Comparative Inactivation of Hepatitis A Virus and Murine Encephalomyocarditis Virus to Various Inactivation Processes (바이러스 불활화 공정에 대한 Hepatitis A Virus와 Murine Encephalomyocarditis Virus의 민감도 비교)

  • Murine encephalomyocarditis virus (EMCV) has been used as a surrogate for hepatitis A virus (HAV) for the validation of virus removal and/or inactivation during the manufacturing process of biopharmaceuticals. Recently international regulation for the validation of HAV safety has been reinforced because of the reported cases of HAV transmission to hemophiliac patients who had received ntihemophilic factors prepared from human plasma. The purpose of the present study was to compare the resistance of HAV and EMCV to various viral inactivation processes and then to standardize the HAV validation method. HAV was more resistant than EMCV to pasteurization (60oC heat treatment for 10 hr), low pH incubation (pH 3.9 at 25oC for 14 days), 0.1 M NaOH treatment, and lyophilization. EMCV was completely inactivated to undetectable levels within 2 hr of pasteurization, however, HAV was completely inactivated to undetectable levels after 5 hr treatment. EMCV was completely inactivated to undetectable levels within 15 min of 0.1 M NaOH treatment, however, residual infectivity of HAV still remained even after 120 min of treatment. The log reduction factors achieved during low pH incubation were 1.63 for HAV and 3.84 for EMCV. Also the log reduction factors achieved during a lyophilization process of antihemophilic factor VIII were 1.21 for HAV and 4.57 for EMCV. These results indicate that HAV rather than EMCV should be used for the virus validation study and the validation results obtained using EMCV should be precisely reviewed.

  • An asymptotic distribution of the ratio of two normal vectors is estimated. Using the estimated asymptotic distribution, an approximation method to estimate confidence interval of passenger's value of travel time is proposed. As a result of empirical study the 95% confidence interval of value of travel time of home-to-work trips in city of Seoul is estimated at ₩7341.25$\pm$1945.05/hr.

Cloud-cell Tracking Analysis using Satellite Image of Extreme Heavy Snowfall in the Yeongdong Region (영동지역의 극한 대설에 대한 위성관측으로부터 구름 추적)

  • This study presents spatial characteristics of cloud using satellite image in the extreme heavy snowfall of the Yeongdong region. 3 extreme heavy snowfall events in the Yeongdong region during the recent 12 years (2001 ~ 2012) are selected for which the fresh snow cover exceed 50 cm/day. Spatial characteristics (minimum brightness temperature; Tmin, cloud size, center of cloud-cell) of cloud are analyzed by tracking main cloud-cell related with these events. These characteristics are compared with radar precipitation in the Yeongdong region to investigate relationship between cloud and precipitation. The results are summarized as follows, selected extreme heavy snowfall events are associated with the isolated, well-developed, and small-scale convective cloud which is developing over the Yeongdong region or moving from over East Korea Bay to the Yeongdong region. During the period of main precipitation, cloud-cell Tmin is low ($-40{\sim}-50^{\circ}C$) and cloud area is small (17,000 ~ 40,000 $km^2$). Precipitation area (${\geq}$ 0.5 mm/hr) from radar also shows small and isolated shape (4,000 ~ 8,000 $km^2$). The locations of the cloud and precipitation are similar, but in there centers are located closely to the coast of the Yeongdong region. In all events the extreme heavy snowfall occur in the period a developed cloud-cell was moving into the coastal waters of the Yeongdong. However, it was found that developing stage of cloud and precipitation are not well matched each other in one of 3 events. Water vapor image shows that cloud-cell is developed on the northern edge of the dry(dark) region. Therefore, at the result analyzed from cloud and precipitation, selected extreme heavy snowfall events are associated with small-scale secondary cyclone or vortex, not explosive polar low. Detection and tracking small-scale cloud-cell in the real-time forecasting of the Yeongdong extreme heavy snowfall is important.

Review of PM-related Air Quality Improvement Policies of United States for PM-related Air Quality Improvement of Metropolitan Region in Korea (수도권 미세먼지 환경 개선을 위한 미국의 대기환경정책 사례 조사 연구)

  • Several fine particle-related policies in Northeast United States were investigated in support of the execution of special measures to improve air quality of Seoul Metropolitan Area (SMA). The definition of particulate matter (PM) in the Clean Air Act (CAA), components and procedures of Regional Haze Rule (RHR), and Air Quality Management (AQM) were reviewed. Several State Implementation Plans (SIPs) were also reviewed as the way to attain required air quality under the Clean Air Act. $PM_{2.5}$ attainment SIP of Maryland, 8-hr Ozone attainment SIP of New Jersey, and Regional Haze Rule attainment SIP of MANE-VU were analyzed in detail as case studies. We realized that "Special Measures for Air Quality Improvement in the Seoul Metropolitan Area" has many similarities with its US counterparts in terms of purpose, components, procedures, and implementation methods. US policies, however, have more advanced features, such as standardized procedures and methods, transparent guidelines, and stable relationship among federal/state/local governments and stakeholders, which would be helpful to improve air quality in SMA.

