Comparison of Clinical Efficacy between an HPV DNA Chip and a Hybrid-Capture II Assay in a Patient with Abnormal Colposcopic Findings (질 확대경상 비정상 소견을 보인 환자에서 HPV DNA chip과 Hybrid-Capture II assay의 임상적 유용성 비교)

  • This study was performed to compare the efficacy between a DNA chip method and a Hybrid-Capture II assay (HC-II) for detecting human papillomavirus in patients with intraepithelial lesions of the uterine cervix. From May, 2005, to June, 2006, 192 patients with abnormal colposcopic findings received cervical cytology, HC-II and HPV DNA chip tests, and colposcopic biopsy or conization. We compared the results of HC-II and HPV DNA chip in conjunction with liquid based cervical cytology (LBCC) and confirmed the results of biopsy or conization. The sensitivity of the HPV DNA chip test was higher than HC-II or LBCC. The HPV DNA chip in conjunction with LBCC showed higher sensitivity than any single method and higher sensitivity than HC-II with LBCC. We confirmed that the HPV DNA chip test was more sensitive for detecting HPV in cervical lesions than HC-II, and that it would provide more useful clinical information about HPV type and its multiple infections.

Evaluation of p16INK4a/Ki-67 Dual Immunostaining in Liquid-based Cytology for Diagnosis of Uterine Cervical Dysplasia and Cancer (자궁경부 이형성증과 암의 진단을 위한 액상세포 검체에서 p16INK4a/Ki-67 이중면역염색의 평가)

  • Recently, $p16^{INK4a}$/Ki-67 dual immunostaining has been introduced as a new biomarker protocol for early detection of uterine cervical dysplasia and cancer in liquid-based cytology (LBC). We performed the $p16^{INK4a}$/Ki-67 dual immunostaining using a CINtec$^{(R)}$ PLUS kit in a total of 109 LBC cases of cervicovaginal smear and compared its results with those from LBC, HPV hybrid capture II (HC II) test and histological diagnosis. Expression of $p16^{INK4a}$ and Ki-67 was significantly associated with cases of LSIL or higher in cytological diagnosis and cases of cervical intraepithelial neoplasia (CIN) 1 or higher in histological diagnosis (p<0.001 and p<0.001, respectively). Among forty-six cases of atypical squamous cells of undetermined significance (ASCUS) in LBC, $p16^{INK4a}$ and Ki-67 was expressed in 31 (67.4%), which were positively associated with cases of CIN I lesion or higher in histology. The sensitivity of $p16^{INK4a}$/Ki-67 dual immunostaining for finding lesions of CIN 1 or higher was 89.0%, which was higher than LBC. The specificity was 73.5%, which was higher than that of the HC II test. Based on these results, the $p16^{INK4a}$/Ki-67 dual immunostaining method can be a useful diagnostic marker for improving the sensitivity of LBC and the specificity of HC II test.

  • Among 179 specimen diagnosed with ASCUS but did not go through biopsy or other tests, 26 cases (14.52%) showed positivity in HPV Hybrid Capture II Test. There were 25 cases of HPV high risk factor(+), marking 13.96%, and 3 cases of low risk factor(+) at 1.67%. According to cytology manifestation, patients with over LSIL can increase their complete recovery rate with continuous tests, but most patients diagnosed ASCUS do not go through any tests at all. It is considered that in order to prevent and start early treatment on cervical cancer, the HPV Hybrid Capture II Test must be enforced to patients diagnosed with ASCUS. Negative patients should go through routine screening, and positive patients should receive treatment after colposcopy.

Preparation of Iron Nanoparticles Impregnated Hydrochar from Lignocellulosic Waste using One-pot Synthetic Method and Its Characteristics (One-pot 합성 방법을 이용한 나노 철입자가 담지된 폐목재 기반 하이드로차의 제조 및 특성 평가)

  • In this study, iron nanoparticles impregnated hydrochar (FeNPs@HC) was synthesized using lignocellulosic waste and simple one-pot synthetic method. During hydrothermal carbonization (HTC) process, the mixture of lignocellulosic waste and ferric nitrate (0.1~0.5 M) as a precursor of iron nanoparticles was added and heated to 220℃ for 3 h in a teflon sealed autoclave, followed by calcination at 600℃ in N2 atmosphere for 1 h. For the characterization of the as-prepared materials, X-ray diffraction (XRD), cation exchange capacity (CEC), fourier transform infrared spectrometer (FT-IR), Brunauer-Emmett-Teller (BET), transmission electron microscope (TEM), Energy Dispersive X-ray Spectroscopy (EDS) were used. The change of Fe(III) concentration in the feedstock influenced characteristics of produced FeNPs@HC and removal efficiency towards As(V) and Pb(II). According to the Langmuir isotherm test, maximum As(V) and Pb(II) adsorption capacity of Fe0.25NPs@HC were found to be 11.81 and 116.28 mg/g respectively. The results of this study suggest that FeNPs@HC can be potentially used as an adsorbent or soil amendment for remediation of groundwater or soil contaminated with arsenic and cation heavy metals.

Concordance in Cervical HPV Detection between Hybrid Capture 2 and HPV GenoArray Tests

  • HPV type-specific detection may promote cervical screening program and vaccination development worldwide. We conduct a study comparing HPV Hybrid capture II (HC II) Test and Hybribio GenoArray test, a newly developed HPV type-specific assay, in patients with cervical epithelial neoplasm. Results showed a good concordance in cervical HPV detection between two tests (kappa value 0.80, p<0.05, McNemar test). Our study may promote utilization of type-specific HPV detection that is helpful for cervical cancer screening and vaccination.

