Coating Method and Germination Test of Coated Tobacco (Nicotiana tabacum L.) Seeds (담배종자의 피복대형화방법과 발아에 미치는 영향)

  • Experiments were conducted to develop a pelletization method of minute tobacco seeds for easy handling at seeding by hand or for mechanical seeding. Serpentine, zeolite, and talc were tested as coating materials and the resulting pills were compared in size uniformity, hardness, length of time needed for coating, percentage of pills containing a single seed, and speed of disintegration when submerged in water. Talc was the poorest and zeolite was good only in hardness. On the other hand serpentine was good in most characters above, except for brittleness. This defect, however, was overcome by double coating, first with serpentine and followed by zeolite. This new pelletizing method results in optimum hardness, uniform size, and high ratio of pellets containing one tobacco seed. When compared to bare tobacco seeds, the double-coated seeds did not differ in germination test.

Geographical Variation of Galactomannan Composition in the Seeds of Glycine soja (돌콩 종자 함유 Galactomannan 조성의 지리적 변이)

  • In order to investigate the geographical variation of Glycine soja distributed in southern area of Korean peninsula, 8 local populations(Sokcho, Wonju, Mt. Chiak, Cheongju, Andong, Taegu, Ulsan, Sacheon), which located from $34^{\circ}50'00"N$ to $38^{\circ}12'00"N$, were selected according to their latitudes and geographical distances. The seeds of these populations were collected and their contents of mannose and galactose were investigated, Mannose contents in the seeds were variable in the range between the highest 460.00 mg/g (Andong) and the lowest 55.23 mg/g(Sacheon). The contents of galactose were represented remarkable differences from 67.17 mg/g(Sacheon) to 387.50 mg/g(Ulsan) also. The local populations were classified into 3 types such as the middle southern inland type (Andong, Taegu), the middle northern type(Wonju, Mt. Chiak, Cheongju) and the coastal type(Sokcho, Ulsan, Sacheon) according to the ratio of mannose and galactose, which indicate the hardness of seeds in Leguminosae, ranged from 0.41 to 1.73. Particularly, those of middle southern inland populations represented the high values compared with those of other populations.

Effects of Various Wavelength on the Hardness and the Free Amino Acid Contents of Soybean Sprouts (다양한 빛 파장 및 식물 스트레스 유발 화합물이 콩나물 경도 및 유리 아미노산 (Free Amino Acids)에 미치는 영향)

  • BACKGROUND: Effect of various light wavelength and plant defense molecules were evaluated on the hardness and the contents of free amino acid including ${\gamma}$-aminobutyric acid (GABA) in soybean sprouts. METHODS AND RESULTS: Germinating soybean seeds were treated with various single wavelength of light (380, 440, 470, and 660 nm) or without light at $25^{\circ}C$ for six days. Soybean seeds were also treated with stress-signaling molecule ethephon or $H_2O_2$ at the same time. Soybean sprouts treated with 470 nm substantially raised the hardness almost two times than the control. The free amino acid contents were higher in 470 nm and $H_2O_2$ treated soybean seeds than the control. Nutritionally beneficial GABA contents were increased by the treatments of 470 nm, 440 nm, ethephon, and $H_2O_2$. CONCLUSION: These results suggest that the hardness and the contents of amino acids can be regulated by stimuli, which stimuli could be composed of various wavelength and plant defense molecules. Especially, single wavelength 470 nm illumination has the effect of increasing GABA contents with increased hardness.

The Relationship between Hardness and Vitreousity of Korean Wheat Cultivars

  • Milling is an important process that determines the quality of flour and is affected by milling machine type, scale, and tempering conditions. In addition, seed hardness is an important factor in determining the amount of tempering water and has been reported to affects flour yield and flour quality. There are reports that vitreousity is used as a measure to distinguish between soft and hard seeds, and the higher the vitreousity, the higher the protein contents. However, there is no established system for measuring viterousity of seeds and studies on the vitreousity and quality characteristics of flour are insufficient. Therefore, in this study, vitreousity, hardness, and milling characteristics were evaluated for 46 major domestic varieties, and their relationship was confirmed. After cutting the seeds using a seed cutter, vitreousity was measured, and seed hardness and flour particle size was measured using SKCS and PSI, respectively. As for the seed hardness index, 'Joa' was the lowest with 11.6, 'Yeonbaek' was the highest with 78.7. As for the milling yield, 'Saeol' had the lowest at 58.1%, and 'Hcjoong' had the highest at 88.6%. Seed hardness index and wheat flour production showed a high positively correlation, showing a similar to that of previous studies. Also, in flour particle size, 'Gobun' was the largest at 75.5 pm, and 'Joa' was the smallest at 43.1 um. Flour yield and flour particle size showed a high positively correlation. As a result of vitreousity, 'Hwangeumal' (55.2%), 'Saekeumkang' (59.1%), 'Baekkang' (52.3%), 'Goso' (44.6%), and 'Joa' (19.2%) were showed. Seed hardness and vitreousity showed a high positively correlation. Also as the vitreousity increased, the flour yield also showed a tendency to increase. In addition, as the seed hardness increased, particle size of the flour yield also showed a tendency to increase. It is thought that this result can be used as a measure to determine the quality of flour with vitreousity. However, further analysis of wheat varieties and methods of analyzing vitreousity are needed.

