잔토리졸과 어성초 추출물의 항균 효과 (Microbiological Effects of Xanthorrhizol and Houttuynia cordata Thunb. Extract)

  • Anti-microbiological effects of xanthorrhizol, ie., extract from curcuma xanthorrhiza and extract from houttuynia cordata Thunb. against influenza virus and E. coli O157 were tested. From the influenza experiments, the effects were shown above 93 % in case of houttuynia cordata Thunb. extract, however, the effects was not shown in case of xanthorrhizol. The effects were sustained in mixtures of houttuynia cordata Thunb. and curcuma xanthorrhiza extracts. We also tested the anti-microbiological effects of hand sanitizer containing houttuynia cordata Thunb. and xanthorrhizol. The effect of hand sanitizer containing 2,000 ppm of xanthorrhizol and 500 ppm of houttuynia cordata Thunb. extract was better than that of commercialized foreign product.

어성초의 아임계수 추출물의 항산화 활성 및 Acetylcholinesterase 저해 활성 (Antioxidant and Acetylcholinesterase Inhibitory Activities of Subcritical Water Extracts from Houttuynia cordata Thunb)

  • 어성초(H. cordata Thunb) 건조 분말(0.05 g)을 10 mL의 증류수와 함께 스테인리스 용기에 넣고 밀봉한 후 여러 온도(50, 100, 200, $300^{\circ}C$)에서 10, 30, 60분간 추출하였다. 추출온도와 시간은 어성초 추출물의 중요 성분, 항산화 활성, acetylcholinesterase 저해 활성에 크게 영향을 미쳤다. $300^{\circ}C$에서 30분간 처리한 어성초 추출물에서 가장 높은 페놀 함량($33.34{\pm}0.96\;{\mu}g$ gallic acid equivalents/mL), quercetin 함량($15.75{\pm}0.20\;{\mu}g/mL$), DPPH 라디칼 소거능($44.07{\pm}2.65%$), acetylcholinesterase 저해 활성($36.53{\pm}1.14%$)이 나타났다. 플라보노이드 함량은 $100^{\circ}C$ 10분 추출물에서 $2.55{\mu}g$ quercetin equivalents/mL로 가장 높게 측정되었다. $100^{\circ}C$ 이상의 고온에서 밀폐된 증류수는 높은 압력에서 아임계수 형태로 존재하는데 이러한 아임계수는 고품질의 어성초 추출물을 제조하는데 매우 유용하였다.

어성초 분말을 첨가한 설기떡의 품질 특성 (Quality Characteristics of Sulgidduk Prepared with Houttuynia cordata Thunb. Powder)

  • Sulgidduk samples made with additions of 2, 4, 6, 8, and 10% Houttuynia cordata Thunb. powder, and a control were examined for quality characteristics such as moisture content, water activity, color, gelatinization properties, textural characteristics, and sensory qualities in order to determine the optimal ratio of Houttuynia cordata Thunb. powder in the formulation. The moisture contents among the samples ranged from 41.98% to 44.97%, and increased as the content of Houttuynia cordata Thunb. powder increased. The water activities of the samples were not significantly different. As the Houttuynia cordata Thunb. powder content increased, the redness and yellowness of the samples also increased, but lightness decreased. For the gelatinization properties, the additions of Houttuynia cordata Thunb. powder caused decreases in peak viscosity (P) and holding strength viscosity (H). Furthermore, final viscosity (F), setback, and time to peak viscosity decreased with increasing Houttuynia cordata Thunb. powder content; however, breakdown and temperature to peak viscosity were not significantly different among the samples. Hardness and gumminess decreased with increasing Houttuynia cordata Thunb. powder content, and adhesiveness, cohesiveness, chewiness, and resilience also tended to decrease; however, springiness was not significantly different among the samples. In the consumer acceptance test, as the content of Houttuynia cordata Thunb. powder increased, the scores of all evaluated characteristics decreased; while the characteristic intensity ratings showed the reverse effect, and the 2 and 4% Houttuynia cordata Thunb. powder samples obtained fairly good scores. In conclusion, the results indicate that adding $2{\sim}4%$ Houttuynia cordata Thunb. powder to Sulgidduk is optimal, providing good physiological properties and reasonably high overall consumer acceptability.