  • In this study, we reviewed the main strategies and contents of the work style innovation promoted by the Moon Jae-in government, and analyzed the contents and success factors of the work innovation based on the case of a specific local government. In particular, this study analyzes the case of innovation in the way of working through simplification of work, and the local governments, which were the subject of the study, received excellent evaluation in the government innovation evaluation of the Ministry of Public Administration and Security in 2019. In this study, we conducted in-depth interviews with employees of local governments to examine the problems, achievements, and success factors related to business simplification. First, it was found that it is very important to form consensus so that the members of the organization can participate in the change in order to simplify the task. Second, incentives, such as HR, were needed to induce active participation of members in the innovation project called Simplification of Work. Third, in order to induce and support organizational change, it was found that the interest and support of middle managers and directors were the most important factors. The results of this study will help to prepare the policy for innovation in the public sector's working style.

Classification Analysis of the Physical Environment of Bicycle Road -Focused on Chang Won City, Kyung Nam Province, S. Korea- (자전거 도로의 물리적 환경에 대한 등급화 연구 -창원시 사례를 중심으로-)

  • This study is to analyze the physical environment and conduct spatial data for bicycle road system in changwon. Index for evaluation index was developed based on literatures. Then the level of importance and weight have been modified through experts review. Finally, index with eight categories such as greenness(40% over), bicycle road connectivity(1.8, 9.8%), road type bike(bicycle lane, 24.4%), pave type(asphalt 72.5%), illegal parking(none, 93.9%), bike road surface visibility(exist, 46.8%), vehicle speed limits(30km, under), vehicle traffic(500/hr under, 44.3%) have been applied to empirical investigation. Collected data has been hierarchically classification by ArcGIS Program. The Highest grades(score 31-35, level 1) occupied 35% of target destination. High level of greenness and load type has contributed to high score. In addition, average level of greenness of those destination was 35% and higher, which provide high degree of security and freshness for bicycle riding. Meanwhile, lowest level(level 5, which earned 15 point or less) occupied 24.5%. illegal parking, low level of greenness, and no surface sign caused low score.

Estimation or Threshold Runoff on Han River Watershed (한강유역 한강유출량 산정)

  • In this study, threshold runoff which is a hydrologic component of flash flood guidance(FFG) is estimated by using Manning's bankfull flow and Geomorphoclimatic Instantaneous Unit Hydrograph(GcIUH) methods on Han River watershed. Geographic Information System(GIS) and 3' Digital Elevation Model database have been used to prepare the basin parameters of a very fine drainage area($1.02\~56.41km^2$), stream length and stream slope for threshold runoff computation. Also, cross-sectional data of basin and stream channel are collected for a statistical analysis of regional regression relationships and then those are used to estimate the stream parameters. The estimated threshold runoff values are ranged from 2 mm/h to 14 mm/6hr on Han River headwater basin with the 1-hour duration values are$97\%$ up to 8mm and the 6-hour values are $98\%$ up to 14mm. The sensitivity analysis shows that threshold runoff is more variative to the stream channel cross-sectional factors such as a stream slope, top width and friction slope than the drainage area. In comparisons between the computed threshold runoffs on this study area and the three other regions in the United States, the computed results on Han River watershed are reasonable.

A study on the design of tunnel lining insulation based on measurement of temperature in tunnel (터널 온도계측을 통한 라이닝 단열 설계에 관한 연구)

