Effects of Methanol Extracts from Rhynchosia nulubilis on serum Lipid Concentrations in Rats fed High Fat and High Cholesterol Diet (서목태(Rhynchosia nulubilis) 추출물이 고지방 및 고콜레스테롤 식이 급여 흰쥐의 혈청 지질 농도에 미치는 영향)

  • The effects of the Rhynchosia nulubilis extracts on serum lipid concentrations were evaluated in rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10g$ were divided into six groups and fed high fat and high cholesterol diets for four weeks. Experimental groups were administered with following diets; basal fat diet(control) and basal and high fat diets with 1% cholesterol and Rhynchosia nulubilis extract(40mg/kg, 400mg/kg and 4000mg/kg). The concentrations of serum triglyceride in rats fed the Rhynchosis mulubilis extract, high fat, 1% cholesterol, and RN-HFC(-40, -400, -4000mg/kg) groups were lower than those in other groups. The concentrations of total cholesterol in the RN-HFC (-40, -400, -4000mg/kg) groups were lower than those in fat diet groups. The concentrations of HDL-cholesterol in serum of the RN-HFC(-40, -400, 4000mg/kg) groups were significantly higer than those of other groups. The levels of LDL-cholesterol in serum of the RN-HFC(-40, -400, -4000mg/kg) groups were tended to be lower than those of other groups. These results suggest that Rhynchosia nulubilis extract may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.

  • This study investigated the effect of Dolichos lablab extract on the lipid metabolism of rats fed a high fat and cholesterol diet. S-D male rats (n=48) were given 40, 400, or 4000 mg/kg of Dolichos lablab extract and fed a high fat and high cholesterol diet for four weeks (DL-HFC group). As a result, serum levels of total lipids, total cholesterol, and triglycerides were higher in the high fat and high cholesterol group (HFC-group) than the control, and significant differences were observed between the control and the DL-HFC (40, 400, 4000 mg/kg) group. The fat absorption rate increased with a high fat diet feeding, but was higher in the DL-HFC (40, 400, 4000 mg/kg) group.

Effect of Marine Fish Diet on Serum Cholesterol Metabolism in Normal Subjects (Marine Fish Diet이 정상 성인이 혈중 콜레스테롤 대사에 미치는 영향)

  • It has been established that hypercholesterolemia is a major risk factor for atherosclerosis. Recent data showed that the incidence of hypercholesterolemia increase in Korea. n-3 Fatty acids lower serum triglyceride profoundly but the effect on serum cholesterol is not clear. This study was performed to assess the effects of low and moderate supplementation of marine n-3 fatty fish on serum cholesterol in young healthy korean woman. Nineteen subjects were divided into two groups. Each group receive an experimental diet supplemented with either 100g (group I) or 200g mackeral(Scomber japonicus) fish(group II) for 1-week. The diet was designed to avoid in which the amount of n-6 fatty acids would be much greater than that of n-3 fatty acids. MUFA, SFA intakes were similar in the two diets. The ratio of n-6/n-3 fatty acids was 1 : 1.l for group I, 1 : 2.51 for group II. The average daily n-3 fatty acids consumption from fish was 3.87g/day (1.03g EPA, 2.84g DHA)for group I, 7.74g/day (2.06g EPA, 5.68g DHA) for group II. Blood samples were obtained 2 times before experimental diet, immediately after experimental diet for 1-week. After experimental diet for 1-week, the serum total-cholesterol levels decreased significantly (16.4$\pm$15.1mg/dl, p<0.01) in group II and lowered slightly (13.7$\pm$25.8mg/dl)in group I. There were no significant changes from baseline to the end of the study in serum HDL-cholesteol, LDL-cholesterol, HDL-C/T-C ratio, and LDC-C/HDL-C ratio. The results suggest that the moderate levels of marine n-3 fatty fish consumption could improve serum cholesterol in normal subjects, therefore in might be of value in the prevention of atherosclerosis. However the clinical usefulness of moderate n-3 fatty fish consumption of hypercholesterolemic subjects will require further study. Also further studies are required to elucidate the long-term effects of low n-3 fatty fish consumtion.