Development of Seed Pelleting Technology for Rice and Cabbage (벼 및 배추종자 Pelleting을 물질채색 및 기술개발)

  • Seed pelleting have successfully been used in many crops for better crop estab-lishment and for mechanizing seeding process for small crop seeds in developed countries. In this experiment various pelleting materials and binders were tested to get basic information on the shape, hardness and germination of pelleted seesds of cabbage (cv. Seoul Beachoo) and rice (cv: Ilpoom). PLL-11, paper clay, lime and coal ash were good materials to make smooth shape of the pellets with pel gel and AG-11 as binders, and PLL-ll as material and pelgel and AG-11 as binders were the best among them in consideration of shape and hardness together. The hardness of the pelleted seeds were differed with each other depending on both of the pelleting materials and binders. Pelleted cabbage seeds coated by pelgel as binder with different materials showed lower germination percentge than control in general, but the seeds pelleted by PLL-11 with different binders showed no restraint effects. When the cabbage seed pelleted by PLL-11 with pelgel as binder showed almost same germination percentage as control. The pH and electrical conductivity of the extract from bentonite and zeolite were very higher than other materials tested and germination percentage showed a little lower than control when the cab-bage seed planted on the filter paper damped with the extract. As a result, PLL-11 as pelleting material and pel gel and AG-11 as binder appeared the good materials to make pellets of cabbage seeds and rice in consideration of shape, hardness and germination.

Characteristics of Pellet Seed on Germination and Emergence in Onion(Allium cepa L.) (양파 Pellet 종자의 발아 및 포장출아 특성)

  • This study was conducted to evaluate development of seed pellet technique such as pellet polymer search, the shape formation and hardness, the germination and emergence rate of the pellet seeds for labor-saving and reducing production cost in onion(Allium cepa L.) cultivation. The pellet seeds shape formation was good such as kaolin, clay, ash, and gypsum, and clay was good shape formation but surface of pellet seed was cracked during the drying. PG(pearlite + gypsum) as pellet material and PVA as binder were the best among the material in consideration with shape and hardness together. The hardness of the pellet seeds was affected by polymers, the kinds and concentration of binders, and that degree was large at polymer. The high hardness polymers were gypsum and coal ash, but burned lime was the lowest hardness among the pellet material. The germination(GP) and emergence percentage(EP) of pellet seed with PG in vitro were the highest among the material, and that was 93.6, 91.8%, respectively. The EP of pellet seed with PG at 20, $25^{\circ}C$ were 91.3, 92.0%, respectively, The EP of pellet seed were over the 91%, and those did not show difference with field moisture capacity , and that of 5 and 6mm size seed were the highest as 92%, respectively. and other size seeds showed over 90%, too. The EP of pellet seed with PG was decreasing as increasing the sowing depth, and that of at 10mm sowing depth was the highest as 92.7%. The time to 50% emergence of that under 70% field moisture capacity was 158h, and that was delayed at 20h compare to control seed.

  • Influence of fungi living on the endocarp surface of depulped seeds of Panax ginseng on the dehiscence was investigated with a fungicidal treatment of the seeds and sterilization of the sand at the beginning of stratification. 1. Isolation frequency of the fungi living on the endocarp surface of de-pulped seed was reduced and hardness of the endocarp did not change significantly in seed treated with a fungicide Captan wp. 50%. A significant negative correlation $(r=-0.984^**)$ was found between the frequency of fungi isolation and the hardness of the endocarp. 2. Increase of water content in the seed treated with the fungicide was delayed 20days compared to the untreated. 3. Growth of the embryo and dehiscence of the seed was suppressed by the fungicide treatment. The length of the embryo was inversely proportional to the hardness of the seed. It is suggested that the fungi facilitate the softening of the endocarp thereby enhancing the supply of oxygen and water necessary for the embryo development, therefore, accelerate the growing of embryo and cause the dehiscence.

