Phylogenetic Relationships among Diverse Dinoflagellate Species Occurring in Coastal Waters off Korea Inferred from Large Subunit Ribosomal DNA Sequence Data

  • We analyzed the nuclear-encoded large subunit ribosomal RNA gene (LSU rDNA) sequences of 19 dinoflagellates occurring in costal waters off Korea and reconstructed a phylogenetic tree containing 74 representative species from 37 distinct genera. Of these, the LSU rDNA sequences of Amylax triacantha (Jörgensen) Sournia, Gonyaulax verior Sournia (= Amylax diacantha Meunier), Gyrodinium fissum (Levander) Kofoid et Swezy, Katodinium glaucum (Lebour) Lebour III, Noctiluca scintillans (Macartney) Kofoid et Swezy, Oxyphysis oxytoxoides Kofoid, and Pyrophacus steinii (Schiller) Wall et Dale are reported for the first time. Our LSU rDNA tree consistently placed Oxyrrhis marina Dujardin and N. scintillans at the most primitive positions, giving rise to a strongly supported monophyletic group of typical dinoflagellate species belonging to the Dinophyceae. The phylogenetic relationships among the typical dinoflagellates, however, were not resolved in the higher taxonomic levels in general. Only genera at terminal branches were usually supported with high confidence. The Dinophysiales, represented by Dinophysis species and O. oxytoxoides, formed a strongly supported monophyletic assemblage. The Gymnodiniales and Peridiniales were recovered as polyphyletic groupings. Members of the Gonyaulacales were consistently grouped together, but lacked statistical support. Within this order, the Ceratiaceae and Goniodomataceae each formed a monophyletic group, but the Gonyaulacaceae was polyphyletic. The phylogenetic relationships of the Gonyaulacaceae were generally congruent with differences in the combinations of the apical pore complex, hypothecal organization and thecal formula.

  • Twenty-three cultures of harmful algal bloom (HAB)-(causing dinoflagellates were isolated from the coastal waters of Korea. For each of the 14 morphospecies, the nuclearencoded small subunit (SSU) rDNA was analyzed to determine the phylogenetic relatedness of the species. Despite temporal and spatial isolation, 3-4 clonal cultures of Alexandrium catenella, Cochlodinium polykrikoides, and Gymnodinium catenatum had 100% identical SSU rDNA sequences. In contrast, heterogeneities in the SSU rDNA sequences were observed in Akashiwo sanguinea and Lingulodinium polyedrum strains. Extreme sequence polymorphism was shown within the SSU rRNA genes of an Al. tamarense clonal culture. A homology search in GenBank revealed that 11 dinoflagellate species were located in clusters corresponding to their morphological classification. The SSU rDNA sequences of C. polykrikoides, Gyrodinium instriatum, and Pheopolykrikos hartmannii, which were determined for the first time in this study, showed the following phylogenetic relationships: C. polykrikoides formed an independent branch separated from other dinoflagellates; Gyr. instriatum was placed in a monophyletic group with Gyr. dorsum and Gyr. uncatenum; and Ph. hartmanii, which forms a distinct two-celled pseudocolony, belonged to Gymnodinium sensu Hansen and Moestrup.

Effect of Thermal Stratification and Mixing on Phytoplankton Community Structure in the Western Channel of the Korea Strait

  • The profile of a fixed site at station M ($34.77^{\circ}N,\;129.13^{\circ}E$) in the Korea Strait was studied from March 2006 to February 2007. The aim was to understand the relationship between the annual thermal stratification pattern and seasonal variation in phytoplankton community structure. Physicochemical factors including temperature, salinity and nutrient concentrations, which strongly influence the proliferation and diversity of phytoplankton, were measured. The study period was divided into three due to the characteristic of thermohaline structures; mixed I (March-May 2006), stratified (June-November 2006) and mixed II(December 2006-Feburuary 2007). Diatoms dominated during the mixed I (89%) and II (48%) periods, while nanoplankton group occupied over 83% of total population during the stratified period. The dominant species during the mixed I and II was Chaetoceros socialis (47% and 29%, respectively), while during the stratified period Gyrodinium sp.(4%) was the most dominant. Averaged total chl a concentrations during the mixed I and II periods were 0.61 mg $m^{-3}$ and 0.72 mg $m^{-3}$, respectively, which were at least two-fold higher than that during the stratified period (0.30 mg $m^{-3}$). The vertical mixing and convection process of the water column induced nutrient supply from the bottom layer to the euphotic zone. It also led to the dominance of diatoms during the mixed periods, whereas small phytoplankton prevailed over large phytoplankton as stratification blocked the upward movement of nutrients to subsurface during the stratified period. During the mixed I and II periods, microplanktonic chl a dominated concentrations (50% and 48%, respectively), while picoplanktonic chl a occupied over 37% of total chl a during the stratified period.