The Effects of Lowering Dietary Fat and Cholesteol on Hypercholessterolemic Men (지방과 콜레스테롤 섭취 제한이 고콜레스테롤혈증에 미치는 효과)

  • In this study, we investigated the effects of lowering the fat and cholesterol in the diets of 26 Korean hypercholesteolemic men($\geq$240mg/dl). They consumed 2378kcal/day with 20.9% of the energy deriving from fat, and a cholesterol intake of 282mg(118mg/1000kcal). The experimental diet consisted of 2400kcal, the same as their usual diet, but the fat content was restricted to 15%, and the cholesterol level to below 100mg/1000kcal. The subjects kept to this diet for four weeks and were asked to maintain their usual life activities during the experimental period. The dietary intake and levels of plasma lipid, lipoprotein-cholesterol and apoprotein of the subjects were analyzed before, two weeks into, and after four weeks dietary intervention. After two of the dietary intervention, there were no significant changes of plasma total cholesterol or triglyceride levels but there was some changes of phospholipid level. However, after four weeks, the levels of plasma total cholesterol, triglyceride, and phospholipid had decreased significantly: 18.2%, 32.9%, and 11.9%, respectively. And the LDL-cholesterol and VLDL-cholesterol levels also showed a marked reduction of 18.1%, and 33.0% respectively without change of HDL-cholesterol level. There were no changes in the levels of Apo-A 1, Apo-B, or Lp(a). The changes of the plasma lipid levels were significantly associated with the changes in dietary fat intake but not the cholesterol intake. In conclusion, although the responses to the dietary intervention varied among the individual subjects, the lowering of dietary fat component from 21% to 15% of energy intake seems to be an effective way to reduce plasma cholesterol and triglyceride levels without decreasing HDL-cholesterol level. It was also found that the restriction of dietary cholesterol to below 100mg/1000kcal not seem to be effective for the hypercholesterolemic patient who already consuming below 300mg/day of cholesterol.

Effects of Artemisia iwayomogi Kitamura Ethanol Extract on Lowering Serum and Liver Lipids in Rats (흰쥐의 혈청 및 간지질 저하에 미치는 쑥 에탄올 추출물의 영향)

  • Artemisia iwayomogi(A. iwayomogi) is a major edible vegetable in Korea. A iwayomogi containing a strong bitter taste, particularly, is known to manifest an effect on cough, abdominal pain, indigestion and bleeding. In this experiment, ethanol extract of A. iwayomogi(50mg/kg body weight) was fed to growing male Sprague-Dawley rats for four weeks in order to examine the lipid lowering effect of A. iwayomogi in the live and serum. High fat diet included 10% lard, 1% cholesterol and 0.25% sodium cholate. The administration of A. iwayomogi extract decreased serum cholesterol compared with the control. At the same time, serum triglyceride(TG) was decreased to 25%. Although atherogenic index and HTR tended to decrease, phospholipid level in the serum was shown to increase by A. iwayomogi administration. At the same token, total liver lipids were decreased to 40%. The group fed both high fat and ethanol extract showed 28% decrease of liver lipids compared with the group fed only high fat diet. On the other hand, HDL cholesteol level of high fat and extract fed group was increased to 30% compared with the non-extract-fed control. Liver TG also was decreased to about 21% in the extract fed groups. Liver phospholipids, particularly, exceeded more than double of the level of high fat control. The above results indicate than A. iwayomogi exert a strong hypocholesterolemic and hypolipidemic effect which may lead to the protection from hypertention, obesity, stroke and many other circulatory diseases.

Anti-Obesity Effects of Fermented Samjung-hwan in Hign Fat Diet Rats (고지방 식이를 섭취한 흰쥐에서 발효 삼정환의 항비만 효과)

  • Objectives: This study was performed to evaluate the effects of fermented Samjung-hwan (SJH) extracts on weight, serum lipids and blood glucose. Methods: SJH was fermented using three different probiotic bacterial strains (Lactobacillus plantarum [LP], Leuconostoc mesenteroides [LM], Bifidobacterium longum [BL]) separately. Thirty-six rats were divided into normal, control (high fat diet), SJH-UF (high fat diet+unfermented SJH 200 mg/kg), SJH-LP (high fat diet+LP fermented SJH 200 mg/kg), SJH-LM (high fat diet+LM fermented SJH 200 mg/kg) and SJH-BL (high fat diet+BL fermented SJH 200 mg/kg). For 8 weeks later, we examined body weight, total cholesterol, high-density lipoprotein (HDL)-cholesterol and blood glucose. Results: The control group showed significantly increased weight gain compared with normal group and SJH-LP and BL groups had less weight gain than control group, significantly. In the lipid serum tests, control group showed significantly increased total cholesterol levels compared with normal group and only SJH-LP represented decreased total cholesterol levels compared with control group. However there was no significant change in the HDL-cholesteol levels. In the blood glucose tests, that of control group significantly incereased more than that of normal group, SJH-BL showed significantly decreased blood glucose levels compared with control group. Conclusions: SJH-LP, SJH-BL showed weight control effect, SJH-LP decreased TC and SJH-BL reduced blood glucose.

