The Parallel Measurements of Stomatal Apertures and Apoplastic pH on Guard Cells from Epidermal Strips and Intact Leaves of Commelina communis

  • The effects of light and darkness on stomatal aperture and guard cell apoplastic pH in the intact leaf and in the isolated epidermal strips of Commelina communis have been investigated. Stomata in the intact leaf opened wide in the light. In contrast, stomata in the isolated epidermal strips did not respond clearly to light. To eucidate the relationship between the stomatal aperture and the guard cell apoplastic pH, apoplastic pH was measured. In the light the guard cell wall of intact leaf was acidified by pH 1.9 units, falling from pH 7.3 to pH 5.4 in the first 10 minutes. On the contrary, apoplastic pH of isolated epidermal strips changed slowly from pH 7.3 to pH 6.9 at 20 min. Stomata in the intact leaf closed rapidly in the dark. On the other hand, stomata in the isolated epidermal strips failed to close in dark. There was a slow increase in apoplastic pH on transfer to the dark after incubation for 1.5 h in the light and the level observed before the experiment was regained after around 40 min. When the isolated epidermal strips were transferred to the dark, apoplastic pH maintained a uniform level of around pH 7.2-7.4. These results indicate that the mechanism of stomatal opening and closing from isolated epidermal strips and intact leaves could be different.

Syntheses and Thermal Behaviors of Rb(FOX-7)·H2O and Cs(FOX-7)·H2O

  • Two new energetic organic alkali metal salts, 1,1-diamino-2,2-dinitroethylene rubidium salt [Rb(FOX-7)${\cdot}H_2O$] and 1,1-diamino-2,2-dinitroethylene cesium salt [Cs(FOX-7)${\cdot}H_2O$], were synthesized by reacting of 1,1-diamino-2,2-dinitroethylene (FOX-7) and rubidium chloride or cesium chloride in alkali methanol aqueous solution, respectively. The thermal behaviors of Rb(FOX-7)${\cdot}H_2O$ and Cs(FOX-7)${\cdot}H_2O$ were studied with DSC and TG methods. The critical temperatures of thermal explosion of the two compounds are 216.22 and $223.73^{\circ}C$, respectively. Specific heat capacities of the two compounds were determined with a micro-DSC method, and the molar heat capacities are 217.46 and $199.47\;J\;mol^{-1}\;K^{-1}$ at 298.15 K, respectively. The adiabatic times-to-explosion were also calculated to be a certain value of 5.81 - 6.36 s for Rb(FOX-7)${\cdot}H_2O$, and 9.92 - 10.54 s for Cs(FOX-7)${\cdot}H_2O$. After FOX-7 becoming alkali metal salts, thermal decomposition temperatures of the compounds heighten with the rise of element period, but thermal decomposition processes become intense.

Antimicrobial Activity of Garlic Juice against Escherichia coli O157:H7 (마늘즙의 Escherichia coli O157:H7에 대한 항균작용)

  • The antimicrobial activity of fresh garlic juice against Escherichia coli O157:H7 was investigated. When E. coli O157:H7 was cultured for 18 hr in the trypticase soy broth containing 1%, 3%, and 5% garlic juice, viable cell number of E. coli O157:H7 was reduced to $2.3{\times}10^2\;CFU/mL$ at 5% from $7{\times}10^8\;CFU/mL$ at the non-treated culture, respectively. The inhibitory effects of the ground beef treated with 3%, 6%, and 10% garlic juice against E. coli O157:H7 was significantly enhanced with approximate 2 log-reduction compared to that of ground beef without garlic. There was no significant difference in the inhibition of E. coli O157:H7 among the groups with different amounts of garlic juice (p<0.05). These results suggest that garlic juice may function well as a natural preservative in food system.

Effect of Incubation Temperature and pH on Chlamydospores Germination of Cylindrocarpon destructans Causing Root Rot of Panax ginseng (인삼 뿌리썩음병균 Cylindrocarpon destructans의 후막포자 발아에 미치는 배양온도 및 pH의 효과)

