Secretion and Expression of Matrix Metalloproteinase-2 and 9 from Bone Marrow Mononuclear Cells in Myelodysplastic Syndrome and Acute Myeloid Leukemia

  • Background: Matrix metalloproteinase -2 (gelatinase-A, Mr 72,000 type IV collagenase, MMP-2) and -9 (gelatinase-B, Mr 92,000 type IV collagenase, MMP-9) are key molecules that play roles in tumor growth, invasion, tissue remodeling, metastasis and stem-cell regulation by digesting extracellular matrix barriers. MMP-2 and -9 are well known to impact on solid cancer susceptibility, whereas, in hematological malignancies, a paucity of data is available to resolve the function of these regulatory molecules in bone marrow mononuclear cells (BM-MNCs) and stromal cells of myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML). Objectives: The present study aimed to investigate mRNA expression and gelatinase A and B secretion from BM-MNCs in vitro and genotypic associations of MMP-2 (-1306 C/T; rs243865), MMP-9 (-1562 C/T; rs3918242), tissue inhibitor of metalloproteinase -1 (TIMP-1) (372T/C; rs4898, Exon 5) and TIMP-2 (-418G/C; rs8179090) in MDS and AML. Results: The study covered cases of confirmed MDS (n=50), AML (n=32) and healthy controls (n=110). MMP-9 mRNA expression revealed 2 fold increased expression in MDS-RAEB II and 2.5 fold in AML M-4 (60-70% blasts). Secretion of gelatinase-B also revealed the MMP-9 mRNA expression and ELISA data also supported these data. We noted that those patients having more blast crises presented with more secretion of MMP-9 and its mRNA expression. In contrast MMP-9 (-1562 C/T) showed significant polymorphic associations in MDS (p<0.02) and AML (p<0.02). MMP-9 mRNA expression of C/T and T/T genotypes were 1.5 and 2.5 fold increased in MDS and AML respectively. In AML, MMP-2 C/T and T/T genotypes showed 2.0 fold mRNA expression. Only MMP-9 (-1306 C/T) showed significant 4 fold (p<0.001) increased risk with chemical and x-ray exposed MDS, while tobacco and cigarette smokers have 3 fold (p<0.04) risk in AML. Conclusions: In view of our results, MMP-9 revealed synergistic secretion and expression in blast crises of MDS and AML with 'gene' polymorphic effects and is significantly associated with increased risk with tobacco, cigarette and environmental exposure. Release and secretion of these enzymes may influence hematopoietic cell behavior and may be important in the clinical point of view. It may offer valuable tools for diagnosis and prognosis, as well as possible targets for the treatments.

헌구두솔버섯균의 균학적 특성 및 목질계 섬유소의 분해 특성 분석 (Analysis of Mycological Characteristics and Lignocellulose Degradation of Gyrodontium sacchari)

  • 참나무와 소나무 목재에서 분리한 Gyrodontium sacchari 균주에 대한 균학적 특성과 목질계 섬유소 분해력을 검정하였다. 균주는 참나무와 소나무 목재에서 분리하였으며 배지는 potato dextrose agar (PDA)에서 가장 생장이 좋았고 생장온도는 참나무에서 분리한 NAAS02335 균주는 $25^{\circ}C$에서, 소나무 목재에서 분리한 NAAS05299 균주는 $30^{\circ}C$에서 가장 우수한 생장을 보였다. 섬유소 분해 효소인 cellulase와 xylanase, amylase의 활성은 G. sacchari NAAS05299 균주가 6.7~12.8배 더 높았으며 리그닌 분해 효소는 G. sacchari NAAS02335 균주가 3.7~138.5배 활성이 더 높았으며 목질계 섬유소를 탄소원으로 첨가하였을 때 효소의 활성은 월등히 증가하였다. 목질계 바이오매스 분해력을 검정한 결과 G. sacchari NAAS05299 균주는 filter paper를 4주만에 완전히 분해하였고 볏짚과 미송, 참나무를 분해하였으나 G. sacchari NAAS02335 균주는 볏짚에서만 분해력을 나타내어 G. sacchari NAAS05299 균주가 더 우수한 바이오매스 분해 효과를 나타내었다.