The Significance of Hyperlipidemia as a Predictive Factor of Relapse in Corticosensitive Nephrotic Syndrome (스테로이드에 반응을 보인 신증후군 환아에서 재발 예측인자로서 고지혈증의 중요성)

  • Purpose : One of the most difficult problems in the care of children with nephrotic syndrome remains the occurrence of relapses, despite initial response to steroids. Constantinescu reported that rapidity of initial response to steroid therapy could predict fewer relapses in the first year. So we evaluated the changes in serum lipid abnormalities in children with corticosensitive nephrotic syndrome before steroid treatment and the correlation between serum lipid levels and renal function, days to remission. Methods . We analyzed the Medical records of children who were managed by us between October 1994 and August 2000. In 33 patients with corticosensitive nephrotic syndrome, we evaluated the correlation between serum lipid levels and renal function [Creatinine clearance(Ccr)] and proteinuria before steroid treatment, and days to remission defined as the third day when the patient's urine becomes protein free. Results : There were 21 males and 12 females. Median age at presentation was 6.4 years (range: 1.8-17.3 years). Median days to remission were 15.4 days (range 4-42 days) on Prednisolone $60mg/m^2$ daily. The increased levels of triglyceride, total cholesterol, LDL cholesterol, apolipoprotein B, total cholesterol/HDL cholesterol, Lipoprotein(a) were observed. But the level of HDL cholesterol was not increased. Serum albumin was decreased a]id proteinuria was increased before steroid treatment. But Ccr was not decreased. There were negative correlation between serum albumin and total cholesterol (r = -0.5157, P<0.005), LDL cholesterol (r = -0.5543, P<0.005), total cholesterol/HDL cholesterol (r = -0.4506, P<0.01), lipoprotein(a) (r = -0.4570, P<0.025), apolipoprotein B (r = -0.5297, P<0.025), apolipoprotein B/apolipoprotein Al (r = -0.5851, P<0.01), apolipoprotein B/HDL cholesterol (r = -0.4961, P<0.05) before steroid treatment. There was no correlation between proteinuria and serum lipid profiles. Also Ccr and serum lipid profiles were not correlated. There was positive correlation between days to remission and HDL cholesterol (r = +0.4511, P<0.05), apolipoprotein B (r = +0.5190, P<0.05), apolipoprotein B/HDL cholesterol (r = +0.7169, P<0.005). Conclusions : This results reveal that HDL cholesterol, apolipoprotein B and apolipoprotein B/HDL cholesterol can be used as a predictive factor in corticosensitive nephrotic syndrome. We could not determine the significant level of these lipids for insufficient patients number, but these level may predict future relapses of corticosensitive nephrotic syndrome patients and thus may allow to better management and treatment protocols.   • The effects of phaseolus vulgaris extracts on serum lipid concentrations were evaluated in rats. Forty-two male Sprague-Dawley rats weighing $100{\pm}10\;g$ were divided into six groups and fed high fat diets for four weeks. Experimental groups were administered with following diets; basal fat diet(control) and basal and high fat diets with 1% cholesterol and phaseolus vulgaris extract(40 mg/kg, 400 mg/kg and 4000 mg/kg). The concentrations of serum triglyceride in rats fed the phaseolus vulgaris extract, high fat, 1% cholesterol, and PVHC-40, -400, -4000 mg/kg were lower than those in other groups. The concentrations of total cholesterol in the PVHC-40, -400, -4000 mg/kg groups were lower than those in fat diet groups. The concentrations of HDL-cholesterol in serum of the PVHC-40, -400, 4000 mg/kg groups were significantly higer than those of other groups. The levels of LDL-cholesterol in serum of the PVHC-40, -400, -4000 mg/kg groups were tended to be lower than those of other groups. These results suggest that phaseolus vulgaris extract may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.

Effects of Glycosyl-Sucrose on Blood Glucose, Serum Lipids and Activities of Carbohydrate Hydrolysing Enzymes in Rats (Glycosyl-Sucrose가 흰쥐가 혈당 및 지질농도와 당 가수분해 효소의 활성에 미치는 영향)

  • This study was designed to investigate the effects of dietary glycosyl-sucrose on blood glucose and serum lipids concentrations and carbohydrate hydrolysing enzyme activities in rats. Male Sprague-Dawley rate were divided into 3 experimental groups, cornstarch group(68% cornstarch diet), sucrose group (20% sucrose + 48% cornstarch diet) and glycosyl-sucrose group(20% glycosyl sucrose + 48% cornstarch diet). After 6 weeks, the mean values of food efficiency ratio of 3 groups were not significantly different. There were no significant differences in mean fasting blood glucose levels of three groups, while the glucose response at 30 and 60 minutes and the group as compared with sucreose area showed a tendency to decrease in glycosyl-sucrose group as compared with sucrose group. Serum lipids (T. G., phospholipids, HDL-cholesteol and total lipid) contents showed no significant differences among 3 groups but a tendency of jejunum and ileum were significantly reduced in glycosyl-sucrose group as compared with cornstarch group. The maltose hydrolysing enzyme activity, especially in jejunum was lower in glycosyl-sucrose group than in sucrose group.
