Survival and Physiological Response of Hybrid Grouper (Epinephleus bruneus♀ × Epinephleus lanceolatus♂) Exposed to Different Salinity Water (교잡바리(대왕자바리)의 생존과 생리학적 반응에 미치는 염분의 영향)

  • The profer salinity range for aquaculture of hybrid grouper (Epinephleus bruneus♀ × Epinephleus lanceolatus♂) were evaluated in terms of their survival, growth, feed intake and physiological responses after 30 days of exposure to different salinities (0 psu, 3 psu, 5 psu, 10 psu, 15 psu, 20 psu, 25 psu, 30 psu). Hybrid grouper all died at 0 psu salinity on the 4th day of exposure but all hybrid survived at a salinity of 5 psu or higher during the exposure period. Body length and weight increased above 5 psu salinity and the growth rate was the highest at 30 psu. Hybrid exposed to 3 psu did not eat the feed pellet. Feed intake did not show a significant difference between salinity 20 psu, 25 psu and 30 psu. Plasma osmolality of the juvenile hybrids exposed to different salinities was 341~368 mg Osmol/kg above 5 psu. The oxygen consumption rate was the highest at 30 psu with 163.6±22.3 mg O2/kg fish/h. SOD, CAT and GSH-PX were significantly higher at 15 psu. Therefore, the critical salinity for survive of the juvenile hybrid was 5 psu, and the proper salinity for optimal culture was analyzed with 20~30 psu.

  • Methods to separate 14 rare earth elements (REEs) and yttrium by the $AG^{(R)}$ 50W-X8 cation exchange resin, and to determine REEs by inductively coupled plasma atomic emission spectrophotometry (ICP-AES) were described. Ion exchange capacities of REEs on the resin were so high that the REEs were quantitatively ion exchanged under the flow rate of 0.3~1.0 mL/min at pH 1~6. The breakthrough capacity curve of the REEs showed that ion exchange capacities of light REEs (Cerium group) were greater than that of the heavy REEs (Yttrium group). When $200{\mu}g$ of each REEs was ion exchanged on 100 mg of resin, most of the heavy REEs were quantitatively desorbed with 10 mL of 2.0 M of $HNO_3$, while most of the light REEs with 30 mL. The method was applied to the monazite sample. The REEs could be separated from matrix, since ion exchange capacities of matrix ions of Ca, Ti, Mg, Mn were much lower than that of the REEs. However the relative standard deviations of the analytical results by the present method were not improved, as high as 1~5%.

Effect of Korean Ginseng on the Expression of Transferrin Receptor in the Liver Cell Membrane of Rat (백서의 간세포막에서 Transferrin Receptor의 발현에 미치는 인삼의 영향)

  • To investigate the effects of korean ginseng on the expression of transferrin receptor (TfR) in the liver cell membrane, we had carried out the experiments of $[^{3}H]thymidine$ uptake, $^{125}I-transferrin$ binding, and TfR mRNA expression in the liver after partial hepatectomy of normal and 3'-methyl-4-dimethylaminoa-zobenzene (3'-Me-DAB) treated rat with or without treatment of korean gingseng. $[^3H]thymidine$ uptake was not changed in the liver of 3'-Me-DAB or ginseng treated rat compared to that of control rat, but increased in that of partial hepatectomy of normal or 3'-Me-DAB treated rat. And this increased $[^{3}H]thymidine$ uptake was lowered slightly by the treatment of ginseng. Transferrin binding sites in the liver plasma membrane of ginseng treated rat with or without partial hepatectomy were similar, but increased in that of 3'-Me-DAB treated rat with or without partial hepatectomy compared to those of each control rat and these increased binding sites were reduced by ginseng treatment. Transferrin binding affinity (l/kd) was not changed by ginseng treatment, but tended to decrease in the liver of 3'-Me-DAB treated rat or in those after partial hepatectomy of all groups and reverse by ginseng treatment in 3'-Me-DAB treated rat. The expression of TfR mRNA was increased in the liver of 3'-Me-DAB treated rat with partial hepatectomy (peak at 24 hours), but lowered by ginseng treatment in this rat. From these results, it is suggested that korean ginseng has no effect on the increased expression of TfR with decreased affinity in the cell membrane of regenerated liver after partial hepatectomy of rat, but could inhibt that of 3'-Me-DAB treated rat through the regulation of DNA synthesis or TfR mRNA in partial.

