Broad Spectrum Anti-bacterial Activity of the Leaves, Stem and Root Barks of Myristica subabulata

  • A number of Myristica species are used in herbal medicine for a variety of ailments. The methanol extracts of the leaves and the stem and root barks of Myristica subabulata were fractionated into petrol, dichloromethane and finally ethyl acetate. These were then screened against a total of 31 micro-organisms comprising of 13 G +ve, 12 G ve, one protozoa and 5 moulds. A broad-spectrum anti-bacterial activity was observed. As compared to fractionated the un-fractionated extracts exhibited much better level of activity.

  • The Bismarck Sea represent a unique region in the equatorial western Pacific where one can explore the relationship between tectonic and magmatic processes associated with back-arc opening. The sea, located north of Papua New Guinea and just south of the equator, formed during the final stages of a long, complex geological development of the Melanesian Borderland. The development resulted from the Cenozoic convergence between the Australian and Pacific- Caroline Plates and the opening of back-arc basins. At present, the Bismarck Sea straddles two oppositely facing trenches, the inactive Manus trench and the active New Britain trench, and covers two basins, the New Guinea Basin (NGB) to the west and the Manus Basin (MB) to the east. The two basins are separated by the shallow Willaumez-Manus Rise (WMR), which trends roughly from WNW to ESE. The origin of these major structural units and their relationship with the presentday zone of major seismicity along the Bismarck Sea Seismic Lineation (BSSL) remains unclear and is the main focus of our study.

  • Acute toxicity study was conducted on a Hepatitis B vaccine (Hepaccine-B-inj.) with mice, guinea pigs, and rabbits, in accordance with the norms suggested by the F.D.A. in U.S.A. Dose ranges were 2 doses/mouse, 5 doses/guinea pig, 10 doses/rabbit. They received the vaccine subcutaneously and intraperitoneally. Thereafter, all animals injected were observed of general signsdaily, and of body weight for two weeks. At the end of the observation period (or at the time of death), all animals received the highest dose group were autopsied and gross observation was made on various organs and tissues. No significant toxicity was noted.

  • PEG-hemoglobin SB1 (SB1), which is a hemoglobin-based oxygen carrier, is intended to use as a safe blood substitute against brain ischemia and stroke. The general pharmacological profiles of SB1 were studied. The doses given were 0, 5, 10, 20 ml/kg and drugs were administered intravenously. The animals used for this study were mouse, rat and guinea pig. SB1 showed no effects on general behavior, motor coordination, spontaneous locomotor activity, hexobarbital sleeping time, anticonvulsant activity, analgesic activity, blood pressure and heart rate, left ventricular peak systolic pressure, left ventricular end diastolic pressure, left ventricular developing pressure, double product, heart rate, coronary flow rate, smooth muscle contraction using guinea pig ileum, gastrointestinal transport, gastric secretion, urinary volume and electrolyte excretion at all doses tested except the decrease of body temperature. These findings demonstrated that SB1 possesses no general pharmacological effects at all doses tested.

Muscarinic Receptor Subtype Controlling the Carbachol-Induced Muscle Contraction in Guinea Pig Gastric Antrum

  • Stimulation of muscarinic receptors by carbachol (CCh) in the circular smooth muscle of the guinea pig gastric antrum causes muscle contraction. In the present study, muscarinic receptor subtype controlling the muscle contraction in response to CCh was studied using putative muscarinic receptor antagonists. Isometric force of the isolated circular muscle strips was measured in an organ bath. CCh contracted the muscle in a dose-dependent way, and each of the three muscarinic receptor antagonists, 4-diphenylacetoxy- N-methylpeperdine methiodide (4-DAMP), methoctramine and pirenzepine shifted the concentration- response curves to the right without significantly reducing the maximum force. The affinities of the muscarinic antagonists $(pA_2\;values)$ obtained from Schild plot analysis were 10.15, 7.05 and 6.84 for 4-DAMP, methoctramine and pirenzepine, respectively. These results suggest that the $M_3-subtype$ mainly mediate the muscle contraction in response to CCh in guinea pig gastric antrum.