파뿌리 열수추출물을 이용한 요구르트의 발효 및 품질특성 (Quality and fermentative characteristics of yogurt added with hot water extract of Welsh onion root)

  • 대파 뿌리 열수추출물을 아용하여 유산균의 성장특성과 요구르트의 발효 특성을 조사 하였다. 파 뿌리 열수 추출물은 pH 5.63, 고형분 함량 10%, 환원당함량 1.12 mg/g, 총 폴리페놀 함량 135.09 mg/g, DPPH 라디칼 소거능 45.24%이었다. 추출액을 이용하여 제조한 BHI배지(WR50, WR100)에서 유산균의 성장은 배양 24시간 이후 대조구에 비해 WR100에서 약 1 log cycle 성장이 촉진되었다. 파 뿌리 열수 추출물 원액(WY100), 증류수로 2배 희석한 추출액(WY50) 그리고 증류수(control)에 각각 12% 환원탈지유를 용해시켜 제조한 요구르트의 발효 중 pH는 추출물의 농도가 증가함에 따라 낮아졌으며, 산도 변화 역시 pH 변화와 유사하였다. 점도는 대조구를 제외하고 발효 8시간 이후로부터 급격히 증가하여 발효기간과 추출물의 농도가 증가함에 따라 유의적으로 증가하였다. 발효 기간 중 유산균수의 변화는 첨가구가 대조구에 비해 높은 생균수를 나타내었으며 발효 24시간 발효 후 각 처리구별 생균수는 8.03(control), 8.77(WY50), 8.84(WY100) log CFU/mL 이었다. DPPH 라디칼 소거능은 추출물의 농도가 증가할수록 증가하였다. 종합적 기호도는 추출물 첨가구가 대조구에 비해 유의적으로 높았으며(p<0.05), 각각 3.00(control), 3.50(WY50), 3.17와(WY100)으로 50% 파 뿌리 열수 추출물로 제조한 요구르트의 기호성이 가장 양호 하였다. 추출물로 제조한 요구르트를 $4^{\circ}C$에서 10일간 냉장 저장 동안 대조구와 첨가구 모두 pH는 낮아지고 유산균수는 감소하였으나 첨가구의 경우 $10^8CFU/mL$을 유지하였다. 파 뿌리 열수추출물은 요구르트의 발효 및 기능성 개선을 위해 부재료로 사용이 가능할 것으로 판단된다.

Comparison of Radiological Tumor Response Based on iRECIST and RECIST 1.1 in Metastatic Clear-Cell Renal Cell Carcinoma Patients Treated with Programmed Cell Death-1 Inhibitor Therapy

