We introduce to the notions of $GT_1$, $GT_2$, G-regular and G-normal on a GTS. And we investigate characterizations for such notions and relationships among $GT_1$, $GT_2$, $GT_3$ and $G_4$.

The specific genetic modification in porcine somatic cells by gene targeting has been very difficult because of low efficiency of homologous recombination. To improve gene targeting, we designed three kinds of knock-out vectors with ${\alpha}1,3$-galactosyltransferase gene (${\alpha}1,3$-GT gene), DT-A/pGT5'/neo/pGT3', DT-A/NLS/pGT5'/neo/pGT3' and pGT5'/neo/ pGT3'/NLS. The knock-out vectors consisted of a 4.8-kb fragment as the 5' recombination arm (pGT5') and a 1.9-kb fragment as the 3' recombination arm (pGT3'). We used the neomycin resistance gene (neo) as a positive selectable marker and the diphtheria toxin A (DT-A) gene as a negative selectable marker. These vectors have a neo gene insertion in exon 9 for inactivation of ${\alpha}1,3$-GT locus. DT-A/pGT5'/neo/pGT3' vector contain only positive-negative selection marker with conventional targeting vector. DT-A/NLS/pGT5'/neo/pGT3' vector contain positive-negative selection marker and NLS sequences in upstream of 5' recombination arm which enhances nuclear transport of foreign DNA into bovine somatic cells. pGT5'/neo/pGT3'/NLS vector contain only positive selection marker and NLS sequence in downstream of 3' recombination arm, not contain negative selectable marker. For transfection, linearzed vectors were introduced into porcine ear fibroblasts by electroporation. After 48 hours, the transfected cells were selected with $300{\mu}g/ml$ G418 during 12 day. The G418-resistant colonies were picked, of which 5 colonies were positive for ${\alpha}1,3$-GT gene disruption in 3' PCR and southern blot screening. Three knock-out somatic cells were obtained from DT-A/NLS/ pGT5'/neo/pGT3' knock-out vector. Thus, these data indicate that gene targeting vector using nuclear localization signal and negative selection marker improve targeting efficiency in porcine somatic cells.

This study was carried out to investigate the self-reported health and eating habit of the elite female adolescent athletes in Incheon. The subjects were 102 elite female athletes of middle and high schools located in Inchon. A self-administered Questionnaire was used for this cross-sectional survey. Statistical analysis by two-way ANOVA was performed to discriminate between the main effects of age (as middle and high school athletes) and sports type, and their interaction. As use of nutritional supplements, 35.8% of the total subjects took vitamin and iron supplements and 62.5% of field and track athletes took vitamin and iron supplements. While mental stress related to exercise was much in fencing > shooting > field and track > swimming athletes, physical burden during training was higher in swimming > field and track > fencing > shooting athletes. More than half of the subjects showed irregular menstrual cycle. As the most severe problem of eating habit, they reported skipping meal > overeating > prejudiced meal > preference to salty and spicy foods. More than half of the subjects skipped meal over three times per week. Fencing and shooting athletes showed very significantly higher frequency of skipping meal and more irregular breakfast time compared to those of other sport types. As for snack, they ate cookies, carbonated drink, chocolate, candy > milk, yogurt, ice cream > cake, bread, rice cake > fried noodle > hamburger, pizza. Therefore, nutritional counseling and education by sport type are necessary for health and good eating habit of these elite female adolescent athletes.
Free amino acids and polysaccharide content of submerged mycelial culture of Fomitopsis pinicola using $2\%$ citrus peel water extracts(CP), $2\%$ green tea water extracts(GT) and yeast-malt broth(YM) were investigated. Cultivations were conducted at $30^{\circ}C$ and 150 rpm for 10 days. Yields of the mycelium as fresh weight basis in GT-, CP- and YM-broth were $42.3\%,\;34.2\%\;and\;9.89\%$, and their turbidity(OD at 660 nm) of the broth without mycelium were 0.14, 0.16 and 0.22, respectively. Total free amino acid content in the mycelium were 928.19 $mg\%$ in YM, 1060.53 $mg\%$ in CP, 764.83 $mg\%$ in GT, and the major free amino acid was lysine in YM, glutamic acid in CP and GT. Total free exo-amino acid contents were 659.75 $mg\%$ in YM, 954.55 $mg\%$ in CP, 838.69 $mg\%$ in GT, and the major in the all broths was glutamic acid Total amino acid derivatives content of the mycelium were in order of CP>GT>YM, and the major was cystathionine in YM, hydroxy proline in CP and GT. The major among exo-amino derivatives was hydroxy proline in the all broth. Exo-AIS content was in order of CP>GT>YM. Acid soluble polysaccharide content of the mycelium was GT($0.69\%$)>YM($0.39\%$)>CP($0.18\%$). The exo-polysaccharide content was in order of GT($0.87\%$)>CP($0.69\%$)>YM($0.09\%$). Alkali soluble polysaccharide content of the mycelium was in order of CP($5.21\%$)>GT($5.18\%$)>YM($4.56\%$), and exo-polysaccharide was in order of GT($6.79\%$)>YM($3.57\%$)>CP($3.01\%$). The alkali soluble polysaccharide eluted from mycelium cultivated in CP broth was supposed to polysaccharide(about 500,000 daltons) composed of hexose and uronic acid bounded with protein(below 10,000 daltons).