어성초 분말을 첨가한 국수의 품질특성 (Quality characteristics of noodles added with Houttuynia cordata Thunb. powder)

  • 국수를 만들 때 다양한 기능성을 가진 어성초의 적절한 첨가 비율을 알아보기 위하여 어성초 분말을 첨가한 복합분의 점도 특성, 국수를 만든 다음 조리시험, 관능검사를 실시하였다. 어성초 분말을 첨가한 복합분은 대조구에 비하여 호화개시 온도, 최고점도, $95^{\circ}C$에서의 점도가 낮아졌다. 그러나 최고점도와 $95^{\circ}C$에서 15분 후의 점도와의 차이는 그다지 크지 않았다. 어성초 분말을 첨가한 조리면은 대조구에 비하여 중량과 부피가 감소하였으나 어성초 분말 0.5%, 1.5% 첨가구는 중량에 있어서 대조구와 유의적인 차이를 나타내지는 않았다. 어성초 분말을 0.5% 첨가한 국물의 흡광도는 대조구보다 낮았으나 다른 어성초 분말 첨가구는 대조구보다 높았으며, 유의적인 차이를 나타내었다. 어성초 분말의 첨가량이 많을수록 조리면의 L값은 감소하였으나 a값과 b값은 증가하는 것으로 나타났다. 관능검사에서는 어성초 분말을 첨가한 시험구의 색도가 대조구보다 낮았으며, 유의적인 차이를 보였으나 전체적인 기호도에 있어서는 대조구, 어성초 분말 0.5%, 1.5%가 유의적인 차이를 보이지는 않는 것으로 보아 어성초 분말을 첨가한 국수 제조시에 0.5%까지 첨가하는 것은 국수의 품질에 큰 영향을 주지 않을 것으로 생각된다.

  • Houttuynia cordata Thunb.[H.cordata]belonging to Saururaceae, is a wild medicinal herb of perennial plants, and grows well in a place with a lot of shade and moisture. The medical action of H.cordata is reported to have an antitumer effect, toxicity-suppressive effect, antifungal effect, diuretic effect, and antioxidative action, but its effect hasn't been reported on cardiovascular diseases, such as ateriosclerosis and hypertension yet. This study intended to confirm the effect of the water extract of H.cordata on the migration and proliferation of rat aortic smooth muscle cells. Such results show that the water extract of H.cordata suppresses the migration and proliferation of rat aortic smooth muscle cells. It is believed that a useful clue will be offered later to the prevention of cardiovascular diseases such as ateriosclerosis and hypertension, and the development of their medicines on the basis of the fact.

어성초 분말 및 즙의 급이가 고콜레스테롤혈증 흰쥐의 간장, 뇌 및 신장의 지질성분에 미치는 영향 (Effects of Houttuynia cordata Thunb Powder and Juice on Lipid Composition of Liver, Brain and Kidney in Dietary Hypercholesterolemic Rats)

  • For the investigation to the effects of H. cordata on prevention of hypercholesteromia, dietary hypercholesteromic rats were fed for 4 weeks with basial diet containing 5, 10, 15, 20% H. cordata powder, 5, 10 and 15% H. cordata juice. Total cholesterol and triglyceride contents of livers were significantly lower in experimental groups than in control group. Phospholipid contents in livers were not significant difference between control and experimental groups. Contents of free cholesterol in livers were ranged from 0.4$\pm$0.2 to 0.7$\pm$0.5mg/kg and cholesteryl ester contents were lower in H. cordata juice added groups than H. cordata powder added groups. Total cholesterol content in brains were not significant difference between cholesterol not added, H. cordata powder added at 15, 20% and its juice added group at 15%. The more powder and juice of H. cordata were added to basial diets, the lower contents of triglyceride and phospholipid were detected in brains. Contents of free cholesterol and cholesteryl ester in brains were not significance in all experimental groups. In kidneys, total cholesterol contents were not significant in control group and H. cordata powder of 5% was added group. Triglyceride, phospholipid and cholesteryl ester contents in kidney were not significant in all groups. Contents of free cholesterol in kidney were equally significant between cholesterol free and 15% H. cordata juice treated group.

어성초즙 및 추출물의 항돌연변이 효과 (Antimutagenic Effects of the Juice and Boiling Water Extract of houttuynia cordata Thunb)

  • The inhibitory effects of the juice and boiling water extract of Houttuyina cordata Thunb., which has been known as a traditional folk antitumor agent, on the genotxicity induced by mutagens were investigated . For this purpose, Salmonella typhimurium TA98 and TA100, and the transheterozygous (mwh/+) third larvae of Drosophila melanogaster were used. The juice of fresh H. cordata 9JHC) and boiling H. cordata extract (BHC) showed antimutagenicities toared alfatoxin B₁ in S. typhimurium TA 98 and TA100. The JHC's inhibitory effect increased by 76% to 98% as the adding concentrations increased, and the BHC had a 86-97% inhibitory effect. however, in the case of N-methyl-N'-nitro-N nitrosoguanidine (MNNG), the JHC's inhibitory effect was 52-60% irrespectively of its concentrations and the BHC had a low inhibitory effect. The JHC and BHC slightly reduced the somatic chromosomal mutagenicity by MNNG in the wing hair spot test system (mwh/+) of D. melanogaster, which suggests that they can inhibit gene mutation , deletion and mitotic chromosomal recomgination.