  • Effects of incubation temperature and pH on chlamydospore germination of Cylindrocarpon destrcutans (isolate CY-9802) causing root rot of Panax ginseng were studied. Germination rate of the chlamydospores on Czapek solution agar(CSA) was higher than on potato dextrose agar(PDA) at the incubation temperatures tested. The chlamydospores were able to be germinated at range of 5$\^{C}$ to 30$\^{C}$ after 48 hours incubation on CSA. Germination rate was 53.2∼6.27% at range of 15$\^{C}$ to 25$\^{C}$, and the optimum temperature was 20$\^{C}$, whereas they were very low at 30$\^{C}$ on PDA. Germination rate was 43.6% to 47.9% at range of 10$\^{C}$ to 20$\^{C}$, and the optimum temperature was 20$\^{C}$ as well. They were able to be germinated at pH of 5.2 to 8.1 on CSA and 5.2 to 7.2 on PDA. Optimum pHs for the germination on CSA and PDA were from 6.4 to 8.2 and from 5.2 to 6.0, respectively. Mycelial color of the fungus on CSA was pale brown at pH from 5.2 to 6.0 and white from pH 6.4 to 8.1, while it was typical dark brown ar range of pH 5.2 to 7.1 and brown at pH 7.2 on PDA after 21 days incubation.

$\alpha$- and $\beta$-Amylase Isozyme Expresser Native Proteins in Tropical Silkworm Bombyx mori L.

  • Amylase isozyme based three multivoltine viz., N+p, Np, N+ $p^{cho}$ and two bivoltine-D6+p, D6p syngenic lines (Syn. L) were developed from germplasm (GP) stocks Nistari (N) and D6 respectively. haemolymph isozyme pattern at pH 7.0 and 8.5 depicted a total 11 number (Am $y_{1 to 6}$ at pH 7.0 and Am $y^{l to 5}$ at pH 8.5) of native proteins (NP) of various sizes are amylase isozyme expressers. Among eleven NPs, two NPs of 770 kDa (Am $y^{6}$ at pH 7.0) and 376 kDa (Am $y^3$ at pH 8.5) are $\alpha$-amylase expressers and remaining NPs of 370, 364, 350, 329 and 274 kDa at pH 7.0 and 206, 292, 416, 725 kDa at pH 8.5 are $\beta$-amylase expressers. Accordingly, digestive juice amylase isozyme pattern at aforesaid pH also depicted a total number of 10 NPs (Am $y^{1 to 5}$) at each pH 7.0 and 8.5 are amylase expressers of which NP of 387 kDa (Am $y^4$ at pH 7.0) and 780 kDa (Am $y^{5}$ at pH 8.5) are a-amylase expresser. Remaining NPs of 338,297 & 216 kDa at pH 7.0 and 370, 341, 329 &302 kDa at pH 8.5 are $\beta$-amylase expresser. Recurrent backcross lines (RBL) viz., N+pRBL and NpRBL were developed through introgression of high shell weight character (a multigenic trait) to be used further for congenic line (Con. L) development and to understand any association with introgressed character. Isozyme pattern in haemolymph of RBLs depicted only one $\alpha$-amylase of 770 kDa at pH 7.0 and 376 kDa at pH 8.0 with three and four respective $\beta$-amylase bands but in bivoltine lines numbers of $\beta$-amylase bands vary between 1 to 2 at aforesaid pH. Variability was also observed in digestive juice of multivolitine and its RBLs but bivoltine lines express null activity at both pH except appearance of one very week $\alpha$-amylase band D6+p at pH 8.5. Overall study suggests that not a single NP at both pH is common for expression of any band of amylase isozyme i.e., a totally different set of proteins are the amylase isozyme expresser at specific pH and no molecular factor of amylase is associated in developed RBLs which showed improvement on survival, single cocoon shell weight (SCSW) and single filament length over receptor parents.s.s.s.

A Study for Kinetics and Oxidation Reaction of Substituted Benzyl Alcohols Using (C10H8N2H)2Cr2O7 ((C10H8N2H)2Cr2O7를 이용한 치환 벤질 알코올류의 산화반응과 반응속도에 관한 연구)

  • $(C_{10}H_8N_2H)_2Cr_2O_7$ was synthesized by reacting 4,4'-bipyridine and chromium (VI) trioxide. The structure of the product was characterized with FT-IR (infrared spectroscopy) and elemental analysis. The oxidation of benzyl alcohol using $(C_{10}H_8N_2H)_2Cr_2O_7$ in various solvents showed that the reactivity increased with the increase of the solvent dielectric constant, in the order of DMF (N,N'-dimethylformamide) > acetone > chloroform > cyclohexane. In the presence of DMF, an acidic catalyst such as $H_2SO_4$ $(C_{10}H_8N_2H)_2Cr_2O_7$ oxidized benzyl alcohol (H) and its derivatives ($p-OCH_3$, $m-CH_3$, $m-OCH_3$, m-Cl, $m-NO_2$). Electron donating substituents accelerated the reaction rate, whereas electron acceptor groups retarded the reaction rate. Hammett reaction constant (${\rho}$) was -0.70 (308 K). The observed experimental data were used to rationalize the hydride ion transfer in the rate determining step.