Differences of Water Absorption Property and Seed Viability according to Morphological Characters in Soybean Genotypes

  • The impermeable seed coat is valuable trait in soybean because impermeable seed retain viability for longer period than permeable seed under adverse conditions such as delayed harvest or prolonged storage. Soybean seeds of various size showing different seed hardness were examined for their water absorption and seed viability under adverse storage conditions. Of one hundred thirty nine genotypes, eight types of seeds having different seed hardness and seed size were used as material. Soybean genotypes showing high hard seed rate, GSI13125 (89%), GSI10715 (54%), and GSI10284 (42%), were slow in water absorption and low in the electroconductivity of seed leachate in distilled water. Germination of GSI10284 and GSI13125 that have higher hard seed rate was less affected by CSVT and artificial aging treatment indicating higher seed storability. The higher storing ability of both collections was confirmed by electroconductivity test for leachate. GSI10122 showed low seedling emergence when the seeds were artificially aged. This genotype was considered as to having a poor storing ability based on difference of electroconductivity before and after artificial aging. Among tests conducted in the experiment, CSVT could be used for determining storage life in legumes. In conclusion, water absorption property of seed was strongly related to the hardness that is directly related to the seed viability and storing ability in soybean seed.

Influence of film-coated Materials on Germination and Seedling Vigor of film-Coated Chinese Cabbage Seeds (필름코팅 소재들이 배추 종자의 발아 및 활력에 미치는 영향)

  • The purpose of this study was to identify for extenders, adhesives, colorants, and fungicides suitable for the production of high-quality film-coated seeds of Chinese cabbage. In addition, the effect of various additives used for film coating on the germination of coated seeds and seedling vigor was investigated to establish appropriate treatment conditions for film coating efficiency. The germination rate and germination speed in Chinese cabbage film-coated seeds did not differ significantly depending on the type and concentration of the extender, while the use of Talc resulted in superior granulation capacity and adhesion of the seed coat in comparison to Zeolite. There was no significant difference in the germination rate depending on the type of adhesive used for film coating of Chinese cabbage seeds, but the germination rate was slightly slower in the film-coated seeds using carboxymethyl cellulose as an adhesive. On the other hand, the seeds coated with a film using 1% PVA as an adhesive not only maintained hardness enough to withstand external pressure, but also showed a high germination rate. In addition, the suitable disinfectant for film coating was venoram, and the treatment concentration was good at 250 mg·L-1. The type of colorant used in the film-coated seeds of Chinese cabbage did not significantly affect the germination rate, germination speed, and seedling vigor. The seeds film-coated using the liquid colorant were superior in appearance to the seeds film-coated with the solid colorant, which implies the possibility of reduction in the manufacturing costs by using the water-soluble paints as colorants for film-coated seeds.

Intraspecific Variation of Glycine max According to the Ratio of Mannose to Galactose in the Seeds (콩(Glycine max)의 종자 함유 Mannose와 Galactose 함량비에 따른 종내변이)

  • In order to investigate the geographical variation of Glycine max distributed in southern area of Korean peninsula, 19 local strains(Goseng, Pyeongchang, Ganghwa, Pocheon, Geumsan, Seocheon, Jincheon, Danyang, Tongyeong, Sancheong, Gumneung, Wolseong, Woolneung, Wando, Naju, Gochang, Jangsu, Jeju, Bukjeju), which located from $33^{\circ}15'N$ to $38^{\circ}11'N$, were selected according to their latitudes and geographical distances. The seeds of these strains were collected and their contents of mannose and galactose were investigated. Mannose contents in the seeds were variable in the range between the highest 12.207mg/g(Gochang) and the lowest 6.648mg/g(Geumsan). The contents of galactose were represented remarkable differences from 9.967mg/g(Danyang) to 16.949mg/g(Bukjeju) also. The local strains were classified into 4 variation types such as the inland type I(Wolseong, Jangsu, Danyang), the coastal type I(Gochang, Naju, Jeju, Goseng, Woolneung, Bukjeju, Ganghwa, Seocheon, Tongyeong), the inland type II(Geumsan, Pocheon, Pyeongchang) and the coastal type II(Wando, Jincheon) and 2 strange strains(Gumneung, Sancheong) according to the geographical climatic type and the ratio of mannose/galactose, which indicate the hardness of seeds in Leguminosae, ranged from 0.63 to 0.78. The variation types are very significant genecologically as an evidence for microevolution related to speciation in cultivated plants.