Expression of major piroplasm protein(p33)of Theileria sergenti (Korean isolate) and its immunogenicity in guinea pigs

  • To investigate the development of a subunit vaccine against theileriosis in cattle, the DNA fragments encoding piroplasm surface protein (p33) of Theileria sergenti of a Korean isolate were expressed in baculoviruses. The expressed p33 was characterized by indirect fluorescent antibody (IFA) and western blotting analysis. The expression of p33 was mainly detected on the surface of infected Sf21 cells by IFA. The immunoblotting analysis revealed the presence of a same molecular weight protein band of p33. The antigenicity of expressed polypeptide was further examined through the inoculation of a guinea pig. The sera of guinea pigs immunized with p33 expressed cell Iysate showed similar fluorescent antibody patterns and reacted with the same molecular weight protein of T. sergenti in immunoblotting analysis, thus indicating that this protein can be a promising candidate for a subunit vaccine in the future.

  • Antigenicity of DA-3285, human recombinant erythropoietin which was produced from mammalian cells, was examined in guinea pigs and mice. In active systemic anaphylactic test, mild anaphylactic signs were observed in guinea pigs sensitized subcutaneously with DA-3285 or DA-3285 incorporated with complete Freund's adjuvant(CFA) when challenged with high dose(1000 IU/Kg) of DA-3285. Other groups showed negative responses. In mouse-rat passive cutaneous anaphylaxis test, 20% sera of mice immunized with DA-3285 or DA-3285 mixed with aluminum hydroxide(alum) showed mild positive responses. In the case of indirect haemagglutination reaction(IHA) test, when sheep red blood cells coated with DA-3285 was incubated with mouse serum, all the serum samples were showed negative responses. These results suggest that DA-3285 has a very weak antigenic potential and probably would not induce systemic allergic reactions in clinical uses.

  • This experiment was performed in order to investigate the effect of radioiodine upon the uterus and the ovary of guinea pigs. The animals were injected single dose of 4. 5 mCi of radioiodine per kg of body weight. They were sacrificed on various time intervals; 1, 7, 14, 28, 42 and 56 days after the injection. The results were as follows: 1. In the ovary, the follicle cells and the ova were degenerated with lapse of time after the injection. 2. In the uterus, hyperplasia of the lamina propria of the endometrium and atrophy of the myometrium were observed in accordance with time after the injection.

  • Waver 와 Bray (1930) 가 cochlear microphonic을 처음 발견한 이래 Derbyshire 와 Davis (1935)는 summating potential을 각각 처음 기술하여 이 세가지 전위를 electrocochleogram 이라 칭하였고 이는 감음신경성 난청의 감별진단 및 청각생리연구에 이용되어 왔다. 저자들은 정상 guinea pig 10 마리를 대상으로 DANAC 7E ERA청각계기를 사용하여 정원창에서 action potential과 summating potential을 측정하였으며 주파수에 따른 역치 자극간 간격 및 두 potential의 상호관계를 분석하여 다음과 같은 결과를 얻었다. 1) 주파수가 증가함에 따라 그 역치는 점차 감소하였다. 2) 자극음의 강도와 action potential의 $N_1$ component 진폭은 상호 비례관계를 보여 주었으며 주파수 증가에 따라 $N_1$ component 진폭은 점차 증가하였다. 3) Action potential 의 $N_1$ component latency는 주파수가 증가할수록 역비례 관계를 보여 주었다. 4) N$_1$ component의 진폭과 자극간 간격(interstimulus interval, ISI)과의 관계는 ISI가 80~160m sec사이에서 plateau를 형성하였다. 5) summating potential은 자극음의 강도가 증가함에 따라 그 진폭이 증가하였으며 action potential도 증가하였으나, SP/AP는 감소하는 경향을 보였다.

Studies of the Activities of Protein Methylases in Pancreatic Tissues (취조직내 Protein Methylases 활성도에 관한 연구)

  • Among the many protein modifications methylation is being investigated actively with regard to bacterial chemotaxis, gene regulation, muscle contraction, cytochrome c methylation, and the synthesis of the acyl transporter, carnitine. In this study the activities of protein methylase I, II, and III in pancreatic tissues of rat, mouse, and guinea pig were examined. Furthermore, the effect of cholinergic agents on the activity of protein methylases in pancreatic fragment of guinea pig was also examined in order to test the relationship between protein methylation and pancreatic secretion. The results are as follows. 1) The activities of protein methylases were generally high in pancreatic tissues of guinea pig and mouse but low in the tissue of rat. 2) The cholinergic stimulants, acetylcholine and carbachol at a concentration of $10^{-5}M$ decreased the activities of protein methylase I, II, and III compared with unstimulated control. 3) The inhibitory effect of the cholinergic stimulant on the activities of protein methylases was not blocked by atropine at a concentration of $10^{-5}M$.