  • Objective: To evaluate the radiological tumor response patterns and compare the response assessments based on immune-based therapeutics Response Evaluation Criteria in Solid Tumors (iRECIST) and RECIST 1.1 in metastatic clear-cell renal cell carcinoma (mccRCC) patients treated with programmed cell death-1 (PD-1) inhibitors. Materials and Methods: All mccRCC patients treated with PD-1 inhibitors at Henan Cancer Hospital, China, between January 2018 and April 2019, were retrospectively studied. A total of 30 mccRCC patients (20 males and 10 females; mean age, 55.6 years; age range, 37-79 years) were analyzed. The target lesions were quantified on consecutive CT scans during therapy using iRECIST and RECIST 1.1. The tumor growth rate was calculated before and after therapy initiation. The response patterns were analyzed, and the differences in tumor response assessments of the two criteria were compared. The intra- and inter-observer variabilities of iRECIST and RECIST 1.1 were also analyzed. Results: The objective response rate throughout therapy was 50% (95% confidence interval [CI]: 32.1-67.9) based on iRECIST and 30% (95% CI: 13.6-46.4) based on RECIST 1.1. The time-to-progression (TTP) based on iRECIST was longer than that based on RECIST 1.1 (median TTP: not reached vs. 170 days, p = 0.04). iRECIST and RECIST 1.1 were discordant in 8 cases, which were evaluated as immune-unconfirmed PD based on iRECIST and PD based on RECIST 1.1. Six patients (20%, 6/30) had pseudoprogression based on iRECIST, of which four demonstrated early pseudoprogression and two had delayed pseudoprogression. Significant differences in the tumor response assessments based on the two criteria were observed (p < 0.001). No patients demonstrated hyperprogression during the study period. Conclusion: Our study confirmed that the iRECIST criteria are more capable of capturing immune-related atypical responses during immunotherapy, whereas conventional RECIST 1.1 may underestimate the benefit of PD-1 inhibitors. Pseudoprogression is not rare in mccRCC patients during PD-1 inhibitor therapy, and it may last for more than the recommended maximum of 8 weeks, indicating a limitation of the current strategy for immune response monitoring.

Preparation and Characterization of Anti-GP73 Monoclonal Antibodies and Development of Double-antibody Sandwich ELISA

  • Background: Serum Golgi protein 73 (GP73) as a novel and potential marker for diagnosing hepatocellular carcinoma (HCC) have been found to be elevated in HCC patients and associated with clinical variables representing tumor growth and invasiveness. The aim of this study was to prepare a pair of monoclonal antibodys (mAbs) against GP73 and develop a newly designed double-antibody sandwich enzyme-linked immunosorbent assay (s-ELISA), which would be used in the detection of serum GP73 (sGP73) as well as in the diagnosis of HCC. Materials and Methods: Produced by prokaryotic expression, the purified recombinant GP73 (rGP73), produced by prokaryotic expression, was used to immunize the Balb/c mice. Two hybridoma cell lines against GP73 were obtained by fusing mouse Sp2/0 myeloma cells with spleen cells from the immunized mice. The titers of anti-GP73 mAb reached 1:243,000. Western blotting analysis and Immunohistochemistry staining revealed that anti-GP73 mAb could recognize GP73 protein. The double-antibody s-ELISA was successfully established and validated by 119 HCC and 103 normal serum samples. Results: showed that the detection limit of this method could reach 1.56 ng/ml, and sGP73 levels in HCC group (mean=190.6 ng/ml) were much higher than those of in healthy controls (mean=70.92 ng/ml). Conclusions: Results of our study not only showed that sGP73 levels of HCC patients were significantly higher than those of healthy controls, but also indicated that the laboratory homemade anti-GP73 mAbs could be the optimal tool used in evaluating sGP73 levels, which would provide a solid foundation for subsequent clinical applications.

Effect of Feeding Strategies on Milk Production of Holstein Dairy Cows Managed by Small-Farmers Alpine Grassland in Korea

  • The main objective of this experiment was to investigate effects of two different feeding systems on body weight, milk yield, milk composition, and mineral and fatty acids content of Holstein dairy cows' milk. Sixteen of 25 months-old Holstein dairy cows were assigned to two groups (n=8) to study effects of the feeding system for 150 days. Two feeding systems were compared for five months; Group 1 was housed indoors and mainly fed a concentrate diet, Group 2 was maintained outdoors for five-seven hours/day on various kinds grass in a pasture. The experiment was conducted June-October 2017. Results revealed the indoor-fed cows had higher body weight, that was significant compared with the outdoor-based feeding system of Holstein dairy cows (p<0.05). Indoor-raised milking cows had higher milk yield (32.45 kg) as compared with pasture-raised milk yield (26.44 kg). Cows fed indoors significantly increased milk yield, total protein content, lactose, citric acid level, and lowered level of total solid and free fatty acids relative to the pasture-fed milking cows (p>0.05). There were higher levels of mineral content and fatty acid content in the milk of indoor-fed dairy cows than the pasture-raised dairy cows (p>0.05). Our study results demonstrated the potential benefits of the indoor feeding system for increased body weight, milk yield, mineral and fatty acids content summer through autumn when low pasture growth rates and quality may otherwise limit production.