Dynamics of Water Environmental Factors and Phytoplankton Before and After Inflow of Seawater in Shingwa Reservoir (시화호에서 해수유입 전.후의 수환경 요인과 식물플랑크톤 동태)

  • The dynamics of water quality and phytoplankton population had examined by monthly sampling from the upper to the lower part of watergate in an artificial Shihwa Reservoir in which situated near newly cities and incustrial complex on the west coast of Korea from January 1997 to December 1998. Among environmental factors, yearly average concentration of chl-a, TN and TP seemed to eutrophic or hypertrophic conditions that ranged 146.4~245.8 $\mu\textrm{g}$/$\ell$, 1.6~2.7 mg N/$\ell$, 258~448 $\mu\textrm{g}$ P/$\ell$, 26.9~80.7 $\mu\textrm{g}$/$\ell$, 1.0~2.4 mgN/$\ell$ and 74~239 $\mu\textrm{g}$P/$\ell$ respectively. Water quality was extremely deteriorated to consistently accumulation into inner reservoir by load of pollutants from autochthonous and allochthonous until early July 1997 after embankment. Water pollution of Shihwa Reservoir was remarkble on the biological condition with largely persistent bloom of phytoplankton and increase rate of standing crops was 2.4/yr. The development trend of phytoplankton in water ecosystem were closely related to increse and decrease of physico-chemical factors and those scale seemed to control by nutrient contents. As to see dominant species, composition of those composed to mostly freshwater algae before inflow of seawater such as Selenastrum capricornutum of green algae, cyclotella atomus, C. meneghiniana of diatom and Microcystis spp. of blue-green algae and the other hand brackish algae were dominated after inflow of seawater such as Chaetoceros dicipiens, Skeletonema costatum of diatom, Dinophysis acuminata, Gymnodinium mikimotoi, G. sanguineum, Gyrodinium spirale, Prorocentrum minmum of dinoflagellate and Eutreptiella gymnastica of euglenoid. Moreover, small flagellates including Chroomonas spp. of cryptomonad were abundant throughout the year. The cause of water deterioration during fill of the freshwater were complexly supported with extra and intra parameters. The variation pattern of phytoplankton were related to water temperature and salinity by inflow of seawater based to plentiful nutrients. The dynamics of phytoplankton were assessed to ecosystem that clearly condition of dominant by unique or a few angel species seasonally.

The Influences of Extracts from Ceratium spp, on the Growth of Harmful Microalgae (유해성 적조생물에 대한 Ceratium 추출물 영향 평가)

  • The growth response of the fish-killing dinoflagellate Cochlodinium polykrikoides was studied in cultures, using the treatment of Ceratium extracts by a methanol, a water-soluble, and a cell-free medium. The cell-free medium had the most increasing on the growth of C. polykrikoides cultures, enriched with $\geq$ 25% Ceratium, whereas the methanol and water-souble fractions did not affect the growth of C. polykrikoides exposed to even higher concentration. In particular, the cell-free medium also increased the growth of Gyrodinium impudicum and Chaetoceros sp., similar species to C. polykrikoides. In contrast to C. polykrikoides, G. impudicum and Chaetoceros sp., the growth of Alexandrium tamarense was inhibited significantly, and there was no great effect on the growth of Prorocentrum minimum. These results imply that Ceratium extracts may play an important role in the stimulatory effect of C. polykrikoides, and they have to affect the interaction between C. polykrikoides and Ceratium when co-existing.

