Optimized Medium Improves Expression and Secretion of Extremely Thermostable Bacterial Xylanase, XynB, in Kluyveromyces lactis

  • An extremely thermostable xylanase gene, xynB, from the hyperthermophilic bacterium Thermotoga maritima MSB8 was successful expressed in Kluyveromyces lactis. The response surface methodology (RSM) was also applied to optimize the medium components for the production of XynB secreted by the recombinant K. lactis. The secretion level (102 mg/l) and enzyme activity (49 U/ml) of XynB in the optimized medium (yeast extract, lactose, and urea; YLU) were much higher than those (56 mg/l, 16 U/ml) in the original medium (yeast extract, lactose, and peptone; YLP). The secretory efficiency of mature XynB was also improved when using the YLU medium. When the mRNA levels of 13 characterized secretion-related genes in the K. lactis cultured in YLP and YLU were detected using a semiquantitative RT-PCR method, the unfolded protein response (UPR)-related genes, including ero1, hac1, and kar2, were found to be up-regulated in the K. lactis cultured in YLU. Therefore, the nutrient ingredients, especially the nitrogen source, were shown to have a significant influence on the XynB secretory efficiency of the host K. lactis.

전침자극이 말의 위와 맹장의 운동성에 미치는 영향 (The effects of electroacupuncture on stomach and cecum motility in horses)

  • The effects of electroacupuncture(EA) on gastrointestinal motility were investigated in 6 horses. Three acupuncture points ; Guan Yuan Shu(BL-26), Wei Shu(BL-21) and Da Chang Shu(BL-25) were stimulated for 20 minutes by EA at separate occasions under varying condition ; 2V-1Hz, 2V-5Hz, 2V-30Hz, 4V-1Hz, 4V-5Hz and 4V-30Hz. Myoelectric activity of stomach and cecum was monitored to investigate the gastrointestinal motility. Electromyogram(EMG) recordings were carried out before, 0, 20 minutes after and 40 minutes after the EA stimulation. EMG bipolar electrode was surgically implanted in seromuscular layer of greater curvature in the stomach and between medial band and ventral band in the cecum. The EA stimulation and monitoring were not commenced until 15 days after electrode implantation. The EA stimulation of Wei Shu influenced on stomach motility and that of Da Chang Shu on, cecum motility. However, the EA stimulation of Guan Yuan Shu influenced on both the stomach and the cecum motility. The myoelectrical spike burst amplitude of the stomach and the cecum was significantly(p<0.05) increased by 2V-1Hz stimulation, but the myoelectrical spike burst frequence of the stomach and the cecum was significantly decreased by 2V-30Hz or 4V-30Hz stimulation. The myoelectrical spike burst duration of the stomach and the cecum was significantly lengthened by 4V-30Hz and 2V or 4V-30Hz stimulation, respectively.

  • As compared with acupuncture, The moxibustion occurred differently in the background of formation and also disagreed in the process of development. From the Jin(晉) dynasty to the Song(宋) dynasty, The school that attached importance to moxibustion had occupied superority by far and had schemed the development of moxibustion. But after the Song(宋) dynasty, The school that attached importance to acupuncture rose. Therefore, The school that attached importance to moxibustion, even if the power decreased, still had kept in existence and had achieved the development of moxibustion. Especially, Among the chinese physicians that recognized "Moxibustion can cure all disease, so don't discuss the weakness the firmness the chills the fever and cauterize the skin" till the Song(宋) dynasty, Ge Hong(葛洪) put in order the basic theory for moxibustion in (A handbook of prescriptions for emergencies). Wang Tao(王燾) only respected the moxibustion and said "Moxibustion has a strange effect, then all acupuncture herb-med(medical decoction) herb-ex are unattainable to it" in volume 14 (Medical secrets of an official). 중국내 재스민 차 패키지디자인 선호도 연구 - 중국 10대 재스민 차 브랜드를 중심으로 - (A Study on the Preference Degree of Jasmine Tea Package Design in China: Focused on China's Top 10 Jasmine Tea Brands)

  • 본 연구에서는 중국내 판매되고 있는 재스민 차의 10대 브랜드 "장이위엔"(張一元), "우위타이"(吳裕泰), "이푸탕"(藝福堂), "수밍룬"(蜀茗潤), "티엔푸밍 차"(天福茗茶), "춘룬"(春倫), "징화"(京華), "비탄피야 오쉐"(碧潭飄雪), "샤오관차"(小罐茶), "중차"(中茶)를 중심으로 패키지 디자인의 4가지 요소(로고, 칼라, 레이아웃, 형태)를 요약하였으며, 이를 바탕으로 소비자 선호도에 대한 설문지를 작성하여 신뢰도와 타당성을 분석하였다. 조사 결과에 따르면, 가장 소비자들의 인기를 얻은 로고는 "샤오관차"이고, 총 6가지 칼라에서 가장 소비자의 인기를 얻은 칼라는 회색 계열이다. 재스민 차 패키지의 레이아웃 디자인의 8가지 레이아웃에서 가장 소비자들의 인기를 얻은 레이아웃은 유형2(세로 포장, 2통)이고 가장 큰 인기를 얻은 패키지 형태는 형태2(장방형)이다. 끝으로 설문조사 결과에 따라 각 요소에 대한 소비자들의 선호도를 분석하였다.

