Mammary Comedocarcinoma in a Dog (개에서 발생한 유선 면포암종)

  • An 18-year-old, neutered female, Yorkshire terrier was presented with abdominal mass. Large abdominal mass was located in right 4th and 5th mammary glands. Surgically excised mammary mass was submitted for histological evaluation. Microscopically, multifocal to confluent neoplastic foci were existed from superficial dermis to subcutis. Several different growth patterns such as comedo, solid, cribriform, and micropapillary were present in the neoplastic foci. Large neoplastic mammary ducts showed great invasive tendency to adjacent tissues. Many superficial lymphatic plexuses and dermal lymphatics contained numerous neoplastic cell emboli. Because comedo pattern was demonstrated by more than 50% of the neoplastic cell population, we confirmed this case as mammary comedocarcinoma according to recent classification of canine mammary tumors.

An Antibiotic against Bacillus amyloliquefaciens from Streptomyces sp. KM-48 (Streptomyces 속 KM-48 균주가 생성하는 Bacillus amyloliquefaciens 에 대한 항생물질)

  • One of Streptomyces sp. was screened as an antibiotic producing strain against Gram positive bacteria specifically from about one hundred Actinomycetes collections. The isolated KM-48 strain, indexed in our collection, was secreted an anbitiotic in nutrient broth culture. The active principle was extracted with acetone from the lyophilized solid of cultured filtrate. The nonadsorbed elute in aluminium oxide was isolated as a purified homogeneity which revealed a sing1e spot under ultra-violet beam on avicel thin layer chromatography. The isolated antibiotic was sensitive specifically against the Gram positive bacteria and fungi. About fifty percent inhibition of the control growth on Bacillus amyloliquefaciens was 18$\mu\textrm{g}$/$m\ell$. The antibiotic was lavile against thermotreatment and stabled in acidic condition remarkably. The KM-48 was identified as a similar strain to Streptomyces tsusimaensis on its morphological and cultural characteristics.

"Lead-free" Piezoelectric Ba(Ti0.94Zr0.06)O3 Single Crystals with Electromechanical Coupling Factor (k33) Higher Than 0.8 (0.8 이상의 전기기계결합계수(k33)를 가지는 고효율 무연 압전 Ba(Ti0.94Zr0.06)O3 단결정)

  • Orthorhombic $Ba(Ti_{0.94}Zr_{0.06})O_3$ single crystals are fabricated using the cost-effective solid-state single crystal growth (SSCG) method; their dielectric and piezoelectric properties are also characterized. Measurements show that (001) $Ba(Ti_{0.94}Zr_{0.06})O_3$ single crystals have an electromechanical coupling factor ($k_{33}$) higher than 0.83, piezoelectric charge constant ($d_{33}$) of about 400 [pC/N], and piezoelectric voltage constant ($g_{33}$) higher than 50 [${\times}10^{-3}Vm/N$]. The transition temperature ($T_{OT}$) of the (001) $Ba(Ti_{0.94}Zr_{0.06})O_3$ single crystals between orthorhombic and tetragonal phases is also observed to be about $61^{\circ}C$. Because their electromechanical coupling factor ($k_{33}$) and piezoelectric voltage constant ($g_{33}$) are higher than those of soft PZT ceramics, it is expected that (001) $Ba(Ti_{0.94}Zr_{0.06})O_3$ single crystals can be used as "lead-free" piezoelectric materials in many piezoelectric applications.

Gene Structure and Function of fkhE, a Forkhead Gene in a Filamentous Fungus Aspergillus nidulans (Aspergillus nidulans forkhead 유전자 fkhE의 구조와 기능 분석)

  • A homothallic filamentous fungus Aspergillus nidulans has been used as the a model organism for studying growth and development for eukaryotic system. Various studies about specific transcription factors have been performed for elucidating the molecular mechanisms of growth, asexual and sexual developmental processes. Among them, the fkhE gene (AN2025.3) is located in chromosome VII and contains an ORF encoding 718 amino acid polypeptide intervening with two short introns. The cDNA sequencing revealed that at least four types of alternative splicing events were occurred when the fkhE gene was transcribed. The putative FkhE polypeptide contains a conserved forkhead domain and a bipartite nuclear localization signal at it's N-terminus and C-terminus, respectively. Deletion of fkhE resulted in impaired conidiophore formation in a solid medium. However, the sexual developmental process or cleistothecia formation was normal. Furthermore, fkhE deletion mutant produced conidiophores and conidia under the submerged culture, indicating that the fkhE gene is involved in asexual developmental process similar to the fkhF gene.

