Rhizobacteria are used as inoculants to enhance crop yield and for biological control of fungal pathogens. Fluorescent pseudomonads isolated from the rhizosphere of groundnut showed suppression of the phytopathogen Macrophomina phaseolina that causes charcoal rot of groundnut, an economically important agroproduct. Two strains of fluorescent pseudomonads, designated as PS1 and PS2, were selected as a result of in vitro antifungal activity. After 5 days of incubation at $28{\pm}1^{\circ}C$, both PS1 and PS2 caused clear inhibition zones in dual cultures, restricting the growth of M. phaseolina by 71 % and 74%, respectively. Both the strains were capable of producing siderophores, indole acetic acid, and hydrocyanic acid, and causing phosphate solubilization under normal growth conditions. These strains, when used as inoculants in groundnut, enhanced germination up to 15% and 30% with subsequent increase in grain yield by 66% and 77%, respectively. Conversely, when the pathogen alone was tested 57% decrease in yield was recorded. Thus the studies revealed the potential of the two pseudomonads not only as biocontrol agents against M. phaseolina, but also as a good growth promoter for groundnut.
Allelopathic activity of rotation crop residues to common weeds and selectivity of crops were studied for developing an alternative weed control strategy in upland organic farming. All rotation crop residues with various ratios suppressed weed growth, but the growth of crop species stimulated a little when those were grown with the mixture of rotation crop residues except a few crop species. Among the rotation crop residues used in this study hairy vetch was the most effective, followed by Chinese milkvetch, barley and rye in suppression of weed growth. The 90:10 (crop:soil, v/v) treatments incorporation rate of crop residues such as hairy vetch and Chinese milkvetch suppressed completely the growth of all weed species tested in this study. The effect of crop residues on suppression of weed growth reduced to 90% when the incorporation rates decreased to 60% [60:40 (crop:soil, v/v). It was noticed that broadleaf weed species were more susceptible to rotation crop residues than grass weed species. In the influence of crop residues on the crop growth, no growth inhibition was found in red pepper, lettuce and perllia at any incorporation rates, but tomato, cucumber and corn were slightly inhibited in a few cases at their highest incorporation levels. This study demonstrated that rotation crop residues especially hairy vetch and Chinese milkvetch have high herbicidal effects against weeds without inhibiting the growth of crop species.
The proliferation of bone marrow stem cell compartment is thought to be under both positive and negative controls by cytokines and colony stimulation factors. Macrophage inflammatory $protein-1{\alpha}(MIP-1{\alpha})$ has been assessed for its potential to protect hematopoietic stem cells from cytotoxic effects of a cycle-specific antineoplastic agents. We have tested the ability of $MIP-1{\alpha}$ to suppress the proliferation of stem cell line Du.528.101 in variety of active status by using $[^{3}H]-thymidine$ incorporation test. The results were as follows. 1. The effect of $MIP-1{\alpha}$ on steady-state Du.528.101 cell represented the cell growth suppression at the concentration of 10, 50, 100nM of $MIP-1{\alpha}$(P<0.001). 2. $MIP-1{\alpha}$ stimulated the proliferation of Du.528.101 cells previously treated with IL-1 at the concentration of 5, 50nM of $MIP-1{\alpha}$(P<0.01). 3. The suppression effect of MIP-1 on Du.528.101 cells at the concentration of 5, 50nM was shown when cells were treated with $MIP-1{\alpha}$ before activation with $IL-1{\beta}(P<0.01)$. 4. The growth rate of synchronized cells were slower than that of non-synchronized ones, and $MIP-1{\alpha}$ represented the similar suppression effect on both synchronized and non-synchronized cells.
Excessive extracellular matrix (ECM) accumulation is the main feature of chronic renal disease including diabetic nephropathy. Plasminogen activator inhibitor (PAI)-1 is known to play an important role in renal ECM accumulation in part through suppression of plasmin generation and matrix metalloproteinase (MMP) activation. The present study examined the effect of PAI-1 antisense oligodeoxynucleotide (ODN) on fibronectin upregulation and plasmin/MMP suppression in primary mesangial cells cultured under high glucose (HG) or transforming growth factor (TGF)-${\beta}1$, major mediators of diabetic renal ECM accumulation. Growth arrested and synchronized rat primary mesangial cells were transfected with $1\;{\mu}M$ phosphorothioate-modified antisense or control mis-match ODN for 24 hours with cationic liposome and then stimulated with 30 mM D-glucose or 2 ng/ml TGF-${\beta}1$. PAl-1 or fibronectin protein was measured by Western blot analysis. Plasmin activity was determined using a synthetic fluorometric plasmin substrate and MMP-2 activity analyzed using zymography. HG and TGF-${\beta}1$ significantly increased PAI-1 and fibronectin protein expression as well as decreased plasmin and MMP-2 activity. Transient transfection of mesangial cells with PAI-1 antisense ODN, but not mis-match ODN, effectively reversed basal as well as HG- and TGF-${\beta}1$-induced suppression of plasmin and MMP-2 activity. Both basal and upregulated fibronectin secretion were also inhibited by PAI-1 antisense ODN. These data confirm that PAI-1 plays an important role in ECM accumulation in diabetic mesangium through suppression of protease activity and suggest that PAI-1 antisense ODN would be an effective therapeutic strategy for prevention of renal fibrosis including diabetic nephropathy.