  • In case of tunnels in cold regions, a freeze of groundwater around tunnel may act as a barrier of tunnel drainage in winter, or may cause the inner extrusion of lining. In spite of that, a design of insulation for preventing the frost damage of tunnel lining has not been introduced in Korea, while foreign countries such as Norway and so on have a standard on insulation. In this study, a few freezing cases of road tunnels have been reviewed, and the results show that the freezing protection is necessary. In order to characterize the thermal distribution in the tunnel, following measurements have been performed at Hwa-ak tunnel; the temperature distribution by longitudinal lengths, the internal temperature of lining and the temperature distribution of the ground under pavement. From these measurements, the characteristics of the tunnel's internal temperature distribution due to temperature change in the air has been analyzed. Based on the measurement results on the temperature distribution at Hwa-ak tunnel, thermal flow tests on the rock specimen with and without insulation have been performed in the artificial climate chamber to investigate the performance of the insulation. Also, a number of 3D numerical analyses have been performed to propose appropriate insulation and insulation thicknesses for different conditions, which could prevent the frost damage of tunnel lining. As a result of the numerical analysis, air freezing index of 291$^{\circ}C{\cdot}$ Hr has been suggested as the threshold value for freezing criteria of groundwater behind the tunnel lining.

Estimation and evaluation on the return period of flash flood for small mountainous watersheds in the Han River basin (한강유역 산지소하천의 돌발홍수 재현기간 산정 및 평가)

  • The objectives of this study are to estimate the return period of flash flood and evaluate its appropriateness based on the actual observation events for small mountainous watersheds in the Han River basin. For these goals, Flash Flood Guidance (FFG) was estimated from 1-hr duration Threshold Runoff (TR) and Saturation Deficit (SD) of soil moisture which was derived from Sejong University Rainfall Runoff (SURR) model. Then, the return period of flash flood was calculated by comparing the rainfall quantile to the 1-hr duration rainfall that exceeded the FFG during the past period (2002-2010). Moreover, the appropriateness of the estimated return period of flash flood was evaluated by using the observation events from 2011 to 2016. The results of the return period of flash flood ranged from 1.1 to 19.9 years with a mean and a standard deviation of 1.6 and 1.1 years, respectively. Also, the result of the appropriateness indicated that 83% of the return periods derived from observation events were within the return period of flash flood range. Therefore, the estimated return period of flash flood could be considered as highly appropriate.

  • 댐은 우리사회에 다양한 이익을 제공하는 반면 과거의 댐 붕괴 사례에 비추어 볼 때 붕괴로 인한 피해 규모는 막대하다. 세계 각국에서는 기존 댐들에 대한 잠재적 붕괴에 대비하기 위해 비상대처계획(Emergency Action Plan, EAP)과 같은 대책을 수립하여 붕괴로 인한 피해를 최소화하기 위한 노력을 기울이고 있다. 댐붕괴 모델링은 크게 댐 결괴부에 대한 예측과정과 댐 붕괴로 인한 하류부 홍수추적 과정으로 분류할 수 있다. 결괴부 예측과정은 댐붕괴로 인한 저수지 유출량에 가장 큰 영향을 미치는 부분이며 하류부 영향지역의 평가에도 매우 중요한 요소이다. 댐붕괴 예측모형은 결괴부에 대한 예측에 따라 매개변수 기반 모형과 물리기반 모형으로 분류된다. 매개변수 기반모형은 결괴형성과정을 단순 시간에 의한 함수로 고려하고 있으며 결괴부를 통한 흐름은 위어흐름으로 가정하고 있다. 이 모형은 현재 댐 붕괴 실무에서 가장 널리 이용되고 있으며 대표적인 모형은 FLDWAV/ DAMBRK 모형, HEC-RAS 모형 등이 있다. 물리기반 모형은 댐 붕괴진행 과정의 상세한 이해를 위해 시작되었으며 결괴부 흐름상황, 제체 침식, 제체가 불안정해지는 과정을 수리실험과 수리학, 토질역학, 구조역학 등의 이론을 통합하여 예측하는 기법으로 댐붕괴 과정을 좀 더 현실적으로 예측할 수 있으며 기존 붕괴된 댐들에 대한 붕괴 원인 및 진행과정을 규명할 수 있는 장점을 가진다. 최근 개발된 물리기반 댐 붕괴 모형은 HR-BREACH 모형, WINDAM 모형, FIREBIRD 모형 등이 있으며 지속적으로 연구되고 있다. 본 연구에서는 현재 진행 중에 있는 물리기반 댐 붕괴 모형들을 검토하고 현재 USDA, ARS에서 개발 중에 있는 WINDAM 모형을 이용하여 물리기반 모형의 수행에 요구되는 입력변수, 수행과정, 결과물에 대한 검토를 통해 댐 붕괴 관련 연구의 발전 방향을 모색하고 국내에서 물리기반 댐 붕괴 모형을 적용하기 위해 요구되는 사항과 한계점을 파악하여 댐 붕괴 실무로의 적용 방안을 마련하는 데 그 목적이 있다.
