소지환가미방이 고지혈 병리인자 및 동물 병태 모델에 미치는 영향 (Effects of Sojihwangamibang on Hyperlipidemia in Rats Induced by High Cholesterol Diet)

  • This experimental study was designed to investigate the inhibitory effects of Sojihwangamibang on hyperlipidemia in rats induced by high cholesterol diet diet. Sprague- Dawley rats were divided into normal group, control group, SJB treated group. Obese rats were induced by high cholesterol diet treatment for 6 weeks including a oral administration of SJB for 4 weeks. In SJB group, serum total cholesterol, LDL cholesterol, triglyceride and glucose were significantly decreased, and HDL cholesterol was significantly increased compared with untreated control group. In SJB group, HMG-CoA and ACAT concentration of hepatic homogenate were significantly decreased compared with untreated control group. These results provide experimental evidence that SJB, applied currently in the clinical practice, appears to be effective for down-regulating risk factors of hyperlipidemia, and thus may be used as an objective information for the development of therapeutic agents.

Atorvastatin과 Telmisartan의 약물상호작용 (Drug-drug Interactions between Atorvastatin and Telmisartan)

  • Purpose: Atorvastatin, a HMG-CoA reductase inhibitor is widely prescribed in hyperlipidemic patients and telmisartan, an angiotensin receptor blocker is frequently used in the treatment of hypertension. Both drugs are substrates of organic anion transporting polypeptide (OATP) expressed in basolateral membrane in the liver, and undergo high first pass metabolism. Therefore, OATP-mediated hepatic uptake is important for disposition and metabolism of these drugs. The present study was designed to investigate the pharmacokinetic interactions between atorvastatin and telmisartan in rats. Method: Young adult SD rats were divided into three groups (n=6, each) and atorvastatin (10 mg/kg) and telmisartan (4 mg/kg) were orally given alone and together. Heparinized blood was serially taken and plasma concentrations of both drugs were measured using HPLC-MS/MS. Pharmacokinetic parameters of two drugs were calculated. Results: No significant pharmacokinetic change was found except a delay of time to peak of telmisartan when administered with atorvastatin. Each drug at the present dosage seemed to be insufficient to alter the pharmacokinetic parameters of its counterpart drug. Conclusion: Conclusively, co-administration of atorvastatin and telmisartan may lead to negligible clinical consequences.

Free Radical Scavenging Activity and Inhibition of Linoleic Acid Peroxidation of Commercial Tocotrienol Fraction

  • Tocotrienols (T3) are minor plant constituents found abundantly in rice bran, which provide a significant source of vitamin E in animal feeds. T3 was reported to have an intrinsic hypocholesterolemic effect by inhibiting HMG-Co A reductase. It has similar antioxidative properties as tocopherols in food and biological system due to their similar chemical structures. However, the antioxidant activity and mechanism of T3 to scavenge free radicals and to inhibit the peroxidation of linoleic acid are less understood. The purpose of this study was to investigate the scavenging effect of T3 on free radicals and its inhibition of peroxide formation. Free radical scavenging activity was monitored by the DPPH (1,1-diphenyl-2-picrylhydrazyl) method whereas inhibition of linoleic acid peroxidation was evaluated using the thiocyanate method. Thiobarbituric acid (TBA) test was used to determine malonaldehyde formation from linoleic acid peroxidation. Free radical scavenging activity increased with increasing concentration levels of T3. T3 exhibited 38.2, 78.6, 92.7 and 96.2% radical scavenging activity at concentrations of 2, 8, 32 and 128 ppm, respectively. At 128 ppm, it was highly effective in inhibiting linoleic acid peroxidation. The activity of T3 evaluated by the thiocyanate method showed low absorbance values indicating a high level of antioxidant activity. All treatments showed similar trends in antioxidant activity when evaluated by both the thiocyanate method and TBA test.

