and growth retardation, neurologic abnormalities, cardiac and cutaneous anomalies, severe feeding difficulties with postnatal growth failure, and increased risk of tumors. Since Costello first described it in 1971 and again in 1977, over 100 cases have been reported worldwide. It was recently shown that CS is a congenital condition caused by heterozygous de novo missense mutations affecting the codon for glycine 12 or 13 of the HRAS gene. We experienced three unrelated cases with coarse faces, developmental delays, short statures, macrocephaly, and redundant skin with deep palmar and plantar creases, hypertrophic cardiomyopathy and atrial tachycardia, which are characteristic of CS.
