Fast Growing Furious Races for Targeting Fibroblast Growth Factor Receptors

  • Targeting fibroblast growth factor receptors (FGFRs) has been slow compared to other targeted cancer therapies for receptor tyrosine kinases, such as epidermal growth factor receptors. The low efficacy and variable response have limited the growth of FGFR inhibitors in clinical use. Nevertheless, recent systematic and genomic approaches have identified the biological conditions for effectively targeting FGFRs and can accelerate the development of targeted drugs. Under clinical and preclinical trials, the inhibitors started fast growing furious races to target FGFRs. Finally, FGFRs will be more actionable and targetable with more precise and effective drugs at the end of the race, passing the finish line.

  • Squamous cell carcinoma(SCC) of head and neck(SCCHN) is the sixth most common human malignant tumor. However, despite advances in prevention and treatment of SCC, the five-year survival rates for patients remain still low. To improve the outcome for patients with SCCHN, novel treatment strategies are needed. Overexpression of the epidermal growth factor(EGF) and activation of its receptor(EGFR) are associated with progressive growth of SCCHN. Vascular endothelial growth factor(VEGF) signaling molecules are related with neoangiogenesis and vascular metastasis of SCC. In this study, we determined the therapeutic effect of AEE788(Novartis Pharma AG, Basel, Switzerland), which is a dual inhibitor of EGFR/ErbB2 and VEGFR tyrosine kinases, on human oral SCC. At first, we screened the expression of EGFR, c-ErbB2(HER-2) and VEGFR-2 in a series of human oral SCC cell lines. And then we evaluated the effects of AEE788 on the phosphorylation of EGFR and VEGFR-2 in a oral SCC cell line expressing EGFR/HER-2 and VEGFR-2. We also evaluated the effects of AEE788 alone, or with paclitaxel(Taxol) on the oral SCC cell growth and apoptosis. As a result, all oral SCC cells expressed EGFR and VEGFR-2. Treatment of oral SCC cells with AEE788 led to dose-dependent inhibition of EGFR and VEGFR-2 phosphorylation, growth inhibition, and induction of apoptosis. Moreover, AEE788 sensitizes the cells to paclitaxel-mediated toxicity and apoptosis. These data mean EGFR and VEGFR-2 can be reliable targets for molecular therapy of oral SCC, and therefore warrant clinical use of EGFR/VEGFR inhibition in the treatment of patients with recurrent or metastatic oral SCC.

Effects of Epidermal Growth Factor (EGF) and Insulin-like Growth Factor-1 (IGF-1) on Maturation of Bovine Follicular Oocytes In Vitro (Epidermal Growth Factor(EGF)와 Insulin-like Growth Factor-1(IGF-1)이 소 난포란의 체외성숙에 미치는 영향)

  • The purpose of this study was to evaluate the effects of growth factors such as epidermal growth factor (EGF) and insulin-like growth factor-1 (IGF-1) on maturation of bovine follicular oocytes in vitro. Oocytes were recovered from the ovaries of slaughtered Hanwoos. The oocytes were matured in TCM 199 at 39$^{\circ}C$, 5% $CO_2$ in air. Growth factors were added to maturation medium as follows: control (no serum), EGF (10ng/m1, 50ng/ml or 100ng/m1), IGF-1 (100ng/m1) and EGF (50ng/ml) + IGF-1 (100ng/m1). The oocytes were placed onto a slide and stained with aceto-orcein dye. Nuclear maturation was evaluated and classified as germinal vesicle breakdown (GVBD), metaphase-I (MI) or metaphase-ll(Mll). Maturation rates were 37.9% (control), 45.8% (EGF, 10ng/m1), 55.8% (EGF, 50ng/ml), 44.4% (EGF, 100ng/m1), 46.7% (IGF-1, 100ng/m1) and 67.0% (IGF-1+EGF). The highest group developed to Mll stage was IGF-1+EGF treatment group (p<0.05). Therefore, nuclear maturation of bovine oocytes were affected by both of growth factors, and it seems to have a mutual activity between them.

Regulation of Transforming Growth Factor ${\beta}1$, Platelet-Derived Growth Factor, and Basic Fibroblast Growth Factor by Silicone Gel Sheeting in Early-Stage Scarring

  • Background Hypertrophic scars and keloids are associated with abnormal levels of growth factors. Silicone gel sheets are effective in treating and preventing hypertrophic scars and keloids. There has been no report on the change in growth factors in the scar tissue following the use of silicone gel sheeting for scar prevention. A prospective controlled trial was performed to evaluate whether growth factors are altered by the application of a silicone gel sheet on a fresh surgical scar. Methods Four of seven enrolled patients completed the study. Transforming growth factor (TGF)-${\beta}1$, platelet-derived growth factor (PDGF), and basic fibroblast growth factor (bFGF) were investigated immunohistochemically in biopsies taken from five scars at 4 months following surgery. Results In both the epidermis and the dermis, the expression of TGF-${\beta}1$ (P=0.042 and P=0.042) and PDGF (P=0.043 and P=0.042) was significantly lower in the case of silicone gel sheet-treated scars than in the case of untreated scars. The expression of bFGF in the dermis was significantly higher in the case of silicone gel sheet-treated scars than in the case of untreated scars (P=0.042), but in the epidermis, the expression of bFGF showed no significant difference between the groups (P=0.655). Conclusions The levels of TGF-${\beta}1$, PDGF, and bFGF are altered by the silicone gel sheet treatment, which might be one of the mechanisms of action in scar prevention.

