Adventitious root growth inhibition in boron-deficient or aluminum-stressed sunflower seedlings

  • Sunflower(Helianthus annuus L.) seedings were de-rooted and grown in nutrient solutions providing either deficient or sufficient boron supply and supplemented with aluminum. Increasing concentrations of aluminum in the nutrient medium caused progressive inhibition of root growth and a parallel increase in proline level of roots. Elevated boron levels improved root growth under toxic aluminum conditions and produced higher proline contents. Exogenous ascorbate improved adventitions root growth in plants supplied with insufficient boron and aluminum. These findings suggest that root growth inhibition resulting from either boron deficiency or aluminum toxicity may be a result of impaired ascorbate metabolism.

Growth Ingibiton Effect of E. coli O157:H7 and Salmonella typhimurium by Lactic Fermented Milk Products Administrated Orally in Rabbit (토끼에서 유산 발효유제품 급여에 의한 Escherichia coli O157:H7 및 Salmonella typhimurium의 증균억제효과)

  • The growth inhibition effect of Orally administrated yogurt ACE and Metchnikoffupon E. coli O157:H7 and S. typhimurium inoculated into gastric lumen of rabbits was in vestigated. The rabbits challenged with each 1 $m\ell$ of suspension containing 108 CFU/$m\ell$ of the pathogens were divided into 4 groups by the interval of yogurt administration: A group; preadministrated 7 days before inoculation of the pathogens and fed daily; B group; administrated daily after inocjlation of the pathogens, C group; administrated every 3 days after inoculation of the pathogens; Control group, not fed after inoculation of the pathogens. Each 3 $m\ell$ of yogurt containing 109 CFU/$m\ell$ was orally administrated into rabbits. All yogurt administrated groups (A, B, c) chowed growth ingibition effect on E. coli O157:H7 in one day after inoculation of the pathogen by the level of 0.8~1.0 log CFU/g, compared with the result differences between the control group and the yogurt administrated groups. In the control group after 5 days of inoculation, the number of colonized pathogens was 105~106 CFU/g, whereas 103~104 CFU/g was detected in the yogurt administrated groups. After 10 days of inoculation, the viable pathogen number per gram (g) of the rabbit feces was 103 CFU/g in the control group, whereas the number below 101 CFU/g was detected in the group A, and 102 CFU/g in the control group, B and C. The growth inhibition effect of yogurt administration on E. coli O157:H7 was highly increased in the order of A, B, and C group. The same effect on S. typhimurium was observed at the level of 2 log CFU/g in the Metchnikoff yogurt administrated groups, compared with the control group result in one day after inoculation of the pathogen. In 7 days after inoculation of the pathogen, the viable number was increasingly decreased, and finally after 15 days no viable cell of S. typhimurium was discharged into the fecal samples in the group A, and the mean level of 10* CFU/g was detected in the group B, but there was no growth inhibition effect in the group C. The growth inhibition effect on S. typhimurium was observed at the same level of viable cell number between the yogurt ACE administrated groups and the control group in 5 days after inoculation. But, after 10 days of inoclation the viable cell number was started to decrease, and the viable cell of S. typhimurium was not discharged from rabbit intestinal contents after 15 days of inoculation in the yogurt ACE administrated groups. In such a case that yogurt was administrated in order to prevent the pathogens, pre-administration on a daily basis one week before inoculation of the pathogens exerted considerable effect in growth inhibition. In comparison with two kinds of yogurt tested in this study, the growth inhibition effect on two kinds of pathogens was observed more highly in the Metchnikoff administated group than the ACE administrated group.

Growth Inhibition of E. coli O157:H7 and Salmonella typhimurium by Lactic Acid Bacteria and Bifidobacteria (젖산균과 비피더스균에 의한 Escherichia coli O157:H7과 Salmonella typhimurium의 생장억제)