Magnetized inductively coupled plasma etching of GaN in $Cl_2/BCl_3$ plasmas

  • In this study, $Cl_2/BCI_3$ magnetized inductively coupled plasmas (MICP) were used to etch GaN and the effects of magnetic confinements of inductively coupled plasmas on the GaN etch characteristics were investigated as a function of $Cl_2/BCI_3$. Also, the effects of Kr addition to the magnetized $Cl_2/BCI_3$ plasmas on the GaN etch rates were investigated. The characteristics of the plasmas were estimated using a Langmuir probe and quadrupole ma~s spectrometry (QMS). Etched GaN profiles were observed using scanning electron microscopy (SEM). The small addition of $Cl_2/BCI_3$ (10-20%) in $Cl_2$ increased GaN etch rates for both with and without the magnetic confinements. The application of magnetic confinements to the $Cl_2/BCI_3$ inductively coupled plasmas (ICP) increased GaN etch rates and changed the $Cl_2/BCI_3$ gas composition of the peak GaN etch rate from 10% $BCI_3$ to 20% $BCI_3$. It also increased the etch selectivity over photoresist, while slightly reducing the selectivity over $Si0_2$. The application of the magnetic field significantly increased positive $BCI_2{\;}^+$ measured by QMS and total ion saturation current measured by the Langmuir probe. Other species such as CI, BCI, and CI+ were increased while species such as $BCl_2$ and $BCI_3$ were decreased with the application of the magnetic field. Therefore, it appears that the increase of GaN etch rate in our experiment is related to the increased dissociative ionization of $BCI_3$ by the application of the magnetic field. The addition of 10% Kr in an optimized $Cl_2/BCI_3$ condition (80% $Cl_2/$ 20% $BCI_3$) with the magnets increased the GaN etch rate about 60%. More anisotropic GaN etch profile was obtained with the application of the magnetic field and a vertical GaN etch profile could be obtained with the addition of 10% Kr in an optimized $Cl_2/BCI_3$ condition with the magnets.

Effects of Feeding Methods (Feed vs. Water) of Vitamin E on Growth Performance and Meat Quality of Broilers

  • This research was conducted to compare the effects of vitamin E (VE) when supplemented in either feed or water on the performance and meat quality of broilers. For a six-week feeding trial, a total of 330 broiler chicks were allotted to five treatments. The treatments were 1) 0 ppm VE, 2) 10 ppm VE in feed, 3) 20 ppm VE in feed, 4) 5 ppm VE in water and 5) 10 ppm VE in water. During the starter phase (0-3 weeks) chicks on non-supplemented groups grew slower (p<0.05) than the supplemented ones and the same trend was followed during the finisher (4-6 weeks) and overall period (0-6 weeks). The feed intake was significantly higher in feed supplemented groups as compared with water-supplemented groups and at higher levels as compared with lower levels of supplementation. The nutrient digestibility studies conducted after 15 and 35 days on the feeding trial showed that the digestibility of all nutrients was significantly (p<0.05) higher in supplemented groups than the non-supplemented one. The dressing percentage was higher in supplemented groups, when fed in feed and at higher levels when compared with their respective counterparts. Similar trends were noticed with respect to bone resistance. The calcium and phosphorus contents in tibia were also significantly (p<0.05) higher in supplemented, feed fed groups at higher levels than other groups. The TBARS values measured after 5 and 10 days of storage, which reflect the degree of oxidation, showed significantly lower levels in supplemented diets. The plasma and muscle vitamin E levels also showed a positive linear correlation with the levels supplemented both in feed and water. Overall it can be inferred that supplementation of VE was beneficial and there was not much difference observed when fed either in feed or water at the levels measured in the present study.