어성초(Houttuynia cordata Thunb) 유산균 발효물의 항알러지 활성 (Anti-allergic Activity of the Extracts from Houttuynia cordata Thunb Fermented by Lactic Acid Bacteria)

  • This study was performed to evaluate the possibility of application of lactic acid bacteria fermentation to increase the anti-allergic activity of the extracts from Houttuynia cordata Thunb. H. cordata Thunb was fermented on 25, 30, 35 and $40^{\circ}C$ for 5 days by two species of lactic acid bacteria, Leuconostoc mesenteroides 4395 and Lactobacillus sakei 383. The anti-allergic activity of water extracts of H. cordata Thunb was then analyzed both before and after fermentation. Anti-allergic activity was determined in vitro assays by using 5-lipoxygenase (5-LO), cyclooxygenase-2 (COX-2) and ${\beta}$-hexoseaminidase release of RBL-2H3 cells (degranulation marker). The extracts fermented at $35^{\circ}C$ by both bacteria had the highest inhibitory activities against 5-LO, and also higher than the control, and the one fermented at $30^{\circ}C$ by both bacteria had the highest inhibitory activity against COX-2. The degranulation of RBL-2H3 cells induced by IgE-antigen complex was estimated as ${\beta}$-hexoseaminidase release rate as reference of 100%, the release rates were inhibited in $25{\mu}g/ml$ of the extracts fermented at 30, 35 and $40^{\circ}C$ only by L. mesenteroides 4395. These results indicate that anti-allergic activity of H. cordata Thunb is increased by lactic acid bacteria fermentation.

어성초 메탄올 추출물에 의한 A549 인체 폐암세포 사멸유도에 관한 연구 (Induction of Apoptotic Cell Death by Methanol Extract of Houttuynia cordata Thunb. in A549 Human Lung Carcinoma Cells)

  • Houttuynia cordata Thunb, well known as 'E-Sung-Cho' in Korea, is traditional medicinal plant generally used in Oriental medicine therapy. We previously reported that the water extract of H. cordata inhibited cell proliferation and induced apoptosis in human breast carcinoma cells. In the present study, we investigated the biochemical mechanisms of anti-proliferative effects by the methanol extract of H. cordata (MEHC) in human lung carcinoma A549 cells. It was found that MEHC could inhibit the cell growth in a dose-dependent manner, which was associated with morphological change and apoptotic cell death as determined by formation of apoptotic bodies, DNA fragmentation and increased populations of apoptotic-sub G1 phase cells. Apoptosis of A549 cells by MEHC was also connected with a down-regulation of anti-apoptotic Bcl-2 and inhibitor of apoptosis proteins (IAPs) expression. MEHC treatment induced the proteolytic activation of caspase-3, caspase-8 and caspase-9, and a concomitant inhibition of poly(ADP-ribose) polymerase (PARP), ${\beta}$-catenin and phospholipase (PLC)-${\gamma}$1 protein expression. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of H. cordata.

Increased Flavonoid Compounds from Fermented Houttuynia cordata using Isolated Six of Bacillus from Traditionally Fermented Houttuynia cordata

  • Flavonoids, which form a major component in Houttuynia cordata Thunb., display a wide range of pharmacological activities. The expression of plant flavonoids is partly regulated by fermentation. Therefore, we studied the effects of fermentation on H. cordata in order to identify the strains present during the fermentation process, and to determine whether fermented H. cordata could be used as a probiotic. Our results showed that all 6 of the bacterial strains isolated from fermented H. cordata (FHC) belonged to the genus Bacillus. As expected, fermenting H cordata also increased the flavonoid content as increases were observed in the levels of rutin, quercitrin, and quercetin. To test the effects of fermentation, we treated LPS-stimulated RAW264.7 cells with non-fermented H. cordata extracts (HCE) or FHC extracts (FHCE). Compared to the HCE-treated cells, the FHCE-treated cells showed increased viability. No cytotoxic effects were detected in the FHCE-treated groups in the 2 cell lines used in the study, namely, RAW264.7 and RBL-2H3. FHCE-treated HepG2 cells showed decreased growth, compared to HCE-treated HepG2 cells. These results indicate that the fermented H. cordata predominantly contained Bacillus strains. Furthermore, FHCE are able to prevent LPS-induced inflammatory effects and inhibit the growth of HepG2 cells.