Mouse Nerve Growth Factor Facilitates the Growth of Interspinal Schwannoma Cells by Activating NGF Receptors

  • Objective : Nerve growth factor (NGF) is a member of the neurotrophic factor family and plays a vital role in the physiological processes of organisms, especially in the nervous system. Many recent studies have reported that NGF is also involved in the regulation of tumourigenesis by either promoting or suppressing tumor growth, which depends on the location and type of tumor. However, little is known regarding the effect of NGF on interspinal schwannoma (IS). In the present study, we aimed to explored whether mouse nerve growth factor (mNGF), which is widely used in the clinic, can influence the growth of interspinal schwannoma cells (ISCs) isolated from IS in vitro. Methods : ISCs were isolated, cultured and identified by S-100 with immunofluorescence analysis. S-100-positive cells were divided into five groups, and separately cultured with various concentrations of mNGF (0 [phosphate buffered saline, PBS], 40, 80, 160, and 320 ng/mL) for 24 hours. Western blot and quantantive real time polymerase chain reaction (PCR) were applied to detect tyrosine kinase A (TrkA) receptor and p75 neurotrophin receptor ($p75^{NTR}$) in each group. Crystal violet staining was selected to assess the effect of mNGF (160 ng/mL) on ISCs growth. Results : ISCs growth was enhanced by mNGF in a dose-dependent manner. The result of crystal violet staining revealed that it was significantly strengthened the cells growth kinetics when cultured with 160 ng/mL mNGF compared to PBS group. Western blot and quantantive real time PCR discovered that TrkA receptor and mRNA expression were both up-regualated under the condition of mNGF, expecially in 160 ng/mL, while the exoression of $p75^{NTR}$ demonstrated no difference among groups. Conclusion : From these data, we conclude that exogenous mNGF can facilitate ISC growth by activating both TrkA receptor and $p75^{NTR}$. In addition, patients who are suffering from IS should not be administered mNGF in the clinic.

Effect of bovine theileriosis on the growth hormone and insulin-like growth factor-I (소의 theileriosis가 성장 hormone과 insulin-like growth factor-I에 미치는 영향)

  • Bovine theileriosis caused by Theileria sergenti is the tick-borne intraery- throcytic piroplasmosis, that occurs in most regions of Korea. It results in severe economic losses on a farm caused by anemia, milk production loss, abortion and death. This study was undertaken to confirm the effects of the growth hormone and the insulin-like growth factor-I which are associated in the growth of cattle infected by T sergenti. The blood of one hundred and twenty ten-month Holstein was collected and the prepared blood smear was stained with acridine orange to investigate their parasitemia. And the hematological profiles were observed. According to the value of the hematocrit, they were categorized into four groups : Group 1 was under 20 percent, groups 2 and 3 were from over 21 to under 30 percent and from over 31 to under 35 percent and group 4 was over 36 percent. As the value of the hematocrit decreased, parasitemia(%) in erythrocytes was observed to increase(Y=-1.064X + 30.537, r=0.660). The amounts of the growth hormone and the insulin-like growth factor-I in the serum were measured by the radioimmunoassay. The growth hormone in serum of the group 1, group 2, group 3 and group 4 were observed as $0.238{\pm}0.043nmol/l$, $0.21{\pm}0.024nmol/l$, $0.366{\pm}0.035nmol/l$ and $0.646{\pm}0.223nmol/l$, respectively. The quantitative of the insulin-like growth factor-I in the same groups were observed also as $209.686{\pm}18.94ng/ml$, $250.9{\pm}12.609ng/ml$, $279.3{\pm}8.883ng/ml$ and $365.9{\pm}22.45ng/ml$, respectively. It can be concluded that the growth hormone and the insulin-like growth factor-I were observed to decrease in severe anemia due to theileriosis.

EGF, IGF-I, VEGF and CSF2: Effects on Trophectoderm of Porcine Conceptus

  • The majority of early embryonic mortality in pregnancy occurs during the peri-implantation stage, suggesting that this period is important for conceptus viability and the establishment of pregnancy. Successful establishment of pregnancy in all mammalian species depends on the orchestrated molecular events that transpire at the conceptus-uterine interface during the peri-implantation period. This maternal-conceptus interaction is especially crucial in pigs because in them non-invasive epitheliochorial placentation occurs, in which the pre-implantation phase is prolonged. During the pre-implantation period, conceptus survival and the establishment of pregnancy are known to depend on the developing conceptus receiving an adequate supply of histotroph, which contains a wide range of nutrients and growth factors. Evidence links growth factors including epidermal growth factor (EGF), insulin-like growth factor-I (IGF-I), vascular endothelial growth factor (VEGF), and colony-stimulating factor 2 (CSF2) to embryogenesis or implantation in various mammalian species; however, in the case of pig, little is known about such functions of these growth factors, especially their regulatory mechanisms at the maternal-conceptus interface. Our research group has presented evidence for promising growth factors affecting cellular activities of primary porcine trophectoderm (pTr) cells, and we have identified potential intracellular signaling pathways responsible for the activities induced by these factors. Therefore, this review focuses on promising growth factors at the maternal-conceptus interface regulating the development of the porcine conceptus and playing pivotal roles in implantation events during early pregnancy in pigs.

Serum Levels of Insulin-Like Growth Factor-I in Flounder, Parlichthys olivaceus (넙치 (Parlichthys olivaceus) 혈액중 Insulin-like growth factor-I의 함유수준)

  • Insulin-like growth factor-1 (IGF-I) is a mitogenic peptide with molecular mass of 7kDa. It is produced mainly in the liver and has important functions in the regulation of development and somatic growth. Recently, several investigations were undertaken to examine the biological actions and structures of IGF-I in fish. In this study, the serum levels of IGF-I were estimated from flounder, Parlichthys oilvaceus, before, during and after fasting, and the levels were accounted for 47 ng/ml, 40 ng/ml and 45 ng/ml, respectively. These results suggest that food deprivation primarily reduces IGF-I level in the blood.