This study evaluates the utility of an antibacterial microneedle composed of green tea extract (GT) and hyaluronic acid (HA), for the efficient delivery of GT. These microneedles have the potential to be a patient-friendly method for the conventional sustained release of drugs. In this study, a fabrication method using a mold-based technique to produce GT/HA microneedles with a maximum area of ${\sim}60mm^2$ with antibacterial properties was used to manufacture transdermal drug delivery systems. Fourier transform infrared (FTIR) spectrometry was carried out to observe the potential modifications in the microneedles, when incorporated with GT. The degradation rate of GT in GT/HA microneedles was controlled simply by adjusting the HA composition. The effects of different ratios of GT in the HA microneedles were determined by measuring the release properties. In HA microneedles loaded with 70% GT (GT70), a continuous higher release rate were sustained for 72 h. The in vitro cytotoxicity assays demonstrated that GT/HA microneedles are not generally cytotoxic to chinese hamster ovary cells (CHO-K1), human embryonic kidney cells (293T), and mouse muscle cells (C2C12), which were treated for 12 and 24 h. Antimicrobial activity of the GT/HA microneedles was demonstrated by ~95% growth reduction of gram negative [Escherichia coli (E. coli), Pseudomonas putida (P. putida) and Salmonella typhimurium (S. typhimurium)] and gram positive bacteria [Staphylococcus aureus (S. Aureus) and Bacillus subtilis (B. subtilis)], with GT70. Furthermore, GT/HA microneedles reduced bacterial growth in the infected skin wound sites and improved skin wound healing process in rat model.
Purpose: The purpose of this study was to simplify the traditional manufacturing method of Jeungpyun by developing a new process for Jeungpyun preparation using meringue and to also investigate the quality characteristics of Jeungpyun made with green tea powder. Methods: The Jeungpyun was manufactured as 4 sample preparations (Jeungpyun: J; Jeungpyun+0.5% green tea: J+0.5% GT; Jeungpyun+1% green tea: J+1% GT; and Jeungpyun+1.5% green tea: J+1.5% GT) through mixing rice flour, meringue, green tea powder (0-1.5%), dry yeast, water, and milk, steaming for 4 min at $180^{\circ}C$ and 3 min at $220^{\circ}C$, and then standing for 1 hr at room temperature. The quality characteristics of Jeungpyun were evaluated based on physicochemical properties, total mold, texture analysis, and sensory evaluation. Results: The pH of Jeungpyun batter was significantly lower in J+1.0% GT and J+1.5% GT than J. The specific gravity, weight, volume, and specific volume of J+0.5% GT and J+1% GT was not significantly different, as compared to J. In color, L value decreased and a and b value increased based on the increase of green tea powder. The antifungal activity was the highest in J+1.5% GT, followed by J+1.0% GT, J+0.5% GT, and J, in order. Samples with added green tea powder showed decreased hardness, as compared with J; its effect was significant in J+1.0% GT and J+1.5% GT after storage for 1 day. In sensory evaluation, the color of Jeungpyun was darkened by the addition of green tea powder. The egg smell and hardness significantly decreased by addition of green tea powder. Based on quantitative description analyses, overall acceptability was the highest in J+1% GT. Conclusion: Therefore, 1% green tea powder was the optimal amount for preparing Jeungpyun with meringue. For simplification of the traditional manufacturing method, Jeungpyun could be produced with meringue and green tea powder, which has potent physiological activities.
The study was conducted to investigate the effect of added green tea and black tea powder on the quality of bread during storage. Tea bread was prepared with 0.1% and 2% concentration of green tea (GT) and black tea(BT) powder. The crude fat contents of GT 2% group and BT 2% group were significantly higher than those of control group. GT l% group and BT l% group. The loaf weight of the control group was the lowest and its loaf volume was the highest. Especially, GT 2% group had the lowest loaf volume. The internal lightness value of control group was the highest. the redness value of BT 2% group and the yellowness value of GT 2% group were the highest. GT 2% group was significantly higher in hardness and gumminess and BT l% group was higher in cohesiveness and springiness. After 5 day storage, the retrogradation of GT 2% group was the highest and that of BT 2% group was the lowest among groups. After 5 day storage. pH. titratable acidity and total colony count of GT 2% group was the lowest and those of control group was the highest. The sensory score of control, GT l% and GT 2% group were evaluated higher than others. The shelf-life of tea breads were prolonged to 1~3 days by adding green tea powder and black tea powder as compared with the control group. but It must be considered the way to increase the quality of bread in loaf volume and retrogradation.
The purpose of this thesis is to develop a micro-computer application for GT/MRP (group technology/material requirement planning) integrated system for efficient management of production scheduling. GT and MRP system have been found to have several drawbacks in practice. By GT and MRP system, however, it should be able to construct a GT/MRP integrated system that possesses the advantages of both concepts while alleviating their individual limitations.
To develope a domestic machinery industry, international competition through reducing cost by increasing productivity is as important as evolving technology and increasing quality. In this study, we have studied in application of GT technology. GT is a skill that acquires high productivity using part's resemblance for multi part and small size production like mass production. We have classified group, such as design, layout and the others, to be applied systematically. The design group includes retrieval for drawings and study for GT design. The layout group includes GT layout, analysis of production process and study for composition method. And others include scheduling, standardization and standard process.

As an approach to high-efficiency of SOFC system, SOFC/GT Hybrid system is effective. However, if the output size of the system belongs to the marine class of dozens MWs, the introduction of the cooling system of GT system, which is used as sub-system, makes its related devices complicated and also makes its control difficult. Accordingly, for the marine use, SOFC/GT (non-cooling)Hybrid system looks more suitable than SOFC/GT(cooling)Hybrid system. This study established the SOFC/GT (non-cooling)Hybrid system, and examined the operating temperature & current density of the stack for the system, pressure ratio of the gas turbine, the influence of TIT(Turbine Inlet Temperature) on system performance, etc. through the simulation process. Through this research process, this study was able to confirm that electrical efficiency rises in spite of the increase in the required power for the air compressor, and there exists a limited range of temperatures for operation in TIT.