Partition of Amino Acids Requirement for Maintenance and Growth of Broilers I. Lysine

  • Purified diets containing five graded levels of lysine were fed to a total of 125 growing chicks (25 chicks per treatment) to evaluate lysine requirements for growth and maintenance. A model was developed to separate lysine requirement for maintenance from requirement for growth. Based on weight gain response, the daily lysine requirement for growth was 12.06 mg/g gain and the daily lysine requirement for maintenance was 0.332 times metabolic body size ($W^{0.75}$). Similarly, the lysine requirement for growth was 0.457 mg/mg nitrogen gain and the daily lysine requirement for maintenance was 0.344 times metabolic body size. The plateau of plasma lysine concentration was reached at 354.75 mg intake/day. The total lysine requirement was 414.27 mg/day or 1.0% of the diet, 420.11 mg/day or 1.01% of the diet based on weight gain response and N gain response, respectively. Previous lysine requirements for growing chicks of 1-28 days old were in close agreement with these estimates. As a percentage of protein, lysine requirement was calculated to be 7.3% and the reported lysine content of chick muscle crude protein of 7.46% was closely related.

Effect of Casein Phosphopeptides on In Vitro Solubilization of Calcium in Fortified Cheese Pizza

  • Cheese pizzas fortified with casein phosphopeptide (CPP) and calcium were subjected to an in vitro digestion to assess whether CPP could prevent the precipitation of calcium. The total calcium content of the cheese pizzas was adjusted to 1,000 mg per pizza (~370 g) with the addition of calcium originating from eggshells. Two levels of trypsin-digested caseins (367 and 459 mg), with a CPP content of ~20%, were added to each pizza. The in vitro digested pizzas were then centrifuged and the supernatant was mixed with Na2HPO3 at 37℃ to estimate the possible soluble effect of CPP on calcium. After 24 h of reaction, the solution was centrifuged and the calcium content in the resultant supernatant was analyzed by inductively coupled plasma-atomic emission spectroscopy. One-way statistical analyses showed that CPP had a positive effect on the solubilization of calcium against phosphate (p<0.05). Cheese pizza supplemented with 459 mg of CPP powder was able to prevent precipitation of calcium by 98.8%, whereas no CPP-added cheese pizza solubilized 86.4% of the calcium. A sensory test was also carried out, revealing that panelists could not discern the bitter taste of the CPP added to the pizzas.

Attenuation of ROS Generation by KCNE1 Genes in Cisplatin-treated Auditory Cells

  • Potassium is essential for the proper functioning of the ears. The inner ear's endolymph differs from all other extracellular fluids (in its positive potential) and in the ionic compositions in the various parts of the endolymphatic space. Ion concentration of the endolymph is 150 mM of potassium, which is comparable to the concentrations in other organs. Cisplatin (cis-diamminedichloroplatinum II: CDDP) is one of the most effective anticancer drugs, widely used against various tumors. However, its clinical use is limited by the onset of severe side effects, including ototoxicity and nephrotoxicity. For ototoxicity, a number of evidences in cytotoxic mechanism of cisplatin, including perturbation of redox status, increase in lipid peroxydation, and formation of DNA adduct, have been suggested. Therefore, in this study, the author investigated the relationship between the potassium ions on cisplatin-induced cytotoxicity in HEI-OC1 cells associated with reactive oxygen species (ROS). KCNE1 gene expression by the concentration of intracellular potassium appeared in the plasma membrane and increased the concentration of intracellular potassium. Cisplatin decreased the viability of HEI-OC1 cells, but the KCNE1 gene increased. Also, the KCNE1 gene significantly suppressed generation of intracellular ROS by cisplatin. Western blot analysis showed that the KCNE1 gene increased phase II detoxification enzymes markers such as superoxide dismutase 1 (SOD1), superoxide dismutase (SOD2), NAD(P)H:quinine oxidoreductases (NQO1), which were associated with the scavenger of ROS. These results suggest that the KCNE1 gene for intracellular potassium concentration ultimately prevents ROS generation from cisplatin and further contributes to protect auditory sensory hair cells from ROS produced by cisplatin.