  • 밀에 대한 느타리버섯 균사체 배양 특성 및 균사체 배양밀가루를 첨가한 제빵 적성에 대하여 조사하였다. 느타리버섯 균사체를 접종하기 위해서는 온수를 이용한 침지가 냉수보다 유용하였으며, 균사체가 배양된 밀은 통밀보다 수분함량이 높은 것으로 나타났으나 일반 성분에는 큰 차이를 보이지 않았다. 이러한 균사체배양 밀가루의 첨가량을 달리하여 식빵을 제조한 결과, 첨가량이 증가할수록 식빵의 무게, 부피 및 반죽의 발효 팽창력은 모두 대조구에 비하여 전체적으로 감소하였다. Crust와 crumb의 색도는 균사체 배양 밀가루 첨가량이 증가함에 따라 전체적으로 감소하였다. 관능 평가결과, 향과 맛 그리고 종합적인 기호도에서 균사체 배양 밀가루 5% 첨가군이 높은 기호도를 나타내어, 균사체 배양 밀가루를 실제 제빵에 이용할 경우 5% 미만이 적절할 것으로 사료된다. 이러한 연구는 기능성 소재를 이용한 제빵 제조에 유용한 정보를 제공하며 또한 균사체 배양 밀가루의 식품소재로의 이용에 도움을 줄 것으로 사료된다.

NCI-H157 폐암 세포주에서 Caspase Cascade 활성을 통한 Arsenic Trioxide와 Sulindac 병합요법의 세포고사효과 (Inducing Apoptosis of NCI-H157 Human Lung Carcinoma Cells via Activation of Caspase Cascade by Combination Treatment with Arsenic Trioxide and Sulindac)

  • 연구배경 : Arsenic trioxide($As_2O_3$)은 재발성 또는 불응성 급성전골수성백혈병의 치료제로 쓰이는 항암제로서 비소세포폐암을 포함한 다른 암세포주에도 효과가 있는 것으로 되어있다. NSAIDs는 항암 예방약제로 사용되고 있고, 세포고사를 통해 다른 항암제나 방사선치료의 반응성을 강화시키는 것으로 알려져 있다. 저자들은 NCI-H157 세포주에서 $As_2O_3$와 sulindac의 병합치료가 그것들의 세포고사를 배가시키는지 여부를 알아보고자 하였다. 방 법 : 세포 독성은 MTT 방법으로 측정하였고, 세포고사를 알아보기 위해 핵산 염색과 유식세포 분석을 시행하였다. 세포고사의 기전을 보기 위해 caspasefamily의 활성을 보았고, PARP와 ICAD의 분절을 western blotting으로 확인하였다. 또한 Fas와 Fas-L의 발현유무를 western blotting을 통해 관찰하였다. 결 과 : NCI-H157 폐암세포에 $As_2O_3$와 sulindac을 병합치료시 단독치료군에 비해 생존율이 의미 있게 감소하였고, 이러한 세포사는 핵산염색을 통한 염색사의 응축과 핵 분절 유도와 유식세포 분석에 의한 $sub-G_0/G_1$ DNA분획의 증가현상을 통해 세포고사에 의해 매개됨을 알 수 있었다. 세포고사의 유도에는 caspase 3, 8, 9를 통한 활성화와 이에 의한 PARP와 ICAD의 절단을 확인하였다. 또한 caspase-8 protease의 활성화에는 Fas와 Fas/L 단백질의 발현증가가 유도되었음을 알 수 있었다. 결 론 :NCI-H157 폐암세포주에 $As_2O_3$와 sulindac의 병합요법은 Fas/FasL 신호전달계의 활성화와 caspase 단백질 활성화 의해 세포고사가 유도되었다.