Effects of $Dy_2$$O_3$ composition for the photoluminescence and long-phosphorescent characteristics of stuffed tridymite $SrAl_2$$O_4$ : $Eu^{2+}$ phosphors (Stuffed tridymite계 $SrAl_2$$O_4$ : $Eu^{2+}$ 형광체의 발광 및 장잔광특성에 미치는 $Dy_2$$O_3$의 영향)

  • We investigated photoluminescence, long-phosphorescent and crystalline properties with various $Dy_2$$O_3$ compositions (0.0~9.5mol%) in $SrAl_2$$O_4$ : $Eu^{2+}$,$Dy^{3+}$ phosphor powders prepared by the solid state reaction. The highest emission wavelength (520nm) of photoluminescence spectra was not affected by the Dy doping concentrations. The$SrAl_2$$O_4$single phase which was determined by X-ray diffraction and photoluminescence was appeared for the concentrations of $Dy_2$$O_3$$\leq$1.0 mol%. After removal of the pulsed Xe-lamp excitation (360nm), also, excellent long phosphorescent properties of the phosphors were obtained with the concentrations of $Dy_2$$O_3$$\leq$1.0mol%, although the decay time for all phosphors decrease exponentially.

The Cultivation of Anchorage-Dependent Animal Cell, Vero-6, on Macroporous Collagen Microcarrier (다공성 콜라젠 미립담체를 이용한 부착성 동물세포 Vero-6의 배양)

  • The comparison of the capabilities of cell growth of four different kinds of commercially available microcarriers was carried out by culturing anchorage-dependent animal cells, Vero-6, in a spinner flask. Using 3 g/l of Cytodex 3, the maximum final cell density was about $1.4{\times}10^6$ cells/ml and increased up to $2.0{\times}10^6$ cells/ml by increasing microcarrier concentration up to 5 g/l. The macroporous collagen microcarriers, VX-100, informatrix, and Cultispher-G showed the final cell concentration of $4{\times}10^6$ cells/ml, $2.1{\times}10^6$ cells/ml, and $3.2{\times}10^6$ cells/ml, respectively at the microcarrier concentration of 5g/1. According to this result, VX-100 showed better cell growth than informatrix and cultispher-G and also showed about 2 fold increase in final cell density comparing to Cytodex 3 solid bead. When the intermittent bead-to-bead transfer technique was introduced in the culture using Cytodex 3 bead and cultispher-G, the result was very successful and the cells grew out very well. The recovered cells by dissolving collagen microcarrier using collagenase enzyme were mostly viable and grew out very well on the surface of the fresh microcarriers.

Medium Composition Affecting In Vitro Regeneration of Matteuccia struthiopteris (청나래고사리의 기내 포자체 재생에 미치는 배지 구성물질의 영향)

  • This study was carried out to investigate the efficient in vitro mass propagation methods for juvenile sporophytes of Matteuccia struthiopteris. Chopped segments of pinnae, petiole and rhizome were cultured on 1/2MS with 0.1% activated charcoal. Among these explant sources only rhizome segments produced young sporophytes, regenerating vigorously on 1/2 MS medium. Adjusting sucrose concentration to 2% and supplement to $50mgL^{-1}$ $NaH_2PO_4$ in 1/2MS medium proved to be more efficient for plant regeneration. Various combinations of growth regulators such as kinetin, BA, NAA, and IBA were added to the growing media, and the best sporophyte regeneration was obtained by $1{\mu}M$ kinetin. The BA addition resulted in vigorous proliferation of meristematic tissues, but without differentiation to sporophytes. Three types of culture methods, solid using agar, liquid stationary, and liquid shaking culture, were employed with or without activated charcoal. The addition of 0.1% activated charcoal to modified 1/2MS media (2% sucrose, $50mgL^{-1}$ $NaH_2PO_4$, $1{\mu}M$ kinetin, pH 5.8 and 0.8% agar) yielded highest sporophyte regeneration in liquid shaking culture.