Study on the Antithiamin Activities of Synthetic Antioxidants (합성항산화제 항Thiamin성에 대한 연구)

  • Antithiamin activities of BHA, BHT, PG and TBHQ of synthetic antioxidants on the effect of temperature and pH was determined by means of HPLC. The influence of synthetic antioxidants on the degration of thiamin was found to be dependent on temperature and pH. The degradation of thiamin was considerably more rapid at pH 7 than pH 4. The influence on the heat of synthetic antioxidants at pH 4 and 38* was extremely slight, but the degradation of thiamin at pH 7 was much more rapid at 60* than at 38*. After 24 hours of incubation both PG and TBHQ at pH 7 and 60* nearly completely destroyed thiamin. Tests of antithiamin activities showed that TBHQ, which was decomposed completely in 72 hours, was more effective than PG at pH 7 and 38* but BHA and BHT hardly had antithiamin activities which was evaluated under various reactions of pH and temperature. Thiamin degradation, at pH 7 and 6$0^{\circ}C$, was proportional to the concentration of PG. When the ratio of PG to thiamin was increased from 0.15:1 to 2:1, the degradation rate also increased. However, the change between ratio of 1:1 and 2:1 was negligible.

Detection of E. coli O157:H7 and Listeria monocytogenes, and Appraisal for Microbiological Qualities in the Commercial Frozen Yogurt Products in Korea (국내 시판용 Frozen Yogurt의 병원성 미생물 검출 및 미생물학적 품질 평가에 관한 연구)

  • Recently the high outbreaks of intestinal disease caused by the consumption of frozen dairy foods containing pathogenic bacteria has generated considerable interest in the causative agent such as Listeria monocytogenes and E. coli O157:H7. This study was carried out to detect the above pathogens and compare the microbiological qualities of three commercial forzen yogurt products. The main results obtained were as follows. L. monocytogenes coliforms and E. coli O157:H7 were not detected in the total of seven frozen yogurt samples. For microbiological qualities the viable lactic counts of products manufactured by FA company were about 2.9$\times$108 -1.6$\times$109cfu/ml 1.7$\times$106 cfu/ml for FB's and 1.2$\times$106 cfu/ml for FC's The PH values of FA's FB's and FC's products was in the range of pH 4.1~5.3 and the values of FA's were 4.1~4.6 compared by the pH 5.2~5.3 of FB's and FC's products. During refrigeration of the test samples the survival rates of L. monocytogenes spiked into thawed frozen yogurt sample(FA's FB,s and FC's) were 0.55% 15.61% and 16.89% respectively. On the other hand E. coli O157:H7 and L. monocytogenes were 12.4% and 25.0% for FA's 10.8% and 20.8% for FB's and 10.26% and 22.7% for FC's under 37$^{\circ}C$ storage, As the results described above each frozen yogurt products were different in microbiological qualities. The survival rates of pathogens spiked into the samples increased with the pH of the products. This indicates that the pH or any other factors pre-sumable supressed the growth of E. coli O157:H7 and L. monocytogenes in frozen yogurt products.

Inhibition of adhesion and biofilm formation in Escherichia coli O157:H7 by diosmin (다이오스민(diosmin)에 의한 병원성 대장균 세포부착 및 생물막 형성 억제)

  • Escherichia coli O157:H7 is one of the most common foodborne pathogens responsible for outbreaks of hemorrhagic colitis, which can lead to the life-threatening hemolytic-uremic syndrome. In this study, we identified phytochemicals that specifically inhibit the expression of LEE operon in E. coli O157:H7. Among phytochemicals, diosmin decreased the adherence of E. coli O157:H7 towards Caco-2 cells in vitro (p<0.01) and its biofilm formation activity (p<0.05). Quantitative RT-PCR analysis revealed that the transcripts of Ler-regulated genes and genes related to curli production were significantly reduced in the presence of diosmin. However, diosmin does not affect bacterial viability, indicating that the resistance rate to diosmin was remarkably low. Overall, these results provide significant insights into the development of a novel anti-infective agent that is different from conventional antibiotics.