Community Dynamics and Distribution of Dinoflagellates and Their Cysts in Masan-Chinhae Bay, Korea

  • The community dynamics and distribution of dinoflagellates and their cysts were monitored monthly from June 1996 to May 1997 at six stations in Masan-Chinhae Bay, one of the famous bays for red-tide occurrence in Korea. The dinoflagellate standing crops ranged from the minimum of 306 cells $ml^{-1}$ in February to the maximum of 37,959 cells $ml^{-1}$ in May. The species causing massive blooms were Prorocentrum micans Ehrenberg and Ceratium furca (Ehrenberg) Claparede & Lachmann in July, Gymnodinium sanguineum Hirasaka in October, Alexandrium tamarense (Lebour) Balech in April and Proroeentrum minimum (Pavillard) Schiller in May. Twenty-seven taxa of dinoflagellate cysts were isolated, including 20 identified species and 7 unidentified species. The mean cyst abundance ranged from the minimum of 556 cysts $cm^{-3}$ in June to the maximum of 5,727 cysts$cm^{-3}$ in February. The spatial pattern of cyst distribution showed the gradual increase in abundance from offshore to inshore stations. The cyst genera of Protoperidinium, Gymnodinium, Serippsiella, Gyrodinium and Alexandrium were abundant throughout the year. The vertical distribution of cysts showed the surface miximum at the 0-2cm layer and the sub-surface maximum at the 2-4cm layer. Total abundance of cysts showed the most significant relationships with water temperature, and some minor relationship with dissolved oxygen, salinity, pH, phosphate and total inorganic nitrogen of surface and bottom layer in the water column. The number of dinoflagellate species decreased, while the abundance of cysts increased 4.5 times as compared with the observation of 10 years prior to the present study at the same stations of Masan-Chinhae Bay.

Feeding by common heterotrophic protist predators on seven Prorocentrum species

  • Species belonging to the dinoflagellate genus Prorocentrum are known to cause red tides or harmful algal blooms. To understand the dynamics of a Prorocentrum sp., its growth and mortality due to predation need to be assessed. However, there are only a few Prorocentrum spp. for which heterotrophic protist predators have been reported. We explored feeding by the common heterotrophic dinoflagellates Gyrodinium dominans, Oxyrrhis marina, Pfiesteria piscicida, Oblea rotunda, and Polykrikos kofoidii and the naked ciliate Strombidinopsis sp. (approx. 90 ㎛ cell length) on the planktonic species Prorocentrum triestinum, P. cordatum, P. donghaiense, P. rhathymum, and P. micans as well as the benthic species P. lima and P. hoffmannianum. All heterotrophic protists tested were able to feed on the planktonic prey species. However, O. marina and O. rotunda did not feed on P. lima and P. hoffmannianum, while G. dominans, P. kofoidii, and Strombidinopsis sp. did. The growth and ingestion rates of G. dominans and P. kofoidii on one of the seven Prorocentrum spp. were significantly different from those on other prey species. G. dominans showed the top three highest growth rates when it fed on P. triestinum, P. cordatum, and P. donghaiense, however, P. kofoidii had negative growth rates when fed on these three prey species. In contrast, P. kofoidii had a positive growth rate only when fed on P. hoffmannianum. This differential feeding on Prorocentrum spp. between G. dominans and P. kofoidii may provide different ecological niches and reduce competition between these two common heterotrophic protist predators.