  • Lactobacillus acidophilus NCFM, Lactobacillus casei YIT 9018, Bifidobacterium longum 8001, and Bifidobacterium longum 8025 at the level of 106 cfu/$m\ell$ were cultured with 104 cfu/$m\ell$ of Escherichia coli O157:H7 KSC 109 or Salmonella typhimurium ATCC14028, in order to verify the effects of lactic acid bacteria and bifidobacteria on the growth of the pathogens. In the mixed culture of lactic acid bacteria with E. coli O157:H7 KSC 109, Growth inhibition and atypical microcolonies of E. coli O157:H7 KSC 109 were observed. The pathogens inoculated grew for 5 hors (pH 5.3), by the time L. acidophilus NCFM reached the exponential growth phase, and then the surviving pathogens were decreased to 101 cfu/$m\ell$ after 35 hours. When L. caseiYIT 9018 was grown with the pathogens, they grew for 10 hours (pH 4.6), by the time L. casei YIT 9018 reached the end of exponential growth phase, and then the surviving pathogens were decreased drastically. Up to the stationary growth phase of lactic acid bacteria, L. acidophilus NCFM exhibited stronger inhibition against the pathogens than L. casei YIT 9018 did, which might be attributed to its faster growth. Likewise bifidobacteria inhibited the growth of the pathogens tested, bifidobaceria was weaker in the inhibitory activity than lactic acid bacteria. When Bifidobacterium longum 8001 was cultured with the pathogens, E. coli O157:H7 KSC 109 was gradually ingibited at the stationary growth phase of bifidobacteria, atypical microcolonies were formed on Levine EMB medium after 48 hours, and Salmonella grew up to 106 dfu/$m\ell$, then was drastically ingibited at the exponential growth phage of Bifidobacterium longum 8001. But when Bifidobacteriuam longum 8025 was cultured with the pathogens, the pathogens grew to the same level of Bifidobacteriuam longum 8025 was cultured with the pathogens, the pathogens grew to the same lever of Bifidobacteriuam longum 8025 after 10 hours, then the surviving pathogens were decreased drastically.

  • The inhibition rate of bacteria growth per molecular weight was higher according as the molecular weight increased, the rate was the highest at the molecular weight 200,000. Microcapsule of ionized calcium was able to be produced by molecular weight 15,000, 30,000, 50,000 and 200,000 of chitosan which was dried for 48 hours after melting it in 2% of acetic acid, adding ionized calcium and controlling pH 1.2. The size of ionized calcium microcapsule was between 200 and $300\;{\mu}m$, the solvency, concentration and the content showed big difference by the molecular weight of chitosan. The inhibition rate of bacteria growth of microcapsule designated high in Gram positive, which was high in S. aureus, S. epidermidis and Bacillus subtilis, low in S. mutans, high in C. albicans in fungi, low in A. niger. The inhibition rate of bacteria growth of chitosan was comparatively high in Gram positive, low in S. mutans and it showed high numerical value in C. albicans of fungi. The rate recorded good result at molecular weight 200,000 relatively, there was no difference according to the molecular weight. The inhibition rate of bacteria growth according to the concentration of the microcapsule increased differently between $1.000{\sim}10,000\;{\mu}g/ml$, it showed antibacterial activity close to the inhibition rate of growth of chitosan rather than ionized calcium. The minimum inhibitory concentration marked the highest in the mixture of chitosan and ionized calcium for all kind of bacteria generally, there was a little difference between yeast and fungi.

Antitumor Activity of Gamdutang Aqua-Acupuncture Solution (감두약침액의 암세포 성장 저해 효과)

  • Gamdutang aqua-acupuncture solution (GAS), Gamdutang water-extracted solution (GWS) and Dae-Gamdutang aqua-acupuncture solution (DGAS) were prepared and tested for antitumor activities. It was shown to possess considerable toxicity toward various tumor cell lines. Concentration of 5 $\times$ and 10$\times$ of GAS resulted in more than 70% inhibition of growth in Ehrlich ascites tumor cells (EATC), Hepa1c1c7 and A549. GAS at concentrations of 5$\times$ and 10$\times$ revealed more than 60% inhibition in HeLa. GWS showed more than 50% inhibition of growth with EATC and HeLa at concentrations of 5$\times$ and 10$\times$, respectively. Toxicity assay with GWS in Hepa1c1c7 and A549 revealed that more than 80% inhibition of growth at the concentration of 5$\times$ and 10$\times$. In morphological study, the number of cells were decreased, and the shape of cells was round-form in EATC, Hepa1c1c7, A549, HeLa with GAS.

  • Attempts were made to investigate the antagonistic activity of soil borne microorganisms Pseudomonas spp. and Trichoderma spp. against to the pathogens of turf diseases Rhizoctionia solani spp. and Pythiom spp. in vitro by a dual culture bioassay. Inhibition zone between the edge of the my-celium and the margin of each antagonistic bacteria, Pocudontonas, on potato dextrose agar was measured 3 days after incubation at 28˚C. Psudomonas spp. showed relatively high inhibition of mycelium growth of R. solani AG-i and Pythium spp. which cause brown patch and pythium blight, respectively. Antagonistic fungi Trichodenma spp. also showed effective inhibition against mycelium growth of both pathogens, more proper methods of measuring the inhibition effects were required because of fast growth of Trichodenna hypae. Brown patch and pythium blight both, re-quire most higher rate of fungicide use to control in golf curses in Korea. Application of antagon-istic microorganisms are useful as biological resources an approach to sole environmental contamination.