Biological control agents (BCAs) from different microbial taxa are increasingly used to control bacterial wilt caused by Ralstonia solanacearum. However, a quantitative research synthesis has not been conducted on the role of BCAs in disease suppression. Therefore, the present study aimed to meta-analyze the impacts of BCAs on both Ralstonia wilt disease suppression and plant (host) growth promotion. The analysis showed that the extent of disease suppression by BCAs varied widely among studies, with effect size (log response ratio) ranging from -2.84 to 2.13. The disease incidence and severity were significantly decreased on average by 53.7% and 49.3%, respectively. BCAs inoculation also significantly increased fresh and dry weight by 34.4% and 36.1%, respectively on average. Also, BCAs inoculation significantly increased plant yield by 66%. Mean effect sizes for genus Pseudomonas sp. as BCAs were higher than for genus Bacillus spp. Among antagonists tested, P. fluorescens, P. putida, B. cereus, B. subtilis and B. amyloliquefaciens were found to be more effective in general for disease reduction. Across studies, highest disease control was found for P. fluorescens, annual plants, co-inoculation with more than one BCA, soil drench and greenhouse condition were found to be essential in understanding plant responses to R. solanacearum. Our results suggest that more efforts should be devoted to harnessing the potential beneficial effects of these antagonists, not just for plant growth promoting traits but also in mode of applications, BCAs formulations and their field studies should be considered in the future for R. solanacearum wilt disease suppression.
As the trench width in the interconnect technology decreases down to nano-scale below 50 nm, superconformal gap-filling process of Cu becomes very critical for Cu interconnect. Obtaining superconfomral gap-filling of Cu in the nano-scale trench or via hole using MOCVD is essential to control nucleation and growth of Cu. Therefore, nucleation of Cu must be suppressed near the entrance surface of the trench while Cu layer nucleates and grows at the bottom of the trench. In this study, suppression of Cu nucleation was achieved by treating the Ru barrier metal surface with capacitively coupled hydrogen plasma. Effect of hydrogen plasma pretreatment on Cu nucleation was investigated during MOCVD on atomic-layer deposited (ALD)-Ru barrier surface. It was found that the nucleation and growth of Cu was affected by hydrogen plasma treatment condition. In particular, as the plasma pretreatment time and electrode power increased, Cu nucleation was inhibited. Experimental data suggests that hydrogen atoms from the plasma was implanted onto the Ru surface, which resulted in suppression of Cu nucleation owing to prevention of adsorption of Cu precursor molecules. Due to the hydrogen plasma treatment of the trench on Ru barrier surface, the suppression of Cu nucleation near the entrance of the trenches was achieved and then led to the superconformal gap filling of the nano-scale trenches. In the case for without hydrogen plasma treatments, however, over-grown Cu covered the whole entrance of nano-scale trenches. Detailed mechanism of nucleation suppression and resulting in nano-scale superconformal gap-filling of Cu will be discussed in detail.
To study the influence of the rotifer Brachionus calyciflorus on harmful algal bloom suppression, we focused on assessing the rotifer's abilities using several prey species : Microcystis aeruginosa, Synechocystis sp., Chlorella vulgaris and Coelastrum sp. of the warm-weather species and the cold-weather centric diatom Stephanodiscus hantzchii. Grazing effects and growth rates of rotifers B. calyciflorus were 94.5% and $1.29d^{-1}$, respectively, for Synechocystis sp., 87.4% and $0.60d^{-1}$, respectively, for M. aeruginosa, 95.2% and $0.65d^{-1}$, respectively, for C. $vulgaris^{TM}$, 78.6% and $0.45d^{-1}$, respectively, for C. vulgaris UTEX., 86.5% and $0.99d^{-1}$, respectively, for Coelastrum sp., and 82.6% and $0.40d^{-1}$, respectively, for S. hantzchii. Of these, although the growth of Synechocystis and Coelastrum was effectively suppressed by rotifer grazing, efficient suppression effects on Stephanodiscus blooms were unexpected. The present study revealed that reproduction of B. calyciflorus was greatly influenced by its food types in the initial stages and the efficiencies of bio-agents as sole food sources vary depending on the target algae and the agent.
Among the root colonizing and plant growth promoting bacteria isolated from the bacterial wilt suppressive soil, five strains were detected to produce siderophores by CAS agar assay. The most effective isolate, TS3-7 strain induced significant suppression of bacterial wilt disease in tomato and pepper plants. Seed treatment followed by soil drench application with this strain resulted in over 80% reduction of bacterial wilt disease compared with the control. Significant disease suppression by TS3-7 strain was related to the production of siderophore. Besides iron competition, induction of resistance of the host plant with siderophore was suggested to be another mode of action that suppress bacterial wilt, based on the lack of direct antibiosis against pathogen in vitro. According to Bergey's Manual of Systemic Bacteriology and 16S rDNA sequence data, TS3-7 stain was identified as Pseudomonas sp. TS3-7.