  • The effects of garlic extracts, lactic acid bacteria, copper and selenium as cholesterol lowering material on productivity and content of egg cholesterol in layer were investigated. The layers were divided into seven treatment groups and fed commercial diet or experimental diets containing probiotics, garlic powder, selenium and copper for 6 weeks. There was no significant difference in egg production between groups. The content of egg yolk cholesterol by combination feeding of probiotics, garlic powder, selenium and copper were significantly reduced by 13.5-29.8% as compared to that of control. The levels of mRNA expression of HMG-CoA reductase were significantly decreased by dietary treatments.

  • This study was undertaken to determine whether dehulled defatted flour has an effect on postprandial plasma lipoprotein composition, hepatic lipid composition, enzyme and hormone levels in rats. Control(casein) and experimental (dehulled defatted soy flour)diets were fed to rats for 7 weeks. all animals (S. D. rats, male) were sacrificed 2 hrs after the feeding of 5g of each diet. Defatted soy flour feeding significantly lowered postprandial plasma total cholesterol, chylomicron/VLDL-cholesterol, hepatic cholesterol and triglyceride(TG) as compared with casein feeding, whereas no significant effect on plasma TG was observed. Intestinal kipase activity was elevated , whereas trypsin activity was suppressed in the dehulled defatted soy flour group. Plasma glucagon, thyroid hormone and hepatic HMG-coA reductase levels were not affected by diet treatment. These results hypothesize that dehulled defatted soy flour affects cholesterol digestion and absorption in guts, thus delaying the appearance of chylomicron cholesterol in plasma or affecting the disappearance of chylomicron remnant to high-density-lipoprotein(HDL).

  • The extracts from stem and fruit of Cactus (Opuntia ficus-indica var. saboten Makino) were applied to confirm the antiatherosclerotic effects in subacute alcoholic hyperlipidemic rats. It was observed that several indications of hyperlipidemia were prevented or changed by the treatments of the extracts but activities of hepatic HMG-CoA reductase and alcohol dehydrogenase were not affected, which suggest that the extracts may promote the fecal excretion and delay the absorption of alcohol in hyperlipidemic rats.

구기자 품종과 교배종의 부위별 심혈관관련 질환 예방 기능성 및 항산화 활성비교 (Cardiovascular Biofunctional Activity and Antioxidant Activity of Gugija (Lycium chinensis Mill) Species and Its Hybrids)

  • 새로운 고부가가치의 구기자 품종을 개발하기 위하여 구기자 표준 품종들과 이들의 교배종들의 순과 잎, 열매와 뿌리등의 물 추출물과 메탄올 추출물을 제조한 후 심혈관 질환 예방생리기능성과 노화억제 항산화 활성을 조사하여 비교하였다. 구기자 표준품종들의 생리기능성 가운데, 항산화 활성은 청운품종의 구기순의 메탄올 추출물이 93%로 가장 높았고 항고혈압성 안지오텐신 전환효소 저해활성은 청양7호의 지골피의 물 추출물에서 84.1%로 제일 높았다. 구기자 교배종의 생리 기능성중 항산화 활성은 명안A-2의 지골피의 메탄올 추출물이 93.1%로 제일 높았고 ACE저해활성은 DO148-72 (A11) 교배종 열매의 메탄올 추출물이 96.9%로 제일 높았다. 그러나 혈전용해활성과 HMG-CoA reductase 저해활성은 표준품종과 교배종 모두에서 대체로 미약하거나 없었다. 이상의 결과를 종합하였을 때 청운 표준품종의 구기순과 명안A-2 교배종의 지골피가 우수한 항산화 자원으로, DO148-72 (A11)교배종의 구기열매가 훌륭한 항고혈압성 ACE저해제 자원으로 각각 기능성 제품생산에 유용하게 활용될것으로 사료된다.