Isolation of Bacillus amyloliquefaciens H41 Producing Growth Inhibition Factor against Vibrio anguillarum (어병균 Vibrio anguillarum 생육 저해 인자를 생산하는 Bacillus amyloliquefaciens H41의 분리)

  • To investigate the possible use of probiont in fish farming industry, a bacterium with inhibitory effect against Vibrio anguillarum was isolated from gastrointestinal tract of the marine fish of yellow tail. It was identified to be Bacillus amyloliquefaciens H41 based on biochemical and physiological characterization. The optimal growth conditions of the isolated strain were 1% peptone, 1.5% yeast extract, 1% sucrose, 0.5% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, pH 7.0-8.0, and 20 hr of incubation between $28-35^{\circ}C$ under aeration. The culture supernatant of the isolated strain showed inhibition activity against V. anguillarum. Inhibition activity was cleared by forming a clear zone by a paper-disk method. The maximal production of growth inhibition factor was induced by cultivation under 1% peptone, 1.5% yeast extract, 1% sucrose, 1% NaCl, 0.05% $MgSO_4{\cdot}7H_20$, pH 7.5 and at $35^{\circ}C$. The highest growth inhibition factor production was observed after 16-24 hr cultivation under aeration. The culture supernatant of the isolated strain showed inhibition activity whereas no inhibition activity was shown from the standard B. amyloliquefaciens KCTC 1724 strain. The growth inhibition affected only against V. anguillarum among other pathogenic Vibrios tested here.

Studies on the Precipitation of Lead Ion and the Inhibition of Plant Growth (연(Pb) 이온의 침전과 식물생장의 억제에 관한 연구)

  • This study was carried out to investigate the formation of precipitates between lead ion and the essential anions of plants, the effects of lead concentration on seed germination and plant growth in water and soil culture, and the germinating and growing recovery of inhibited seed germination and plant growth by lead. Four kinds of the seeds (Glycine max M., Triticum vulgare V., Setaria viridis (L) P. De Beauvois, and Digitoria sanguinalis (L) Scopoli var) were germinated and growth in water and soil culture included the different concentrations of lead for five days. The seeds and plants inhibited germination and growth by lead were transferred to lead free Hoagland solution and the growing recovery was observed. The precipitates of lead ion were observed in the solution of both acidity and alkalinity included each anion of $H_2PO_4^-, HPO_4^{2-}, PO_4^{3-}, SO_4^{2-} and MoO_4^{2-}$ in a room temperature, whereas the precipitates between lead ion and other anions were observed largely in the solution of alkalinity, so that it seemed that lead could be remained in the state of non-soluble in plant and soil. The inhibition of germination and growth in the water culture was observed in 100ppm of lead, whereas the inhibition in the case of the soil culture was observed in 10000ppm of lead. The difference of the effected concentration between water and soil culture in germination and the growth was 100 times. When the seed and plant inhibited the growth in 5000ppm or 10000ppm of lead for five days were transferred to lead free Hoagland solution, the recovery of germination and growth was observed in three days. This growing recovery was different according to the kinds of plant and concentrations of lead. It seemed that plant growth could be inhibited by the inhibition of the metabolism concerned with the precipitates between lead iion and other anions.

  • In order to investigate antimicrobial substances from natural spices, the effects of garlic juice and ginger juice on the growth of microorganisms were investigated. It was showned that garlic juice inhibited the growth of bacteria, yeasts and fungi but ginger juice did not inhibit the growth of bacteria and yeasts. As result of concentration effect of garlic juice on the growth of microorganisms, the higher concentration of garlic juice was employed, the more growth inhibition was obtained. Comparing the specific growth rate at various concentration of garlic juice, the inhibition effect on yeast appeared much higher than that on bacteria.

Extracts from Pinus densiflora Siebold et Zuccarini Sawdust Inhibit the Mycelial Growth of Lentinus edodes (Berk.) Sing., Edible Mushroom

  • The extracts of Pinus densiflora sawdust by n hexane, ethyl acetate and methanol solvent were investigated to identify their mycelial growth inhibition against Lentinus edodes. The yields of n hexane soluble fraction, ethyl acetate-soluble fraction, and methanol soluble fraction from P. densiflora sawdust were obtained 1.36%, 2.21% and 4.03% using organic solvent, respectively. The mycelial growth inhibition of L. edodes was the greatest for n hexane extract, ranging from 36.5% to 47.6% at concentrations of 125 ppm to 1,000 ppm, with the values for all concentrations significantly different from one another. After direct extraction of P. densiflora sawdust using n hexane, ethyl acetate and methanol, each extract was separated into three fractions by silica gel column chromatography and then the fractions were isolated on the values of $R_f$ by thin layer chromatography. The mycelial growth inhibition against L. edodes was recognized in the fractions II (33.5%) and III (37.6%) of n hexane extract, the fraction II (21.4%) of ethyl acetate extract and the fraction II (26.4%) of methanol extract. The fractions III of n-hexane extract showed the highest growth inhibition among the nine fractions of the organic solvent extract.