EFFECTS OF CONDENSATION TECHNIQUES AND CANAL SIZES ON THE MICROLEAKAGE OF ORTHOGRADE MTA APICAL PLUG IN SIMULATED CANALS (모조 근관의 크기와 충전 방법이 orthograde MTA apical plug의 미세누출에 미치는 영향)

  • The purpose of this study was to compare the dye leakage of MTA (mineral trioxide aggregate) apical plug produced by two orthograde placement techniques (hand condensation technique and ultrasonically assisted hand condensation technique). To simulate straight canal, 60 transparent acrylic blocks with straight canal were fabricated. These transparent acrylic blocks were divided into 2 groups (Group C; hand condensation technique (HC) and Group U; ultrasonically assisted hand condensation technique (UAHC)) of 30 blocks with each MTA application method. Each group was divided into 2 subgroups (n=15) with different canal size of #70 (subgroup C70 and subgroup U70) and #120 (subgroup C120 and subgroup U120). After apical plug was created, a wet paper point was placed over the MTA plug and specimen was kept in a humid condition at room temperature to allow MTA to set. After 24 hours, remaining canal space was backfilled using Obtura II. All specimens were transferred to floral form socked by 0.2% rhodamine B solution and stored in 100% humidity at room temperature. After 48 hours, resin block specimens were washed and scanned using a scanner. The maximum length of micro leakage was measured from the scanned images of four surfaces of each resin block using Photoshop 6.0. Statistical analysis was performed with Mann-Whitney U test. Group U of UAHC had significantly lower leakage than Group C of HC in #70-size canal (subgroup U70) (p<0.05).

The Effects of High Torque Starters on the Starting Characteristics of a Micro-hybrid Engine (고토크 스타터에 의한 마이크로 하이브리드 엔진의 시동특성 개선 효과)

  • It is requested to shorten the starting duration for idle stop function equipped cars without harmful effects on the environment. Higher cranking speeds can be achieved with high torque starter. The object of this study is to develope the high torque starter and evaluate its effect on the exhaust emissions. The test was conducted on a 1.5 L, 4-cylinder, 16 valve, multipoint-port-fuel-injection gasoline engine. Engine out emissions such as HC, CO, $CO_2$, and the excess air ratios, lambda were measured using MEXA-554JK. The result showed that a high torque starter, HTS-II shortened the starting duration and reduced engine out emissions of HC, CO and improved starting performance with larger excess air ratio than that of the original starter, Org. S and a high torque starter, HTS-I.

  • The purpose of this study was to evaluate four different composite resins in vitro for microleakage in Class II box type restorations that have gingival margins apical to the cementoenamel junction. Forty caries free extracted human molars were used in this study. The Class II cavities were prepared 1.0mm below cementoenamel junction with a #701 carbide bur. The teeth were randomly divided into four groups, each group comprising 20 treated cavities according to adhesives and filling materials ; Group 1: Scotchbond Multipurpose/Z 100. Group 2: Ariston Liner/Ariston pHc, Group 3: One Step/Pyramid, Group 4: Prime & Bond NT/SureFil. To simulate the clinical situation during restoration placement, a restoration template was fabricated and composite resin was filled using a three sited light-curing incremental technique. The specimens were stored in the 100% humidity for 7 days prior to thermocycling. The specimens were immersed in 2% methylene blue dye solution for 24 hours and then embeded in transparent acrylic resin and sectioned mesiodistally with a diamond wheel saw. The degree of marginal leakage was scored under the stereomicroscope($\times$20) and the data were analyzed by Kruskal Wallis test and Mann Whitney test. (omitted)

  • This paper describes briefly the effect of ultrasonic fuel supply device on the performance of four stroke cycle gasoline engine. Experiments were carried out to clarify the effect of ultrasonic fuel supply device on the engine output, traveling fuel consumption ratio, exhaust emissions. The results were obtained as follows: 1.Engine output was increased 9-14% in comparison with that of the conventional injector. 2.Travelling fuel consumption ratio was improved 17-29% in comparison with that of the conventional injector. 3. CO, HC exhaust emissions was decreased compared to the value of the conventional injector. 4.Fuel consumption ratio in highway driving test was improved about 10% in comparison with that of the conventional injector.

Risk Factors Associated with HPV Infection in the Female Employees and Employees' Partner (여성 직장인 및 직장인 배우자의 인유두종 바이러스 감염 위험인자)

  • Objectives: Human papillomavirus (HPV) infection is highly associated with cervical cancer. So, the modification of the risk factors of HPV infection is essential for prevention of cervical cancer. This study was performed to evaluate the risk factors of HPV infection. Methods: HPV test of 12,337 study population conducted using Hybrid-Capture II assay(HC-II) and self-administered questionnaires were collected. The study population was people who visited hospital-based medical screening center from January to December 2007 and all were female employees or employees' partner. Results: In logistic regression analysis, smoking and alcohol drinking were significant factors, with odds ratios of 1.328 (95% CI 1.010~1.746) and 1.644 (95% CI 1.309~2.066), respectively. Nutritional supplements was also significant factor, which odds ratio was 1.161 (95% CI 1.004~1.343). Oral contraceptives was positive association with HPV infection (odds ratio 2.108; 95% CI 1.217~3.652), whereas condom was negative association (odds ratio 0.851; 95% CI 0.740~0.979). Conclusion: HPV Prevalence of 12,377 study population was 11.4%. Smoking, alcohol drinking, nutritional supplements and oral contraceptives were possible risk factors of HPV infection, and condom had possible preventive effect on HPV infection. Further prospective and comprehensive studies about HPV risk factors are required.