Effect of phosphorus application on appearance of algal water bloom and rice yield in rice-barley double cropping system

  • Algal communities are important to maintain the aquatic ecosystems function. Algae have short life cycles, they respond quickly to environmental change and their diversity and density can indicate and the quality of their habitat. The bloom forms before the rice seedings have emerged, it may present a physical barrier that prevents the seedlings from penetrating the floodwater. Wind may also move the algal bloom, pushing the young plants beneath the surface. Another harmful action develops when the water dries up and the algae form a layer at the bottom of the field. The layer envelops the seedlings, which are not yet deeply rooted, and drag them to the surface when the water is let in again. Soil utilization pattern can be the mail facter affecting soil physico-chemical properties, especially in soil phosphorus (P). Solid content of the algae culture solution increased with the increase in the nitrogen rather the phosphors concentration. Phosphoric acid was treated with conventional treatments (100-0%, before transplanting time-tillering stage), 50-50%, 0-100%, and un-treated. The herbicide was treated on the 7 DAT (day after transplanting). Green algae samples were collected 20 DAT. Total phosphoric acid was the highest at 0.06 in 50-50% treatment in 20 DAT. The amount of green algae was about twice (9.8 mg/20ml) that of un-treated. Total number of green algae was 54 species(Green algae 35 species, Euglena 9 species, Stone wheel 10 species). Among the phosphoric acid treatment methods, the number of occurrences of green algae were the highest with 39 species in 0-100%, followed by 50-50%, 28 species, conventional treatments, 22 species, non-treatment, 18 species, respectively. Rice Yield was not significantly different by phosphoric treatment time, but slightly higher than un-treated. The maximal algal biomass was observed about 2weeks or 1 month after transplanting; the subsequent decrease of the biomass was related to the consumption by grazers and to a deficient light under the rice canopy. Maximal algal growth was observed just before tillering. To estimate the suitable method of phosphorus application in puddled-soil drill seeding of rice, available phosphorus appearance of algal water bloom, and rice yield were investigated in paddy soil of rice-barley double cropping system.

결과지 길이에 따른 복숭아 '수미'의 신초 생장 및 과실 특성 (Shoot Growth and Fruit Characteristics of 'Soomee' Peach according to Length of Fruit Bearing Branch)

  • 국내에서 육성된 복숭아인 '수미'의 고품질 과실을 생산을 하는데 적합한 결과지 길이를 선발하기 위하여 결과지 길이와 신초 및 과실 특성들과의 상관성을 분석하였다. 결과지의 길이와 직경은 각각 평균 26.1cm와 6.1mm이었고, 결과지 당 신초 및 잎 수는 각각 평균 3.2개와 38.6장이었다. 그리고 변이계수는 이들 중 신초 및 잎 수에서 매우 높았다. 조사된 결과지에서 생산된 과실 무게는 평균 298.6g, 과실 당도는 12.2Brix였고, 변이계수는 과실 무게에서 18.0%로 당도보다 높았다. 나무 당 10-20cm와 20-30cm 길이의 결과지가 각각 27.1%와 25.4% 분포함으로서 단 중과지의 비율이 50% 이상이었다. 그리고 나무 당 250-350g과 11.0-13.0Brix의 과실이 각각 68.6%와 74.0% 분포하였다. 조사된 특성 간 상관분석 결과, 과실 무게와 결과지 당 신초 수는 결과지 길이의 영향을 받는 것으로 나타났다. 특히 과실 무게와는 $y=-0.0482x^2+2.4512x+277.36$의 관계를 나타내어 최대 과실 무게를 생산하는 결과지의 길이는 25.4cm로 분석되었다. 따라서 복숭아 '수미'의 결과지 길이는 20-30cm 정도가 적합할 것으로 판단된다.