Outbreak of Red Tides in the Coastal Waters off the Southern Saemankeum areas, Jeonbuk, Korea 1. Temporal and Spatial Variations in the Phytoplankton Community in the Summer-fall of 1999 (전북 새만금 남쪽 해역의 유해성 적조 발생연구 1. 1999년도 여름-가을 식물플랑크톤의 시공간적 변화)

  • We investigated the outbreak of red tides dominated by harmful dinoflagellates from August to November 1999 in the coastal waters off the southern Saemankeum areas where a huge red tide dominated by Cochlodinium polykrikoides had been first observed in 1998. We took water samples from 2~5 depths of 4 stations (with 3-4 additional stations during red tides) in this study period and then measured the abundance of phytoplankton, water temperature, salinity, and the concentrations of nutrients. In the study period harmful dinoflagellates Alexandrium tamarense, C. polykrikoides, Gymnodnium catenatum, Gyrodinium aureolum, Gymnodnium impudicum were present, and of these G. aureolum and C. polykrikoides formed red tide patches on September 16 and October 18, respectively. The date of the outbreak of red tide dominated by C. polykrikoides in the study area was approximately 50 days later than that off the Kohung areas in 1997 and the surface water temperature when the red tides outbroke in the former area was 6$^{\circ}C$ lower than that fur the latter area. The maximum abundance of C. polykrikoides on September 16, October 7 and 18 were 5, 14, and 463 cells $m\ell$$^{-1}$ , respectively. The growth rate of C. polykrikoides, isolated from the study area, was 0.3~0.4 d$^{-1}$ at 20~$25^{\circ}C$, which enable this species to reach the maximum concentration without being transported from the adjacent waters containing already made red tide patches. The outbreaks of red tides dominated by C. polykrikoides in the study area and off Kohung have occurred when and/or where the concentrations of diatoms were low. This evidence suggests that the outbreak of red tides dominated by C. polykrikoides is adversely affected by the high diatom concentrations or the conditions favorable for the growth of diatoms.

  • Physical characteristics and nutrient distributions in seawater were investigated to understand the mechanism of red tide outbreak in coastal waters off Kohung area. To identify any physical and chemical differences before, during and after the red tide outbreaks, 6 times of field observations have been carried out from May to October, 1997. We found that major environmental properties of the seawater in the study area were determined not by the local meteoric conditions or nearby-land influences, but by the intrusion of seawater from offshore. In particular, extreme environmental variations in seawater were found during the period of red tide outbreak from August to September. Before the red tide outbreak, high concentrations of DIN(Dissolved Inorganic Nitrogen)were found in seawater, but they decreased during the outbreak. Whereas no significant variations of DIP(Dissolved Inorganic Phosphate) were found. For the water mass in the semienclosed northern part of the study area, local nutrient sources originated from nearby-lands were estimated to cover 70% of total DIN input, but the rest part of the water mass in the study area, sporadic intrusion of offshore water mass could account for the major source of DIN supply. An offshore water mass influenced by Yangtze river effluent was suggested as one of the candidates with its high nitrate contents, high seawater temperature and low salinity. A red tide related phytoplankton, Gyrodinium impudicum, was found in seawater on the 21th day of August and, on the same day, a unique water mass with high temperature and extremely low salinity suddenly appeared in the study area. On the 22th day of September, after one and half month duration of red tide we found that red tide had disappeared simultaneously with the intrusion of new water mass with different characteristics.

  • We investigated the phytoplankton community from June to September 1997 in the waters off Kohung, Korea where red tides dominated by harmful dinoflagellates had occurred from August to September or October since 1995. We took water samples five times from 5 depths at 6 or less stations in this study period. The most dominant harmful dinoflagellate during the red tide which had outbroken on August 24, 1997 was Gyrodinium impudicum, not Cochlodinium polykrikoides. On August 21 just before the harmful red tide occurred the abundance of G. impudicum at the inner bay station, 90cells $ml^{-1}$, was higher than that at the outer bay station. However, on August 27 just after the red tide had outbroken, the abundance of G. impudicum at the inner bay station did not increase, whereas that at the outer bay increased rapidly and reached to the maximum of 30,000 cells $ml^{-1}$. Instead, diatoms such as Skeleltonema costatum, Chaetoceros pseudocurvisetus, Pseudonitzschia pungens rapidly increased at the inner bay station where fresh water from lands has reached. The high abundance of diatoms might have inhibited the growth of red tide dinoflagellates at this station. The transport of already formed red tide patches from offshore areas, aggregation of scattered cells driven by physical forces, and/or competition between diatom and dinoflagellates might be responsible for this appearance of dense red tide patches at the outer bay station.