Bacterial Microbiome Differences between the Roots of Diseased and Healthy Chinese Hickory (Carya cathayensis) Trees

  • Carya cathayensis is an important economic nut tree that is endemic to eastern China. As such, outbreaks of root rot disease in C. cathayensis result in reduced yields and serious economic losses. Moreover, while soil bacterial communities play a crucial role in plant health and are associated with plant disease outbreaks, their diversity and composition in C. cathayensis are not clearly understood. In this study, Proteobacteria, Acidobacteria, and Actinobacteria were found to be the most dominant bacterial communities (accounting for approximately 80.32% of the total) in the root tissue, rhizosphere soil, and bulk soil of healthy C. cathayensis specimens. Further analysis revealed the abundance of genera belonging to Proteobacteria, namely, Acidibacter, Bradyrhizobium, Paraburkholderia, Sphaerotilus, and Steroidobacter, was higher in the root tissues of healthy C. cathayensis specimens than in those of diseased and dead trees. In addition, the abundance of four genera belonging to Actinobacteria, namely, Actinoallomurus, Actinomadura, Actinocrinis, and Gaiella, was significantly higher in the root tissues of healthy C. cathayensis specimens than in those of diseased and dead trees. Altogether, these results suggest that disruption in the balance of these bacterial communities may be associated with the development of root rot in C. cathayensis, and further, our study provides theoretical guidance for the isolation and control of pathogens and diseases related to this important tree species.

  • 동양의학에서 "상한론(傷寒論)"의 발전은 대체로 삼단계(三段階)의 과정을 거쳤으나 "삼강정립(三綱鼎立)"설이 대세를 이루고 있었다. 이러한 관점에 대항하여 변증논치규율(辯證論治規律)을 연구한 학파(學派)가 나타나게 되었는데 우이(尤怡)가 그 중 한사람이다. 우이(尤怡)의 생애(生涯), 저서(著書), 학술사상(學術思想) 및 후세에 미친 영향 등을 조사하고 특히 "상한론(傷寒論)"을 안법류증(按法類?)하여 육경(六經)에 따른 정치법(正治法) 권변법(權變法) 알선법(斡旋法) 구역법(救逆法) 유병법(類病法) 명변법(明辨法) 잡치법(雜治法) 등의 치법(治法)에 대하여 연구하여 보고하는 바이다.

Dynamic Deformation Behavior of Aluminum Alloys Under High Strain Rate Compressive/Tensile Loading

  • Mechanical properties of the materials used for transportations and industrial machinery under high strain rate loading conditions such as seismic loading are required to provide appropriate safety assessment to these mechanical structures. The Split Hopkinson Pressure Bar (SHPB) technique with a special experimental apparatus can be used to obtain the material behavior under high strain rate loading conditions. In this paper, dynamic deformation behaviors of the aluminum alloys such as A12024-T4, A1606 IT-6 and A17075-T6 under both high strain rate compressive and tensile loading conditions are determined using the SHPB technique.

Robust Digital Image Watermarking Algorithm Using RBF Neural Networks in DWT domain

  • This paper proposes a new watermarking scheme in which a logo watermark is embedded into the discrete wavelet transform (DWT) domain of the original image using exact radial basis function neural networks (RBF). RBF will learn the characteristics of the image, and then watermark is embedded and extracted by the trained RBF. A watermark is added to the coefficients at the low frequency band of the DWT of an image and a watermark is embedded into the DWT domain using the trained RBF. The trained RBF also used in watermark extracting process. Experimental results show that the proposed method has good imperceptibility and high robustness to common image processing attacks.

Study of Propagation of Light along an Acupuncture Meridian

  • Propagation of light along a pericardium meridian is measured and compared with that along a non-meridian path one cm away from the meridian. Source-light is illuminated on the Nei-Guan(PC6) and propagated light is detected at three points along the meridian. Four reference points are one cm away from the corresponding meridian points, respectively. Light propagates better along the meridian than the reference path with more than twenty percent difference for all the subjects tested.

  • 많은 센서 네트워크 응용분야에서 센서 노드는 개방된 환경에 놓이게 된다. 공격자는 개방된 환경에 놓인 센서 노드를 물리적으로 획득할 수 있으며, 포획한 노드를 이용하여 허위보고서를 센서네트워크에 삽입 시킬 수 있다. 삽입된 허위 보고서는 제한된 센서노드 에너지를 고갈 시키며, 허위 경보를 일으켜 심각한 문제를 야기 시킬 수도 있다. 이러한 공격을 막기 위해 Yu와 Guan은 Dynamic En-route Filtering(DEF) 방법을 제안 하였다. DEF는 인증키 재분배를 통하여 필터링 효율을 강화 시키지만, 키 분배 효율을 높일수록 인증키를 분배에 사용되는 에너지 소비가 커지는 문제점을 가지고 있다. 본 논문에서는 인증키 분배에 소비되는 에너지를 줄이면서 효율적인 키 분배를 위해 퍼지 시스템 기반의 키 분배 거리 결정 방법을 제안하였다.
