원유로 오염된 지역으로부터 분리한 생물계면활성제 생산균주, Pseudoalteromonas sp. HK-3의 특성조사 (Characterization of the Biosurfactant-Producing Bacterium, Pseudoalteromonas sp. HK-3 Isolated from the Crude-Oil Contaminated Areas)

  • 본 연구의 목적은 원유에 오염된 지역의 토양시료로부터 분리한 생물계면활성제 생산균주의 특성을 조사하기 위하여 실시하였다. 1% 원유를 포함하는 배지에서 균주 HK-3의 배양기간 동안, 생장, 생물계면활성제의 생산, pH의 변화 등을 조사하였다. 생물계면활성제의 생산능력이 뛰어난 균주인 HK-3를 선별하여, 이 균주의 배양기간에 따른 생장 변화와 생물계면활성제의 생산량, 그리고 pH에 대하여 관찰하였다. HK-3는 배양 36시간이 경과하였을 때, 가장 많은 양의 생물계면활성제를 생산하였다. 생물계면활성제 생산에 대한 부가탄소원의 효과를 알아보기 위하여 HK-3를 다른 부가탄소원(예, glucose, dextrose, mannitol, citrate, acetate)과 함께 배지에서 배양하였다. 그 결과, 생물계면활성제의 최대생산량은 mannitol을 포함하는 BH 고체평판배지에서 관찰되었으며, 투명대의 면적은 직경은 약 7.64 cm2로 측정되었다. 분리균주의 생리학적 및 생화학적 특성을 조사하였으며, 주사전자현미경을 통하여 불규칙한 막대형의 세균으로 관찰되었다. BIOLOG 시스템과 16S rRNA 염기서열을 이용한 계통유전학적 분석을 통하여 동정하여 Pseudoalteromons 종으로 확인되어 Pseudoalteromons sp. HK-3로 명명하였으며, GenBank에 [FJ477041]로 등록하였다.

청국장에서 분리한 Bacillus licheniformis HK-12의 혈전용해활성과 프로테옴 분석 (Fibrinolytic Activity and Proteomic Analysis of Bacillus licheniformis HK-12 Isolated from Chungkuk-Jang)

  • 자연발효된 청국장으로부터 혈전용해활성을 가지는 세균 Bacillus licheniformis HK-12를 농화분리하여, 배양기간 동안 생산된 혈전용해활성을 가지는 단백질에 대해 프로테옴 분석을 실시하였다. B. licheniformis HK-12를 액체 영양배지에 접종하여 얻어진 배양상등액을 피브린 평판법(fibrin plate method)을 사용하여 효소활성을 측정하였다. 그 결과, HK-12의 혈전용해 활성은 대조구인 plasmin보다 약 2.3배정도 높은 활성을 나타내었다. 효소는 배양상등액을 ammonium sulfate 침전, DEAE-cellulose chromatography, Sephadex chromatography 등을 수행하여 분리정제하였으며, 정제된 혈전용해효소의 분자량은 SDS-PAGE를 통해 약 23 kDa로 측정되었다. 배양시간에 따른 HK-6의 세포외 단백질의 변화를 2-D PAGE 분석을 통하여 분석하였다. 그 결과 36시간배양 후에 가장 현저하게 유도된 spot #1을 분리하였으며, MALDI-TOF MS를 이용하여 단백질 동정을 실시한 결과, 유도된 단백질의 아미노산 서열은 $^1EKKIEKYREEEORLK^{15}$으로서, serine protein kinase (PrkA) (AAU22526)로 확인되었다.

  • The antiulcer effects of newly synthesized prostaglandin derivatives were investigated in various experimental ulcer models and on gastric secretion in rats. HK-3 and HK-4, PG $E_2$derivatives, prevented the formation of acute gastric ulcer induced by ethanol or aspirin in pylorus-ligated rats. The ulcer formation was moderately inhibited by HK-1 and HK-2, PG $F_{2{\alpha}}$ derivatives, and aggravated by SK-1, SK-2 and SK-3, PG $F_{2{\alpha}}$ derivatives. HK-3 and HK-4 reduced the volume, acid output and pepsin output of gastric juice in pylorus-ligated rats. The gastric perfusion with physiologic saline(pH 6.0) showed relatively constant acid secretion and indomethacin increased the acid secretion. The acid secretion was markedly decreased by PG $E_2$but PG $F_{2{\alpha}}$ caused little change. Prostaglandin derivatives, especially HK-3 arid HK-4, significantly inhibited the acid secretion induced by indomethacin. The results show that, PG $E_2$ derivatives, HK-3 and HK-4, inhibit acid secretion and also have protective effects on gastric ulceration induced by ethanol or aspirin.

