소의 theileriosis가 성장 hormone과 insulin-like growth factor-I에 미치는 영향 (Effect of bovine theileriosis on the growth hormone and insulin-like growth factor-I)

  • Bovine theileriosis caused by Theileria sergenti is the tick-borne intraery- throcytic piroplasmosis, that occurs in most regions of Korea. It results in severe economic losses on a farm caused by anemia, milk production loss, abortion and death. This study was undertaken to confirm the effects of the growth hormone and the insulin-like growth factor-I which are associated in the growth of cattle infected by T sergenti. The blood of one hundred and twenty ten-month Holstein was collected and the prepared blood smear was stained with acridine orange to investigate their parasitemia. And the hematological profiles were observed. According to the value of the hematocrit, they were categorized into four groups : Group 1 was under 20 percent, groups 2 and 3 were from over 21 to under 30 percent and from over 31 to under 35 percent and group 4 was over 36 percent. As the value of the hematocrit decreased, parasitemia(%) in erythrocytes was observed to increase(Y=-1.064X + 30.537, r=0.660). The amounts of the growth hormone and the insulin-like growth factor-I in the serum were measured by the radioimmunoassay. The growth hormone in serum of the group 1, group 2, group 3 and group 4 were observed as $0.238{\pm}0.043nmol/l$, $0.21{\pm}0.024nmol/l$, $0.366{\pm}0.035nmol/l$ and $0.646{\pm}0.223nmol/l$, respectively. The quantitative of the insulin-like growth factor-I in the same groups were observed also as $209.686{\pm}18.94ng/ml$, $250.9{\pm}12.609ng/ml$, $279.3{\pm}8.883ng/ml$ and $365.9{\pm}22.45ng/ml$, respectively. It can be concluded that the growth hormone and the insulin-like growth factor-I were observed to decrease in severe anemia due to theileriosis.

성장호르몬 방출인자를 발현하는 형질전환 생쥐에서 소변분석 (Urine Analysis in Transgenic Mice Expressing the Growth Hormone-releasing Factor)

  • 생쥐에서 페르몬으로 알려진 소수성의 분자와 결합하는 major urinary proteins(MUPs)는 부분적으로 성장 호르몬(GH)의 조절을 받는다. 본 연구에서는 MT-GRF로 형질전환되어 성장호르몬이 증가된 생쥐에서 MUP의 발현을 조사하였다. 이 MT-GRF로 형질전환된 생쥐의 소변에서 MUP이 대조군보다 심하게 저하되어 나타났고, 암컷보다 수컷에서 더 저하되어 나타났다. 또한 MT-GRF를 근육에 주사한 생쥐에서도 MUP는 저하되어 나타났다. 부가하여, MT-GRF로 형질전환시킨 생쥐의 소변에서 전기영동상에서 albumin과 동일하게 이동하는 고분자의 단백질이 다량으로 관찰되었는데 신장의 사구체 여과가 손상되었음을 암시하고 있다. 이 혈장 단백질의 손실도 암컷보다 수컷에서 심하게 나타났다. 결과적으로 GRF의 과다생성은 성별에 차별화된 영향을 미치면서 GH로 형질전환된 동물모델에서 관찰되는 MUP의 발현과 사구체 기능의 변화를 동일하게 유발하고 있었다.

Clinical and Laboratory Features to Consider Genetic Evaluation among Children and Adolescents with Short Stature

  • Conventional evaluation method for identifying the organic cause of short stature has a low detection rate. If an infant who is small for gestational age manifests postnatal growth deterioration, triangular face, relative macrocephaly, and protruding forehead, a genetic testing of IGF2, H19, GRB10, MEST, CDKN1, CUL7, OBSL1, and CCDC9 should be considered to determine the presence of Silver-Russell syndrome and 3-M syndrome. If a short patient with prenatal growth failure also exhibits postnatal growth failure, microcephaly, low IGF-1 levels, sensorineural deafness, or impaired intellectual development, genetic testing of IGF1 and IGFALS should be conducted. Furthermore, genetic testing of GH1, GHRHR, HESX1, SOX3, PROP1, POU1F1, and LHX3 should be considered if patients with isolated growth hormone deficiency have short stature below -3 standard deviation score, barely detectable serum growth hormone concentration, and other deficiencies of anterior pituitary hormone. In short patients with height SDS <-3 and high growth hormone levels, genetic testing should be considered to identify GHR mutations. Lastly, when severe short patients (height z score <-3) exhibit high levels of prolactin and recurrent pulmonary infection, genetic testing should be conducted to identify STAT5B mutations.

영양소 결핍으로 유도한 성장장애 흰쥐에서 용옥고(龍玉膏)가 성장 및 학습효과에 미치는 영향 (Effects of Yongohkgo on Growth and Learning Ability in Growth Deficiency Rat With Linsufficient Nutrition Diet)

  • Effects of Kyungohkgo Ga Nokyong(Yongohkgo) on growth development and learning ability were investigated growth and intellectual impairment rat with insufficient nutrition diet. We divided male Sprague-Dawley rats into 4 groups. They were Normal group, Growth deficiency rat with insufficient nutrition diet group, Growth deficiency rat with 0.1% Yongohkgo group and 0.2% Yongohkgo group. They were administered for 5 weeks. We measured body weight, and morris water maze test in escape distance, escape time and escape speed, serum growth hormone, insulin-like growth factor and thyroid stimulating hormone, RBC, concentration of Hb and PCV ratio, total WBC and its composition, the values of GOT and GPT activities. The results are as follows that Yongohkgo 0.1%, 0.2% groups were showed significantly different than control groups in body weight and the counts of RBC. In the morris water maze test, in escape distance and escape time, in concentration of Hb and PCV ratio, 0.2% Yongohkgo group were significantly different than control groups. Serum growth hormone, insulin- like growth factor and thyroid stimulating hormone showed a tendency to increase in Yongohkgo groups. The counts of total WBC and its composition, GOT, GPT activities showed no significantly different in all treatment groups. These results suggested that Yongohkgo have an effect of promoting growth and learning ability of rats and might be effect to treat various kinds of growth and learning ability delay in children.