A Study on the Antifungal Properties of Ranunculaceae Herbal Medicines (미나리아재비과 한약재의 항진균성 검증 및 비교 연구)

  • The purpose of this study was to verify the antifungal properties of various herbal medicines belonging to the Ranunculaceae family and to find an extraction method effective in inhibiting fungal growth. When antifungal activity was measured in a liquid medium with extracts obtained by either hot water extraction or organic solvent extraction of the herbal medicines Clematis apiifolia, Coptis chinensis, and Pusatilla chinensis, effective results were obtained from the chloroform extract. In addition, fungal growth inhibition experiments were performed on unicellular fungi, Candida albicans, Candida tropicalis, and Candida lusitaniae, and on filamentous fungi, such as Pythium ultimum, Aspergillus fumigatus, and Fusarium oxysporum, using disk diffusion experiments on solid media. It was confirmed that P. chinensis extract has excellent antifungal properties against Candida spp. and C. apiifolia extract against filamentous mold. Finally, GC-MS analysis was performed to explore the useful antifungal substances present in the extract. As a result of the study, thurbergenone from C. apiifolia and 16-hydroxycleroda-3, 13(14)-dien-15, 16-olide (16-HCDO) from C. chinensis were confirmed as antifungal candidates. In conclusion, it was confirmed that C. apiifolia, C. chinensis, and P. chinensis have antifungal activity against various fungi, and in GC-MS analysis, all herbal medicines were confirmed to have different antifungal candidates. These results indicate that the Ranunculaceae family has evolved in several directions for fungal resistance traits.

Studies on the Isolation and Identification of Lactic Acid Bacteria and its Utilization for Pharmaceutical Preparation (유산균의 분리와 동정 및 제제화에 관한 연구)

  • A microbial strain capable of producing lactic acid was isolated and identified as Streptococcus faecium. During the incubation of the isolated bacterium in a synthetic medium (Petterson broth), the optimal temperature was 36 to 39$^{\circ}C$ and the cell concentration at stationary growth phase was 2.1${\times}$10$\^$8/ viable cells/ml. When it was dried in vacuum and diluted with avicel, the Viability of Streptococcus faecium in physiological saline solution was decreased to 80% after incubation for 48 hr at 37$^{\circ}C$, whereas the viability was above 90% after incubation for 1 hr at 40$^{\circ}C$ and the viability in M buffer solution (pH 4.5∼9.0) at 37$^{\circ}C$ was above 95%. From these data it was concluded that the isolated microbe must be adoptible for pharmaceutical preparation such as solid dosage form.

Fungicidal and Hemolytic Activity of Cecropin A-Magainin 2 Analogue Peptides against Tri-chospoon beigelii and Human Red Blood Cells (Cecropin A-Magainin 2 유도체 펩티드의 Trichosporon beigelii에 대한 항진균 활성 및 인간 적혈구 세포에 대한 용혈활성)

  • In order to design a novel synthetic peptide with improved fungicidal activity but low hemolytic activity, a hybrid peptide, cecropin A(l-8)-magainin 2(1-12), and its analogue peptides were synthesized by the solid phase method. Antifungal and hemolytic activities of the synthetic peptides were assessed by the growth inhibition against Trichosporon beigelii and the cell membrane lysis against human red hlood cells, respectively. Analogue 2 in which Lys at position 12 in cecropin A(1-8)-magainin 2(1-12) was substituted with Ala showed most potent antifungal activity (MIC: 2.5.$\mu$g/ml) with minimal hemolytic activity (0.5% hemolysis at the (200.$\mu$g/ml peptide). This peptide (A2), therefore, could be useful as a model for further designing potent antifungal peptides without cytotoxicity.