폭약 Hexahydro-1,3,5-trinitro-1,3,5-triazine(RDX)에 노출된 분해세균 Pseudomonas sp. HK-6의 세포반응과 형태변화 (Cellular Responses and Morphological Changes of RDX-degrading Bacterium, Pseudomonas sp. HK-6 Exposed by Explosive Hexahydro-1,3,5-triaitro-1,3,5-triazine (RDX).)

  • 환경 오염원으로서 폭약 hexahydro-1,3,5-trinitro-1,3,5-triazine(RDX)에 대한 RDX 분해세균 Pseudomonas sp. HK-6의 세포반응과 형태변화에 대하여 조사하였다. 아치사조건의 RDX농도와 노출시간에 따른 균주 HK-6의 생존율을 분석한 결과, 이 세균의 생존율은 스트레스 충격 단백질의 생성과 비례하였다. 총세포 지방산 조성분석에서 균주 HK-6는 trypticase soy agar(TSA)에서 자랄 때 보다 RDX배지에서 자랄 때 여러 가지 종류의 지방산이 생성되거나 사라지는 것이 밝혀졌다. Anti-DnaK와 anti-GroEL을 이용하여 SDS-PAGE와 Western blot을 통한 분석으로 균주 HK-6는 70 kDa DanK와 60 kDa GroEL을 포함하는 몇가지 스트레스 충격단백질을 생성하는 것으로 밝혀졌다. RDX에 노출된 HK-6배양에서 수용성 단백질 분획에 대하여 2-D PAGE를 실시하였으며, pH 3에서 pH 10 범위에서 약 300 spots가 silver로 염색된 gel상에서 관찰되었다. 그 결과, RDX에 대한 반응으로 10여개의 spots가 현저히 유도 발현되었다. RDX(0.135mM, 12시간)에 노출된 세포는 구멍이 나타나고 표면의 불규칙적인 형태 변화가 일어나 죽게되는 것이 주사 전자현미경을 통하여 관찰되었다.

  • The principal objective of this study was to determine the optimal conditions for the production of biosurfactant by the indigenous bacterium, Pseudoalteromonas sp. HK-3, originating from oil-spilled areas. The relationship between total biosurfactant production and the factors affecting biosurfactant production were evaluated by statistical analysis using SPSS software. The effects of various supplemental carbon sources (e.g., glucose, dextrose, mannitol, citrate, acetate) on the maximal production of biosurfactant by the test culture of Pseudoalteromonas sp. HK-3 was then evaluated. As a result, mannitol was found in this study to be the best supplemental carbon source for the production of biosurfactant. A spot inoculation of crude cultural liquid containing the HK-3 cells generated the largest clear zone, whereas only small clear zones appeared around the spots inoculated with either supernatant only or cell pellets following centrifugation. Our results demonstrated that the HK-3 test culture supplemented with 2% mannitol at an initial pH of 6 generated the maximal amount of biosurfactant within 72 h of incubation.

  • 이 연구의 목적은 자연 천연한 청국장에서 분비되는 혈전용해효소를 생산하는 Bacillus Iicheniformis HK-12의 혈전용해활성과 특성을 조사하기 위하여 실시한 것이다. 먼저 균주 HK-12의 생리생화학적 특성에 대하여 조사하였다. BIOLOG GP2 MicroPlate system과 16S rRNA 염기서열 분석을 통하여 균주를 동정하였고, 그 결과 Bacillus licheniformis로 확인되었고, B. licheniformis HK-12로 명명하였으며, 이 균주는 GenBank에 [Eu288193]로 등재하였다. 16S rRNA 염기서열 분석에 근거하여, B. licheniformis HK-12의 계통수를 작성하였다. B. licheniformis HK-12의 배양기간동안 세균의 생장, 혈전용해활성, pH의 변화를 모니터링하였다. 36시간배양 후, 배양의 최대 혈전용해활성은 대조군으로서 플라스민과 비교하여 약 2.25배로 나타났다. 혈전용해활성과 관련하여, B. licheniformis HK-12의 생장에서 최적 pH와 온도는 각각 초기 pH 7.0과 40$^{\circ}C$로 나타났다.