성장호르몬 활성화 신발에 대한 운동역학적(지면반력, 최대압력) 평가 및 성장호르몬 분비 효과 검증 (Verification for the Effect of Growth Hormone Promotion and Kinetic Factor Evaluation on Growth Hormone Activated Shoes)

  • 본 연구는 성장호르몬 활성화와 뼈 형성에 효과가 있도록 고안된 성장호르몬 활성화 신발과 일반 런닝화에서의 운동역학적 변인과 성장호르몬 분비에서 어떠한 차이가 나타나는 지에 대한 평가 및 검증을 수행하였다. 연구 수행 결과 다음과 같은 결론을 얻었다. 첫째, 성장호르몬 활성화 신발이 일반 런닝화에 비해 달리기 시 2km 이내 달리기시 통계적으로 성장호르몬 분비가 유의하게 증가되었다. 둘째, 4km 이내의 걷기 운동에서 청소년기에서는 일반 런닝화보다는 성장호르몬 활성화 신발에서 평균적으로 성장호르몬이 많이 분비되는 것으로 나타났다. 셋째, 성장호르몬 활성화 신발은 걷기동작에서는 일반 런닝화보다 더 큰 부하를 신체에 전이시키고, 달리기에서는 일반 런닝화 보다 더 큰 충격량을 신체에 전달하고, 동시에 신체 일부분에 집중되는 부하를 분산시키고 작은 부하율을 통한 부상 발생원인을 제거함으로써 부상 없이 몸에 자극을 크게 전달, 성장호르몬 분비를 활성화 시킬 것으로 판단된다.

The role of p21/CIP1/WAF1 (p21) in the negative regulation of the growth hormone/growth hormone receptor and epidermal growth factor/epidermal growth factor receptor pathways, in growth hormone transduction defect

  • Purpose: Growth hormone transduction defect (GHTD) is characterized by severe short stature, impaired STAT3 (signal transducer and activator of transcription-3) phosphorylation and overexpression of the cytokine inducible SH2 containing protein (CIS) and p21/CIP1/WAF1. To investigate the role of p21/CIP1/WAF1 in the negative regulation of the growth hormone (GH)/GH receptor and Epidermal Growth Factor (EGF)/EGF Receptor pathways in GHTD. Methods: Fibroblast cultures were developed from gingival biopsies of 1 GHTD patient and 1 control. The protein expression and the cellular localization of p21/CIP1/WAF1 was studied by Western immunoblotting and immunofluorescence, respectively: at the basal state and after induction with $200-{\mu}g/L$ human GH (hGH) (GH200), either with or without siRNA CIS (siCIS); at the basal state and after inductions with $200-{\mu}g/L$ hGH (GH200), $1,000-{\mu}g/L$ hGH (GH1000) or 50-ng/mL EGF. Results: After GH200/siCIS, the protein expression and nuclear localization of p21 were reduced in the patient. After successful induction of GH signaling (control, GH200; patient, GH1000), the protein expression and nuclear localization of p21 were reduced. After induction with EGF, p21 translocated to the cytoplasm in the control, whereas in the GHTD patient it remained located in the nucleus. Conclusion: In the GHTD fibroblasts, when CIS is reduced, either after siCIS or after a higher dose of hGH (GH1000), p21's antiproliferative effect (nuclear localization) is also reduced and GH signaling is activated. There also appears to be a positive relationship between the 2 inhibitors of GH signaling, CIS and p21. Finally, in GHTD, p21 seems to participate in the regulation of both the GH and EGF/EGFR pathways, depending upon its cellular location.

자전거 운동이 비만 중년여성의 염증관련인자 및 성장호르몬에 미치는 영향 (Effects of Bicycle Exercise on Inflammation Related Factor and Growth Hormone in Obese Middle aged Women)

  • 본 연구는 비만 중년여성을 대상으로 8주간 자전거 운동을 통한 염증관련인자와 성장호르몬의 변화를 분석함으로써 자전거 운동의 효과를 규명하고 효율적인 운동을 위한 과학적 근거를 제시하는데 그 목적이 있다. 15명의 비만 중년 여성들은 자전거 운동군과 통제군으로 구분하여 8주간 주당 3회 60분간 자전거를 이용한 유산소 운동 프로그램을 실시하였다. 측정변인으로는 염증관련인자와 성장호르몬의 변화를 분석하였으며 통계처리는 이원 반복측정 분산분석(two-way ANOVA with repeated measure)을 실시하였다. 분석 결과 염증관련인자는에서 $TNF-{\alpha}$ 및 IL-6는 운동군에서 유의한 감소를 보였으며 통제군에서 증가하였다. 성장호르몬은 운동군에서 증가하였으나 유의한 차이가 나타나지 않았으며 통제군에서 감소하였다. 이와 같은 결과는 본 연구에서 실시한 자전거 운동이 비만 중년여성들의 심혈관계 및 만성질환을 예방 할 수 있는 운동 프로그램이라고 사료된다.