Electron Emission Properties of Hetero-Junction Structured Carbon Nanotube Microtips Coated With BN And CN Thin Films (탄소 나노튜브 위에 붕소 및 탄소 질화 박막이 코팅된 이종접합 구조 미세팁의 전자방출 특성)

  • Boron nitride (BN) and carbon nitride (CN) films, which have relatively low work functions and commonly exhibit negative electron affinity behaviors, were coated on carbon nanotubes (CNTs) by magnetron sputtering. The CNTs were directly grown on metal-tip (tungsten, approximately 500nm in diameter at the summit part) substrates by inductively coupled plasma-chemical vapor deposition (ICP-CVD). The variations in the morphology and microstructure of CNTs due to coating of the BN and CN films were analyzed by field-emission scanning electron microscopy (FE-SEM). The energy dispersive x-ray (EDX) spectroscopy and Raman spectroscopy were used to identify the existence of the coated layers (CN and BN) on CNTs. The electron-emission properties of the BN-coated and CN-coated CNT-emitters were characterized using a high-vacuum field emission measurement system, in terms of their maximum emission currents ($I_{max}$) at 1kV and turn-on voltage ($V_{on}$) for approaching $1{\mu}A$. The results showed that the $I_{max}$ current was significantly increased and the $V_{on}$ voltage were remarkably reduced by the coating of CN or BN films. The measured values of $I_{max}-V_{on}$ were as follows; $176{\mu}A$-500V for the 5nm CN-coated emitter and $289{\mu}A$-540V for the 2nm BN-coated emitter, respectively, while the $I_{max}-V_{on}$ of the as-grown (i.e., uncoated) emitter was $134{\mu}A$-620V. In addition, the CNT emitters coated with thin CN or BN films also showed much better long-term (up to 25h) stability behaviors in electron emission, as compared with the conventional CNT emitter.

The Antiulcer Effects of Alove vera on the Stomach ulcer Induced by Stress in rats (흰쥐의 스트레스성 위궤양에 대한 Alove vera의 항궤양작용)

  • This was performed to investigate the antiulcer effects of Aloe vera on the stomach ulcer induced by restraint and water-immersion stress in rats. For this experiment, 60 male Sprague-Dawley strain were used. The experimental groups were divided into five: a control(CA) and 4 aloe treatment groups. Each dose of aloe was 50 mg/kg BW(AA), 100mg/kg BW(AB), 200 mg/kg BW(AC), and 400 mg/kg BW(AD). The rats were allocated to each group by 12 and observed for 4 weeks. The results are as following. 1. The stomach surface pH in each group showed no significant difference, byt the values of aloe treatment groups were higher than the value of the control group. 2. The gastric wall mucus was significantly increased in all aloe treatment groups(p<0.05) compared with the control group. Especially in AC and AD group the differences were higher(p<0.01). 3. At shear rate rate 11.25, 45.0, 90.0, 225.0 sec-1, whole blood viscosity and plasma viscosity were measured. Most of the values of aloe treatment groups were significantly low compared with those of the control group(P<0.05). 4. The ulcer index of aloe treatment groups were significantly low campared with control group(p<0.05). Especially in AC and AD group the differences were more significant(p<0.01). 5. Less severe ulcers were observed in AA and AB group than in the control group. Tissues of AC and AD group had only slight ulcers and necrosis of tissue was not observed in these groups. Especially in AD group, there was more mucus than other groups and it seemed that alove vera stimulated the epithelial regeneration. From the results of this study, it can be concluded that the oral administration of Alove vera results in protection of stomach ulcer by stimulating the secretion of gastric mucus and the circulation of blood.