키누레닌 대사산물에 의한 신경세포 손상에 대한 Magnolol의 보호효과에 대한 연구 (Magnolol Attenuates Neuronal Cell Death Induced by Kynurenine Metabolite)

  • This study investigated the protective roles and mechanism of magnolol, from the stem bark of Magnolia officinalis against potential neurotoxin 3-hydroxykynurenine (3-HK)-induced neuronal cell death. For the evaluation of protective role of magnolol, we examined cell viability, apoptotic nuclei, change of mitochondrial membrane potential and caspase activity in human neuroblastoma SH-SY5Y cells. It was found that 3-HK induces neuronal cell death in the human neuroblastoma SH-SY5Y cell line. The reduced cell viability produced characteristic features such as cell shrinkages, plasma membrane blebbing, chromatin condensation, and nuclear fragmentation. The cells treated with 3-HK showed an increase in the concentration of reactive oxygen species (ROS) as well as in caspase activity. In addition, both are involved in the 3-HK-induced apoptosis. Magnolol attenuated the cell viability reduction by 3-HK in both a dose- and time-dependent manner. Optical microscopy showed that magnolol inhibited the cell morphological features in the 3-HK-treated cells. Furthermore, the increase in the ROS concentration and the caspase activities by 3-HK were also attenuated by magnolol. These results showed that magnolol has a protective effect on the 3-HK induced cell death by inhibiting ROS production and caspase activity.

HK-1000 약침이 흰쥐의 고지혈증 및 동맥경화증에 미치는 영향 (A Study on the Effect of Herbal-acupuncture with HK-1000 Solution on Hyperlipidemia and Arteriosclerosis Induced Rats)

  • Objective&Methods: The purpose of this study is to observe the effects of HK-1000 Herbal-acupuncture solution at GB34(Yangleungchean) on hyperlipidemia and arteriosclerosis in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum, urine and liver, as well as the histological change of liver and aorta. Results: 1. HK-1000 herbal-acupuncture infusion increased the cell viability rate, DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. The levels of total cholesterol, free cholesterol, LDL-cholesterol, phospholipid in serum and AI(atherogenic index) were decreased, and the ratio of HDL to TCL(HDL/TCL) and the level of TG in serum were increased as compared with those of the control group. 3. In the HK-1000 group, serum AST and ALT were significantly lower than those of the HG group and saline group. 4. Hepatic GSH and catalase activities were significantly increased as compared with those of the saline group. Conclusions: From the above results, it is suggested that HK-1000 herbal-acupuncture at GB34 has a therapeutic effect on hyperlipidemia and arteriosclerosis.

  • In order to obtain a good microorganism capable of degrading microcrystaline cellulose (avicel), the screening test was carried out from soil and brown-rot wood. 8 strains which had good avicel-hydrolyzing activity were isolated. Among them, HK 47 which exhibited the highest avicel hydrolyzing activity was identified as Trichoderma sp. HK 47. Maximum avicel-hydrolyzing enzyme production from Trichoderma sp. HK 47 was obtained with the optimum medium contained carboxymethylcellulose 1.5% as carbon source, NaN030.75% as nitrogen source, KH2P040.5%, MgSO4.7H2O 0.1%, Tween 800.005% (V/V) during stationary cultivation at pH 6.0, 3$0^{\circ}C$ In this case, the production of avicel-hydrolyzing enzyme was 0.028 U/ml.

MRI 보어 구경에 따른 검사 시 실효 단면적 분석 (Analysis of Actual Cross-Sectional Area During Scanning According to MRI Bore Size)

  • In this study, we tried to quantify the actual cross-sectional area inside the bore when scanning by the MRI system with various bore sizes. To this end, a comparative analysis was conducted by both of blueprint of each MRI equipment and actual measurement in the field. As a result of analysis, ACSA(Actual Cross-Sectional Area) in Ingenia CX, Elition X, uMR 780, Omega, Vida, Lumina, Architect, Premier is recorded as 171230, 232150, 242100, 309332, 230760, 230760, 229380 and 235990 ㎟, respectively ACSA% was 60.6, 60.3, 73.0, 70.0, 60.0, 60.0, 59.6, and 61,3%. In addition, DTB (Distance from Table top to Bore top) recorded 400, 407, 445, 495, 405, 405, 405, 403, and 412 mm. Through this study, it was confirmed that there is a difference between the bore size according to each MRI system and the actual cross-sectional area during MRI scanning. Accordingly, if we consider the internal actual area just not bore size at the clinical site, useful diagnostic images can be obtained in the end with better convenience.
