Synthesis and Biological Activity of Fungal Metabolite, 4-Hydroxy-3-(3'-Methyl-2'-Butenyl)-Benzoic Acid

  • 4-Hydroxy-3-(3'-methyl-2'-butenyl)-benzoic acid (HMBA) was previously isolated from Curvularia sp. KF119 as a cell-cycle inhibitor. However, the present study used a novel and practical synthetic method to prepare a large quantity of HMBA. The synthetic HMBA was found to inhibit the cell-cycle progression of HeLa cells with a comparable potency to the natural fungal metabolite. The inhibition of the cell-cycle progression by the synthetic HMBA involved both the activation of $p21^{WAF1}$ and the inhibition of cyclin D1 expression in the cells. Consequently, this new synthetic procedure provides an easy and convenient way to produce or manipulate the original fungal metabolite.

  • 본 연구는 비만세포의 일종인 RBL-2H3 세포주에 대한 hexamethylene bisacetamide (HMBA)의 세포독성 유도 기전이 telomerase의 활성과 관계가 있다는 것에 대한 보고이다. RBL-2H3 세포주에 5mM HMBA를 처리시 그 세포주의 세포성장이 억제 되었다. 기전연구로서 HMBA는 telomerase활성을 억제 하였으며 RT-PCR에 의한 결과 telomerase mRNA발현의 억제에 의한 것이었다. Madl은 telomerase의 발현을 억제하는 억제인자로 잘 알려져 왔으며 이를 확인해본 결과 Madl은 발현은 유의성있게 증가 되었다. 이를 종합해보면, HMBA에 의한 RBL-2H3 세포주에 대한 세포독성은 Madl 발현의 증가에 따른 telomerase발현의 억제에 기인하는 것으로 밝혀졌다.

  • Gastrodia elata (GE) is an oriental medicinal herb which has been used traditionally for the treatment of various brain diseases including convulsion and epilepsy. The purpose of this study was to determine pharmacokinetic parameters of 4-hydroxy-3-methoxybenzaldehyde (HMBA) and p-hydroxybenzaldehyde (PHBA), constituents of GE, in rats. Male rats were cannulated in the femoral vein, femoral artery, bile duct and ureter. They received a single i.v. bolus dose of either HMBA or PHBA through the femoral vein. The concentration of HMBA or PHBA in plasma, bile and urine samples were analyzed by reversed-phase HPLC. HMBA and PHBA have very short half-lives, i.e. 4.03 and 2.26 minutes respectively. Most of HMBA and PHBA were thought to be eliminated through metabolism as the metabolized fraction approaches unity. Derivatives of HMBA or PHBA with longer biological half-lives should be designed to develop better anticonvulsants and more complete qualitative and quantitative understanding of the overall pharmacokinetic fate of these compounds awaits further investigation.

  • It's important to study about suitable substitutional material of expensive silica fume because it used widely for silica fume as a high-strength mortar binding agent. The main purpose of this study is to check which is the most efficient binder for the expensive silica fume's alternative material. And this study also present basic data about to make high-strength mortar when we use alternative material instead of silica fume through research outcome. Also writers analzed flow properties respectively, so it was founded out the substitutional goods fare like meta-kaolin, HMBA which are less expensive than silica fume because they are in domestic enough.

Synthesis of N,N'-Bisacrylamide Derivatives (N,N'-비스아크릴아미드 유도체의 합성)

  • Four kinds of N,N'-bisacrylamide derivatives were synthesized by Scotten-Baumann reaction from acryloyl chloride and several diamines in the presence of triethylamine, and their chemical structures were identified by IR, $^1H-NMR$, and elemental analysis. The yields of the synthesized N,N'- hexamethylenebisacrylamide and N,N'-dodecamethylenebisacrylamide were 56.0% and 70.4%, respectively, and were increased with the increased number of methylene group in the dimines. The yields of the synthesized N,N'-1,4-phenylenebisacrylamide and N,N'-2,6-pyridinebisacrylamide were 80.3% and 83.1%, respectively, which were higher than those of the alkylenebisacrylamide derivatives. The synthesized N,N'-bisacrylamide derivatives can be used to prepare various crosslinked polymers.

Changes of Phenolic Compounds and Pectin in Asian Pear Fruit during Growth (배의 생장시기에 따른 페놀성물질과 펙틴의 변화)

  • The changes in phenolic compounds and pectin content were investigated during the growth of 3 cultivars (Hosui, Niitaka and Chuwhangbae) of Asian pear (Pyrus pyrifolria) fruits. The amounts of total phenolic compounds in peel, flesh and core was 20.61-22.98mg/g, 0.87-1.23mg/g and 6.39-37.96mg/g during early growth, respectively, and decreased with pear growth. Arbutin, chlorogenic acid and epicatechin were detected in each part of all three cultivars during early growth; of these, arbutin content was the highest. Caffeic acid, catechin and 4-hydroxymethyl benzoic acid were detected in the peel and core during the ripening process. The total pectin content in the flesh of Hosui, Niitaka, and Chuwhangbae cultivars decreased from 5.93mg/g, 5.99mg/g and 5.40mg/g to 1.07mg/g, 1.60mg/g and 1.63mg/g, respectively. Of the soluble pectins, the hydrochloric acid-soluble pectin content was the highest, 3.21-3.45mg/g, and decreased during growth.