  • 비알코올성 지방간은 만성 간 질환으로 비만, 고혈압, 비만, 이상지질혈증과 같은 다양한 대사증후군과 연관되어 있다. 꾸지뽕은 한국을 포함한 동아시아 국가에서 다양한 질병에 사용되는 약용작물로 본 연구에서는 유리지방산에 의해 지방축적이 유도된 세포 내에서 꾸지뽕 열매 추출물의 비알코올성 지방간 개선 효과와 기전을 규명하였다. 꾸지뽕 열매 추출물은 지방 축적 및 중성지방, 콜레스테롤 생성 및 HMG 환원효소의 활성을 억제하였다. 또한 지방생성과 관련된 유전자인 SREBP-1, FAS, SCD-1, SREBP-2의 발현을 억제 하였으며 AMPK의 활성화를 억제하였다. 본 연구결과를 통해서 꾸지뽕 열매 추출물이 유리지방산에 의해 유도된 지방 축적을 억제하고 AMPK/SREBP 신호전달 경로를 조절하여 억제 활성을 나타냄을 밝히며, 비알코올성 지방간의 예방 및 개선을 위한 천연물 소재로 활용될 수 있을 것으로 사료된다.

스타틴 그리고 배아줄기세포에서의 작용 (Statins and Their Effects on Embryonic Stem Cells)

  • 배아줄기세포를 이용한 치료법 개발을 위해서는 배아줄기세포의 자가재생산 및 분화과정을 조절하는 분자적 기전을 이해하는 것이 매우 중요하다. 지질합성경로(Mevalonate pathway)에 작용하는 HMG-CoA 환원효소(Hydroxymethylglutaryl-coenzyme A reductase)의 억제제인 스타틴은 콜레스테롤 저하제로 잘 알려져 있으며, 콜레스테롤 이외에 단백질 isoprenylation의 기질로 작용하는 아이소프레노이드(Isoprenoids)(Farnesyl pyrophosphate(FPP), Geranylgeranyl pyrophosphate(GGPP))의 생성을 억제하는 효능을 가지고 있다. 스타틴에 의해 매개되는 표적단백질의 isoprenylation 억제는 다양한 세포내 신호전달과정에 영향을 미치게 되며, 결과적으로 세포기능을 조절하는데 핵심적인 역할을 하게 된다. 스타틴이 첨가된 배양배지에서 배양된 배아줄기세포는 자가재생산능이 억제되고 분화가 촉진되는데, 특히 지방/골세포 직계열로의 분화가 촉진된다. 배아줄기세포에서의 스타틴의 효과 및 작용기전에 대한 이해가 아직은 미비한 수준이나, 최근 우리 연구팀에서는 스타틴이 콜레스테롤 작용과는 무관하게 RhoA G-단백질의 세포내 분포 및 활성을 억제함으로써 배아 줄기세포의 자가재생산능을 억제하고 있음을 규명하였다. 스타틴 다면효과와 그 작용에 대한 이해는 배아줄기세포의 미분화 및 분화상태를 조절하는데 관여하는 분자적 조절기전을 이해하는데 중요한 모델이 될 수 있을 것으로 추정된다.

목단피 약침이 고지혈증과 동맥경화에 미치는 영향에 대한 실험적 연구 (The Effect of Herbal-acupuncture with Moutan Cortex Radicis Extract)

  • Objective & Methods : The present study was to investigate the effect of Moutan Cortex Radicis Herbal-acupuncture (MCR-HA) at GB34 (Yangleungchean) on hyperlipidemia and arteriosclerosis induced by high fat diet in rats. Results : 1. Moutan Cortex Radicis herbal-acupuncture infusion solution (MCR-HAS) increased DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. MCR-HA at GB34 decreased serum total cholesterol level, and increased serum phospholipid/total cholesterol in rats with high fat diet. 3. MCR-HA at GB34 decreased atherogenic index in rats with high fat diet. 4. MCR-HA at GB34 increased hepatic catalase activity in rats with high fat diet. Conclusion : These results suggested that Moutan Cortex Radicis herbal-acupuncture at GB34 has therapeutic effects on hyperlipidemia and related complications in rats with high fat diet.
